Survival of adult Daphnia magna was assessed after acute (<96h) exposure to UV312 under various temperatures (6, 12 and 18°C)or oxygen concentrations (5.6, 8.5 and 14.1 mg O2 l–1)in the laboratory. The surviving animals were screened for theenzymes catalase (CAT) and glutathione transferase (GST), whichmay protect against UV-induced oxidative damage. In addition,the same two enzymes were assayed in separate experiments afteracute exposure to UV312 (6 h, 0.014 mW cm–2) and the differentlevels of temperature and oxygen. No differences were observedin either CAT or GST activity after exposure to the three levelsof oxygen, but there was a tendency for decreasing specificactivity with decreasing temperature for both enzymes. CAT activitywas not influenced by UV radiation, whereas GST activity displayeda slight increase. Oxygen concentration did not influence survivalduring UV exposure but, contrary to expectations, survival testsat different temperatures clearly showed that reduced temperatureincreased survival. The results indicate that temperature effectsmust be considered when comparing dose effect relationshipsin situ, and suggest that low temperature is not a major causeof UV susceptibility in cold-adapted alpine and Arctic populationsof Daphnia.  相似文献

Seasonal changes in the structure of the zooplankton communitiesof two large (18 500 m3) experimental enclosures (‘LundTubes’) are reviewed. The diets of the two most prominentcrustaceans, Daphnia hyalina and Diaptomus gracilis, were investigatedby direct analyses of gut contents and in a series of shortterm experimental manipulations of communities isolated in 6m3 bags, placed in one of the enclosures. The relative abundanceof Daphnia hyalina generally coincided with warmer water temperaturesbetween late spring and late summer, but its numbers fluctuatedwithin that period, apparently in response to the availabilityof smaller algae (<104 µm3 unit volume: chiefly cryptomonads,chlorellids and Ankyra). Daphnia numbers declined steadily throughthe summer in the enclosure overwhelmingly dominated by Microcystis,although it did feed on smaller colonies (< 105 µm3in volume). Diaptomus gracilis and most rotifers were also prominentin the first half of the year, but peak abundances of Keratellaquadrata followed the presence of large populations of small(<103 µm3) algae. 1. The predation impact of underyearling perch (20 mm total length) on the dynamics of Daphnia galeata was studied in three 6.5-m3 enclosures during a 17-day experiment in June 1995. These data were compared with zooplankton succession in three fish-free control enclosures and in the pelagic zone of Bautzen reservoir, Germany. 2. Due to individual growth during the experiment, fish biomass in the enclosures increased from 210 mg wet body mass (w.b.m.) m–3 to 830 mg w.b.m. m–3, equivalent to an increase from 20 kg ha–1 to 75 kg ha–1. 3. In the enclosures with fish, biomass of daphnids decreased steadily to values below 1 mg wet weight (w.w.) l–1 within 17 days, whereas in the fish-free controls and in Bautzen reservoir the Daphnia biomass fluctuated around 8 mg w.w. l–1. Other zooplankton species exhibited little or no change. Approximately 60 kg ha–1 was calculated as the critical underyearling perch biomass which may induce a drastic decline of the Daphnia galeata population in Bautzen reservoir. Comparison with values from other lakes is difficult due to differences in water depths and Daphnia biomasses. 4. Mean individual biomass of daphnids, egg ratio and proportion of adult daphnids were significantly lower in the enclosures with fish compared with the control enclosures at the end of the experiment. This may be explained partly by preferential predation of the large, egg-carrying daphnids by fish. However, no difference was found in clutch size and size at first reproduction, possibly due to the short duration of the experiment. 5. It must be assumed that the ‘summer depression’ of daphnids observed in many waters is not the exclusive effect of direct reduction of daphnids by fish predation. Even the high biomass of underyearling perch stocked in the enclosures did not completely account for Daphnia mortality. Instead, the selective loss of large size classes, combined with low food resources and reduced reproduction rates, may induce the marked declines in daphnids.  相似文献

