The development of new approaches to study the affinity between ligands and G‐protein‐coupled receptors proves to be of growing interest for pharmacologists, chemists, and biologists. The aim of this work was to determine the binding of seven drugs to β2‐adrenoceptors by frontal analysis using immobilized receptor stationary phase. The dissociation constants (Kd) were determined to be (3.16 ± 0.09) × 10?4 M for salbutamol, (4.29 ± 0.12) × 10?4 M for terbutaline, (6.19 ± 0.16) × 10?4 M for methoxyphenamine, (2.11 ± 0.07) × 10?4 M for tulobuterol, (1.82 ± 0.11) × 10?4 M for fenoterol, (9.75 ± 0.24) × 10?6 M formoterol, and (9.84 ± 0.26) × 10?5 M for clenbuterol. These results showed a good correlation with the data determined by radioligand binding assay. Further investigations revealed that the dissociation constant mainly attributed to the number of hydrogen bonds in the structures of ligands. This study indicates that affinity chromatography using immobilized receptor stationary phase can be used for the direct determination of drug‐receptor binding interactions and has the potential to become a reliable alternative for quantitative studies of ligand–receptor interactions. Intact human platelets bind cytochalasin B (CB) with a capacity of 100– 120 p mols CB/mg protein or approximately 7 × 104 molecules/cell and dissociation constants (KD) ranging from 2 × 10?8 to 10?6 M. Up to 85% of this saturable binding is displaced by 10?5 M cytochalasin E (CE). This CE-sensitive binding also appears heterogeneous with KD similar to those of the overall binding. The CE-insensitive binding, however, appears as a single component with KD ≌ 4 × 10?7 M. The sedimentable constituents from frozen, thawed, and washed cells also bind CB with KD ranging from 2.4 × 10?8 to 1.5 × 10?6 M and a total capacity of approximately 39 p mols/mg protein which accounts for only 4% of the ligand binding to the intact cell. The major portion (60–80%) of this CB binding is displaceable by 500 mM D-glucose and has a KD of 1.5 × 10?6M, while only 10–15% is CE-sensitive with a KD of 2.4 ± 10?8 M. It is concluded that 95% of the saturable CB binding in platelets is associated with the cytosol of which 80–85% is sensitive to CE and that only 3% of the cellular binding is glucose sensitive, membrane-associated binding. If the CE-sensitive binding associated with the cytosol is entirely to actin, the stoichiometry of this binding is approximately one CB to 30 actin monomers, which is greater by an order of magnitude than that for CB binding to muscle actin.  相似文献

Abstract: We identified and characterized 125I-endothelin-1 (125I-ET-1) binding sites in tumor capillaries isolated from human glioblastomas, using the quantitative receptor autoradiographic technique with pellet sections. Quantification was done using the computerized radioluminographic imaging plate system. High-affinity ET receptors were localized in capillaries from glioblastomas and the surrounding brain tissues (KD = 4.7 ± 1.0 × 10?10 and 1.6 ± 0.3 × 10?10M, respectively; Bmax = 161 ± 38 and 140 ± 37 fmol/mg, respectively; mean ± SEM, n = 5). BQ-123, a selective antagonist for the ETA receptor, potently competed for 125I-ET-1 binding to sections of the microvessels with IC50 values of 5.1 ± 0.3 and 5.1 ± 1.5 nM, and 10?6M BQ-123 displaced 84 and 58% of ET binding to capillaries from tumors and brains, respectively. In addition, competition curves obtained in the presence of increasing concentrations of ET-3 showed two components (IC50 = 5.7 ± 2.5 × 10?10 and 1.4 ± 0.2 × 10?6M for tumor microvessels, 1.8 ± 0.6 × 10?10 and 1.1 ± 0.3 × 10?6M for brain microvessels, respectively). Our results indicate that (a) the method we used is simple and highly sensitive for detecting and characterizing various receptors in tumor capillaries, especially in the case of a sparse specimen, and (b) capillaries in glioblastomas express specific high-affinity ET binding sites, candidates for biologically active ET receptors, which predominantly belong to the ETA subtype.  相似文献

