Clonal replicates of different transformed potato plants expressing transgene constructs containing the constitutive Cauliflower Mosaic Virus (CaMV) 35S promoter, and sequences encoding the plant defensive proteins snowdrop lectin (Galanthus nivalis agglutinin; GNA), and bean chitinase (BCH) were propagated in tissue culture. Plants were grown to maturity, at first under controlled environmental conditions, and later in the glasshouse. For a given transgene product, protein accumulation was found to vary between the different lines of clonal replicates (where each line was derived from a single primary transformant plant), as expected. However, variability was also found to exist within each line of clonal replicates, comparable to the variation of mean expression levels observed between the different clonal lines. Levels of GNA, accumulated in different parts of a transgenic potato plant, also showed variation but to a lesser extent than plant–plant variation in expression. With the majority of the clonal lines investigated, accumulation of the transgene product was found to increase as the potato plant developed, with maximum levels found in mature plants. The variation in accumulation of GNA among transgenic plants within a line of clonal replicates was exploited to demonstrate that the enhanced resistance towards larvae of the tomato moth, Lacanobia oleracea L., caused by expression of this protein in potato, was directly correlated with the level of GNA present in the plants, and that conditions under which the plants were grown affect the levels of GNA expression and subsequent levels of insect resistance.  相似文献

Nitrogen fixation (acetylene reduction) rates were measured over an annual cycle in meadows of the seagrass Z. noltii and uncolonised sediments of the Bassin d'Arcachon, south-west France, using both slurry and whole core techniques. Measured rates using the slurry technique in Z. noltii colonised sediments were consistently higher than those determined in isolated cores. This was probably due to the release of labile organic carbon sources during preparation of the slurries. Thus, in colonised sediments the whole core technique may provide a more accurate estimate of in situ activity. Acetylene reduction rates measured by the whole core technique in colonised sediments were 1.8 to 4-fold greater, dependent upon the season, in the light compared with those measured in the dark, indicating that organic carbon released by the plant roots during photosynthesis was an important factor regulating nitrogen fixation. In contrast acetylene reduction rates in uncolonised sediments were independent of light.Addition of sodium molybdate, a specific inhibitor of sulphate reduction inhibited acetylene reduction activity in Z. noltii colonised sediments by > 80% as measured by both slurry and whole core techniques irrespective of the light regime, throughout the year inferring that sulphate reducing bacteria (SRB) were the dominant component of the nitrogen fixing microflora. A mutualistic relationship between Z. noltii and nitrogen fixing SRB in the rhizosphere, based on the exchange of organic carbon and fixed nitrogen is proposed. In uncolonised sediments sodium molybdate initially severely inhibited acetylene reduction rates, but the level of this inhibition declined over the course of the year. These data indicate that the nitrogen fixing SRB associated with the Zostera roots and rhizomes were progressively replaced by an aerobic population of nitrogen fixers associated with the decomposition of this recalcitrant high C:N ratio organic matter.Acetylene and sulphate reduction rates in the seagrass beds showed distinct summer maxima which correlated with a reduced availability of NH 4 + in the sediment and the growth cycle of Z. noltii in the Bassin. Overall, these data indicate that acetylene reduction (nitrogen fixation) activity in the rhizosphere of Z. noltii was regulated both by release of organic carbon from the plant roots and maintenance of low ammonium concentrations in the root zone due to efficient ammonium assimilation.Nitrogen fixation rates determined from acetylene reduction rates measured by the whole core technique ranged from 0.1 to 7.3 mg N m–2 d–1 in the Z. noltii beds and between 0.02 and 3.7 mg N m–2 d–1 in uncolonised sediments, dependent upon the season. Nitrogen fixation in the rhizosphere of Z. noltii was calculated to contribute between 0.4 and 1.1 g N m–2 y–1 or between 6.3 and 12% of the annual fixed nitrogen requirement of the plants. Heterotrophic nitrogen fixation therefore represents a substantial local input of fixed nitrogen to the sediments of this shallow coastal lagoon and contributes to the overall productivity of Z. noltii in this ecosystem.  相似文献

