长角血蜱雌蜱感染嗜菌异小杆线虫后血淋巴的变化   总被引:1,自引:0,他引:1  
用嗜菌异小杆线虫Heterorhabditis bacteriophora E-6-7(Hb E-6-7)感染长角血蜱Haemaphysalis longicornis雌蜱,测定感染后雌蜱血淋巴总蛋白含量、酯酶活性及酯酶同工酶的变化,以探讨昆虫病原线虫对蜱的致病机理。结果显示,雌蜱血淋巴总蛋白含量在Hb E-6-7感染后12 h显著增加,达到最大值92.21 μg/μL;在感染后24 h明显下降(49.06 μg/μL);至感染后48 h降至34.25 μg/μL。雌蜱血淋巴酯酶活性在线虫感染后0~12 h内无显著变化;在感染24 h后迅速增加(OD24h=0.1840,OD36h=0.1940,OD48h=0.2165),与对照组差异显著。PAGE结果表明,线虫感染后导致雌蜱血淋巴酯酶同工酶电泳图谱发生变化,主要为电泳图谱两端a带和b带的消失以及一条c带的增加。上述结果表明,长角血蜱雌蜱被Hb E-6-7感染后其血淋巴总蛋白含量和酯酶发生变化,这种变化可能与蜱的防御和对昆虫病原线虫的适应有关。  相似文献

用高效液相色谱法和放射免疫测定法测定了银盾革蜱滞育雌虫的合神经节、血淋巴、卵巢和整体中蜕皮激素的含量,并与正常发育的雌虫做比较。结果表明,滞育雌虫血淋巴和卵巢中蜕皮激素的含量在饱血后前10天与正常发育雌虫基本上相同。饱血10天以后,滞育雌虫蜕皮激素的含量下降,较正常发育雌虫少得多。蜕皮激素的缺乏影响了卵母细胞的发育。为阐明蜕皮激素对滞育雌虫的影响,在不同时间向银盾革蜱滞育雌虫体内注入不同剂量的β-蜕皮素。刚饱血者注入大剂量(1.375和10000ng/蜱)的β-蜕皮素引起雌虫死亡。饱血后20—30天,注入一定剂量(50,70,100ng/蜱)的β-蜕皮素可以促进卵黄形成,并解除了雌虫的生殖滞育,但其所产的卵量明显少于正常发育的雌虫。应用外源蜕皮激素解除硬蜱雌虫的生殖滞育现象尚属首次报道。  相似文献

【目的】通过分析宿主皮下注射伊维菌素对长角血蜱生长发育及生殖的影响,进而评价伊维菌素对长角血蜱的防治效果。【方法】以新西兰白兔为宿主,皮下注射不同剂量伊维菌素,观察幼蜱、若蜱及雌蜱的叮咬率、死亡率、吸血期、饱血体重、产卵量和孵化率等生物学参数,分析伊维菌素对长角血蜱生殖和生长发育的影响。【结果】伊维菌素对长角血蜱各期蜱虫的叮咬率均无显著影响。与对照组相比,伊维菌素可显著增加幼蜱和若蜱死亡率,延长若蜱和雌蜱的吸血期并降低其饱血体重,降低雌蜱产卵量及卵孵化率,抑制卵巢及卵的发育。【结论】宿主皮下注射伊维菌素能够显著抑制长角血蜱的发育及生殖,可作为防治长角血蜱的候选杀虫剂。  相似文献

用昆虫病原线虫小卷蛾斯氏线虫(Sc BJ)、夜蛾斯氏线虫(Sf Otio)、拟双角斯氏线虫(Sc D43)、格氏斯氏线虫(Sg NC32)和嗜菌异小杆线虫(Hb E-6-7)对长角血蜱雌蜱进行感染试验,所用线虫剂量为4 000 Ijs/dish。结果表明,5种线虫均对长角血蜱雌蜱有致死效应。Hb E-6-7和Sc BJ两种线虫对雌蜱各发育期致病力最强,导致雌蜱的累积死亡率和半致死时间分别为饥饿雌蜱82.5%,9.0天和75.0%,8.8天;吸血雌蜱90.0%,8.0天和82.5%,8.0天;饱血雌蜱93.3%,7.3天和86.7%,7.3天。Sc D43对饱血雌蜱有较高的致死效应,为80.0%,但半致死时间较长,为11.7天。Sf Otio和Sg NC32对长角血蜱雌蜱的致死效应较低。饱血雌蜱较饥饿雌蜱和吸血雌蜱更易被线虫感染。  相似文献

