The fate of contaminant carbon was monitored during aerobic biodegradation in the presence of a mixed indigenous microbial consortium in order to calibrate a microbial-growth-based biokinetic model. The methodology simultaneously monitored mineralization, substrate depletion and microbial population evolution in biomass extract spiked with14C-labeled hexadecane. Hexadecane depletion and hexadecane-degrader population were monitored using sacrificed microcosms by centrifuging the extract so that the supernatant and the residue contained residual hexadecane and microbial population, respectively. This methodology allowed verification of the carbon mass balance (average14C-carbon recovery of 90.33 ± 1.62% for biotic microcosms) and calibration of a biokinetic model. Four biokinetic parameters and three yield coefficients were identified (Haldane kinetic parameters:μS = 1.3639 d-1, Ks = 0.4295 mg-C, KI = 6.6457 mg-C; decay kinetic parameter:μd = 1.3.102 d-1; substrate/biomass, carbon dioxide/ biomass during growth and carbon dioxide/biomass during decay yield coefficients: Ys = 1.5948 mg-C/mg-C, YP g = 0.4554 mg-C/mg-C, YP d = 1.3263 mg-C/mg-C) and compared with the literature data. The methodology can facilitate the identification of biodegradation models by decoupling the intrinsic ability of microorganisms to degrade contaminant from restrictions imposed by limiting conditions. 相似文献

通过营养液栽培,研究不同矿质营养的缺乏对黄瓜叶片组织生理生化特性的影响。结果表明,缺铜对植株生理影响最大,在各处理中,叶片组织的可溶性糖含量最低,木质素含量也处于较低水平,而相对电导度则最高,所测定的4种保护酶(POD、PPO、PAL、SOD)活性也最低。其次是锌,缺锌造成了相对电导度的增加,可溶性糖含量和木质素含量的下降,除PAL活性外,其余3种酶活性也处于次低的位置。缺锰主要导致了叶片组织可溶性糖含量、木质素含量以及PAL活性的较大幅度的下降。缺钙主要降低了叶片组织中可溶性糖含量和SOD活性。比较而言,硼和铁的缺乏影响较小。 相似文献

为探究谷胱甘肽和没食子酸对紫淮山花色苷的辅色作用,本文研究了谷胱甘肽和没食子酸对紫淮山花色苷降解率、热稳定性及色差的影响。试验表明,谷胱甘肽和没食子酸能有效抑制花色苷的降解,且最佳添加量分别为0.03%和0.2%。在此添加量条件下,紫淮山花色苷在50、70、90℃水浴中的热降解均符合一级降解反应动力学规律。添加谷胱甘肽和没食子酸的紫淮山花色苷降解速率常数(k)小于对照组,半衰期(t 1/2)和活化能(Ea)高于对照组,说明谷胱甘肽和没食子酸能够增强花色苷的热稳定性。色差测定结果表明,经谷胱甘肽和没食子酸辅色后的紫淮山花色苷,其明度指数(L*)和色品指数(a*、b*)较对照组变化缓慢,颜色的稳定性增强。 相似文献

下载免费PDF全文 The kinetics of alternative oxidase (AOX) of Arum italicum spadices and soybean (Glycine max L.) cotyledons were studied both with intact mitochondria and with a solubilized, partially purified enzyme. Ubiquinone analogs were screened for their suitability as substrates and ubiquinol-1 was found to be most suitable. The kinetics of ubiquinol-1 oxidation via AOX in both systems followed Michaelis-Menten kinetics, suggesting that the reaction is limited by a single-step substrate reaction. The kinetics are quite different from those previously described, in which the redox state of ubiquinone-10 was monitored and an increase in substrate was accompanied by a decrease in product. The difference between the systems is discussed. Pyruvate is a potent activator of the enzyme and its presence is essential for maximum activity. The addition of pyruvate to the solubilized enzyme increased the maximum initial velocity from 6.2 [plus or minus] 1.3 to 16.9 [plus or minus] 2.8 [mu]mol O2 mg-1 protein min-1 but had little effect on the Michaelis constant for ubiquinol-1, an analog of ubiquinol, which changed from 116 [plus or minus] 73 to 157 [plus or minus] 68 [mu]M. It is concluded that pyruvate (and presumably other keto acids) increases the activity of AOX but does not increase its affinity for its substrate. In agreement with this is the finding that removal of pyruvate (using lactate dehydrogenase and NADH) leads to an 80 to 90% decrease in the reaction rate, suggesting that pyruvate is important in the mechanism of reaction of AOX. The removal of pyruvate from the enzyme required turnover, suggesting that pyruvate is bound to the enzyme and is released during turnover. 相似文献

Indazoles are valuable because of their biological activities. A series of 3-aryl-1H-indazoles have been synthesized by condensing bisulfite adduct of aromatic aldehydes with phenyl hydrazine in good yield. Different concentrations of indazoles were employed to check their effects on seed germination and early growth. The arylindazoles were proved to be growth inhibitors of root and shoot lengths of wheat and sorghum, especially at a high concentration (100 ppm). At lower concentrations growth inhibition was found to be less prominent. Seed germination and early growth of plantlets also depended on the nature of substitution on the aryl group. 相似文献
