Pochonia chlamydosporia (Pc123) is a fungal parasite of nematode eggs which can colonize endophytically barley and tomato roots. In this paper we use culturing as well as quantitative PCR (qPCR) methods and a stable GFP transformant (Pc123gfp) to analyze the endophytic behavior of the fungus in tomato roots. We found no differences between virulence/root colonization of Pc123 and Pc123gfp on root-knot nematode Meloidogyne javanica eggs and tomato seedlings respectively. Confocal microscopy of Pc123gfp infecting M. javanica eggs revealed details of the process such as penetration hyphae in the egg shell or appressoria and associated post infection hyphae previously unseen. Pc123gfp colonization of tomato roots was low close to the root cap, but increased with the distance to form a patchy hyphal network. Pc123gfp colonized epidermal and cortex tomato root cells and induced plant defenses (papillae). qPCR unlike culturing revealed reduction in fungus root colonization (total and endophytic) with plant development. Pc123gfp was found by qPCR less rhizosphere competent than Pc123. Endophytic colonization by Pc123gfp promoted growth of both roots and shoots of tomato plants vs. uninoculated (control) plants. Tomato roots endophytically colonized by Pc123gfp and inoculated with M. javanica juveniles developed galls and egg masses which were colonized by the fungus. Our results suggest that endophytic colonization of tomato roots by P. chlamydosporia may be relevant for promoting plant growth and perhaps affect managing of root-knot nematode infestations.  相似文献

An experiment was conducted to test the effect of different doses of 2, 4 and 8?g/2?kg of soil of Pochonia chlamydosporia against the root-knot nematode (Meloidogyne incognita) on Phaseolus vulgaris. It was observed that inoculation of plant with the nematode alone, and 15?days prior to fungal inoculation, reduced the plant growth when compared with the plant with fungal application followed by the nematode. Plant length, fresh and dry weight, chlorophyll, carotenoid, protein contents and nitrate reductase activity decreased in nematode-infested plants. Application of higher dose of 8?g/2?kg of soil of P. chlamydosporia increased all the plant growth parameters as well as biochemical parameters. Highest number of galls per root system was recorded on the plants infested with nematode but not treated with the fungus. However, application of fungus prior to nematode inoculation improved the plant growth and reduced the number of galls and the number of egg masses per root system.  相似文献

Fungi were isolated from Meloidogyne spp. eggs and females on 102 field-collected root samples in China. Of the 235 fungi isolated (representing 18 genera and 26 species), the predominant fungi were Fusarium spp. (42.1% of the isolates collected), Fusarium oxysporum (13.2%), Paecilomyces lilacinus (12.8%), and Pochonia chlamydosporia (8.5%). The isolates were screened for their ability to parasitise Meloidogyne incognita eggs in 24-well tissue culture plates in two different tests. The percentage of eggs parasitised by the fungi, the numbers of unhatched eggs and alive and dead juveniles were counted at 4 and 7 days after inoculation. The most promising fungi included five Paecilomyces isolates, 10 Fusarium isolates, 10 Pochonia isolates and one Acremonium isolate in test 1 or test 2. Paecilomyces lilacinus YES-2 and P. chlamydosporia HDZ-9 selected from the in vitro tests were formulated in alginate pellets and evaluated for M. incognita control on tomato in a greenhouse by adding them into a soil with sand mixture at rates of 0.2, 0.4, 0.8 and 1.6% (w/w). P. lilacinus pellets at the highest rate (1.6%) reduced root galling by 66.7%. P. chlamydosporia pellets at the highest rate reduced the final nematode density by 90%. The results indicate that P. lilacinus and P. chlamydosporia as pellet formulation can effectively control root-knot nematodes.  相似文献

The effects of Glomus mosseae and Paecilomyces lilacinus on Meloidogyne javanica of tomato were tested in a greenhouse experiment. Chicken layer manure was used as a carrier substrate for the inoculum of P. lilacinus. The following parameters were used: gall index, average number of galls per root system, plant height, shoot and root weights. Inoculation of tomato plants with G. mosseae did not markedly increase the growth of infected plants with M. javanica. Inoculation of plants with G. mosseae and P. lilacinus together or separately resulted in similar shoots and plant heights. The highest root development was achieved when mycorrhizal plants were inoculated with P. lilacinus to control root-knot nematode. Inoculation of tomato plants with G. mosseae suppressed gall index and the average number of galls per root system by 52% and 66%, respectively, compared with seedlings inoculated with M. javanica alone. Biological control with both G. mosseae and P. lilacinus together or separately in the presence of layer manure completely inhibited root infection with M. javanica. Mycorrhizal colonization was not affected by the layer manure treatment or by root inoculation with P. lilacinus. Addition of layer manure had a beneficial effect on plant growth and reduced M. javanica infection.  相似文献

