Lactating Friesian dairy cows (2nd-4th parity) which calved in spring (N = 7) or autumn (N = 15) were used. Their ovaries were examined by ultrasound scanning and blood samples were obtained daily for progesterone and oestradiol concentrations from the 5th day after calving until the first post-partum ovulation occurred. Five autumn-calving cows selected at random were bled every 15 min over a 6-h period on 1 day each week for 4 weeks after calving to assess the patterns of LH secretion. Follicular development during the post-partum anoestrous period was characterized by the growth and regression of small (less than or equal to 4 mm) and medium-sized (5-9 mm) follicles, until a dominant follicle (greater than 10 mm) was detected. The first detected dominant follicle ovulated in 14 cows, became cystic in 4 cows (all in autumn), and failed to ovulate in 1 cow. It was not possible to detect a dominant follicle in 3 cows due to scanning difficulties. The post-partum interval to detection of the first dominant follicle (mean +/- s.d.) was shorter (P less than 0.05) in autumn (6.8 +/- 1.8 days) than in spring (20 +/- 10.1 days). However, there was no significant difference between the respective intervals to first ovulation (autumn 27.4 +/- 25.9 and spring 27.3 +/- 18.9 days). Autumn-calved cows which had cysts had longer (P less than 0.001) intervals to first ovulation (58.2 +/- 23.5 days) than did normal cows (12.0 +/- 2.5 days). All cows with cysts had twin ovulations at their first post-partum ovulation. A pulsatile pattern of LH secretion was detected in the first week post-partum and LH pulse frequency was 2-3 per 6-h period in Weeks 1 and 2 post partum and increased to 5-7 pulses per 6-h period in the presence of a dominant or cystic follicle. Concentrations of progesterone in plasma during post-partum anoestrus were usually low (less than 0.2 ng/ml); oestradiol concentrations were also low (less than 5 pg/ml), but higher values (5-110 pg/ml) were observed in cows that had a dominant or a cystic follicle.  相似文献

Effects of restricted suckling frequency and bromocryptine treatment on return of ovarian cycles after calving were studied on 151 Angus and Friesian Angus crossbred cows. A control group consisted of cows and calves grazed together. In two groups, calves were allowed to suckle for one hour only each morning (1 x S) or one hour each morning and afternoon (2 x S). A further group of cows, grazed with their calves, were treated with 10 mg bromocryptine per day from days 2-11 post-partum (BCT). Interval from calving to resumption of ovarian cycles, assessed from oestrous records and weekly progesterone samples, did not differ between control and 2 x S cows. Interval from calving to a recorded progesterone sample greater than 1.7 ng/ml associated with oestrus in 1 x S cows was 13 days shorter than for control cows (63 vs 76 days, SED = 1.4, p < 0.05) although, the difference in interval to first oestrus of 11 days was not significantly different in these groups (64 vs. 75 days, SED = 4.5). Time from calving to resumption of ovarian cycles did not differ between BCT and control cows. Collectively these data indicate that suckling frequency per se is not a major factor influencing resumption of ovarian cycles after calving. Instead they suggest the association of the cow and its calf may influence the long post-partum anoestrous intervals in these animals.  相似文献

