A sediment core located on the West Spitzbergen margin in the Fram Strait (78°54.931′N, 6°46.005′E, water depth: 1497 m) was analyzed for its dinocyst content in order to reconstruct hydroclimatic variations of the last 2500 years. The relative abundance of dinocyst taxa and principal component analysis show a major transition at about 300 cal. years BP. It is characterized by the disappearance of thermophilic taxa Spiniferites mirabilis-hyperacanthus and Impagidinium sphaericum and the increase of polar–subpolar taxa Impagidinium pallidum and Pentapharsodinium dalei. Sea-surface temperature (SST) estimates suggest warmer conditions than present (anomaly~+2 °C) averaging at 7 °C in summer until 300 cal. years BP, although cooling pulses are recorded around 1700, 1500, 1200 and 800 cal. years BP. The last 300 years were marked by a cooling from 7.6 to 3.5 °C and sea-ice cover increasing up to 7 months/yr. The results demonstrate that the Fram Strait area is sensitive to hydroclimatic variations, notably with respect to sea-ice and SSTs, which are linked to the relative strength of northward flow of North Atlantic waters to the East and southward outflow of cold and fresh waters from the Arctic Ocean. Based on our data, the warmest part of our record around 1320 cal. years BP is the only interval of the last 2500 years that provides a possible analogue for the modern post-AD 2000 interval, which is characterized by sea-ice free conditions.  相似文献

In this work, three novel genes encoding di-d-fructofuranose-1,2′:2,1′-dianhydride (DFA I)-forming inulin fructotransferases (IFTases) from Nocardiaceae family, including Nocardioides luteus, Nocardioides sp. JS614, and Nocardioidaceae bacterium Broad-1, were cloned and expressed in Escherichia coli. The recombinant IFTases from N. luteus (NoluIFTase), Nocardioides sp. JS614 (NospIFTase), and N. bacterium Broad-1 (NobaIFTase) were purified, identified, and characterized. SDS-PAGE analysis showed that they had molecular weights of approximately 41–42 kDa, while gel filtration analysis indicated that their native molecular weights ranged from 50 to 62 kDa, suggesting that the three enzymes may be monomers. Their optimum pH values ranged from 5.5 to 6.0, similar to other DFA I-forming IFTases or di-d-fructofuranose-1,2′:2,3′-dianhydride (DFA III)-forming IFTases. NoluIFTase, NospIFTase, and NobaIFTase exhibited maximal activities at 55 °C, 50 °C, and 45 °C and were stable at 70 °C (for 15 min), 70 °C (187 min), and 55 °C (239 min), respectively. Furthermore, by comparing with our previously reported DFA I-forming IFTase, namely CcIFTase, a probable mechanism for the formation of DFA I by the three new enzymes was speculated, and CcIFTase will be selected for future structural resolution to illustrate the catalytic mechanism of DFA I-forming IFTases toward inulin.  相似文献

A new strain of the parasitoid Trichogramma pretiosum, was collected in Rio Verde County, State of Goiás, Central Brazil, and designated as T. pretiosum RV. This strain was then found to be the most effective one among several different strains of T. pretiosum tested in a parasitoid selection assay. Therefore, its biological characteristics and thermal requirements were studied, aiming at allowing its multiplication under controlled environmental conditions in the laboratory. The parasitoid was reared on eggs of Pseudoplusia includens and Anticarsia gemmatalis at different constant temperatures within an 18–32 °C temperature range. The number of annual generations of the parasitoid was also estimated at those temperatures. Results have shown that T. pretiosum RV developmental time, from egg to adult, was influenced by all temperatures tested within the range, varying from 6.8 to 20.3 days and 6.0 to 17.0 days on eggs of P. includens and A. gemmatalis, respectively. The emergence of T. pretiosum RV from eggs of A. gemmatalis was higher than 94% at all temperatures tested. When this variable was evaluated on eggs of P. includens, however, the figures were higher than that within the 18–30 °C range (more than 98%), and were also statistically higher than the emergence observed at 32 °C (90.2%). The sex ratio of the parasitoids emerged from eggs of A. gemmatalis decreased from 0.55 to 0.29 at 18–32 °C, respectively. However, for those emerged from eggs of P. includens, the sex ratio was similar (0.73, 0.72 and 0.71) at 20, 28 and 32 °C, respectively. The lower temperature threshold (Tb) and thermal constant (K) were 10.65 °C and 151.25 degree-days when the parasitoid was reared on eggs of P. includens; and 11.64 °C and 127.60 degree-days when reared on eggs of A. gemmatalis. The number of generations per month increased from 1.45 to 4.23 and from 1.49 to 4.79 when the parasitoid was reared on eggs of P. includens and A. gemmatalis, respectively, following the increases in the temperature.  相似文献