Arcachon Bay is characterized by extensive meadows of the seagrass Zostera noltii. Moreover, as a consequence of eutrophication, massive proliferations of the macroalga (Monostroma obscurum) have occurred since the beginning of 1990s.This paper describes the anaerobic decomposition of biomass of both species under experimental conditions by two methods. Firstly, the dynamics of decomposition were studied in situ using litter bags. The remaining biomass and the elemental composition of the decomposing macrophytes were monitored. Secondly, degradation was studied in experimental containers under anoxic conditions in which the release of inorganic nutrients and the development of fermentative and sulfate-reducing bacterial populations were followed.The decomposition rate of total biomass was faster for macroalgae than for the vascular plants, thus corroborating previous observations. However, both in situ and laboratory experiments showed that the anaerobic decomposition of the seagrass Z. noltii resulted in rapid release of inorganic N and P, and increasing C/N and C/P ratios of the residual biomass. As a result, the recycling of inorganic nitrogen and phosphorus compounds was slightly more efficient for Z. noltii than for M. obscurum. Recycling of inorganic nutrients appears to be of a great importance to the whole ecosystem, because of the extensive spreading of Z. noltii in the bay.  相似文献

The role of epiphytes in an intertidal Zostera noltii seagrass bed in Marennes-Oléron Bay was assessed in comparison with the other main benthic primary producers (Z. noltii, microphytobenthos) at two bathymetric levels and on a seasonal basis. Assemblage and biomass of epiphytes were studied using scanning electron microscopy (SEM). Z. noltii and its detrital matter followed a typical seasonal pattern: microphytobenthos was present in large quantities throughout the year representing 21% of the total biomass while detrital matter, above-ground parts and below-ground parts accounted for 65, 9 and 5%, respectively. Only two species of epiphytic diatoms, Cocconeis scutellum and Cocconeis placentula, were observed on seagrass leaves. Epiphyte biomass was very low, representing on average less than 0.001% of that of microphytobenthos or leaves. This low epiphyte biomass is linked with the absence of macroalgae and also with the low biovolume of Cocconeis, which formed a monolayer of cells on leaves. This can be explained by the severe conditions of the intertidal and the high leaf turn-over of Z. noltii leaves.  相似文献

The density, biomass and shoot morphology of two populations of Zostera noltii were monitored from January 1998 to July 1999 at two shallow Mediterranean lagoons of Biguglia and Urbino, which differ in hydro-morphological conditions and nutrient loading. Monitoring included the principal biological and foliar parameters (shoot density, aboveground and belowground biomass, length, width and number of leaves, LAI and coefficient A: percentage of leaves having lost their apex), the organic matter contents of the sediment and the environmental conditions (salinity, turbidity, temperature, nutrient concentrations and dissolved oxygen levels). The two populations of Z. noltii displayed seasonal changes in density (1600–19600 m2), aboveground biomass (11–153 g. DW. m−2), leaf length (33–255 mm), and leaf width (0.9–1.8 mm). Temperature and turbidity were significant environmental factors influencing the temporal changes observed in the Z. noltii meadows studied. Conversely, the belowground biomass, the number of leaves per shoot and the LAI did not undergo any seasonal changes. In the Biguglia lagoon, the functioning dynamics of the Z. noltii seagrass beds are determined by the catchment area and the inputs of nutrients derived from it, whereas in the Urbino lagoon the dynamics of the Z. noltiibeds depend on low levels of water turbidity.  相似文献

