We used mitochondrial [cytochrome c oxidase subunit I (CO I ), cytochrome b , and 16S] and nuclear [internal transcribed spacer (ITS) phylogenies of Skistodiaptomus copepods to test hypotheses of Pleistocene divergence and speciation within the genus. Mitochondrial (mt)DNA sequence divergences do not support hypotheses for Pleistocene speciation and instead suggest much more ancient speciation events in the genus. Skistodiaptomus oregonensis and Skistodiaptomus pygmaeus (i.e. two morphologically similar and parapatric species) exhibited uncorrected mtDNA sequence divergences exceeding 20%. Similarly, we identified three divergent clades of Skistodiaptomus pallidus that exhibited mtDNA sequence divergences exceeding 15%, suggesting that even intraspecific divergence within this morphospecies predates the Pleistocene. We found clear evidence of CO I pseudogenes in S. pygmaeus , but their presence did not lead to significant overestimates of sequence divergences for this gene. Substitution saturation and strong purifying selection have most likely led to underestimates of sequence divergences and divergence times among Skistodiaptomus . The widespread phenomenon of morphological stasis among genetically divergent copepod groups indicates that speciation often occurs with little or no morphological change. Instead, morphological evolution may occur idiosyncratically after speciation and create discordant patterns of morphological similarity, shared ancestry and divergence time. The reef building vermetid gastropod Dendropoma petraeum inhabits the warmest waters of the Mediterranean Sea and is considered a threatened marine species. The aim of this study was to characterize its genetic structure throughout its whole distribution range using mitochondrial and nuclear sequence data. Because of its sessile adult lifestyle and lack of a pelagic larval stage, we expected a markedly subdivided population structure with limited levels of gene flow. Fragments of the mitochondrial genes cytochrome c oxidase subunit I (COI) and 16S rRNA (16S), were sequenced, along with the nuclear ribosomal cluster (internal transcribed spacer; ITS) in specimens from 18 localities. Our analyses identified four highly distinct phylogroups separated by a mean divergence of > 14% according to the COI sequence data or > 9% according to 16S, but differing only slightly in morphology. The nuclear data (ITS) indicated a lower substitution rate (divergence among groups of around 1%). These large genetic distances among the four lineages clearly point to the existence of a cryptic species complex within D. petraeum comprising at least four species. Differences in the characteristics of intracapsular larval development and protoconch were also detected among these lineages. The spider genus Weintrauboa new genus (Araneae, Pimoidae) is described to place two species of pimoids from Japan and adjacent islands that were formerly classified in the linyphiid genus Labulla . Weintrauboa contortipes (Karsch) new comb., the type species, and W. chikunii (Oi) new comb. are redescribed. Parsimony analysis of morphological characters provides robust support for the monophyly of the genus Weintrauboa and corroborates the monophyly of Pimoa , Pimoidae, and the clade Linyphiidae plus Pimoidae. Due to the pronounced morphological variation and geographical distribution of Galápagos' Opuntia cacti, numerous hypotheses have been advanced regarding their radiation, diversification, and classification. The currently accepted classification is based on morphology and recognizes six species and fourteen varieties, but the plasticity of many of the characteristics renders any morphological taxonomy problematic. Our analysis of previously published morphological data agrees only partially with the current classification. We present the first molecular phylogeny of these plants. Multiple DNA sequences indicate little genetic distinction among the currently identified species, despite restricted gene flow and limited long distance dispersal within the archipelago. No clear relationship exists between morphological and genetic differences. Systematics of the genera of Bodotriidae (Crustacea: Cumacea)   总被引:1,自引:1,他引:0  
The cumacean family Bodotriidae includes 382 species in 31 genera grouped in three subfamilies: Bodotriinae, Mancocumatinae and Vaunthompsoniinae. Generic diagnoses are based on few characters that often have overlapping states among genera, complicating the understanding of the relationships within the group. The goals of this study are to illuminate the phylogenetic relationships among the genera of the Bodotriidae using morphological characters and to review the systematics of the family. For this purpose, all species within each genus were studied from the literature to code all the variability of genera for 109 variable morphological characters. Phylogenetic analyses show that there is independent reduction of the pleopods in two clades from a plesiomorphic state of five pairs, while the number of exopods of peraeopods has been reduced gradually in more derived groups of bodotriids. The subfamily Bodotriinae is the most derived and the Vaunthompsoniinae the most basal, and is paraphyletic with the Mancocumatinae embedded within it. No discriminatory characters were found between the subfamilies Mancocumatinae and Vaunthompsoniinae and they are not clearly separated in the phylogeny. Mancocumatinae