The interactions of chymotrypsin, subtilisin and trypsin with a low MW proteinase inhibitor from potatoes were investigated. The Ki value calculated for the binding of inhibitor to chymotrypsin was 1.6 ± 0.9 × 10?10M, while the second-order rate constant for association was 6 × 105 M?1/sec. Although binding was not observed to chymotrypsin which had been treated with diisopropyl fluorophosphate or with l-tosylamide-2-phenylethyl chloromethyl ketone, the 3-methylhistidine-57 derivative bound inhibitor with a Ki value of 9.6 × 10?9 M. The inhibitor also exhibited a tight association with subtilisin (Ki < 4 × 10?9 M). In contrast, little inhibition of trypsin was observed, and this was believed to be due to low levels of a contaminant in our preparations. No evidence for reactive site cleavage was observed after incubation of the inhibitor with catalytic amounts of chymotrypsin or subtilisin at acid pH.  相似文献

ABSTRACT

Chondroitin sulfate (CS) used for treatment of osteoarthritis exerts distinct effects on human articular chondrocytes in vitro. We performed a binding analysis with 99mTc-labeled CS (Condrosulf, a commercial CS preparation containing calcium stearate) and cultured human chondrocytes in order to evaluate the presence of specific receptors. Saturation binding at 37°C for 2?h revealed the presence of high-affinity binding sites for CS with a Kd of 2.3 × 10?9?mol/L and a Bmax of 5.0 × 108. Extensive dialysis of Chondrosulf led to a decrease of the binding affinity by 52.5 ± 19.5% and of the number of CS binding sites/cell by 62.0 ± 14.0%, demonstrating that the additive present in the Condrosulf preparation enhances CS binding. The nature of the binding site is not yet known but evidence exists in the literature that the scavenger receptor CD36, thoroughly investigated on macrophages, is also found on chondrocytes and might be involved in CS binding. Therefore, we undertook a comparative binding study with human monocytes and labelled LDL and oxidized LDL, the latter being a postulated atherogenic agent in atherosclerosis. For [125I]-LDL binding we found a Kd of 0.45 × 10?8?mol/L and a Bmax of 0.14 × 106 on quiescent monocytes and for [125I]-(ox)LDL binding a Kd of 1.8 × 10?8?mol/L and a Bmax of 1.3 × 106 using LPS-activated monocytes. These data are comparable to the binding affinity found for lipoprotein–proteoglycan-complexes and hence are an indication but not a proof that CD36 is involved in CS binding to human chondrocytes.  相似文献

Green tea is rich in several polyphenols, such as (?)-epicatechin-3-gallate (ECG), (?)-epigallocatechin (EGC), and (?)-epigallocatechin-3-gallate (EGCG). The biological importance of these polyphenols led us to study the major polyphenol EGCG with human serum albumin (HSA) in an earlier study. In this report, we have compared the binding of ECG, EGC, and EGCG and the Cu(II) complexes of EGCG and ECG with HSA. We observe that the gallate moiety of the polyphenols plays a crucial role in determining the mode of interaction with HSA. The binding constants obtained for the different systems are 5.86?±?0.72?×?104 M?1 (K ECG-HSA), 4.22?±?0.15?×?104 M?1 (K ECG-Cu(II)-HSA), and 9.51?±?0.31?×?104 M?1 (K EGCG-Cu(II)-HSA) at 293?K. Thermodynamic parameters thus obtained suggest that apart from an initial hydrophobic association, van der Waals interactions and hydrogen bonding are the major interactions which held together the polyphenols and HSA. However, thermodynamic parameters obtained from the interactions of the copper complexes with HSA are indicative of the involvement of the hydrophobic forces. Circular dichroism and the Fourier transform infrared spectroscopic measurements reveal changes in α-helical content of HSA after binding with the ligands. Data obtained by fluorescence spectroscopy, displacement experiments along with the docking studies suggested that the ligands bind to the residues located in site 1 (subdomains IIA), whereas EGC, that lacks the gallate moiety, binds to the other hydrophobic site 2 (subdomain IIIA) of the protein.  相似文献