Hoplitomeryx Leinders was originally described only on cranial characters. The type specimens were found during the 1970's in karstic fissure fillings, most likely of Messinian age, in Gargano (Apulia, southeastern Italy), between Poggio Imperiale (41°49′30′ N, 15°21′58′ E) and Apricena (41°47′06′ N, 15°26′41′ E). During the 1990's, Hoplitomeryx remains were also discovered in the lower Tortonian layered calcarenites near Scontrone (Abruzzo, central Italy; 41°45′15.55′ N, 14°02′13.23′ E). The skull fragments, teeth, and jawbones from both localities have been examined. The dental characters had never been described before, and also some maxillaries and jawbones were not part of the original sample that was analyzed to establish the genus. Because they possess two lacrimal orifices and closed metatarsal gulley, hoplitomerycids have been linked more closely with Cervids, and accommodated in Cervoidea. A cladistic analysis of a character-taxon matrix of 121 morphological features (48 cranial, 51 dental and 22 postcranial characters) is performed. The analysis shows that hoplitomerycids stem either between antilocaprids and bovids, or antilocaprids and giraffids. They are not linked directly with cervids. Hoplitomerycids likely stemmed from a primitive ruminant stock, perhaps around 29 Ma when a land bridge connected the Abruzzo-Apulia platform with the Balkans across the Adriatic Sea. In the new land, hoplitomerycids developed a mosaic of apomorphic and homoplastic (convergent) character states that recall those found in other higher ruminants.  相似文献

During phytoplankton monitoring in the Beagle Channel (≈54°52′ S, 67°32′ W) a previously undetected Alexandrium species was observed in coincidence with mouse bioassay toxicity. Detailed thecal plates analysis using epifluorescence and scanning electron microscopy revealed the presence of the Alexandrium ostenfeldii species complex, showing a mixture of the diagnostic features usually used to discriminate between the morphospecies A. ostenfeldii and A. peruvianum. Cells of the A. ostenfeldii complex were commonly observed during spring after the main annual diatom bloom, when temperatures and salinities were respectively around 7.5–10 °C and 30–30.5 psu, and nutrients showed a seasonal decrease. Toxin analysis by liquid chromatography–mass spectrometry revealed the production of 13-desmethyl spirolide C and 20-methyl spirolide G in cell cultures. The cellular contain of spirolides during exponential phase growth was 0.5906 ± 0.0032 and 0.1577 ± 0.0023 pg cell−1 for 13-desMe-C and 20-Me-G, respectively. A third unknown compound, with a structure resembling that of spirolides was also detected in culture. Moreover, an additional compound with a similar m/z (692) than that of 13-desMe-C but presenting a higher retention time (Rt = 40.5 min) was found in high proportions in mussel samples. PSP toxins were present at low concentration in mussels but were not detected in cultures. These results extend the world-wide distribution of toxic strains of the A. ostenfeldii complex to the Beagle Channel (southern South America), where toxic events have been traditionally linked to the presence of Alexandrium catenella. This is the first confirmed occurrence of spirolides in mussels and plankton from Argentina, which highlights the importance of monitoring these toxins and their producing organisms to protect public health and improve the management of shellfish resources.  相似文献