<正> 边缘革蜱Dermacentor marginatus Sulzer和刻点血蜱Haemaphysalis punctata Canestrini etFanzago是人类某些疾病的重要传播媒介。在我国北方和西北地区有广泛分布。为观察这两种蜱的产卵过程,我们于1974年从新疆新源那拉堤地区的牛和羊体外采集了一批成蜱,从其中称取不同程度饱血的雌蜱,边缘革蜱29只,重量146.1—748.5毫克;刻点血蜱17只,重量111.7—483.3毫克。按每管1只,分别装入玻璃管,置于温箱内。现将观察结果简报如下。  相似文献

亚东璃眼蜱用家兔喂血后放在30℃、各种湿度和无光条件下观察产卵情况。从宿主体自然掉下的饱血雌蜱,体重平均为1,321.3±462.6毫克;总产卵数平均为13,392±5,041.4粒;总产卵数/饱血体重平均为10.08±1.33:总产卵重/饱血体重平均为 0.599±0.06;产卵前期平均为 4.9±0.52天;产卵期平均为18.1±2.93天;一天内产卵最多平均为2,173.1±745.8粒;产卵高峰产卵开始后的第3天。饱血体重与总产卵数和产卵天数间以及总产卵数与一天内产卵最多数目间都有非常显著的正相关。饱血体重和总产卵数与雌蜱吸血前的体重有关。从宿主体人工摘下未饱血雌蜱的体重为 136—395毫克,其产卵过程中各种相关关系与自然掉下的饱血雌蜱相似,但产卵高峰所需天数缩短和体重最轻者(136—155毫克)其产卵前期所需天数延长。  相似文献

长角血蜱Haemaphysalis longicornis的交配行为包括7个时期,行为的完成依赖于性信息素的调节。生物测定表明:雄蜱的行为反应受雌蜱分泌的性信息素影响。堵塞雌蜱盾窝其行为受到抑制,点滴2,6-DCP或雌蜱盾窝腺提取物则被恢复。用气相色谱法测定了雌蜱盾窝腺中2,6-DCP的含量;吸血后1~2天含量最高(11.12 ng/只);吸血后3~5天即交配前下降交维持在一较恒定的水平;吸血后6~7天即交配后明显降低;饱血后检测不到2,6-DCP。2,6-DCP是长角血蜱性信息素的一种成分。  相似文献

为阐明蜕皮激素在硬蜱发育和生殖中的作用,用高效液相色谱法(HPLC)和放射免疫测定法测定了银盾革蜱和长角血蜱中饱血和产卵前后雌虫整体、血淋巴、合神经节及卵巢中蜕皮激素含量的变化.此外,还用HPLC测定了长角血蜱的卵和幼虫中蜕皮激素的含量.结果表明:在饱血前雌虫整体中蜕皮激素含量的变化不大,饱血后迅速增加.血淋巴中的蜕皮激素在饱血后第5天(即产卵前1天)达到高峰,可做为产卵的信号.合神经节中蜕皮激素含量的高峰在饱血后第5天,与血淋巴中高峰出现的时间一致,这与神经分泌细胞的分泌活性变化相吻合.从饱血后第3天起,卵巢中蜕皮激素含量增加很快,直到产卵时止,在长角血蜱的卵中也测到α-蜕皮素,来源于卵巢,随胚胎发育进程其含量逐渐增加.卵产出后第4天到第12天,蜕皮激素的含量增加十几倍.  相似文献

森林革蜱 (DermacentorsilvarumOlenev)雌蜱唾液腺由唾液腺管和大量的腺泡组成。从假头基到唾液腺末端 ,唾液腺管分为三部分 ,即中央腺管、主分支腺管和小叶管。球状的腺泡分布在各级腺管上。气管和中央腺管并行。腺泡呈圆形或近圆形 ,表面呈褶皱状 ,并有细小的气管分布。饥饿雌蜱唾液腺长度短 (5 4 7 3 3 μm) ;吸血后长度增加 ,吸血后 3天达到最大值 (1 1 0 9 40 μm)。从吸血后 3天到饱血后前 3 天 ,无明显变化 ,饱血后 4天明显缩短 (5 0 0 0 0 μm)。饥饿雌蜱的腺泡直径短 (45 2 4 μm) ,吸血期逐渐增大 ,吸血后 5天达到最大值 (74 1 0 μm)。饱血后腺泡逐渐萎缩并于饱血后 4天退化。  相似文献