Superovulation and embryo recovery from peripubertal Holstein heifers   总被引:1,自引:0,他引:1  
The use of peripubertal donors in embryo transfer (ET) programs presents significant opportunity to accelerate genetic gain in domestic livestock by reducing the generation interval. These studies were designed to evaluate feasibility of superovulation and embryo recovery in peripubertal heifers (starting at 7.8 months of age), and to determine whether subsequent reproductive and lactational performance of donor heifers were impaired. Study 1 utilized 10 pairs of contemporary full-sibling heifers in which one heifer in each pair was assigned to receive a superovulation regimen and her full-sibling contemporary received placebo. Treated heifers were artificially inseminated at estrus and embryos were flushed transcervically 4-6 days later. Based on recovery of oocytes and/or embryos, 9 of 10 heifers responded to the hormonal regimen and 12 total embryos were recovered. Seven embryos (58%) were transferred into recipients resulting in five pregnancies. Control and treated heifers remained in the herd and were bred at a natural estrus by AI at 15 months of age. Lactation records, i.e., 305 days mature equivalent (305 d ME) were obtained, and all animals were evaluated for udder conformation traits between 32 and 38 months of age. Reproductive traits (age at first calving and days to conception) and lactational traits of heifers subjected to embryo transfer and their non-treated full-siblings did not differ (P > 0.05). Study 2 was conducted to establish the commercial feasibility of hormonally programming peripubertal heifers ranging in age from 7.8 to 9.9; 10 to 11.9; 12 to 13.9 and >/= 14 months. In total, 3982 embryos were recovered from 520 heifers, with 2419 (60.7%) of those categorized as viable (transferable). The number of ova/embryos obtained per flush (5.6 +/- 1.0) and the number of transferable embryos (2.8 +/- 0.5) was reduced (P < 0.05) in heifers of age 7.8-9.9 months compared to all other age groups. There was no difference (P > 0.05) in the number of ova/embryos recovered (7.8 +/- 0.3), or the number of transferable embryos (4.8 +/- 0.2), among heifers that were >/=10 months of age. The number of unfertilized ova did not differ by age, however, more degenerate embryos tended to be recovered from heifers <10 months of age compared to heifers >/=14 months of age. These data indicate that transferable embryos can be safely recovered from heifers beginning at 10 months of age without compromising subsequent reproductive or lactational performance of the donor.  相似文献

Somatotropin and FSH act synergystically on insulin-like growth factor-I (IGF-I) synthesis in ovarian follicles; IGF-I regulates several granulosa cell specific functions and may thereby be beneficial in bovine superovulation. In a series of 3 experiments we investigated the effects of recombinant bovine somatotropin (rBST) on several parameters of the superovulatory response in dairy cows. A total of 81 Holstein Friesian crossbred dairy cows received either 640 mg rBST or the vehicle (controls) on Day 4 or 13 of the superovulation schedule. Superovulation was induced with 2500 IU PMSG on Day 9. The cows were artificially inseminated on Day 13. In Experiment 1, on Days 4, 8, 11, 13 and 17 4 to 5 animals each were slaughtered to obtain follicular fluid, endometrium and plasma. The rBST application increased IGF-I contents in plasma and follicular fluid on Days 8, 11 and 13 (P < 0.05) in the treated cows when compared with that of the controls. Plasma and follicular IGF-I contents were correlated closely (rBST: r = 0.90, n = 10; control: r = 0.94, n = 9). The number of antral follicles increased following rBST treatment, and on the day of artificial insemination (AI) twice as many follicles > 4 mm were counted in the rBST treated animals than in the control group. In Experiment 2, the flushing of 38 donors on Day 7 after AI resulted in more transferable embryos in the rBST group than in the control group (4.2 +/- 1.0 vs 2.5 +/- 0.7; P < 0.05). In contrast, in Experiment 3 involving 21 animals when rBST was administered at the time of AI the superovulation response was not altered. It is concluded that rBST increases follicular and plasma IGF-I contents and thereby has profound effects on follicular and early embryonic development.  相似文献

To understand the causes for poor response to superovulation in mature cows of high genetic potential, endocrine and follicular events during and after superovulation were compared in heifers (<2 yr old) yielding large numbers of embryos and cows (9 to 13 yr old) known to be poor embryo donors. Follicular development was monitored by daily ultrasonography. Blood samples were taken 2 to 3 times a day for the measurements of P4, E2, FSH and LH by RIA. Intensive blood collections at 15-min intervals for 6 h were also performed during preovulatory and luteal phases. The number of embryos produced in the heifers (15.2 +/- 2; mean +/- SEM) and the cows (0.6 +/- 0.4), was similar to the number of ovulatory follicles derived from ultrasonographic observations in the heifers (16.2 +/- 3.7), but not in the cows (7.8 +/- 2.8). Contrary to that observations in heifers, there was no increase in the number of 4- to 5-mm follicles in cows during superovulation. The number of larger follicles (>5 mm) increased during superovulation in both cattle groups, but it was significantly lower in cows than in heifers. During superovulation, the maximal E2 concentration was greater (P < 0.0001) in heifers than in cows. One cow showed delayed luteolysis during superovulation, while another had abnormally high FSH (>10 ng/ml) and LH (>3 ng/ml) concentrations following superovulation. All the cows had a postovulatory FSH rise which was not detected in the heifers. The results showed that attempts to improve superovulatory response in mature genetically valuable cows are hampered by a number of reproductive disorders that are not predictable from the study of the unstimulated cycle.  相似文献