A combination of numerical modeling results with measurement and satellite imagery data was used during the biologically active period for the determination of the coastal zone extent in the central and eastern Gulf of Finland. Adopting the approach that the coastal zone can be identified by the spatial distribution of biotic parameters, spatial variations and gradients of chlorophyll a (chl-a) concentrations were analyzed. The results showed that chl-a concentrations vary in a wide range over the biologically active period. During heavy blooms, the coastal zone may appear occasionally and depend on the spatial distribution of the bloom. On average, clear limits of the coastal zone could be defined for the central and eastern Gulf of Finland. In the central Gulf of Finland, water and material exchange are rather intensive, and the coastal zone is narrower than in the eastern Gulf. In the easternmost part of the Gulf of Finland, chl-a concentrations were permanently high in an area of about 100 km width due to the discharge of the Neva River. The study has shown that gradients of chl-a spatial distribution can be applied for determining limits of the coastal zone extent. The standardized gradient of zero is shown to be a threshold separating the coastal zone (standardized gradients > 0) from the open sea (standardized gradients < 0). Guest editors: J. H. The trophic status of the eastern Gulf of Finland, where the largest Baltic metropolis St. Petersburg sits at the mouth of the largest Baltic river Neva, is elevated but existing recommendations on water protection measures are controversial. In this study, the effects of nutrient load reductions on this ecosystem were estimated with the aid of a three-dimensional coupled hydrodynamic-biogeochemical model. As a reference, the contemporary seasonal dynamics were simulated with nutrient inputs corresponding to the recent estimates of point and riverine sources. In order to eliminate the effects of natural inter-annual variations, the computations were run under recurrent annual forcing for 3 years, until quasi steady-state seasonal dynamics were reached. Reasonable comparability of simulated concentrations and biogeochemical fluxes to available field estimates provides credibility to scenario simulations. These simulations show that substantial reductions of nutrient point sources in St. Petersburg would affect only the Neva Bay as the immediate receptor of treated sewage waters, where primary production could decrease by up to 20%. Eutrophication in the other parts of the Neva Estuary and in the entire eastern Gulf of Finland would change insignificantly owing to increased nutrient import from the offshore waters. Therefore, more significant changes can occur only via a reduction in nutrient pools in the open Gulf of Finland and the Baltic Proper, which would require a longer time. Guest editors: J. H. Seagrass ecosystems are expected to benefit from the global increase in CO 2 in the ocean because the photosynthetic rate of these plants may be Ci‐limited at the current CO 2 level. As well, it is expected that lower external pH will facilitate the nitrate uptake of seagrasses if nitrate is cotransported with H+ across the membrane as in terrestrial plants. Here, we investigate the effects of CO 2 enrichment on both carbon and nitrogen metabolism of the seagrass Zostera noltii in a mesocosm experiment where plants were exposed for 5 months to two experimental CO 2 concentrations (360 and 700 ppm). Both the maximum photosynthetic rate (Pm) and photosynthetic efficiency (α) were higher (1.3‐ and 4.1‐fold, respectively) in plants exposed to CO 2‐enriched conditions. On the other hand, no significant effects of CO 2 enrichment on leaf growth rates were observed, probably due to nitrogen limitation as revealed by the low nitrogen content of leaves. The leaf ammonium uptake rate and glutamine synthetase activity were not significantly affected by increased CO 2 concentrations. On the other hand, the leaf nitrate uptake rate of plants exposed to CO 2‐enriched conditions was fourfold lower than the uptake of plants exposed to current CO 2 level, suggesting that in the seagrass Z. noltii nitrate is not cotransported with H+ as in terrestrial plants. In contrast, the activity of nitrate reductase was threefold higher in plant leaves grown at high‐CO 2 concentrations. Our results suggest that the global effects of CO 2 on seagrass production may be spatially heterogeneous and depend on the specific nitrogen availability of each system. Under a CO 2 increase scenario, the natural levels of nutrients will probably become limiting for Z. noltii. This potential limitation becomes more relevant because the expected positive effect of CO 2 increase on nitrate uptake rate was not confirmed.  相似文献