A pollen study at Survilly (2235m asl, 06° 49′ 12″ E, 45° 59′ 24″ N), a small peatbog located on the Anterne mountain (Upper-Arve Valley, French north-western Alps) highlights the local role of human activities in Holocene vegetation dynamics of the currently treeless subalpine belt and the consecutive resumption of erosion. As early as 8890 cal. years BP (± 122), Pinus cembra grew close to the site. Grasslands without shrubs were established at around 4624 ± 86 cal. years BP. Due to human activities, spruces extended little after 3600 cal. BP. The intense grazing that resulted in the current alpine meadows goes back to 1436 cal. years BP (± 81). After 4624 cal. BP three clay layers show that from this period, the erosion became as active as during the first steps of the colonization of the vegetation prior to 10,050 cal. BP. During peat growth only a millimetre of clay at the end of the 9400–9050 cal. BP climatic event was recorded.  相似文献

In the Porcelana Hot Spring (Northern Patagonia), true-branching cyanobacteria are the dominant primary producers in microbial mats, and they are mainly responsible for carbon and nitrogen fixation. However, little is known about their metabolic and genomic adaptations at high temperatures. Therefore, in this study, a total of 81 Fischerella thermalis strains (also known as Mastigocladus laminosus) were isolated from mat samples in a thermal gradient between 61–46 °C. The complementary use of proteomic comparisons from these strains, and comparative genomics of F. thermalis pangenomes, suggested that at least two different ecotypes were present within these populations. MALDI-TOF MS analysis separated the strains into three clusters; two with strains obtained from mats within the upper temperature range (61 and 54 °C), and a third obtained from mats within the lower temperature range (51 and 46 °C). Both groups possessed different but synonymous nifH alleles. The main proteomic differences were associated with the abundance of photosynthesis-related proteins. Three F. thermalis metagenome assembled genomes (MAGs) were described from 66, 58 and 48 °C metagenomes. These pangenomes indicated a divergence of orthologous genes and a high abundance of exclusive genes at 66 °C. These results improved the current understanding of thermal adaptation of F. thermalis and the evolution of these thermophilic cyanobacterial species.  相似文献

Adults of Trissolcus basalis and Telenomus podisi were stored either at 15 or 18 °C after their immature development had been completed at 18 or 25 °C. Longevity of the parasitoids in the storage temperatures was evaluated, as well as fecundity and longevity following their return to 25 °C after different periods in reproductive diapause. Temperature during immature development influenced female longevity and highest mean longevity was obtained for females that developed to the adult stage at 25 °C and then were stored at 15 °C (ca. 13 months for T. basalis and 10 months for Te. podisi). For adults of T. basalis that developed at 25 °C, storage periods of 120 or 180 days at 15 or 18 °C did not affect fecundity. The fecundity of T. basalis females that developed at 18 °C and were stored for 120 days at 15 or 18 °C was not affected; however, after remaining for 180 days, fecundity was reduced in ca. 30 and 50%, respectively. Storage of Te. podisi adults at 15 or 18 °C significantly reduced fecundity. It is concluded that adults of T. basalis can be stored in the adult stage at 15 or 18 °C between two soybean crop seasons for mass production purposes, aiming the biological control of stink bugs.  相似文献

Rapana venosa hemocyanin (RvH), a circulating glycoprotein of the marine snail, has a complex structure. To provide details on the stability of the protein, one functional unit, RvH2-e, was compared with the native molecule and the structural subunits, RvH1 and RvH2, via pH–T diagrams, typical phase portraits for stability and denaturation reversibility. By analyzing the T transition curves of RvH2-e at different pH values, several parameters of the thermodynamic functions were obtained. Increasing the temperature from 25 °C to 55 °C, the reversibility of the molecule of protein also increases, opening a reversibility window within the range of pH 4.0–8.0. On analyzing the pH transition curves, the start of the acid denaturation (below pH 6) and alkaline denaturation (above pH 9) was determined to be between 20 °C and 35 °C. For this range, the thermodynamic functions ΔH° and ΔG° for a standard temperature of 25 °C were calculated.  相似文献