Inhibition of bovine erythrocyte acetylcholinesterase (free and immobilized on controlled pore glass) by separate and simultaneous exposure to malathion and malathion transformation products which are generally formed during storage or through natural or photochemical degradation was investigated. Increasing concentrations of malathion, its oxidation product malaoxon, and its isomerisation product isomalathion inhibited free and immobilized AChE in a concentration-dependent manner. KI, the dissociation constant for the initial reversible enzyme inhibitor-complex, and k3, the first order rate constant for the conversion of the reversible complex into the irreversibly inhibited enzyme, were determined from the progressive development of inhibition produced by reaction of native AChE with malathion, malaoxon and isomalathion. KI values of 1.3 × 10? 4 M? 1, 5.6 × 10? 6 M? 1 and 7.2 × 10? 6 M? 1 were obtained for malathion, malaoxon and isomalathion, respectively. The IC50 values for free/immobilized AChE, (3.7 ± 0.2) × 10? 4 M/(1.6 ± 0.1) × 10? 4, (2.4 ± 0.3) × 10? 6/(3.4 ± 0.1) × 10? 6 M and (3.2 ± 0.3) × 10? 6 M/(2.7 ± 0.2) × 10? 6 M, were obtained from the inhibition curves induced by malathion, malaoxon and isomalathion, respectively. However, the products formed due to photoinduced degradation, phosphorodithioic O,O,S-trimethyl ester and O,O-dimethyl thiophosphate, did not noticeably affect enzymatic activity, while diethyl maleate inhibited AChE activity at concentrations > 10 mM. Inhibition of acetylcholinesterase increased with the time of exposure to malathion and its inhibiting by-products within the interval from 0 to 5 minutes. Through simultaneous exposure of the enzyme to malaoxon and isomalathion, an additive effect was achieved for lower concentrations of the inhibitors (in the presence of malaoxon/isomalathion at concentrations 2 × 10? 7 M/2 × 10? 7 M, 2 × 10? 7 M/3 × 10? 7 M and 2 × 10? 7 M/4.5 × 10? 7 M), while an antagonistic effect was obtained for all higher concentrations of inhibitors. The presence of a non-inhibitory degradation product (phosphorodithioic O,O,S-trimethyl ester) did not affect the inhibition efficiencies of the malathion by-products, malaoxon and isomalathion.  相似文献

A method has been developed for the study of somatostatin binding to anterior pituitary plasma membranes. When 5×10?9M [125I]Tyr1-somatostatin (SA 18 Ci/mmol) was incubated with isolated pituitary plasma membranes (protein = 100 μg), 13.6% of total radioactivity was bound excluding nonspecific binding. The Scatchard plot could be resolved into two distinct components and analyzed to yield: K1diss = 3.3×10?8M and K2diss = 7.7×10?6M. This binding was shown to be specific for somatostatin.  相似文献

Abstract: Pridefine (AHR-1118) is a pyrrolidine derivative with clinically established antidepressant efficacy. Previous work from this laboratory indicates that pridefine is a reuptake blocker of catecholamines and serotonin with weak releasing activity. This study characterized the mode of amine uptake inhibition by pridefine as noncompetitive. The uptake experiments were performed utilizing ouabain instead of zero-degree controls to differentiate between the passive and active components of uptake. Furthermore, the passive component was resolved into diffusion and binding of substrate. Correction was made for the effects of ouabain on binding. Kinetic constants determined from Lineweaver-Burk plots were: Km= 3 × 10?7 M for NE, Km= 9 × 10?8 M for DA, and Km= 3 × 10?8 M for 5-HT. Dixon analyses of uptake at various pridefine concentrations indicated noncompetitive inhibition with Ki= 2.5 × 10?6 M for NE uptake, Ki= 2.0 × 10?6 M for DA uptake, and Ki= 1 × 10?5 M for 5-HT uptake. These constants compare well with IC50 values for the same transmitters: NE, IC50= 2.4 × 10?6 M; DA, IC50= 2.8 × 10?6 M; 5-HT, IC50= 1.0 × 10?5 M. The in vitro results indicate that pridefine is relatively specific as a catecholamine uptake blocker. It differs from tricyclic antidepressants which are reportedly competitive inhibitors of monoamine uptake. The possible mechanisms by which pridefine acts as a noncompetitive inhibitor are discussed.  相似文献

Specific and saturable binding of 125I-bovine albumin to rat adipocytes in suspension was observed (apparent Kd 2.09 ± 0.52 × 10?6 M; 8.58 ± 2.49 × 106 sites per cell; mean ± SEM). The binding was rapid and reversible for at least 10 min, suggesting that endocytosis of albumin was minor under assay conditions. Pre-incubation of cells with epinephrine bitartrate caused an apparent increase in number and decrease in affinity of the adipocyte binding sites for albumin. These findings suggest that a specific and saturable interaction of albumin with the adipocyte surface may play a role in the cellular uptake and release of free fatty acids.  相似文献