Abstract

The effects of no. 20 diesel oil exposure, 2,4-dichlorophenol (2,4-DCP) exposure and combined exposure on the antioxidant defences of Daphnia magna have been studied systematically for the first time. Daphnia magna was exposed for 1 day or 10 days to several concentrations of 0, 0.005, 0.01, 0.05, 0.1, 0.5 or 1.0 mg L?1 solutions. Antioxidant defences consisting of the levels of reduced glutathione (GSH) and glutathione disulfide (GSSG) and activities of superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), selenium dependent glutathione peroxidase (Se-GPx) and glutathione S-transferase (GST) of daphnids were determined to evaluate their protective roles and to analyse the occurrence of oxidative stress. The possible antioxidant defence mechanisms are discussed. Furthermore, GST can be a potential biomarker and an early-warning index for the pollutants in waters in that GST responded sensitively to 1 day and 10 days of exposure to diesel oil and 2,4-DCP and 10 days of combined exposure. Crossover comparisons showed an antagonistic action about the no-observed-effect concentration (NOEC) against Daphnia magna, which needs further studies.  相似文献

The development of the Daphnia longispina (O. F. Müller)population in a highly humic boreal lake was followed throughoutone growing season, and the amount of secondary production wasestimated in relation to primary production and available foodresources. The growth rate method was applied in the secondaryproduction measurements. Daphnia longispina did not appear inthe water column until 16 May, after which the animals werepresent throughout the growing season. The population showedthree density peaks; the first appeared in early June, and thesecond and third in mid-July and at the beginning of September,respectively. Somatic production followed a seasonal pattern,with highest production rates in midsummer. The maximum valueof 127 mg C m–2day–1 was recorded at the beginningof July. The total annual net production of D. longispina was7.9 g C m–2. During most of the growing season, the primaryproductivity in the lake was well below 100 mg C m–2 day–1and the total annual productivity of photosynthetic algae was5.0 g C m–2. We conclude that in this lake the zooplanktonpopulation did not rely on phytoplankton primary productionas a sole carbon source, but that most of the carbon must haveoriginated from bacterial production either directly or througha microbial loop.  相似文献

Primary production, and bacterial production as measured byincorporation of [3H-methyl]thymidineinto ice cold TCA insolublematerial were investigated during 1984 in Lake Kvernavatnet,west Norway. Primary production averaged 222 mg C m–2day–1 and bacterial production averaged 163 mg C m–2day–1. The bacterial production in the euphotic pelagiczonecontributed -60% of the total pelagic bacterial production.The zooplankton was dominated byDaphnia longispina. From growthexperiments with animals fed only natural food in coarse filteredlake water, the population daily growth increments were calculated.The average production of D.longispina was 151 mg C m–2day–1 during the period investigated. The estimated primaryproduction was too low to sustain both the bacterial productionand the zooplankton food requirements. These results imply thatthe carbon cycle of the lake is dependent on the supply of allochtonousmaterial, or that the current methods for measuring productionrates in aquatic environments are systematical erratic.  相似文献

This study examined the partitioning of organic matter intoparticulate organic carbon (POC) and dissolved organic carbon(DOC) pools in nutrient-enriched enclosures containing naturalplankton from the Gulf of Trieste (northern Adriatic), an areaaffected by mucilage. The strategy of nutrient additions wasto introduce a pulse of new nutrients in concentrations thatmimic natural inputs and to survey community structure and organicmatter fluxes long enough so that plankton became nutrient-limited.Maximal bacterial biomass attained roughly double the initialvalue, while autotrophic carbon increased by nearly an orderof magnitude. The microflagellate-dominated community releasedmore DOC per unit biomass (5.5 ± 7.2 to 50.6 ±28.0 µg C µg Chl a-1 day-1 versus 3.4 ± 3.4to 10.8 ± 4.6 µg C µg Chl a-1 day-1 for diatom-dominatedphytoplankton), POC increase was modest (~300 µg C l-1)and there was little change in DOC. Organic carbon partitioningduring two experiments in which diatoms prevailed was dominatedby POC (>800 µg C l-1) in the exponential growth phasewith an increasing contribution of particulate carbohydratesthat paralleled gradual nutrient depletion. Transition to thestationary phase and the decay of autotrophic communities wereaccompanied by the net accumulation of a carbohydrate-rich DOC.  相似文献