The effects of a host plant on reproduction/abundance of fungal populations in relation to soil nutrients released by plants in the rhizosphere were studied. Abundance in the soil and potato rhizosphere of the fungi Paecilomyces lilacinus, Monographella cucumerina (CABI 380408) and Pochonia chlamydosporia var. chlamydosporia (Pc280, potato cyst nematode biotype) and P. chlamydosporia var. catenulata (Pc392, root‐knot nematode biotype) were assessed. The different ability of break crops (oilseed rape, sugarbeet and wheat) in the potato rotation to support Pa. lilacinus, Pochonia isolates Pc280 and Pc392 and abundance of the latter two isolates in soil and rhizosphere of potato plants infected with Meloidogyne incognita were also studied. Potato chits and crop seedlings were planted into boiling tubes containing 5000 chlamydospores or conidia g?1 in acid washed sand (pH 6) and kept in a growth chamber at 20°C, and 16 h of light for up to 9 weeks. The abundance of the fungi in sand (fallow) differed significantly between fungal species, being in general less abundant in the absence than in the presence of the plant, although there was no interaction between plant species and fungal isolate. There was evidence of a different response to Me. incognita for Pc392 than for Pc280 but there was no significant effect of the presence of the nematode on the rate of increase of the fungus.  相似文献

Three control strategies for management of Meloidogyne arenaria were evaluated on carrot under greenhouse conditions. The control strategies tested were: i) incorporation of fresh broccoli; ii) carbofuran (Furadan®); and iii) Pochonia chlamydosporia var. mexicana, isolate Pcp21. Each strategy was evaluated separately and in combination (16 treatment combinations), each with three replicates. The experiment was done between February and April 2014 and repeated over the same time period in 2015. Each replicate experimental unit consisted of a 1?kg pot filled with tyndallised soil, to which broccoli, fungus and carbofuran were added according to treatment. Carrot seeds were sown into each pot and inoculated with M. arenaria eggs according to treatment. Pots were then maintained in the glasshouse for 12 weeks at 25?±?5°C. After this time the fresh root weight, root length, percent reduction in root galling, P. chlamydosporia var. mexicana colony forming units (CFU)/g of soil, and CFU/g root were all measured. Results showed that isolate Pcp21 of P. chlamydosporia var. mexicana significantly reduced root galling percentage by 50 and 78% in 2014 and 2015, respectively. Application of the fungus in combination with broccoli or carbofuran also reduced root galling. The number of CFU/ g of soil or root remained the same over the 12 weeks of the experiment. This isolate of P. chlamydosporia significantly reduced damage caused by M. arenaria on carrot and is a promising agent for management of this nematode.  相似文献

Effects of plant growth promoting rhizobacteria (Pseudomonas putida MTCC No. 3604 and Pseudomonas alcaligenes MTCC No. 493) and parasitic fungi (Pochonia chlamydosporia KIA and Paecilomyces lilacinus KIA) were studied, alone and together with Rhizobium sp. (charcoal commercial culture) on the growth of chickpea and multiplication of Meloidogyne javanica. Individually, P. putida 3604, P. alcaligenes 493 and Rhizobium caused a significant increase in the growth of chickpea in both nematode inoculated and uninoculated plants. Inoculation of Rhizobium with a parasitic fungus or with plant growth promoting rhizobaterium caused a greater increase in the growth of plants inoculated with nematodes than caused by either of them singly. Individually, P. lilacinus KIA caused a greater increase in the growth of nematode inoculated plants than caused by P. putida 3604 or P. alcaligenes 493. P. lilacinus KIA caused a greater reduction in galling and nematode multiplication followed by P. chlamydosporia KIA, P. putida 3604 and P. alcaligenes 493. Combined use of P. lilacinus KIA with Rhizobium was better in reducing galling and nematode multiplication than any other treatment. P. putida 3604 caused a greater colonization of root than P. alcaligenes 493 while P. lilacinus KIA was isolated from more nematodes than P. chlamydosporia KIA.  相似文献

Interaction between arbuscular mycorrhizal fungus Glomus mosseae and plant growth promoting fungus Phoma sp. was studied for its effect on their root colonization and plant growth of cucumber. Two isolates of Phoma sp. (GS8-2 and GS8-3) were tested with G. mosseae. The percent root length colonized by G. mosseae was not adversely affected by the presence of Phoma isolates. In contrast, the root colonization of both isolates GS8-2 and GS8-3 in 4-week-old plants was significantly reduced (80.7% and 84.3%, respectively) by added G. mosseae. Inoculating plants with each Phoma isolate significantly increased the shoot dry weight. However, dual inoculation of each Phoma isolate with G. mosseae had no significant effect on growth enhancement.  相似文献