is synonymized with Vaunthompsoniinae and all the genera of the former Mancocumatinae should be included within Vaunthompsoniinae. Analyses of character evolution justify a few other taxonomic changes. All genera were redescribed based on all the coded characters and a complete list of all valid species for each genus is included. Specimens belonging to Tortula subulata complex ( T. inermis , T. mucronifolia and T. subulata ) were analysed using a combination of morphometric methods based on quantitative characters [principal component analysis (PCA), discriminant analysis (DA); 76 samples)] and molecular methods (ITS1 – 5.8S rRNA gene and ITS2; 47 samples) to assess patterns of morphological and molecular differentiation within this complex of taxa. The study shows that four species can be recognized: T. mucronifolia , T. subulata , T. inermis and T. subulata var. angustata with bistratose border, which is elevated to the species rank as T. schimperi nom. nov. The most valuable quantitative characters for identification of these species are the strata number of the marginal laminal cells, the ratio of middle marginal laminal cell width/middle marginal laminal cell length, basal membrane of peristome length, middle laminal cell width and papillae number on the middle laminal cells. The internal transcribed spacer (ITS) data suggest that this group of taxa is not monophyletic, T. mucronifolia being close to Protobryum bryoides (= Tortula protobryoides ). The remaining species seem to be a monophyletic group, T. schimperi being the sister group of the clade composed by T. inermis and T. subulata . A survey of morphological characters is carried out for Stapeliopsis . The information obtained from this is combined with molecular data from the plastid trn L-F DNA region and ITS1 of the nuclear encoded 18S−26S rRNA cistron, to obtain a hypothesis of the evolutionary relationships among the species. It is shown that Stapeliopsis is monophyletic in a combined molecular and morphological analysis. Stapeliopsis is sister to a clade containing Huernia , Orbea and Tromotriche . The species of Stapeliopsis group into two clades. One contains S. khamiesbergensis , S. neronis and S. urniflora , and this is highly supported. The remaining species fall into an unsupported clade in which S. exasperata is sister to the others. The genera Hermanschwartzia Plowes and Neopectinaria Plowes are rejected. It is shown that a synapomorphy for Stapeliopsis is the laterally flattened inner corona-lobes, which touch the anthers only at their bases. The systematic biology of the subgenus Daphnia s . s . remains confused. Prior attempts at resolution used chiefly postabdominal claw morphology, chromosome numbers and rRNA gene sequences as characters for higher-level relations. Still, several taxa, such as Daphnia curvirostris Eylmann, 1878, have unclear affiliations. We addressed the position of D. curvirostris in this genus by estimating phylogenetic trees from a rapidly evolving protein coding gene (ND2), conducting broad geographical comparisons and carrying out detailed morphological comparisons. The Japanese ' curvirostris' was found to be a new divergent lineage in the subgenus Daphnia , and to possess distinctive morphological characteristics from D. curvirostris . We described this new species as Daphnia tanakai sp. nov. , and redescribed D. curvirostris . The polymorphic postabdominal claw morphology and the distinctive chromosome number of D. tanakai sp. nov. provided evidence for rapid evolution of these traits. Our new morphological, chromosomal and genetic assessment of Daphnia weakened the argument for division of the subgenus Daphnia ( Daphnia ) O. F. Amplified fragment length polymorphism (AFLP) genetic fingerprinting of 14 accessions of Chara curta and Chara aspera Willd., sampled across a range of habitats and morphologies in Britain, suggests that these taxa are part of the variation within a single species complex. Two primer combinations generating 397 fragments (97% of which were polymorphic), analysed by Jaccard's similarity coefficient and principal co-ordinate analysis, did not recover groups which reflect the current taxonomy. By contrast with the genetic study, a Gower general similarity coefficient and principal co-ordinate analysis of 52 morphological characters recovered the currently recognized species groups. A Mantel test showed no significant correlation between the genetic data and the morphological data, supporting the hypothesis that phenotypic variability in Chara L. is either to some extent environmentally induced or represents developmental stages. A new species of Daptonema is described based upon morphological characters and 18S rRNA sequence. Daptonema matrona sp. nov. was collected in Pina Basin (north‐eastern Brazil). It differs from all other species of the genus by the presence of reduced cephalic setae and straight spicules. These features require an adaptation of the generic diagnosis. Moreover, the females are characterized by intra‐uterine development of the offspring, considered herein as their major autapomorphic feature. Molecular systematic analyses supported Daptonema matrona sp. nov. as a distinct genetic and evolutionary lineage. Calyptraeid gastropods are well know for the taxonomic difficulties caused by their simple, phenotypically variable shells. In this paper I demonstrate that what was previously considered to