The objective of this experiment was to determine the effect of prepartum lipid supplementation on the number and quality of embryos recovered following ovarian super-ovulation in postpartum suckled beef cows. Mature cows (n = 40) were assigned to one of two treatments (lipid versus. no lipid) and supplemented for approximately 40 days prior to calving. Supplements provided to cows were isocaloric and isonitrogenous. The treatment group was fed 1.6 kg hd(-1) per day of whole soybeans (WSB; 19.8% ether extract, and 41.8% crude protein) and the control group received a supplement consisting of 1.8 kg hd(-1) day of a soybean meal and soy-hull combination (SBS; 2.15% EE and 36.81% CP). Cows were synchronized using a GnRH [Cystorelin((R)) 100 microg im]-GnRH-PGF(2alpha) [Lutalyse 25 mg im] protocol. Cows were administered two injections of GnRH seven days apart and PG seven days after the second GnRH injection. Twenty-eight cows (WSB, n = 15; SBS, n = 13) responded to estrus synchronization and were superstimulated. Super-ovulation was initiated on day 8-10 of the synchronized cycle by twice-daily injections of pFSH (Pluset) over four days in decreasing doses using a total of 608.4 IU per cow. Prostaglandin F(2alpha) was administered 96 and 108 h after super-stimulation was initiated with FSH. Days postpartum (WSB = 59 days; SBS = 57 days) at initiation of FSH treatments were similar (P > 0.10) for both treatments. Cows were monitored for estrus activity by the HeatWatch Estrus Detection System. Twenty-seven cows (WSB, n = 15; SBS, n = 12) exhibited estrus after FSH and inseminated at 0, 12, and 24 h after the onset of estrus with 1, 2, and 1 units of semen, respectively. Embryos were recovered and evaluated 7-8 days later. Only cows that responded to FSH and that were inseminated were used for statistical analysis. Data were analyzed using the General Linear Models Procedure of SAS. Body condition scores did not differ (P > 0.10) between treatments when cows were evaluated at the initiation of the experiment, two weeks prior to calving, and at initiation of superovulation with FSH. Estrous cyclicity prior to the initiation of estrus synchronization did not differ (P > 0.10) between treatments. There was no difference (P > 0.10) between treatments in recovery of total embryos (WSB, 14.7 +/- 3.5; SBS, 17.5 +/- 3.0), transferable embryos (WSB, 10.3 +/- 2.5; SBS, 13.6 +/- 2.6), degenerate embryos (WSB, 3.3 +/- 1.1; SBS, 1.6 +/- 1.7) or unfertilized ova (WSB, 1.1 +/- 0.5; SBS, 2.3 +/- 1.2). Cows that were supplemented with whole soybeans prior to parturition failed to produce an increased total number of ova or transferable embryos following super-ovulation.  相似文献

Eighty Holstein dairy cows were treated with 25 mg of prostaglandin F2alpha (PGF2alpha) on Days 14 to 16 post calving. Eighty-four herd mates served as saline controls in the double blind study. The reproductive parameters used to measure fertility were mean days to first service on all cows and mean days open, first service conception rates and services per pregnancy on cows that became pregnant. Mean days to first service was similar in both groups (71.8 +/- 27, treated; 68.5 +/- 28.6, controls; P = 0.5352). The mean days open was 98.6 +/- 52.0 d for treated cows compared to 118.8 +/- 71.2 d for controls (P = 0.0680). First service pregnancy rates (treated, 41.3%; control, 35.7%) were not significantly different (P = 0.0630); however, the mean services per pregnancy (treated, 1.64; control, 2.33) were significantly (P = 0.0021) improved in the treated group. Ten cows in the treated group and twelve cows in the control group were diagnosed as having retained fetal membranes and/or metritis. For treated and control cows mean days to first service were 82.2 +/- 34.8 d and 82.8 +/- 58.7 d (P = 0.5652). Days open were 97.0 +/- 32.5 d and 133.4 +/-58.4 d (P = 0.3636); services per conception were 1.83 and 2.50 (P = 0.3178), respectively. In all treated cows reproductive parameters were similar whether cows had high serum progesterone ( > 1 ng/ml) or low serum progesterone ( < 1 ng/ml) on the treatment day. This study suggests that early postpartum treatment of lactating dairy cows with prostaglandin produces an improvement in fertility.  相似文献