Twenty-one isolates of phosphate solubilizing-indole acetic acid producing rhizobacteria (PSIRB), 20 isolates of phosphate solubilizing rhizobacteria (PSRB) and 42 isolates of indole acetic acid producing rhizobacteria (IRB) were isolated from 49 rhizospheric soil samples of tomato (Lycopersicon esculentum Mill.) collected from tomato growing regions of Karnataka. A method combining Pikovskaya’s and Bric’s technique was developed to isolate PSRIB, PSRB and IRB’s. The selected isolates were further analyzed for their ability to solubilize calcium phytate. Based on the root colonization assays and the abilities of bacterial isolates to increase the seed germination and seedling vigor under laboratory conditions, five isolates were selected from each group for further studies. Under greenhouse conditions, all the selected rhizobacteria isolates significantly increased root length, shoot length, fresh weight, dry weight and total phosphorus content of 30-day-old-seedlings with respect to control. Isolate PSIRB1 and IRB36 significantly reduced the Fusarium wilt incidence over other isolates of same and other group, and the control. On the basis of results from laboratory and greenhouse studies, one bacterial isolate from each group was selected for plant growth and yield analysis studies. Isolate PSIRB2 showed increased plant height, fresh weight, number of fruits per plant and average weight of fruit over PSRB9, IRB36 and untreated controls. Studies on the nature of protection offered by these bacterial isolates following split-root technique revealed that the isolates PSIRB2 and PSRB9 had the ability to induce systemic resistance. One isolate, IRB36 appeared to protect the tomato seedlings through direct antagonism.  相似文献

【背景】利迪链霉菌(Streptomyces lydicus)对多种作物均有较好的促生效果,且对病原真菌具有广谱抑制作用,但该菌对细菌性青枯病的防控研究较少。【目的】探究利迪链霉菌M01能否促进番茄生长并抑制番茄青枯病,以及M01对番茄生长的影响是否通过影响根际细菌群落结构实现。【方法】采用温室盆栽试验和扩增子高通量测序技术研究M01对番茄生长、青枯病发病率及根际细菌群落组成的影响。【结果】施用利迪链霉菌M01的番茄植株鲜重、干重、株高、用土壤与作物分析开发(soil and plant analyzer develotrnent, SPAD)方法测量的叶绿素浓度、根系活力和植株P含量比对照分别提高了22.7%、12.5%、16.0%、28.1%、18.4%和17.9%,其中对株高、SPAD值和植株磷含量影响显著(P<0.05)。M01处理延缓了番茄青枯病的发病时间,接种9周后发病率比对照降低了41.8%。此外,M01对番茄根际细菌群落无显著影响(门水平群落组成,P=0.4;属水平群落组成,P=0.4)。【结论】利迪链霉菌M01可促进番茄植株生长并抑制番茄青枯病,利迪链霉菌M01对番茄生长的影响并非通过调控根际细菌群落实现。  相似文献

Application of the tertiary amine bioregulator, 2-(3,4-dichlorophenoxy)triethylamine [DCPTA] to seedling Phalaenopsis plants increased seeding survival, long-term vegetative plant growth, and greatly accelerated flowering when compared to controls. Application of 30 µM (10 ppm) DCPTA during the transfer of orchid seedlings from sterile agar culture to greenhouse community pot culture caused a 2 to 3-fold increase in root and leaf growth of seedling plants over the growth of controls. After 2 years greenhouse culture, the total dry weight of 30 µM DCPTA-treated plants was doubled when compared with controls. Flowering of 30 µM DCPTA-treated plants was increased significantly (p < 0.05) over that of controls after 11 and 18 months plant growth. During a 2 year growth period, the total number of racemes produced by 30 µM DCPTA-treated plants was doubled when compared with the floral performance of controls. In the present study, 43 isolates of Phosphate solubilizing rhizobacteria (PSRB) were isolated from 37 rhizospheric soil samples of tomato collected from tomato growing regions of Karnataka. Among the 43 isolates, 33 were found to be positive for solubilizing both inorganic and organic forms of phosphorous. The isolates were analyzed for their ability to colonize roots of tomato and to increase the seed quality parameters under laboratory conditions. On the basis of above criteria, 16 isolates were selected for further studies. Organic acids from PSRB isolates were analyzed and phytase zymogram for two isolates viz., PSRB21 and 31 was prepared. Under greenhouse conditions, all selected isolates showed increased shoot length, root length, fresh weight, dry weight and phosphorous content of tomato seedlings to various extent with respect to control. Analysis of pH and available phosphorous in rhizosphere soil samples of 30 day-old-seedlings revealed that the available phosphorous content was high in rhizospheric soil samples of plants raised from seeds bacterized with PSRB isolates over control. Even though all selected PSRB’s were able to increase the plant growth, only few of them showed protection against fusarium wilt and none of them against early blight.  相似文献