The aim of this paper is to assess of the stability and repeatability of the biological soil-quality (BSQ) index over time and at short/long distances in agricultural soils of the Po Valley, Northern Italy, to support its correct use in soil-quality monitoring and is based on soil microarthropods representative of the structure, function and composition of ecological systems, but avoiding classification at the species level. Microarthropods are separated from the soil and weighted using the biological form approach to evaluate their level of adaptation to the soil environment. The field test was organized to assess time and scale effects in a landscape within the coordinates 45°00′ to 45°16′N and 9°50′ to 10°15′E for an estimated area of 200 km2. The BSQ had, in different soils cropped with corn and fertilized with sewage sludge, an average value of 76.4 ± 23.5; it appears to be reproducible both over short distances as well as over short periods of time, but no significant differences (p > 0.05) between years nor between farms were observed. In the landscape conditions tested, it appears reproducible and stable at a field and basin scale, though very sensitive to seasonal climatic variations. From a practical point of view, the information reported in this paper contributes to the standardization of the application of BSQ and provides reference values for this index in soils of the Northern Italy cropped with corn.  相似文献

Chrysoperla genanigra Freitas is a common green lacewing associated with melon pests in the Northeastern Brazil. All life stages of this recently described species were studied under a range of constant temperature conditions (17, 21, 25, 29, 33, 35 and 37 °C), a photoperiod of 12 h:12 h (L:D) and 70 ± 10% relative humidity. Adults of C. genanigra were fed on a diet consisting of a 1:1 (v/v) mixture of brewer’s yeast and honey, while larvae were provided with eggs of Sitotroga cerealella (Olivier) ad libitum. The duration of preimaginal development of the species was inversely proportional to temperature and ranged from approximately 63 days at 17 °C to 15 days at 35 °C. The percentage of adult emergence varied from 6.7% at 17 °C to 76.7% at 25 °C, although no larvae were able to complete development at 37 °C. The lower thermal threshold for total preimaginal development was approximately 10.8 °C and the thermal requirement was 336.7 degree-days. Egg production, along with the longevity of both males and females, were significantly affected by temperature. It is concluded that the best temperature for rearing C. genanigra is 25 °C, with the lowest preimaginal mortality and the highest egg production (992.7 eggs/female).  相似文献

The green alga Caulerpa taxifolia was recorded at Malinska in 1994 and this actually represents the highest northern latitude (45°7′30″N) at which this invasive alga has been found in the world. The alga was widespread at four sites from which it was eradicated by suction pumps during 1996 and 1997. However, it immediately and intensively recolonized all but one site. Throughout 1998 and 1999 the renewed vegetation showed consistent seasonal patterns. The alga nearly disappeared in April and May while regenerating from over-wintering parts of the thalli in summer. The maximum development occurred in autumn and winter with values of biomass (around 200 g dry weight m−2) and frond number (around 2000 m−2) generally lower than those reported for the north-western Mediterranean. Values for the frond length (10–18 cm) were in the same range as those in the north-western basin. Throughout 1998 and 1999 the biomass was closely correlated to frond number and length (adjusted R2 = 0.90). During the following years C. taxifolia entered a phase of regression. The total colonized area, which amounted to several thousands of square metres in 1998, spontaneously declined in 2000 and 2001 so that only several thalli were found in 2004 during a detailed survey of the settlement. No major changes in winter seawater temperatures, ranging from 9.5 to 10.5 °C, were observed in the area from 1994 to 2004. Thus, other unknown processes could likely play a role on specific vegetation patterns of C. taxifolia in Malinska. Accordingly, it is difficult to explain why the still surviving thalli did not proliferate during the favourable summer–autumn period.  相似文献