The tolerance to low ambient calcium (Ca) was assessed in twopopulations of Daphnia galeata. There was a threshold for survivalbetween 0 and 2 mg Ca l-1. However, the ability to cope withlow Ca concentrations clearly differed between the two populations,as the population from the low-Ca locality was less tolerantto low Ca. Additionally, neonate individuals had poorer survivalthan adults when reared at ~0 mg Ca l-1, supporting a suggestedjuvenile bottleneck regarding the tolerance to low Ca concentrations.The mean specific Ca content in adult D. galeata was not differentbetween the two populations, but individuals reared in mediumwith 1 mg Ca l-1 only had two-thirds of the Ca content of thosereared in medium with 10 mg Ca l-1. The significant differencesbetween the two populations investigated suggest that inter-populationvariation in tolerance to low ambient Ca concentrations mightbe important to explain the success of Ca-demanding crustaceansin softwater lakes.  相似文献

Neomysis mercedis, an important invertebrate predator in somelakes and estuaries in North America, occurred at high densities(>2 mysids m-3) in Lake Washington in the early 1960s, aperiod when Daphnia were scarce in the lake. Because Neomysisfeed selectively on Daphnia, it was hypothesized that mysidpredation contributed significantly to the scarcity of Daphnia.To evaluate this hypothesis, mysid abundance was monitored inthe lake (July 1989–February 1992), and whole lake predationimpacts on Daphnia were estimated. Mysid abundance varied from319 million, 0.1 mysids m-3 (February 1992) to 4276 million,1.7 mysids m-3 ( June 1991), and mysid biomass ranged from 1153(October 1991) to 4700 kg dry weight (November 1990). A peakin Daphnia consumption was noted during autumn, when total mysidbiomass was high. In late autumn/winter mysid consumption month-1accounted for 28–95% of Daphnia biomass and 13–38%of production. Mysid densities in this study were 18x and 5xlower than in 1962 and 1975, respectively. By extrapolation,at a density of 3.5 mysids m-3 observed in the early 1960s,total Neomysis consumption demand month-1 exceeded 100% of Daphniabiomass (late autumn–early spring), and 100% of Daphniaproduction (late autumn–winter). Estimates of the totalmysid consumption demand in this study are thus in accord withthe hypothesis that Neomysis could control Daphnia abundancein Lake Washington.  相似文献

Cell numbers and fluorescence of the green algae Selenastrum capricornutuum and survival of Daphnia magna exposed to simulated sun-light was assessed along a gradient of DOC (0, 1, 5 10 and 50 mg C l–1). When exposed to UV-doses and spectral distribution (295–750 nm) closely resembling surface solar radiation during mid summer, Selenastrum showed major losses of cell fluorescence. In the absence of DOC, fluorescence was severely depressed, with successively decreasing effects with increased DOC. Surviving cells also required an extensive recovery period (10–12 d) for regrowth after exposure, while an almost immediate recovery was observed at concentrations above 1 mg DOC l–1. For Daphnia, survival was reduced to less than 10% after 4 h exposure, and almost zero after 8 h exposure in the absence of humus DOC, while no effects were observed in treatments with 10 and 50 mg C l–1. Selenastrum and Daphnia that were not directly irradiated, but exposed to UV-irradiated water with the same concentrations of DOC did not reveal negative effects. This indicates negligible indirect effects mediated by long-lived free radicals or other toxic compounds. Irradiation of Daphnia under increased oxygen concentration (200% saturation) did not indicate acute effects, suggesting that effects of ambient radicals and oxidants would be of minor importance relative to intracellular photoproducts.  相似文献