The dwarf seagrass Zostera noltii is an important primary producer in Atlantic coastal ecosystems from Mauritania to southern Norway and the Mediterranean Sea. Sessile intertidal organisms existing at the interface between marine and terrestrial environments may be particularly vulnerable to environmental change. In this study, we asked how near to thermal tolerance limits natural populations of Z. noltii are in the Ria Formosa coastal lagoon system in southern Portugal. We recorded the maximum temperatures in the Ria Formosa during the 2007 summer, and conducted experiments to determine the sub-lethal temperature of Z. noltii shoots sampled at two sites located at different tidal heights. Mortality rates and photosynthetic performance were recorded within a range of heat shock temperatures between 35 and 41°C. Survival was recorded ≤37°C, while higher temperatures led to a sudden drop in photosynthetic capacity followed by mortality (shoot loss) that occurred more rapidly with increasing temperatures. At 39°C and above, the rate of shoot mortality in both sites was close to 100%, occurring between 5 and 13 days after the heat shock. Survival was ca. 95 and 90% at 35 and 37°C, respectively. From these results for Z. noltii populations in the Ria Formosa we estimated sub-lethal temperature to be approximately 38°C for Z. noltii, close to the maximum of 36°C recorded in the summer 2007. Considering predicted trajectories in the coming decades, these results raise concern as to the future viability of intertidal Z. noltii populations near the southernmost edge of their distribution. Leaf dynamics and standing stocks of intertidal seagrasses were studied in the Baie d'Aouatif (Parc National du Banc d'Arguin, Mauritania) in April and September 1988. Standing stocks of Zostera noltii Hornem. suggest a unimodal seasonal curve similar to what is found for populations at higher latitudes. Also, leaf growth rates (0.03 cm2 cm–2 day–1 on average) were similar to those found at higher latitudes in these months. Variation in leaf loss over tidal depth, time and different locations in the Baie d'Aouatif was larger and more often significant than variation in leaf growth. In general, Z. noltii beds in the Baie d'Aouatif had comparable leaf growth rates and standing stocks. In both months losses were almost always higher than or equal to growth.Variation in leaf loss over time was much higher in the plots that were situated high in the intertidal than in lower plots. This is explained by differences in susceptibility to sloughing, which is presumably higher in periods with low tide around noon for shallow depths.In an experiment using artificial shading nets, in situ leaf growth was affected negatively from 94% shading onwards. This shading was observed to reduce the light intensity reaching the seagrass bed to a level below the reported range of light compensation points for Z. noltii. Cymodocea nodosa (Ucria) Ascherson on average had higher leaf area and relative growth rates than Z. noltii and much lower loss rates, resulting in a positive net increase in September. Standing stocks were also higher than for Z. noltii. A mixed seagrass bed containing the above two species and Halodule wrightii Ascherson had the highest observed total biomass: 335 g m–2 ash-free dry weight.  相似文献

The effects of UV-B radiation (290–320 nm) on development of damping-off of spinach (Spinacia oleracea) caused by the fungusFusarium oxysporum were examined in a growth cabinet. The incidence of disease greatly increased when experimental plants were grown in visible radiation with supplementary UV-B radiation. This increase was suppressed by increasing the irradiation of visible radiation.Fusarium oxysporum was isolated from the roots of all damping-off plants and the roots of some unwilted plants, indicating that spinach infected with the pathogen did not necessarily suffer from damping-off in 15d. Supplementary UV-B radiation suppressed the increase in growth components such as the number of leaves, the plant height and the fresh weight of aboveground plant parts, but did not affect the fresh weight of roots. The ratio of the number of plants infected with pathogen to the total number of plants was over 80% irrespective of light conditions. It was suggested that the defense response of spinach to this pathogen was greatly influenced by the physiological state of aboveground plant parts resulting from supplementary UV-B radiation.  相似文献