Beneficial plant-microbe interactions in the rhizosphere are primary determinants of plant health and soil fertility. Some antagonistic fungi have shown great effects toward the growth of plant crops. In this study, two major crops, cotton and potato, were selected to evaluate their growth promotion by the antagonistic fungus Talaromyces flavus. For each plant, five T. flavus isolates were selected from our fungal collection which had shown the highest antagonistic activities against the causal agent of wilt diseases on these plants. In the next step, for every crop, five isolates were used under greenhouse conditions. For evaluation of the plant growth promotion ability of T. flavus isolates, a split-plot trial was arranged in a randomized complete block design with four replications. The main factor was the method of application of T. flavus as a soil treatment, a seed treatment, and a combination of both methods. The subfactor was the use of different fungal isolates. Measured parameters were root length, crown length, plant height, plant fresh weight, and plant dry weight. Results showed that the maximum increase in the above parameters was mediated by the seed treatment method. The most effective isolate for cotton plants was TF-Co-M-23, which increased root length, plant height, plant fresh weight, and plant dry weight by 1.80-, 2.26-, 1.23-, and 1.19-fold, respectively. There were no significant differences among the various treatments affected by T. flavus in terms of crown length. The most effective isolate for potato plants was TF-Po-V-50, which increased root length, crown length, plant height, and plant dry weight by 1.71-, 1.09-, 1.45-, and 3.75-fold, respectively. The overall results of this study suggest that it may be possible to promote cotton and potato growth characteristics by using the antagonistic fungus T. flavus.  相似文献

A greenhouse investigation was conducted to study the influence of the arbuscular mycorrhizal (AM) fungus Glomus mosseae and the plant growth-promoting rhizomicroorganisms (PGPRs) Bacillus coagulans and Trichoderma harzianum on the growth and nutrition of micropropagated Ficus benjamina plantlets. The AM fungus was inoculated either singly or in combination with the PGPRs. Plants showed maximum plant height, biomass, P content, mycorrhizal root colonization, spore numbers and populations of T. harzianum and B. coagulans in root zone soil when all the three organisms were inoculated together. Thus, when G. mosseae co-inoculated with PGPRs enhances growth and nutrition of Ficus benjamina. T. harzianum and B. coagulans are thus designated as mycorrhizal helper organisms.  相似文献

The purpose of this study was to analyze morphological and physiological aspects of Arbutus unedo L. plants treated with paclobutrazol (PAC), compounds characterized by their double activity as plant growth regulators and fungicides, and the ectomycorrhizal fungus Pisolithus tinctorius (Pers.) Coker and Couch, which forms a special type of mycorrhizal colonization called arbutoid mycorrhiza. Native A. unedo L. seedlings were grown in a greenhouse and subjected to four treatments for 4 months: 0 or 60 mg of PAC and inoculated or not with P. tinctorius (Pers.). The arbutoid mycorrhizal inoculation increased in plants treated with PAC. Paclobutrazol reduced shoot and root biomass, plant height, internode length, stem diameter, leaf area, total root length and number of tips. P. tinctorius increased plant height and had a beneficial effect on the root system (increasing root diameter and the number of tips). PAC treatment led to an increase in ion levels in the leaf tissue, while mycorrhizal inoculation induced lower K and higher P contents in the roots. Leaf water potentials (at predawn and at midday) increased with the combined treatment. The absence of water deficit conditions meant there was no osmotic adjustment. Higher photosynthesis (Pn) values were associated with higher stomatal conductance (gs) values in the mycorrhizal plants, which influenced water uptake from the roots. However, gs decreased in the PAC-treated plants, reducing photosynthesis and, as a consequence, growth. The higher hydraulic conductivity (Lp) in the plants treated with PAC may have induced a better water energy status and good water transport. The combined treatment produced beneficial effects in the plants, improving their water and nutritional status.  相似文献