The effect of suckling on post-partum ovarian function was monitored by weekly plasma progesterone determinations using enzyme immunoassay in 16 Small East African Zebu (Bos indicus) cows maintained with a fertile bull. Five cows continuously suckled their calves (COS), but starting 30 d after calving 6 cows remained with their calves which were prevented from suckling using nose plates (NOS) while 5 calves were separated and penned (PEN). The mean (+/- SE) interval from calving to first behavioural oestrus was 81 +/- 11 d and was significantly (P less than 0.001) shorter for PEN (43 +/- 10 d) than NOS (72 +/- 9 d) or COS cows (133 +/- 11 d). COS cows nonetheless weaned heavier (P less than 0.001) calves at 5 months. Ovarian activity determined by plasma progesterone levels greater than or equal to 1.3 ng/ml indicated that "silent ovulation" was initiated in 44% of cows by 40 +/- 6 d post-partum. The results indicate that continuous suckling or constant cow-calf interaction extend the post-partum anoestrous interval in Ethiopian Zebu cows. However, early-weaned calves or those on restricted suckling would need extra nutritional supplementation to ensure continued growth.  相似文献

The ovaries of 18 post-partum beef suckler cows were examined daily, using ultrasound, from Day 5 post partum until a normal oestrous cycle was completed. Periods of growth and regression of medium-sized (5-9 mm) follicles were identified before one medium follicle became dominant (single large follicle greater than or equal to 10 mm). The mean (+/- s.e.m.) number of days from parturition to detection of the first post-partum dominant follicle was 10.2 +/- 0.5. The first post-partum dominant follicle ovulated in 2/18 (11%) cows. The interval from calving to first ovulation (mean +/- s.e.m. = 35.9 +/- 3.3 days) was characterized by the growth and regression of a variable number (mean = 3.2 +/- 0.2; range 1-6) of dominant follicles. The maximum diameter of the dominant follicle increased as the cows approached first ovulation (P less than 0.05). Behavioural oestrus was not detected in 16/18 (89%) cows at first ovulation. Following first ovulation, the length of the subsequent cycle was short (mean = 9.7 +/- 0.5 days; range 8-15 days) in 14/18 (78%) cows and was characterized by the development and ovulation of a single dominant follicle. During oestrous cycles of normal length (mean = 20.6 +/- 0.5 days; range 18-23 days) one (N = 2), two (N = 7) or three (N = 8) dominant follicles were identified. The growth rate, maximum diameter or persistence of non-ovulatory dominant follicles before first ovulation or during oestrous cycles were not different (P greater than 0.05). These data show that, in beef suckler cows, follicular development and formation of a dominant follicle occur early after parturition and the incidence of ovulation of the first dominant follicle is low. The number of dominant follicles that develop before first ovulation is variable; first ovulation is rarely associated with oestrus and short cycles are common after first ovulation. It is concluded that prolonged anoestrus in post-partum beef suckler cows is due to lack of ovulation of a dominant follicle rather than delayed development of dominant follicles.  相似文献

Variability in the superovulation response is an important problem for the embryo transfer industry. The objective of this study was to determine whether FSH treatment at the beginning of the cycle would improve the ovulation rate and embryo yield in dairy cows. Twenty-eight postpartum cyclic dairy cows were allocated at random to 4 treatment groups (A, B, C and D). Group A cows (n = 10) received FSH (35 mg) at a decreasing dose, starting on Day 9 (Day 0 = day of estrus) for 5 days followed by PGF(2alpha) (35 mg) on Day 12. Cows assigned to Groups B, C and D (n = 6 cows each, respectively) were given 35 mg FSH at a decreasing dose from Days 2 to 6 followed by PGF(2alpha) on Day 7. Group C and D cows received PRID inserts from Day 3 to Day 7. Cows in Group D additionally received 1000 IU hCG 60 hours after PGF(2alpha) treatment. Ovaries were scanned daily using a real time ultrasound scanner from the beginning of FSH treatment until embryo recovery, to monitor follicular development, ovulation and the number of unovulated follicles. Embryos were recovered from the uterus by a nonsurgical flushing technique 7 days after breeding. There were no differences (P>0.01) in the number of follicles > 10 mm at 48 hours after PGF(2alpha) treatment among the 4 groups. The mean numbers of follicles were 10.6 +/- 1.2, 9.3 +/- 1.3, 12.2 +/- 1.3 and 15.0 +/- 2.9 for Groups A, B, C and D, respectively. A significantly (P<0.001) higher number of ovulations was observed and a larger number of embryos was recovered in Group A than in the other groups. The results of this study indicate that superovulation with FSH at the beginning of the cycle causes sufficient follicular development but results in very low ovulation and embryo recovery rates.  相似文献