The effect of different overwintering temperatures (2.5 ± 1 °C in a refrigerator or outdoor natural overwintering on wet topsoil with weak frosts) on the freezing temperature and survival rate of turions of 10 aquatic plant species with different ecological traits (free-floating habit or bottom rooting) was studied using mini thermocouples. Dormant, non-hardened turions of 9 species exhibited freezing within a narrow temperature range of ?7.0 to ?10.2 °C, while Hydrocharis morsus-ranae froze at ?3.6 °C. The survival rate of the turions after the measurements was, however, very low (0–38%). In several species, the freezing temperature of turions at the beginning of germination was not significantly different (at p < 0.05) from the dormant ones. The mean freezing temperature of outdoor hardened turions of 6 species was within a very narrow range of ?2.8 to ?3.3 °C and was thus significantly higher by 4–7 °C (p < 0.0002) than that for the non-hardened turions. It is assumed that the freezing temperatures indicate freezing of the extracellular water. The hardened turions of all 7 species were able to survive mild winter frosts under the topsoil conditions at a rate of 76–100%. These characteristics suggest that the turions of aquatic species can be hardened by weak frosts and that their frost hardiness is based on the shift from frost avoidance in non-hardened turions to frost tolerance.  相似文献

Maltase from Bacillus licheniformis KIBGE-IB4 was immobilized within calcium alginate beads using entrapment technique. Immobilized maltase showed maximum immobilization yield with 4% sodium alginate and 0.2 M calcium chloride within 90.0 min of curing time. Entrapment increases the enzyme–substrate reaction time and temperature from 5.0 to 10.0 min and 45 °C to 50 °C, respectively as compared to its free counterpart. However, pH optima remained same for maltose hydrolysis. Diffusional limitation of substrate (maltose) caused a declined in Vmax of immobilized enzyme from 8411.0 to 4919.0 U ml?1 min?1 whereas, Km apparently increased from 1.71 to 3.17 mM ml?1. Immobilization also increased the stability of free maltase against a broad temperature range and enzyme retained 45% and 32% activity at 55 °C and 60 °C, respectively after 90.0 min. Immobilized enzyme also exhibited recycling efficiency more than six cycles and retained 17% of its initial activity even after 6th cycles. Immobilized enzyme showed relatively better storage stability at 4 °C and 30 °C after 60.0 days as compared to free enzyme.  相似文献

Rheological comparisons have been made between preparations of high methoxy pectin (DE  70%) gelled by acidification with d-glucono-δ-lactone (GDL) on holding for 16 h at 25 °C in the presence of 60 wt% sucrose, and otherwise identical preparations gelled by acidification with citric acid at high temperature and cooling from 90 to 25 °C at 1 °C/min. Two series of experiments were carried out for both methods of acidification. In the first series, the concentration of pectin (c) was held constant at 1.0 wt% and the final pH attained after holding (with GDL) or cooling (with citric acid) was varied from 3.75 to 2.25. In the second series, the final pH was held constant at 3.0 and c was varied from 0.25 to 2.00 wt%. All samples were then heated (1 °C/min) from 25 to 90 °C. Rheological changes on cooling/holding and heating were characterised by low-amplitude oscillatory measurements of storage modulus (G′) and loss modulus (G′′) at 1 rad s?1 and 0.5% strain, and mechanical spectra were recorded at 25 °C. Selected samples, gelled with GDL, were also characterised by compression testing (at 25 °C), and a direct linear relationship was found between the logarithm of yield stress and log G′.The concentration-dependence of moduli for the samples acidified to pH 3.0 with GDL had the form typical of biopolymer gels, with log G′ versus log c approaching a limiting slope of 2 as c was raised above the minimum critical gelling concentration (co  0.3 wt%). Under all conditions of pH and pectin concentration studied, the values of G′′ (at 25 °C) for the samples acidified with citric acid were higher than those of the corresponding GDL-induced networks. The values of G′ were also higher, except at very low pH (below ~2.7 at c = 1.0 wt%) or very high concentrations of pectin. At pectin concentrations above ~1.5 wt%, the moduli of the samples gelled with citric acid (at pH 3.0) levelled out, or decreased slightly, with the values of G′ dropping below those of the GDL-induced networks towards the end of the concentration range studied (at c  2 wt%). All samples acidified with citric acid showed gel-like response (G > G′′) at 90 °C, attributed to hydrophobic association. The downturn in moduli at 25 °C for high concentrations of pectin is attributed to formation and disruption of strong networks during mixing with citric acid at high temperature (“pregelation”). It is suggested, however, that “weak gels” formed at lower concentrations or at pH values above ~2.7 may enhance gel properties by preserving a continuous network as hydrophobic junctions dissociate on cooling and are replaced by hydrogen-bonded junctions, in contrast to random percolation during gelation with GDL at 25 °C. On re-heating from 25 to 90 °C, the reverse processes (dissociation of hydrogen-bonded structures and formation of hydrophobic associations) were evident in an initial reduction and subsequent increase in moduli, as observed in previous studies. Similar heating traces were obtained for samples acidified with GDL to pH values above ~3.0 (at c = 1.0 wt%) or with pectin concentrations below ~1.0 wt% (at pH 3.0). However, at higher concentrations or lower values of pH (i.e. conditions favourable to extensive intermolecular association) an abrupt decrease in G′, with an accompanying maximum in G′′, was observed on heating through the temperature range ~60–80 °C. This is attributed to excessive hydrophobic association, causing collapse of network structure. It is further suggested that, for samples acidified with citric acid, there is preferential association of chain sequences of high ester content into hydrophobic junctions at 90 °C, leaving sequences with a high content of unesterified carboxyl groups available to form long hydrogen-bonded junctions during cooling, and thus giving gels that are stronger and more resistant to network collapse.  相似文献

