Construction ofE. coli-yeast shuttle plasmids containing theneo selection gene is described. The protein-coding regions of theE. coli ada orrecA genes under the control of theADH1 promoter and terminator were ligated into theSphI unique site of pNF2 to produce pMSada and pMSrecA, respectively. The plasmids were used for transformation of the haploid and diploidpso4-1 strains ofS. cerevisiœ and their corresponding wild types. Transformants were obtained by selection for geneticin (G418) resistance. Crude protein samples were extracted from the individual transformants. Both the RecA and Ada proteins were present in all strains containing therecA andada genes on plasmids, respectively. Thus the geneticin selection system was successfully used for the preparation of model, yeast strains.  相似文献

The diethyl ether extracts isolated from unfermented milk and milk fermented byEnterococcus fœcium exhibited dose-dependent inhibition of mutagenesis induced by N-methyl-N′-nitro-N-nitrosoguanidine (MNNG), nitrovin (NIT), 5-nitro-2-furylacrylic acid (NFA) and UV-irradiation on the Ames bacterial test (Salmonella typhimurium strains TA97 and TA100) and the unicellular flagellateEuglena gracilis. Overall, the fermented milk extract was the most active against UV-irradiation, less active against NIT and MNNG, and the least active against NFA on bacteria. The highest antibleaching effects were observed against MNNG. The differences between antimutagenic effects from fermented and unfermented milk extracts were determined to be statistically significant at the 0.95 CI level.  相似文献

Polyoxirane (polyoxyethylene) and some of its derivatives stimulate the lignin peroxidase activity in shakenPhanerochœte chrysosporium cultures. The majority of polyoxirane remains in the culture fluid while a minor part diffuses into the fungal pellets where it is distributed between cell solids and cell liquid. The lower the polyoxirane content in the cell liquid, the higher the lignin peroxidase activity of theP. chrysosporium culture.  相似文献

The production of lignin-degrading enzymes by free and entrapped cells ofPhanerochœte chrysosporium in a tubular air-lift bioreactor was studied. Under optimized cultural conditions the production of lignin peroxidase by free cells, calcium-alginate-entrapped cells and scouring-mesh-entrapped cells was in a ratio of 520∶720∶950 mU/mL, while the production of manganese peroxidase was in a ratio of 350∶480∶620 mU/mL. The stability of the entrapped cells by fed-batch systems was highest after 3 feeding experiments which is similarly demonstrated in the repeated use of the preparations in batch system.  相似文献

Summary The thick rigid chorion of the egg of Triatoma secreted by the follicle cells shows two porous layers: an aerial layer in the exochorion, an alveolar one in the endochorion. The anterior part of the eggshell is closed up by an operculum which is heaved up by the hatching larva. The operculum has no alveolar layer. The air enters through the numerous holes of the shell surface into the aerial layer and through the micropyles into the alveolar layer. The egg has no respiratory plastron.The follicle cells produce also a vitelline envelope whose structure shows a rapid condensation at fertilization time. During its development the embryo secretes two layers: serosal and embryonic cuticle.At high humidities, at low temperatures the egg is able to increase its weight during the early stages of embryogenesis, and this increase stops when the serosal cuticle is secreted. In a dry atmosphere the egg loses water but can develop if the temperature is higher than 20°C.The little permeability of the egg is related to the structure of its envelopes. The chorion and the vitelline envelope prevent the water from getting out of the egg. The serosal cuticle seems to be opposed to the penetration of the water into the egg. The role of the embryonic cuticle is probably limited in the transit of water.

Summary

World-wide, our coastal waters have been subject to an increased nutrient input since the latter part of the nineteenth century. This has led to the eutrophication or ‘nutrient pollution’ of many coastal sites, including Langstone Harbour and the Ythan Estuary here in the UK. Eutrophication at these and, indeed, at other nutrient enriched sites is evident by the appearance of large blooms of fast-growing opportunistic macroalgae. Blooms of macroscopic species of green algae (Chlorophyta: Ulvophyceae) are particularly common and the phenomenon is often referred to as the occurrence of green tides.