To determine whether follicular development, superovulation and embryo production were affected by the absence or presence of a dominant follicle, cows were administered injections of FSH twice daily in the early (Days 2 to 6, estrus = Day 0) or middle stage (beginning on Day 10 or 11) of the estrous cycle. Treatment with FSH early in the cycle stimulated follicular development in 83 to 100% of all cows from 4 groups evaluated at different times after PGF2alpha treatment on Days 6 and 7. However, the proportion of cows with > 2 ovulations varied from 31 to 62.5%, indicating that induction of follicular development may occur in the absence of superovulation. When compared with cows treated in the middle of the cycle, no differences were observed in the proportion of cows with > 2 ovulations (31 vs 20%), ovulation rate. (26.0 +/- 6.3 vs 49.6 +/- 25.8), production of ova/embryos (13.3 +/- 3.2 vs 14.4 +/- 3.4), or the number of transferable embryos (8.0 +/- 3.6 vs 5.4 +/- 1.5; early vs middle, respectively). The proportion of the total number of embryos collected that were suitable for transfer was greater (P<0.01) in cows treated early in the cycle (60%) than at midcycle (37.5%). The diameter of the largest follicle observed by ultra-sound prior to initiation of FSH treatment in the early stage of the cycle (10.0 +/- 2.0 mm) was smaller (P<0.05) than at midcyle (16.8 +/- 1.3 mm). These results demonstrate that superinduction of follicular development is highly consistent after FSH treatment at Days 2 to 6 of the cycle and that superovulation and embryo production are not less variable than when FSH is administered during the middle of the cycle. However, superovulation in the early stage of the cycle may increase the proportion of embryos suitable for transfer.  相似文献

Genetic trends for 305-day milk yield (P305) in Brazilian Guzerat herds under selection were compared. Data from 4898 lactations of 3179 purebred and crossbred cows from various regions of Brazil were used. Milk yield was adjusted for mature age and the contemporary groups were defined as herd and calving year. Genetic parameters were estimated using the MTDFREML program. The model included the random effects of animals and permanent environment, and herd-calving year, calving season and genetic composition as fixed effects. Genetic trends were estimated by linear regression of weighted average estimated breeding values as a function of calving year. The average P305 was 2065 +/- 922 kg and the heritability was 0.23 +/- 0.03. The annual genetic trend in estimated breeding values of cows for P305 was 7.09 +/- 0.71 kg between 1987 and 2004, and 6.47 +/- 2.35 kg between 1997 and 2004. For cows born and raised in the multiple ovulation and embryo transfer (MOET) nucleus, this trend was 36.46 +/- 24.54 kg/year between 1997 and 2004, 183.14 +/- 47.94 kg/year between 1997 and 2000, and 9.13 +/- 19.19 kg/year between 2001 and 2004. An average inbreeding coefficient of 0.04 was found for inbred MOET cows in 2004. Increasing the size of the family and introducing new progenies changed reliabilities and predicted transmitting ability estimates of MOET sires. In conclusion, there was a positive genetic trend for milk yield in the MOET nucleus at low inbreeding coefficients due to the increased accuracy and estimated genetic merit, but no changes in the average milk yield were observed.  相似文献