be a single species, Crepidula aculeata , is an ancient (3–15 Myr) cryptic species complex made up of at least eight species, and that this group should be placed in the genus Bostrycapulus . Despite the difficulty in finding diagnostic adult shell and anatomical features upon which species can be unambiguously identified, DNA sequences, protoconch morphology, embryonic morphology and developmental characters clearly differentiate these eight species. A single species with direct development and nurse eggs is present in the South Atlantic, and a species with planktotrophic development occurs in the equatorial Pacific. The species from Japan, Australia, Florida, the Pacific coasts of Mexico and Central America, and the Cape Verde Islands all have direct development. Most of these species are separated by > 15% divergence in COI sequence data. The fossil record of Bostrycapulus goes back to the Miocene, which agrees with genetic estimates of divergences within the genus ranging from 3 to 15 Mya. Surprisingly, these ancient species differ only slightly in morphology from each other and genetic differentiation does not correlate with geographical distance. I revise the genus Bostrycapulus on the basis of differences in adult morphology, embryonic morphology, mode of development, protoconch morphology, and DNA sequence data. The endemic South African velvet worm genus Peripatopsis currently contains eight recognized species described from variable morphological characters and the current taxonomy is unsatisfactory. In an attempt to investigate evolutionary relationships within Peripatopsis, we collected 137 individuals from 34 sample localities for six of the eight species. Sequence data derived from two partial mitochondrial (mt)DNA gene loci (COI and 12S rRNA), as well as partial sequence data from the ribosomal nuclear 18S rDNA locus in combination with gross morphological characters and scanning electron microscopy (SEM), was used to examine evolutionary relationships. Phylogenetic relationships were investigated using minimum evolution (ME) and Bayesian inferences (BI). Additionally, we also undertook a maximum likelihood (ML) analyses on the combined DNA sequence data set. The combined DNA evidence topologies derived from the ME, BI, and ML was highly congruent and was characterized by the presence of multiple lineages within recognized taxa. Peripatopsis clavigera, Peripatopsis moseleyi, and Peripatopsis sedgwicki each comprised two evolutionary lineages; Peripatopsis capensis comprised three; and Peripatopsis balfouri comprised six operational taxonomic units respectively. Genealogical exclusivity at both mtDNA and nuclear DNA among the geographically coherent groups coupled with pronounced sequence divergence suggested a two‐fold increase in the number of species within Peripatopsis. Previously used gross morphological characters (such as the number of leg pairs and colour) were either highly variable within operational taxonomic units, or were invariant, suggesting that alternative morphological characters are necessary for species discrimination. The genus Phora (Diptera, Phoridae) includes more than 50 species widely distributed in temperate regions of Eurasia and Africa. Taxonomic identification of most species is based upon the morphology of the male hypopygium: hence for many species, including the type species of the genus, females are unknown. We used mitochondrial 16S ribosomal RNA sequences to match males with previously unidentified females from Phora atra , P. stictica , and P. holosericea . We then identified morphological characters that allow identification of females of P. atra and P. stictica without recourse to DNA sequencing. Our results show that small scale sequencing can aid in the development of taxonomic characters for use in the field to identify previously cryptic females. This iterative method of identifying populations genetically followed by re-examination of morphology should allow development of better keys for rapid identification of heretofore cryptic populations of insects. We also found that sequences from individual of P. holosericea from Cambridge, England and Malakhovka, Russia, were more similar to each other than to sequences from a conspecific fly also collected in Cambridge. Recent studies have shown the value of complementing standard taxonomy with genetic analyses to reveal cryptic diversity and to aid in the understanding of patterns of evolution. We surveyed variation in the COI mitochondrial gene in members of the three genera of centropagid copepods from the inland waters in Argentina. In general, we found a close association between molecular and morphological systematics in this group. Similar to findings for marine calanoids, genetic distances within Boeckella species were modest (< 4%), while distances among morphospecies were much larger (> 11%). Parabroteas is currently monotypic, although we detected cryptic genetic diversity, with two lineages showing 5.5% divergence. In contrast, Karukinka was not a valid genus, apparently representing an interesting and atavistic offshoot of B. poppei , a result reinforcing the value of considering both morphological and molecular evidence. Moreover, we used combined genetic and morphological information, analysed with maximum likelihood methods, to evaluate the common assumption that evolution tends to proceed via the loss of structures in crustaceans.