Uptake rates of 14C (filtration and the acidification-bubblingmethod—ABM) were measured weekly in a shallow region ofthe Patos Lagoon estuary (3207'S, 5206'W) between March 1989and March 1990. Phytoplankton production varied seasonally,the lowest values occurring in the austral winter (June–August1989) and the highest rates during spring and summer (March1989; September 1989–March 1990). Rates of birth, growth and death are inferred for enclosed populationsof the marine copepods Pseudocalanus elongates, Acartia clausiand Temora longicomis. The rates are determined by fitting apopulation dynamics model to the time series of developmentalstage abundances observed over 87 days in four 310 m3 enclosures.Two enclosures were treated with ‘produced’ waterfrom North Sea oil production platforms and two were untreatedcontrols. The pollutant mainly affected recruitment to the populations.Development times varied considerably between enclosures, thoughnot in any apparent relation to the pollutant. Mortality wasmost strongly related to the population size of herring larvaeintroduced to all four enclosures. Levels of secondary productionderived from the model were compared with those calculated directlyfrom population densities.  相似文献

Phytoplankton composition, density, vertical distribution andprimary production were investigated in the Sea of Okhotsk andin the adjacent northern north Pacific in July–August1994, together with measurements of density and distributionof planktonic microheterotrophs: bacteria, nanoheterotrophsand ciliates. Different phases of phytoplankton seasonal successionwere encountered during the period of investigation in variousregions of this sea. Primary production measured at 144 stationswas found to be greatest (1.5–4 g C m-2day-1) in areasof spring-phase succession along the Sakhalin shelf and theKashevarov bank. Periodic relapses of the spring blooms of ‘heavy’diatoms during the whole growth season were recorded over thisbank. The summer phase of the phytoplankton minimum prevailedin the central and eastern parts of the sea, manifested by thedominance of nanoflagellates in terms of phytoplankton biomass.Primary production was 0.5–1 g C m-2 day-1. The earlyautumn phase of succession was typical of the Kurile straitarea and the adjacent north Pacific. Primary production therevaried from 0.7 to 2 g C m-2 day-1. The integrated phytoplanktonbiomass in the water column varied from 9–12 g m-2 inzones supporting the summer minimum assemblage to 15–20g m-2 in zones of early autumn recovery of phytoplankton growth,and up to 40–70 g m-2 in areas of remnant or relapseddiatom blooms. The numerical density of bacterioplankton wasbetween 1 x 106 and 3 x 106 cells ml-1 and its wet biomass wasbetween 100 and 370 mg m-3. In deep waters it was 8–15mg m-3. The integrated bacterioplankton biomass in the upperwater column varied from 6 to 29 g m-2. The numerical densityof zooflagellates varied in the upper layer between 0.8 x 106and 4 x 106 l-1 and their biomass was between 20 and 50 mg m-3.In deep waters they were still present at a density of 0.05x 106 to 0.2 x 106 cells l-1. The biomass of planktonic ciliatesvaried between stations from 20 to 100 mg m-3. The joint biomassof planktonic protozoa in the water column was between 3 and12 g m-3 at most of the stations.  相似文献