Data on productive and reproductive performance of cows in dual-purpose herds were analyzed to determine the effect of some environmental and genetic factors on saleable milk yield (SMY), lactation length (LL), daily saleable milk per lactation (DMYL), calving interval (CI), and daily saleable milk per calving interval (MYCI) in dual-purpose herds in Yucatan, Mexico. Repeatabilities (re) for these traits were also estimated. Data were obtained from monthly visits to 162 herds from 1996 to 2000. The fixed factors studied were: region (central, eastern and southern), parity number (1 to 6), calving year (1996 to 2000) and calving season (dry, rainy and windy and rainy), genetic group (low- (< 0.50%), medium- (50%) and high- (> 50%) grade cows with European genes). About 2700 to 7700 cows were evaluated for each trait. All factors had significant effects (P < 0.05) on the traits except for region on CI and calving season on DMYL. The overall means for SMY, LL, DMYL, CI, and MYCI were: 1322.3 +/- 80.5 kg, 224.8 +/- 1.3 days, 5.8 +/- 0.1 kg, 555.1 +/- 16.5 days, and 3.0 +/- 0.1 kg, respectively. The re values for SMY, LL, DMYL, CI, and MYCI traits were: 0.19 +/- 0.03, 0.08 +/- 0.04, 0.16 +/- 0.04, 0.00 +/- 0.08, and 0.08 +/- 0.07, respectively. First parity cows had lower SMY, shorter LL, longer CI, and lower MYCI means than cows with more than one parity. Medium grade cows produced more SMY, DMYL and MYCI and had shorter CI than low- and high-grade cows. Therefore, under Yucatan conditions medium-grade cows should be exploited, and more attention should be given to first parity cows in order to improve the productivity in the herd. The relatively high re estimates for SMY and DMYL can be used to calculate most probable producing abilities, in order to identify which cows should be culled.  相似文献

Claims made for prophylactic effects by a progestin (Chlormadione acetate = CAP) on fertility, given peroral daily to dairy cows from Day 15 to 35 post-partum, were investigated in field trials in a total of 1516 cows in Northern Germany, on 12 privately and one government-owned dairy farm, between 1969 and 1973. All animals were inseminated at estrus after Day 50 post-partum. For all treated animals, significant reduction of the calving interval by 11 days (p < 0.01) was recorded. In 942 cows, in which comparisons between treatment and control animals under identical environmental conditions were made, calving intervals were reduced by 10.5 days (p < 0.01) and the percentage of cows remaining infertile over two years was reduced from 12.4 % in the control population to 6.4 % in treated cows (p < 0.05). These advantages of CAP were most pronounced in cows older than 5 years and animals producing less than 25 kg milk/day during the first two months post-partum. In individual farms, calving intervals between treatment and control groups ranged from insignificant differences up to 44 days. Predictability of breeding dates, assurance of desirable short calving intervals and improved fertility makes this a tool for problem farms and well managed dairies alike.  相似文献

The objective of the present study was to determine the possibility to improve the reproductive performance of buffalo cows through the continuous exposure to bull with grazing and free-stall housing management. Sixty-four Egyptian multiparous buffalo cows raised under two different management systems in two farms were used in this study. The cows in the first farm (management system 1, MS1) were loose--housed in a free-stall yard, grazed for 4 h per day, suckled their calves for 2-3 months and were continuously exposed to a fertile bull. The cows in the second farm (management system 2, MS2 ) were confined in an open-fronted tie-stall shed, not grazed, suckled their calves for only 7 days and were exposed to a fertile bull twice per day (30 min per session). All the cows were fed a diet of green berseem (Trifolium alexandrinum), rice straw and concentrates to meet their maintenance and production requirements. The cows during both the treatments were milked twice per day after weaning. The cows in both groups were between the second and the sixth parity, weighed 450-480 kg and had average daily milk yields of 5.0-6.0 kg. In each farm, cows were visually checked twice daily at 07:00 and 17:00 h for the signs of oestrus and animals proved standing heat were naturally mated. Rectal palpation was used to monitor uterine involution and for pregnancy diagnosis. Blood was sampled twice per week from 7 to 150 days post-partum for serum progesterone assay. The results revealed that post-partum intervals to each of first ovulation, first oestrus, conception and next parturition were significantly (P < 0.05) shorter in MS1 group than in MS2 group. In the meantime, MS1 increased (P < 0.01) the conception and calving rates by 21 and 25%, respectively compared to MS2. Percentages of post-partum cyclic animals and animals exhibiting ovulatory oestrus were greater (P < 0.01) in MS1 group than in MS2 group. However, the percentage of animals cycling before day 60 post-partum was significantly (P < 0.01) lower in MS1 group than in MS2 group (13% versus 28%). By day 120 post-partum, only 63% of the buffaloes were cycling in MS2 group versus 94% in MS1 group. Percentage of silent ovulation was insignificantly higher in MS2 group (34%) than in MS1 group (25%). However, the percentage of false oestrus was higher (P < 0.01) in MS1 group than in MS2 group (16% versus 3%). In addition, percentage of short ovulatory cycles (15-17 days) was greater (P < 0.01) in MS1 group than in MS2 group, whereas percentage of long ovulatory cycle (25-28 days) was higher (P < 0.01) in MS2 group than in MS1 group. It was concluded that continuous exposure of buffalo cows to a fertile bull with grazing management under free-stall housing system enhances resumption of post-partum ovarian activity and improves conception and calving rates.  相似文献