Heterodera schachtii is a well-known, destructive pathogen of Chinese cabbage (Brassica rapa pekinensis) in Korea, and several studies have attempted to find a potential control measure against it. This study is the first to investigate the effects of varying temperature on the reproduction and damage potential of H. schachtii to Chinese cabbage. Chinese cabbage plants were inoculated with H. schachtii at different densities (1, 2, or 4 juveniles per gram of soil) and grown under three temperature regimes: constant (15, 20, or 25 °C), increasing (10, 14, and 18 °C), and fluctuating (positive, 16.7–22.0 °C; negative, 21.5–11.5 °C). At a constant temperature after 30 days of inoculation, both Chinese cabbage and H. schachtii performed best at 20 °C. However, after 60 days of inoculation, H. schachtii had a significantly higher population at 20 °C, whereas cabbage growth was best at 25 °C. With increasing temperature, the numbers of cysts and females did not change significantly, and reached maxima at an initial temperature of 14 °C. However, the number of leaves and weights of the Chinese cabbage plants significantly differed at 14 °C. Under fluctuating temperatures, temperature decreases reduced the H. schachtii population.  相似文献

The effect of temperature on the biology of Venturia canescens (Gravenhorst) (Hymenoptera: Ichneumonidae) is well understood under constant temperature conditions, but less so under more natural, fluctuating conditions. Herein we studied the influence of fluctuating temperatures on biological parameters of V. canescens. Parasitized fifth-instar larvae of Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) were reared individually in incubators at six fluctuating temperature regimes (15–19.5 °C with a mean of 17.6 °C, 17.5–22.5 °C with a mean of 19.8 °C, 20–30 °C with a mean of 22.7 °C, 22.5–27.5 °C with a mean of 25 °C, 25.5-32.5 °C with a mean of 28.3 °C and 28.5–33 °C with a mean of 30 °C) until emergence and death of V. canescens adults. Developmental time from parasitism to adult eclosion, adult longevity and survival were recorded at each fluctuating temperature regime. In principle, developmental time decreased with an increase of the mean temperature of the fluctuating temperature regime. Upper and lower threshold temperatures for total development were estimated at 34.9 and 6.7 °C, respectively. Optimum temperature for development and thermal constant were 28.6 °C and 526.3 degree days, respectively. Adult longevity was also affected by fluctuating temperature, as it was significantly reduced at the highest mean temperature (7.0 days at 30 °C) compared to the lowest one (29.4 days at 17.6 °C). Survival was low at all tested fluctuating temperatures, apart from mean fluctuating temperature of 25 °C (37%). Understanding the thermal biology of V. canescens under more natural conditions is of critical importance in applied contexts. Thus, predictions of biological responses to fluctuating temperatures may be used in population forecasting models which potentially influence decision-making in IPM programs.  相似文献