为了研发对番茄灰霉病高效、稳定、安全的生物农药,试验利用自主分离获得的寡雄腐霉菌株制备发酵液,采用盆栽试验研究寡雄腐霉发酵液对番茄生长的影响和对灰霉病的防治效果及机制,并在大田生产中验证其生防效果。结果表明,盆栽试验中,寡雄腐霉发酵液促进健康番茄植株生长,植株总生物量和根系生物量分别增加9.5%和15.4%,提高了植株叶绿素含量、根系活力及氮、磷、钾吸收量,并使带病番茄植株的发病率和病情指数分别降低57.2%和60.3%,相对防治效果达60.3%,施用寡雄腐霉发酵液对番茄叶片细胞膜具有保护性,降低丙二醛含量,提高病原性相关酶""超氧化物歧化酶、多酚氧化酶和苯丙氨酸解氨酶活性。后续田间试验中寡雄腐霉发酵液对番茄灰霉病的防治效果达71.2%。说明寡雄腐霉发酵液能有效防治番茄灰霉病,还具有促进番茄生长的作用,并且可诱导番茄植株对病原菌的防御作用,应用前景广泛。  相似文献

Saline soils constitute a serious production problem for vegetable crops as they are known to suppress plant growth. One of the possible measures to improve crop health in such conditions is to use salt-tolerant bacterial inoculants which can control diseases and promote plant growth. In the present work the ability of Pseudomonas chlororaphis isolate TSAU13 to promote cucumber and tomato plant growth and to improve fruit yield by protecting these plants against soil-borne pathogens in salinated soil were investigated. The bacterial strain stimulated shoot growth (up to 32%), dry matter (up to 43%), and the fruit yield of tomato and cucumber (up to 16%) compared to the uninoculated control plants under saline conditions. The strain was able to survive on the root of 2-month-old plants. 29% of the cucumber and 27% of the tomato plants which had grown in soil to which no Fusarium solani spores had been added were diseased, whereas in the presence of the pathogenic fungus 58% of the cucumber and 52% tomato plants had disease symptoms. P. chlororaphis TSAU13 showed statistically significant disease reduction in comparison to the Fusarium-uninfected and infected control plants. Those results showed that P. chlororaphis TSAU13 has a great biotechnological potential in improvement of vegetable production in commercial greenhouses under saline conditions.  相似文献

In order to study the influence of Arbuscular Mycorrhiza (AM) on the development of root rot infection, tomato plants were raised with or withoutGlomus mosseae and/orPhytophthora nicotianae var.parasitica in a sand culture system. All plants were fed with a nutrient solution containing one of two phosphorus (P) levels, 32µM (I P) or 96µM (II P), to test the consequence of enhanced P nutrition by the AM fungus on disease dynamics. Mycorrhizal plants had a similar development to that of control plants. Treatment withPhytophthora nicotianae var.parasitica resulted in a visible reduction in plant weight and in a widespread root necrosis in plants without mycorrhiza. The presence of the AM fungus decreased both weight reduction and root necrosis. The percentage reduction of adventitious root necrosis and of necrotic root apices ranged between 63 and 89% The enhancement of P nutrition increased plant development, but did not appreciably decrease disease spread. In our system, mycorrhiza increased plant resistance toP. nicotianae var.parasitica infection. Although a contribution of P nutrition by mycorrhiza cannot be excluded, other mechanisms appear to play a crucial role.  相似文献

Bacterial canker of tomato, caused by Clavibacter michiganensis subsp. michiganensis, continues to be a problem for tomato growers in the Souss-Massa Draa valley, South of Morocco. Assuming that biological control is an alternative for the management of this disease, a total of 303 fluorescent pseudomonads strains isolated from roots and rhizospheric soil of tomato plants were in vitro tested against C. michiganensis subsp. michiganensis. Fluorescent pseudomonads strains which showed the highest antagonistic properties were thereafter investigated for their ability to colonize tomato roots. Our results showed that fluorescent pseudomonads are more represented in rhizospheric soils. However, the most efficient fluorescent pseudomonads isolates were found in the rhizoplane soil and the endorhizosphere. Among 42 spontaneous antibiotic resistant mutants obtained by treatment of the wild-type isolates with five antibiotics (rifampicine, nalidixic acid, ampicilline and chloramphenicol), 28 completely colonized the roots of all tomatoes seedlings used in this investigation. The 42 wild type isolates were then used for in vivo screening with the cotyledon test. Using this test, eight isolates from 42 tested induced a significant decrease of disease incidence and disease symptoms. The eight efficient isolates were then tested for their effectiveness in the protection of tomato plants in pots under greenhouse conditions. Results obtained showed that all tested isolates applied as seed and root treatments reduced significantly (P ≤ 0.001) the incidence of bacterial canker.  相似文献