Plasma progesterone and LH concentrations around estrus were determined for both PMSG treated (experimental animals) and non-treated (control animals) dairy cows and heifers of the Holstein Friesian and Jersey breeds, and these hormone profiles were related to the embryo quality. Most experimental animals experienced an increase in progesterone concentrations following PMSG treatment and an abrupt decrease to values below 3 nmol/l after PG injection. The mean (+/-SE) intervals from prostaglandin treatment to estrus were 46.9+/-1.8 h and 64.5+/-4.8 h for experimental and control animals, respectively. At the onset of heat the progesterone concentration in experimental animals with optimal embryo quality (group I) was significantly lower (p<0.01) than in experimental animals which yielded unfertilized eggs (group II) (1.2+/-0.1 versus 3.9+/-0.8 nmol/l) and significantly higher than the level in the control group (0.6+/-0.1 nmol/l). Following estrus the progesterone profiles in all 3 groups were studied and the length of the superovulatory cycle was measured to 26.0+/-4.8 days. The preovulatory LH surge occurred sooner after prostaglandin injection in experimental (41 h) than in control animals (65 h). The LH surge in group I occurred within a narrow range and reached a higher average level than group II (24.2+/-2.2 ng/ml and 16.3+/-3.7 ng/ml, respectively). The control group attained an even higher LH surge (31.8+/-8.8 ng/ml) than did the experimental animals. The data presented in this experiment indicate that plasma levels of progesterone and LH in PMSG-PGF(2)alpha treated animals are related to embryo or egg quality.  相似文献

In post-partum anestrous beef cows suckling calves, neither the choice of hormonal regime to ensure the presence of a healthy dominant follicle at the end of a progestagen treatment nor the optimum hormone to induce estrus and ovulation is clear. Twenty-eight beef cows, in good body condition, 25-30 days post-partum, were assigned to one of four treatments: (i) 3mg norgestomet (N) implant with 5mg estradiol valerate (EDV) and 3mg N injection at the time of insertion (Crestar) for 5 days followed by 600 IU eCG at the time of implant removal; (ii) Crestar for 5 days as in (i) followed by 0.75 mg estradiol benzoate (EDB) 24h later; (iii) Crestar for 9 days followed by 600 IU eCG at the time of implant removal; and (iv) Crestar for 9 days followed by 0.75 mg EDB 24h later. Ovarian scanning was preformed from 4 days before implant insertion until ovulation and 4 days postovulation to detect the CL. Daily blood samples were collected from day 20 post-partum until second ovulation for FSH and E(2) assay. Data were analyzed using analysis of variance. There was no effect of the stage of follicle wave at the time of implant insertion on interval to new follicle wave emergence (range 1-7 days; mean 4.7 days). FSH concentrations were decreased to 5.9+/-2.0 and 7.7+/-1.1 ng/ml for pre- and post-selection cows 1 day after start of treatment; thereafter, they increased on Day 2 to 7.9+/-2.0 and 11.0+/-1.1 ng/ml and on Day 3 to 10.3+/-2.7 and 11.4+/-1.7 ng/ml for pre- and post-selection cows, respectively, despite high-estradiol concentrations at that time. There was no effect of treatment on the interval from implant removal to ovulation (3.2-4.0 days) or on the number of cows detected in estrus (26 of 27 cows). The size of the ovulatory follicle in cows given 0.75 mg EDB 24h post implant removal was decreased in animals at the pre-selection stage (12.2+/-0.1mm) of the follicle wave compared with those at the post-selection stage (15.3+/-0.9 mm) at implant removal. Cows given 600 IU eCG at the pre-selection phase of follicular growth had multiple ovulations (4.0+/-1.1). Cows given EDV at the start of a 5-day implant period had higher estradiol concentrations before and on the day of implant removal than those given EDV at the start of a 9-day implant period. The injection of 0.75 mg EDB 1 day after implant removal tended to increase concentrations of estradiol one day later. In conclusion, 5mg EDV and 3mg N at insertion of a 3mg N implant resulted in variable new follicle wave emergence 1-7 days later in post-partum beef cows nursing calves (22 of 27); both eCG and EDB were equally effective at inducing estrus after implant removal in cows in good BCS, but eCG resulted in a significant increase in ovulation rate in cows treated before dominant follicle selection.  相似文献