Green tides may have a dramatic environmental impact, causing much damage to the local ecosystem. Numerous strategies have hence been employed in order to combat the problem, but to date there has been limited success. For this reason, current research in the UK is aimed at increasing our knowledge of green tide algae in terms of their ecophysiology, whilst further investigation of the nutrient pathways and fluxes within specific ecosystems has been deemed necessary. It is anticipated that this ‘backto basics’ approach will ultimately contribute to the development of new, successful eutrophication management strategies.  相似文献

Jules Cotard was a psychiatrist at the Vanves asylum. Described as a sharp observer and clinician, he also had a passion for clinical research. Jules Cotard was always ready to modify, question, or even abandon a hypothesis if clinical facts and scientific analysis made him deem it necessary to do so. He devoted most of his writing to “negation delirium”, (also known as “nihilistic delusion”): “Du délire hypochondriaque dans une forme grave de mélancolie (“Concerning hypochondriac delusion in serious melancholy”)” in 1880, “Du délire des négations (“Concerning negation delirium”)” in 1882, “Perte de la vision mentale dans une mélancolie anxieuse (“Loss of mental vision in anxious melancholy”)” in 1884 and “Du délire d’énormité (“Concerning delusions of enormity”)” in 1888. Long after his death, there was much controversy and debate about the existence of negation delirium. J. Séglas and J. Régis were the main contributors to the development and clarification of his work. Thus it is this most unusual of syndromes, Cotard’s syndrome, that has caused Jules Cotard to be part of the history of French psychiatry since the end of the 19th century.  相似文献

Using the method for the identification of promoters recognized by the sporulation specific σ factor (σF), we identified a positive 950 pbSau3Al DNA fragment inStreptomyces cœlicolor A3(2). High-resolution S1-nuclease mapping identified a potential promoter, PF35, in theE. coli two-plasmid system similar to the consensus sequence ofBacillus subtilis promoters recognized by the general stress-response σ factor (σB). However, the putativesigF-dependent promoter, PF35, was inactive inS. cœlicolor in the course of diffenentiation and it was located divergently in the promoter region directing expression of thechiC gene encoding chitinase. Sequence analysis of the region potentially governed by PF35 revealed two translationally coupled genes encoding proteins similar to bacterial two-component regulatory systems, and with the highest similarity to the two-component systemchiS, chiR, regulating chitinase activity inStreptomyces thermoviolaceus. However, the genes had a divergent orientation with respect to the PF35 promoter. Disruption of theS. cœlicolor chiR gene appeared to have no obvious effect on growth, morphology, differentiation, and production of pigmented antibiotic actinorhodin and undecylprodigiosin. Moreover, thechiR disruption did not affect the overall chitinase activity.  相似文献

Protoplasts of a strain ofA. fœcalis incapable of utilizing β-sitosterol as carbon source for growth were fused with protoplasts ofA. oxydans — a strain capable of complete degradation of β-sitosterol. Five fusants showing morphology and pattern of transformation of C-19 steroids identical toA. Fœcalis were selected. Analysis of the fermentation broth containing β-sitosterol showed that the fusants were capable of utilizing β-sitosterol for growth but their pattern of metabolite formation from β-sitosterol was different from that ofA. oxydans. The study revealed that the protoplast fusion technique could be used for intergeneric transfer of genetic determinants linked to partial cleavage of β-sitosterol side chain toA. fœcalis fromA. oxydans.  相似文献

Protective effect of the lipopolysaccharide (LPS) antigen ofKlebsiella pneumoniœ was tested against ascending-mode urinary tract infection in BALB/c and LACA strains of mice. LPS was given by two different routes; LPS was found to be protective (whatever the application route) since colonization with the challenge organism was significantly lower in both cases as compared with unimmunized mice. A maximum decrease in bacterial count in the kidney of LPS-treated animals was observed on challenge after a 4-d treatment.  相似文献