Cladocerans from three lakes in northwest Iowa were examinedfor attached diatoms over a period of 1 year to assess the seasonality,substrate preference and attachment site specificities of epizoicdiatoms. Over 90% of the cladocerans had attached diatoms immediatelyfollowing ice thaw, when the lakes were not yet thermally stratified,surface water temperatures were low(10–15°C)and watertransparency was at its annual high. Mean maximum density ofdiatoms on cladocerans averaged among lakes at this time was3000 cells animal–1. The dominant epizoic diatom was Synedracyclopum Brutschy, which was found most often on large, pelagicDaphnia pulicaria and Daphnia galeata mendotae. Heavily infestedanimals had diatoms concentrated on the second antennae andanterior ventral margin of the carapace. Animals with <5epizoic diatoms were more likely to have cells attached to thepostabdomen or tail spine. Bacterial growth and grazing mortality were estimated from Mayto October in two south Swedish oligotrophic lakes, one beinga clearwater lake (water colour 5–10 mg Pt l–1 DOC2.9–3.4 mg l–1, Secchi disk depth 5.0–9.4m) and the other a humic, brownwater lake (water colour 105–165mg Pt l–1, DOC 13.7–22.7mg l–1, Secchi diskdepth 1.3–2.1 m). Specific rates of growth and grazingmortality were generally similar for both lakes. However, theabundance of bacteria was consistently 2–3 times higherin the water of the humic lake, suggesting that the total productionand consumption of bacterial cells were also higher than inthe dearwater lake. The ratio of bacterial secondary productionto primary production was higher in the humic lake than in theclearwater lake, indicating that the bacterioplankton of thehumic lake utilize allochthonous substrates, in addition tosubstrates originating from autochthonous primary production.Most of the bacterial loss in both lakes could be attributedto small protozoan grazers. This implies that allochthonousand autochthonous organic carbon fixed by bacterioplankton isless important in terms of carbon flow to higher trophic levelsthan would be expected if macrozooplankton were the dominantbacterivores, providing a more direct and efficient transferof carbon to larger organisms.  相似文献

Biomass and productivity of microplankton were measured in theCraternaya Bay (Kurile Islands), which is influenced by hydrothermalactivity and volcanic heating. The hydrothermal fields are situatedaround its shores and underwater within the 0–20 m depth.A dense ‘bloom’ of photoautotrophic microplanktonwas observed there, dominated by diatoms, phytoflagellates andthe symbiont-containing ciliate Mesodinium rubrum. The biomassof these ciliates attained 3–11 g m-3 in the upper waterlayer. The total biomass of the phototrophic microplankton reached30–46 g m-3. The primary production in the water columnwas, correspondingly, enormously high: 6–10 g C m-2 day-1.The depth of the euphotic zone was 7 m. Pelagic photosynthesiswas inhibited in the upper 0–1 m by the spreading of alayer of low-salinity hydrothermal water. The numerical densityof bacterioplankton in the upper zone of the water column variedfrom 1 x 106 to 2.9 x 106 cells ml-1, and its wet biomass from250 to 750 mg m-3. Its production varied at stations from 70to 390 mg m-3 day-1. Chemosynthesis contributed up to 30% ofthis production in the sites neighbouring the hydrothermal vents.Outside their direct impact however, its share was negligible.The biomass of heterotrophic planktonic ciliates varied from30 to 270 mg m-3. The mechanisms of possible influence of shallowvolcanic activity on development and function of microplanktonin the Craternaya Bay is discussed.  相似文献