Alcohol dehydrogenase was prepared from germinating soybean seeds. Specific activity was increased from 511 to 31316 units. The coenzyme is NAD with a Km of 10?4M. Allyl alcohol is oxidized faster than ethanol; with the latter substrate, the Km is 1.3 × 10?2M, and the pH optimum 8.7. L-T3 transport has been investigated in human red cell ghosts. Determination of initial T3 uptake revealed two separate saturable uptake systems, one with a Km of 1.6 × 10?8M, the other with a Km of 3.3 × 10?6M. Binding experiments resulted in two dissociation constants, 1.4 × 10?7M.and 2.6 × 10?6M. Uptake was dependent on the ghost volume, indicating an intravesicular location of T3. The T3 was concentrated 6 times by the ghosts. Ouabain reduced the uptake by the low Km system, but was without effect on the high Km system. Thus evidence is provided both of binding of T3 to the ghost membrane and of its uphill transport across the membrane.  相似文献

Intact rat blood platelets are shown to possess benzodiazepine binding sites of the peripheral type, binding of [3H]diazepam being strongly inhibited by Ro5-4864 (Ki = 3.6 ± 0.5 nM) but only weakly inhibited by clonazepam (Ki = 35.1 ± 18.2 μM). Binding of [3H]diazepam is specific and saturable. Scatchard analysis reveals a single class of binding sites with KD = 14.7 ± 1.0 nM and Bmax = 564 ± 75 fmoles/108 platelets. The Hill coefficient is 0.94, indicating a lack of binding site heterogeneity or negative cooperativity. Binding reaches equiliibrium at 6 min, with k+1 = 2.9 × 107 M?1 min?1, and is rapidly reversible (t12 = 2.2 min with K?1 = 0.315 min?1. KD derived from the rate constants agrees with that estimated by Scatchard analysis. KD of the crude membrane fraction of platelets is also close to that of intact platelets. Binding of [3H]diazepam is linear with platelet number (between 0.25–2 × 108 platelets), is temperature sensitive with maximum binding at 0°C, and has a broad optimal pH range between pH 5–9.  相似文献

3H-nicotine binding was performed on intact and solubilized rat brain membranes as well as membranes from the electric organ of the Torpedo fish. The Kd for binding to intact and solubilized rat brain membranes was 5.6 × 10?9 M and 1.1 × 10?8M respectively, and the binding capacity 2.0 × 10?14 and 3.0 × 10?13 moles /mg protein respectively. The Kd for Torpedo membranes was 3.1 × 10?7M and the binding capacity 6.8 × 10?13 moles/mg protein. The binding was stereospecific with the affinity of the (?)-nicotine being about 8 times greater than the (+)-nicotine with all three preparations. The relative affinity for the nicotine binding site of nicotinic cholinergic drugs was considerably less in rat brain than in Torpedo membranes, where the sites are mainly cholinergic. A comparison was made of the ability of a variety of cholinergic drugs and nicotine derivatives to compete with 3H-nicotine binding and their relative pharmacologic potency to produce or inhibit a characteristic prostration syndrome caused by (?)-nicotine administered intraventricularly to rats. From such studies it was concluded that nicotine, in part, may be interacting at noncholinergic sites in rat brain.  相似文献

Agmatine iminohydrolase was purified ca 375-fold from groundnut cotyledons. The enzyme exhibited an optimum pH between 5.5 and 8.5 and the energy of activation was 22 kcal/mol. The Km for agmatine was (7.57 ± 0.77) × 10?4 M. The enzyme was inhibited by tryptamine, putrescine, cadaverine, spermidine and spermine. Inhibition by cadaverine and spermidine was competitive. The Ki values for cadaverine and spermidine were 4.1 × 10?3 and 7.5 × 10?4 M, respectively.  相似文献

4-oxo-N,N,N-trimethylpentanaminium chloride is a competitive inhibitor of eel acetylcholinesterase with KI = 8 × 10?6 M at 25°, 0.1 M NaCl, 0.04 M MgCl2, pH 7.5. Its binding decreases at low pH with pKa = 6.0. N,N,N-trimethylpentanaminium bromide has KI = 4 × 10?4 M under the same conditions. Its binding also decreases with pH with pKa = 5.35. Comparison with literature data indicates that the ketone binds much more strongly than substrates and that its binding shows the pH dependence expected for a transition state analog.  相似文献