A proteomics approach was employed to identify proteins secreted into the hemolymph of Ornithodorus savignyi ticks 2 h after immune-challenge with the yeast, Candida albicans. Profiling of the proteins present in hemolymph of unchallenged ticks versus ticks challenged with heat-killed yeast revealed five proteins to be differentially expressed. The modulated protein spots were subjected to tandem mass spectrometry (MS/MS) analysis, but could not be positively identified. These proteins can be assigned to the immune response as they were not induced after aseptic injury. In an attempt to identify hemolymph proteins that recognize and bind to yeast cells, hemolymph obtained from both unchallenged and challenged ticks was incubated with C. albicans. Elution of the bound proteins followed by SDS–PAGE analysis indicated that three proteins (97, 88 and 26 kDa) present in both unchallenged and challenged hemolymph samples bind to yeast cells. The constant presence of these three proteins in tick hemolymph leads us to believe that they may be involved in non-self recognition and participate in yeast clearance from tick plasma. The analyzed yeast-binding proteins could also not be positively identified, suggesting that all the tick immune proteins investigated in this study are novel.  相似文献

Summary

Vitellogenesis in ticks is known to be induced by engorgement and mating. In this paper, the synthetic pyrethroid cypermethrin (CyM) is shown to induce production of yolk protein precursor, vitellogenin (Vg), and ovarian development in unengorged mated adult female Ornithodoros moubata. The levels of Vg found in the hemolymph and ovarian development induced by CyM were dose-dependent. i.e., CyM doses of more than 0.2 and 1.0 μg/tick were needed for significant increase of Vg titer in the hemolymph and yolk deposition in oocytes, respectively. Immunological and electrophoretical analyses of Vg and Vitellin (Vn) induced by CyM were identical with those induced by engorgement. Vg titer induced by CyM in unengorged females followed approximately the same time course as that in the normal engorged females. However, Vg titer induced by CyM continued to increase after day 8 and reached a maximum (95 μg/μ1) on day 10 after treatment, while Vg titer induced by engorgement decreased again after reaching a maximum (60 μg/μ1) on day 6, correlated with yolk Vn deposition in oocytes. Ovarian development induced by even high doses (10 or 20 μg/tick) of CyM was slow compared to normal development stimulated by engorgement. Oviposition was not observed in females treated with CyM.  相似文献

Hemolymph proteins of a soft tick, Ornithodoros moubata, were analyzed immunochemically and biochemically. The components of tick hemolymph proteins were shown to be totally different from the host (rabbit) serum proteins by polyacrylamide gel electrophoresis with sodium dodecyl sulfate and Coomassie blue or silver stain. However, in the hemolymph of ticks engorged from rabbits immunoglobulin G was detected by immunoblotting analysis with goat anti-rabbit immunoglobulin G. The concentration of rabbit Immunoglobulin G in tick hemolymph changed with the physiological stages after a blood meal. Immunoglobulin G was isolated from tick hemolymph by affinity chromatography on a Protein A-Sepharose 4B column. Analysis of the isolated immunoglobulin G from tick hemolymph with sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Ouchterlony double diffusion test showed it to be composed of the same subunits as heavy and light chains of host (rabbit) immunoglobulin G. Tracer experiments showed that 125I-labeled heavy and light chains of immunoglobulin G were detected in an intact form in hemolymph from ticks that sucked 125I-labeled rabbit immunoglobulin G through an artificial membrane. These facts suggested that the host rabbit immunoglobulin G ingested in the tick midgut passed through the gut wall without digestion. By solid-phase enzyme immunoassay, immunoglobulin in the hemolymph was shown to retain its antibody activity.  相似文献

粘虫幼虫血淋巴脂蛋白的分离鉴定及理化特性分析   总被引:1,自引:0,他引:1  
通过利用NaBr密度梯度超速离心对粘虫Mythimna separata幼虫的血脂蛋白进行了分离及理化特性分析。查明了幼虫血淋巴内主要是高密度脂蛋白(HDLp),而低密度脂蛋白(LDLp)的含量极低。这两种脂蛋白的密度分别为HDlp 1.123g/Ml,LDLp 1.059g/mL。HDLp和LDLp蛋白含量、脂类、氨基酸的种类测定结果表明HDLp,LDLp的蛋白含量分别是25.2μg/μL和11μg/μg。HDLp水解后可获得16种氨基酸。LDLp可获得15种。氨基酸的种类比较一致,其含量变化幅度也不大。HDLp脂类部分占最多的是二酰基甘油酯(DC)和磷脂(PC)。其余的脂类依次是碳氢化合物(HC)和胆固醇?。LDLp主要是磷脂和碳氢化合物。昆虫脂蛋白的主要中性甘油酯是二酰基甘油酯。  相似文献