A gene encoding a novel β-d-galactosidase from the psychrotolerant Antarctic bacterium Arthrobacter sp. 32cB was isolated, cloned and expressed in Escherichia coli. The active form of recombinant β-d-galactosidase consists of two subunits with a combined molecular weight of approximately 257 kDa. The enzyme's maximum activity towards o-nitrophenyl-β-d-galactopyranoside was determined as occurring at 28 °C and pH 8.0. However, it exhibited 42% of maximum activity at 10 °C and was capable of hydrolyzing both lactose and o-nitrophenyl-β-d-galactopyranoside at that temperature, with Km values of 1.52 and 16.56 mM, and kcat values 30.55 and 31.84 s−1, respectively. Two units of the enzyme hydrolyzed 90% of the lactose in 1 mL of milk at 10 °C in 24 h. The transglycosylation activity of Arthrobacter sp. 32cB β-d-galactosidase was also examined. It synthesized galactooligosaccharides in a temperature range from 10 to 30 °C. Moreover, it catalyzed the synthesis of heterooligosaccharides such as lactulose, galactosyl-xylose and galactosyl-arabinose, alkyl glycosides, and glycosylated salicin from lactose and the appropriate acceptor at 30 °C. The properties of Arthrobacter sp. 32cB β-d-galactosidase make it a candidate for use in the industrial removal of lactose from milk and a promising tool for the glycosylation of various acceptors, especially those which are thermosensitive.  相似文献

A thermostable and pH-stable laccase from Klebsiella pneumoniae was cloned and expressed in Escherichia coli. The recombinant laccase (rLac) achieved a specific activity of 7.12 U/mg after purification by Ni-affinity chromatography. Optimal enzyme activity was observed at pH 4.0 and 35 °C for 2,2′-azino-bis (3-ethylbenzthiazoline sulfonic acid) (ABTS) oxidization and pH 8.0 and 70 °C for 2,6-dimethoxyphenol (2,6-DMP) oxidization. Thermostability and pH stability studies showed that the rLac was stable over the range of 30–70 °C and pH 5.0–9.0 using 2,6-DMP as substrate. Circular dichroism analysis suggested that the secondary structure of the rLac mainly consisted of α-helix that played a vital role in maintaining laccase activity and revealed the potential mechanisms for the changes in laccase activity under varying pHs (3.0–11.0) and temperatures (20–90 °C). Finally, the rLac could decolorize the tested dyes with high decolorization efficiency.  相似文献

It is important to improve the quality of the enzyme inulinase used in industrial applications without allowing the treatment to have any adverse effects on enzyme activity. We achieved preferential chemical modification of the non-catalytic domain of endoinulinase (EC 3.2.1.7) to enhance the thermostability of the enzyme. We used pyridoxal 5′-phosphate (PLP) to modify the more accessible lysine residues at the surface of endoinulinase and then performed a necessary step of reduction with ascorbate. Endoinulinase was incubated in the presence of PLP at various concentrations; this step was followed by reduction of the resulting Schiff base and dialysis. The effects of different PLP concentrations and incubation times on enzyme modification were evaluated. Enzyme deactivation was observed immediately after treatment, even at low PLP concentrations, while reactivation was observed for samples treated with low PLP concentrations after a period of time. Structural analysis revealed that the α-helix content increased from 13.60% to 17.60% after applying the modification strategy; consequently, enzyme stabilization was achieved. The melting temperature (Tm) of the modified enzyme increased from 64.1 °C to 72.2 °C, and a comparative study of thermal stability at 25 °C, 45 °C, and 50 °C for 150 min confirmed that the enzyme was stabilized because of increase in its half-life (t1/2) after PLP modification/ascorbate reduction. The modification process was optimized to achieve the optimum mole ratio for the PLP/endoinulinase (1.37). Excess moles of the modifier are thought to be responsible for enzyme deactivation through unwanted/nonspecific and noncovalent interactions, and the optimization ensured that there was no excess modifier after the desired covalent reaction was complete.  相似文献