Recombinant bovine somatotropin (rbST) has been shown to increase follicular growth in cattle and some studies have demonstrated an increase in superovulatory response for rbST-treated cows. Pregnancy rates have also been shown to increase when rbST was administered around the time of insemination or prior to embryo transfer. The application of rbST for the purpose of increasing superovulatory responses of donor cows and increasing pregnancy rates of recipient heifers was tested in a commercial embryo transfer program. In Experiment 1, embryo donor cows (n = 56) underwent three cycles of control superovulation (two before and one after weaning) and subsequently underwent up to four additional superovulations while being treated with either rbST (500 mg sustained-release rbST; Posilac, Monsanto, St. Louis, MO; n = 28) or excipient (control; n = 28) once every 14 days. In Experiment 2, lactating embryo donor cows (n = 37) underwent a control superovulation and then underwent a superovulation while lactating and being treated with either rbST (n = 16) or excipient (n = 21). In Experiment 3, embryo recipient heifers that were being implanted with either in vitro or in vivo produced embryos were treated with either rbST (n = 146) or excipient (n = 143) at the time of embryo transfer. Treatment of non-lactating (Experiment 1) or lactating (Experiment 2) donor cows with rbST during repeated superovulation did not affect the number of corpora lutea, the sum of transferable embryos, degenerate embryos, and unfertilized oocytes, or the number of transferable embryos. Treatment of recipient heifers with rbST (Experiment 3) did not affect pregnancy rates for either in vitro or in vivo produced embryos. We conclude that superovulatory response and pregnancy rates (respectively) are similar to control for rbST-treated cows undergoing repeated superovulations and rbST-treated recipient heifers treated at the time of embryo transfer.  相似文献

Reduced reproductive performance and lower conception rates of lactating cows are closely associated with genetic progress for high milk production. In contrast, the fertility of nulliparous Holstein heifers has remained fairly stable over the years and appears to be markedly higher than that of mature lactating cows. Possible differences in oocyte quality and follicular steroid levels, which could be associated with the low fertility of high-lactating cows, were examined in 13-month-old heifers, cows around the time of first AI (60-95 d post-partum, yielding 49+/-2.4 kg/d) and cows at mid-lactation (120-225 d post-partum, yielding 37+/-2.1 kg/d). Estrus was synchronized by two doses of PGF2alpha and follicles (5-8 mm) were aspirated on days 4, 8, 11 and 15 of the cycle by an ultrasound-guided procedure. Oocytes were morphologically examined, matured in vitro, chemically activated and cultured for 8d. Cleavage rate and the proportion of developing parthenogenetic blastocysts were determined on days 3 and 8 post-activation, respectively. On day 17, heifers and cows received additional PGF2alpha and follicular fluids from preovulatory follicles were collected on day 19 perior to the expected estrus. Follicular-fluid volumes were similar in cows and heifers, as were estradiol, progesterone and androstenedione concentrations in the follicular fluid. Percentages of high-grade oocytes, proportions of cleaved oocytes and developed blastocysts did not differ between the groups. Results suggest that the fertility gap between nulliparous heifers and high-lactating cows is not directly related to steroid content in the preovulatory follicular fluid or oocyte developmental competence.  相似文献

Repeatabilities of total numbers of ova (TO) and morphologically normal embryos (MNE) recovered following superovulation of Brangus cows were estimated from records of 210 attempts involving 75 cows. Repeatability of the number of TO recovered was 0.22 +/- 0.08 and the number of MNE recovered was 0.19 +/- 0.08. Overall mean numbers of TO and MNE were 9.0 and 5.0, respectively. The mean numbers of TO recovered were 8.9 from the first treatment, 9.5 from the second treatment, and 8.6 from the third treatment. The mean numbers of MNE recovered were 5.2 from the first treatment, 4.6 from the second treatment, and 5.2 from the third treatment. Regressions of TO or MNE numbers per collection indicated that the superovulatory response did not decline significantly with repeated superovulation.  相似文献