High concentrations of PGE(2) and PGF(2alpha) were identified by radio-immunoassay (RIA) and/or gas chromatography/mass spectrometry (GC/MS) in the hemolymph, salivary glands and saliva of the lone star tick Amblyomma americanum (L.). Binding studies indicated that PGE(2) was free and not bound to any proteins in the hemolymph. A small amount of 6-keto-PGF(1alpha) (breakdown product of PGI(2); prostacyclin) was also found in the salivary glands but not in the hemolymph or saliva. Neither PGD(2) nor PGA(2)/B(2) was detected in any tick material investigated. Although PGE(2) was found in the gut contents, only small amounts of label crossed the gut into the hemolymph during artificial feeding with labeled PGE(2), indicating that the high amounts of PGE(2) in hemolymph and salivary glands are not sequestered from the host blood meal. Isolated salivary glands and salivary gland homogenates demonstrated robust synthesis of PGE(2) at high concentrations of exogenous arachidonic acid. Synthesis by the salivary glands was monitored by measuring increasing PGE(2) with increasing arachidonic acid by RIA, GC/MS and labeled PGE(2) in the presence of labeled arachidonic acid. Synthesis was inhibited in a dose-dependent manner by indomethacin indicating that the cyclooxygenase synthesizing prostaglandins in ticks shares similarities to the enzyme found in mammals.  相似文献

The main protein of the hemolymph of the cattle tick Boophilus microplus has been isolated and shown to be a heme lipoprotein (HeLp). HeLp has an apparent molecular mass of 354,000 and contains two apoproteins (103 and 92 kDa) found in equal amounts. HeLp presents a pI of 5.8 and a density of 1.28 g/ml and contains 33% lipids, containing both neutral lipids and phospholipids, and 3% of sugars. A remarkable feature of HeLp is the abundance of cholesterol ester (35% of total lipids), a lipid not previously reported in invertebrate lipoproteins. Western blot analysis showed HeLp in hemolymph from adult females and males, but not in eggs. Although HeLp contains 2 heme molecules, it is capable of binding 6 additional molecules of heme. Boophilus feeds large amount of blood, and we recently showed that this tick is unable to perform de novo synthesis of heme (Braz, G. R. C., Coelho, H. S. L., Masuda, H., and Oliveira, P. L. (1999) Curr. Biol. 9, 703-706). Injection of tick females with (55)Fe-labeled heme-HeLp indicated that this protein transports heme from hemolymph to tissues. HeLp is suggested to be an essential adaptation to the loss of the heme synthesis pathway.  相似文献

Trehalose levels were determined over two 24 hr spans in groups of face fly adults 3–4 days after emergence from the puparium. Face fly pupae were placed in rearing chambers at 27° C in a staggered light-dark regimen, LD 16:8, so that at a given clock hour, samples could be obtained at several different hours after lights on (HALO). Trehalose was determined in hemolymph collected from a puncture in the intersegmental membrane of the abdomen. Treated hemolymph samples were passed through a Bio-Rad Amino S-S disaccharide column and a Waters 410 refractive index detector was used to differentiate among sugars. The circadian acrophase derived by cosinor analysis in hemolymph trehalose (when the values were 25.49 and 26.86μg/μ1 on the first and second days respectively) occurred at -226° (ca 15 HALO) and the bathyphase at 24 HALO. The mesor = 11.82μg/μ1 trehalose, the amplitude = 8.57/μg/μ1 trehalose and the P-value for presence of a rhythm was 0.003. Based on these data, differences between control and test flies in a bioassay of hypertrehalosemic activity would be most easily observed at 0-8 HALO, while exogenous hypotrehalosemic activity would be best assayed at 12-20 HALO.  相似文献

Antifungal and antibacterial activities were detected in the hemolymph and gut contents of the cattle tick, Boophilus microplus. A peptide with antibacterial activity from the tick gut contents was purified to homogeneity by reversed-phase chromatography. The molecular mass of the purified peptide was 3,205.7 Da, measured by matrix-assisted laser desorption/ionization mass spectrometry. The amino acid sequence was obtained by Edman degradation and showed that the peptide was identical to a fragment of the bovine alpha-hemoglobin. A synthetic peptide based on the sequence obtained showed characterization data identical to those of the isolated material, confirming its structure. The synthetic peptide was active in micromolar concentrations against Gram-positive bacteria and fungi. These data led us to conclude that the antibacterial activity detected in tick gut contents is the result of enzymatic processing of a host protein, hemoglobin. This activity may be used by ticks as a defense against microorganisms.  相似文献