The seasonal dynamics of metazooplankton biomass was monitoredin an aerated waste stabilization pond during three consecutiveyears (1994–1996). The pond showed a low diversity ofplanktonic metazoans because of elevated pH, relatively highconcentration of free dissolved ammonia and low oxygen concentration.The planktonic community was composed of the anomopod branchiopodDaphnia magna, and the cyclopoid copepods Cyclops vicinus andCyclops strenuus. Both predation by cyclopoids and competitionwith D.magna excluded rotifers from the pond, except duringa short period in spring 1996. Daphnia magna was the dominantorganism from a biomass point of view. In parallel with biomass,demographic parameters, secondary production and the spatialdistribution of D.magna were studied. A significant seasonaland interannual variation in the density, biomass and productionof D.magna was observed. The maximum density of daphnids variedfrom 264 x 103 to 686 x 103 individuals m–2 and the maximumbiomass from 4 to 30 g dry weight (DW) m–2. The annualnet production was high compared with the production of Daphniain natural environments, ranging from 288 to 593 g DW m–2year–1. The annual net production of exuviae accountedfor ~25% of the total annual net production. Harvesting of daphnidsfor commercial applications that took place during the productiveperiod did not have any discernible effect on the populationdynamics of D.magna. Sexual reproduction was not observed duringthe three studied years. Negative mortality rates, occurringduring early spring, however, indicated that recruitment fromephippia was effective in the pond of Differdange and that sexualreproduction took place before 1994. We conducted a long term (4 week) continuous culture studyto measure the chronic effects of UV radiation on the alga,Cryptomonas erosa, using three different fluence rates of UVradiation. We measured carbon allocation into carbohydrate,protein and lipid pools, as well as chlorophyll a concentrationsand algal and bacterial density. After 21 days, algal densityin the control and lowest UV treatment (treatment 1 = 3.4 Wm–2 UVR unweighted) was significantly lower than in thetwo highest UV treatments (treatment 2 = 14.9 W m–2 andtreatment 3 = 16.2 W m–2 UVR unweighted), and did notrecover in the following week of no UV exposure. Chlorophylla and carbohydrate content (ng algal cell–1) for the controland treatment 1 were clearly lower than treatments 2 and 3 byday 15, and did not recover by day 28. Percentage total lipidfor the control and treatment 1 also decreased compared withtreatments 2 and 3 by the end of the exposure period. However,by day 21, protein content for the control and treatment 1 wassignificantly higher than treatments 2 and 3, and demonstrateda further increase by day 28. The results were largely attributedto competition effects between C.erosa and bacteria in thesenon-axenic cultures. Bacterial density was significantly (x4)higher in the control and lowest UV treatment compared withthe two highest UV treatments. Our findings suggest a competitiveadvantage of phytoplankton over bacteria under these conditions.If UV radiation, in general, affects bacteria to a greater extentthan algae, there are likely to be changes in (i) bacterialutilization of dissolved organic matter produced by phytoplankton,(ii) competition between phytoplankton and bacteria for nutrientminerals and (iii) predation rates on bacteria by micro-flagellates.  相似文献

1. Dissolved organic carbon (DOC) concentration was determined for a range of lakes of varying conductivity (30–4000 μS cm−1) in the low Arctic of SW Greenland. DOC concentration range from <1 to >100 mg C L−1, occasionally approaching 200 mg C L−1 in meromictic, oligosaline lakes. DOC concentration was strongly related to [log10] conductivity and total nitrogen. 2. Peak DOC concentrations (>80 mg L−1) occur in lakes located approximately 50 km from the present ice sheet margin, a zone of low effective precipitation; evaporative concentration is the first‐order control on DOC concentration. Lakes at the coast and closer to the ice margin had lower DOC concentrations (<20 mg C L−1). Local factors, notably the presence or absence of an outflow and catchment morphometry, resulted in considerable variability in concentration (20–100 mg C L−1) within the area of maximum concentration around 51°W. 3. Despite their high DOC concentration, these lakes are essentially colourless. Dissolved organic matter (DOM) absorption (a375) was low in most lakes (<10 m−1) with maximum values (approximately 20 m−1) occurring in one humic‐stained lake in the area. Absorption values corrected for DOC concentration () were very low (<0.6 m2 g−1 C) for all lakes apart from those at the coast, perhaps reflecting greater allochthonous inputs at these sites. 4. S, the spectral slope coefficient, ranged from 16 to 27 μm−1 and was weakly correlated with DOC concentration. Both a375 and S showed similar distribution patterns along the sampling gradient as did DOC, with maximum values at approximately 51°W. High and low S may indicate fresher, more rapidly flushed, systems with less degraded DOM or greater inputs from their catchments. 5. The lakes closer to the head of the fjord with higher conductivity, had low (<0.2 m2 g−1 C) and high S (>21 μm−1) and this may reflect increasingly longer lake water residence times, greater DOM age and photochemical degradation.  相似文献