Phytoplankton is one of the biological quality elements included in the EU Water Framework Directive (WFD). Classification of water quality according to the WFD is based on the deviation of the present conditions from reference conditions. Given the lack of data from pristine conditions, this study used approximately 100-year-old measurements of Secchi depths from Danish waters in combination with relationships between Secchi depth and chlorophyll a (as a proxy for phytoplankton biomass) obtained from recent monitoring to calculate ‘historical’ or reference chlorophyll a (Chl-a) concentrations. Historical Secchi depth data were available for 9 out of the 11 Danish WFD intercalibration sites. At eight of the sites, reference summer (May–September) Chl-a concentrations were in the range 0.7–1.2 μg l−1. At one site, west of Bornholm in the western Baltic Sea, historical Secchi depth measurements date back to only the late 1950s corresponding to a calculated Chl-a concentration of 1.3 μg l−1. Transient genetic transformation of plant organs is an indispensable way of studying gene function in plants. This study was aimed to develop an optimized system for transient Agrobacterium-mediated transformation of the Arabidopsis leaves. The β-glucuronidase (GUS) reporter gene was employed to evaluate growth and biochemical parameters that influence the levels of transient expression. The effects of plant culture conditions, Agrobacterial genetic backgrounds, densities of Agrobacterial cell suspensions, and of several detergents were analyzed. We found that optimization of plant culture conditions is the most critical factor among the parameters analyzed. Higher levels of transient expression were observed in plants grown under short day conditions (SDs) than in plants grown under long day conditions (LDs). Furthermore, incubation of the plants under SDs at high relative humidity (85–90%) for 24 h after infiltration greatly improved the levels of transient expression. Under the optimized culture conditions, expression of the reporter gene reached the peak 3 days after infiltration and was rapidly decreased after the peak. Among the five Agrobacterial strains examined, LAB4404 produced the highest levels of expression. We also examined the effects of detergents, including Triton X-100, Tween-20, and Silwet L-77. Supplementation of the infiltration media either with 0.01% Triton X-100 or 0.01% Tween-20 improved the levels of expression by approximately 1.6-fold. Our observations indicate that transient transformation of the Arabidopsis leaves in the infiltration media supplemented with 0.01% Triton X-100 and incubation of the infiltrated plants under SDs at high relative humidity are necessary for maximal levels of expression.  相似文献

In the present study, we compared the clonal architecture between two seagrass species, the dioecious Cymodocea nodosa and the hermaphroditic, self-compatible Zostera noltii, in order to verify the hypothesis that clonal growth strategies and resulting genet architecture are associated with mating system in clonal plants. It is expected that self-incompatible species should be associated to a guerrilla growth form, because of pollen limitation due to obligate outbreeding, while the ecologically advantageous phalanx strategy can be adopted in self-compatible species. Genotypic diversity and heterozygosity were also assessed in the two species. Sampling has been conducted in mixed stands, collecting shoots of the two species at the same points of the sampling grid, in order to even out any effects of environmental heterogeneity. Species-specific microsatellite loci have been used as molecular markers to identify clones and assess their spatial distribution in both species. As expected, we found an intermingled configuration of genets in the dioecious C. nodosa while Z. noltii was characterized by a clumped, `phalanx-type' distribution of clones. C. nodosa was characterized by higher genotypic diversity with regard to Z. noltii, while heterozygosity levels were comparable in the two species. Coordinating Editor: Dr J. Tuomi  相似文献