The interaction of alkylguanidines and decahydrohistrionicotoxin with the membrane-bound and solubilized muscarinic acetylcholine receptor (mAcChR) from porcine atria was described. Alkylguanidines with alkyl chain lengths from one to ten carbons displaced l-[3H]quinuclidinyl benzilate (l-[3H]QNB) competitively from a single class of sites for the membrane-bound mAcChR. From a plot of ?ln Ki versus alkyl carbon chain number, a value of ?(473 ± 30) cal/mol was estimated as the energetic contribution per methylene group to the total binding energy. The binding of alkylguanidines to the digitonin/cholate solubilized mAcChR was complex in nature resulting in titration curves that did not obey the law of mass action for simple competitive inhibition at higher alkyl carbon numbers and a sigmoidal plot of ?ln Ki versus carbon number. Decahydrohistrionicotoxin bound in a competitive manner versus l-[3H]QNB to both the membrane-bound (Ki = (6.9 ± 1.4) × 10?6 M) and the solubilized (Ki = (1.5 ± 0.3) × 10?5 M) preparations.  相似文献

The kinetics of the binding of cyanide to ferric chloroperoxidase have been studied at 25°C and ionic strength 0.11 M using a stopped-flow apparatus. The dissociation constant (KCN) of the peroxidase-cyanide complex and both forward (k+) and reverse (k?) rate constants are independent of the H+ concentration over the pH range 2.7 to 7.1. The values obtained are kcn = (9.5 ± 1.0) × 10-5 M, k+. = (5.2 ± 0.5) × 104 M?1 sec?1 and k- = (5.0± 1.4) sec-1. In the presence of 0 06 M potassium nitrate the affinity of cyanide for chloroperoxidase decreases due to the inhibition of the forward reaction. The dissociation rate is not affected. The nitrate anion exerts its influence by binding to a protonated form of the enzyme, whereas the cyanide binds to the unprotonated form. Binding of nitrate results in an apparent shift towards higher pKa values of the ionization of a crucial heme-linked acid group. Hence the influence of this group can be detected in the accessible pH range. Extrapolation to zero nitrate concentration yields a value of 3.1±0.3 for the pKa of the heme-linked acid group.  相似文献

Streptomyces lividans 1326 carries inducible mercury resistance genes on the chromosome, which are arranged in two divergently transcribed operons. Expression of the genes is negatively regulated by the repressor MerR, which binds in the intercistronic region between the two operons. The merR gene was expressed in E. coli using a T7 RNA polymerase/promoter expression system, and MerR was purified to around 95% homogeneity by ammonium sulfate precipitation, gel filtration and affinity chromatography. Gel filtration showed that the native MerR is a dimer with a molecular mass of 31?kDa. Two DNA binding sites were identified in the intercistronic mer promoter region by footprinting experiments. No evidence for cooperativity in the binding of MerR to the adjacent operator sequences was observed in gel mobility shift assays. The dissociation constants (KD) for binding of MerR were: binding site I, 8.5?×?10?9?M; binding site II, 1.2?×?10?8?M; and for the complete promoter/operator region 1?×?10?8?M. The half-life of the MerR-DNA complex was 19.4?min and 18.8?min for binding site I and binding site II, respectively. The KD value for binding of mercury(II)chloride to MerR, again determined by mobility shift assay, was 1.1?×?10?7?M.  相似文献

Both myoblasts and myotubes in cultures of clonal rat muscle cells have action potential Na+ ionophore activity. The ionophore is activated by batrachotoxin (K0.5 = 3 to 5 × 10?7 M) and veratridine (K0.5 = 4 to 6 × 10?5 M) which compete for the same activation site. As in denervated rat muscle, the ionophore of cultured muscle is 100 fold more resistant to inhibition by tetrodotoxin (K0.5 = 1.5 to 3 × 10?6 M) and 20 fold more resistant to inhibition by saxitoxin (K0.5 = 1.5 to 3 × 10?7 M) than in nerve, innervated muscle, or cultured neuroblastoma cells.  相似文献