There are very few time series documenting clear trends of change in the biomass of total phytoplankton or single taxa that coincide with trends of increasing nutrient concentrations. Weekly or biweekly monitoring since 1997 on a cross section of the central Gulf of Finland (NE Baltic Sea) with similar climatic and hydrographic conditions, but different nutrient levels, provided a uniform dataset. In order to evaluate seasonal (June–September) patterns of phytoplankton succession, more than 1,200 samples were statistically analyzed by selecting 12 dominant taxa using wet weight biomass values. In addition, the continuously measured hydrographic parameters on board the ships of opportunity, and simultaneous nutrient analyses gave high frequency information on the water masses. The objective of this study was to identify the taxa that may prove indicative in the assessment of eutrophication in the appropriate monitoring time periods. None of the most common bloom-forming species (Aphanizomenon sp., Nodularia spumigena, and Heterocapsa triquetra) showed reliable correlations with enhanced nutrient concentrations. The species we suggest as reliable eutrophication indicators—oscillatorialean cyanobacteria and the diatoms Cyclotella choctawhatcheeana and Cylindrotheca closterium—showed the best relationships with total phosphorus concentrations. Their maxima appear toward the end of July or in August–September when phytoplankton community structure is more stable, and less frequent observations may give adequate results. Another diatom, Skeletonema costatum, exhibited stronger correlations with dissolved inorganic and total nitrogen in June, during the period of the summer phytoplankton minimum. Guest editors: J. H. The results of a modelling study to investigate the mechanisms controlling macroalgal growth within the Medway estuary, UK, are presented. Intertidal zone bathymetry, tidal dynamics and turbidity control the time available for nutrient uptake and photosynthesis, and were used as a basis for predicting areas where macroalgae will grow. Tidal bed stress was also considered as a controlling factor for the presence of the less robust green macroalgae species. Two approaches to predicting macroalgal distributions were applied: (1) a simple ‘suitability index’ method based on tidal flooding and drying, taking account of the conflict between time available for nutrient uptake and for photosynthesis; and (2) a biological macroalgal growth model that includes a detailed treatment of nutrient uptake and plant growth. The former approach assigns a value between zero and one for the suitability of a location for macroalgal occurrence, while the latter predicts the full macroalgal growth dynamics over an annual cycle. Tidal bed stress was included in both approaches as an independent modifier of macroalgal occurrence/growth. Results were compared with aerial survey maps of observed vegetation cover and time series of measured biomass density. Both approaches gave good predictions of non-species-specific vegetation cover in the intertidal zone of the Medway. Tidal bed stress was found to be a strong predictor of the specific occurrence of Enteromorpha spp. and Ulva spp., with these species favouring areas of low tidal energy. It was concluded that light and a lack of suitable regions with low tidal bed stress, rather than nutrients, were the main factors limiting excessive growth of Enteromorpha spp. and Ulva spp. in the estuary. Although this study was focussed on the Medway, the results are likely to be applicable to a broad range of relatively turbid, meso- and macro-tidal estuaries. Water movement across cellular membranes is regulated largely by a family of water channel proteins called aquaporins (AQPs). Since several abiotic stresses such as, drought, salinity and freezing, manifest themselves via altering water status of plant cells and are linked by the fact that they all result in cellular dehydration, we overexpressed an AQP (tonoplast intrinsic protein) from Panax ginseng, PgTIP1, in transgenic Arabidopsis thaliana plants to test its role in plant’s response to drought, salinity and cold acclimation (induced freezing tolerance). Under favorable conditions, PgTIP1 overexpression significantly increased plant growth as determined by the biomass production, and leaf and root morphology. PgTIP1 overexpression had beneficial effect on salt-stress tolerance as indicated by superior growth status and seed germination of transgenic plants under salt stress; shoots of salt-stressed transgenic plants also accumulated greater amounts of Na+ compared to wild-type plants. Whereas PgTIP1 overexpression diminished the water-deficit tolerance of plants grown in shallow (10 cm deep) pots, the transgenic plants were significantly more tolerant to water stress when grown in 45 cm deep pots. The rationale for this contrasting response, apparently, comes from the differences in the root morphology and leaf water channel activity (speed of dehydration/rehydration) between the transgenic and wild-type plants. Plants overexpressed with PgTIP1 exhibited lower (relative to wild-type control) cold acclimation ability; however, this response was independent of cold-regulated gene expression. Our results demonstrate a significant function of PgTIP1 in growth and development of plant cells, and suggest that the water movement across tonoplast (via AQP) represents a rate-limiting factor for plant vigor under favorable growth conditions and also significantly affect responses of plant to drought, salt and cold stresses.  相似文献

The chromosome numbers of the five European seagrasses have been determined in material from several sites along the coasts of the Atlantic Ocean, the North Sea and the Mediterranean:Zostera marina L., 2n = 12;Z. noltii Hornem., 2n = 12;Posidonia oceanica (L.)Delile, 2n = 20;Cymodocea nodosa (Ucria)Aschers., 2n = 14, 2n = 28;Halophila stipulacea (Forsk.)Aschers., 2n = 18. The difference in chromosome morphology betweenZ. marina andZ. noltii supports the division of the genus into two subgenera.  相似文献
