The enzymatic activity and the biocontrol ability of two new isolates of Trichoderma spp. (T-68 and Gh-2) were compared in laboratory and glasshouse experiments with a previously studied T. harzianum strain (T-35). In dual culture tests with Fusarium oxysporum f. sp. melonis and F. oxysporum f. sp. vasinfectum, isolates T-68 and Gh-2 overgrew the colonies of Fusarium, whereas T-35 failed to parasitize both wilt pathogens. Under glasshouse conditions, the three isolates of Trichoderma were effective in controlling Fusarium wilt of cotton but only T-35 was effective against F. oxysporum f. sp. melonis on muskmelon. When the three Trichoderma isolates were grown on liquid media containing laminarin, colloidal chitin or F. oxysporum f. sp. melonis cell walls as sole carbon sources, maximum β-1,3-glucanase and chitinase specific activity in the culture filtrates of all fungi was reached after 72h of incubation. When culture filtrates of the three Trichoderma isolates were incubated with freeze-dried mycelium of F. oxysporum f. sp. melonis or F. oxysporum f. sp. vasinfectum, different concentrations of glucose and N-acetyl-D-glucosamine were released. Overall no correlation was found between enzymatic activity and the biocontrol capability against Fusarium wilt on muskmelon and cotton.  相似文献

In autoclaved greenhouse soil without Fusarium oxysporum f. sp. vasinfectum, Meloidogyne incognita did not cause leaf or vascular discoloration of 59-day-old cotton plants. Plants had root galls with as few as 50 Meloidogyne larvae per plant. Root galling was directly proportional to the initial nematode population level. Fusarium wilt symptoms occurred without nematodes with 77,000 fungus propagules or more per gram of soil. As few as 50 Meloidogyne larvae accompanying 650 fungus propagules caused Fusarium wilt. With few exceptions, leaf symptoms appeared sooner as numbers of either or both organisms increased. In soils infested with both organisms, the extent of fungal invasion and colonization was well correlated with the extent of nematode galling and other indications of the Fusarium wilt syndrome.  相似文献

Jojoba [Simmondsia chinensis (Link) Schneider] plantations in Israel originated from vegetative propagation, planted during 1991–92, have shown symptoms of wilting and subsequent death. Verticillium dahliae was only rarely isolated from these plants and artificial inoculation showed only mild disease symptoms. Fusarium oxysporum caused severe chlorosis, desiccation, defoliation and wilt in leaves of jojoba plants, resulting in plant death. Recovery of the fungus from artificially inoculated stem cuttings and seedlings showed for the first time that F. oxysporum was the primary pathogen. Inoculated cuttings exhibited wilt within 3 weeks, while in seedlings wilt occurred 10–24 weeks after inoculation. Seedlings and cuttings of jojoba which were inoculated with other Fusarium isolates originating from different crops (F. oxysporum f. sp. vasinfectum from cotton, F. oxysporum f. sp. dianthi from carnation, F. oxysporum f. sp. lycopersici from tomato and F. oxysporum f. sp. basilicum from basil) did not develop symptoms. Moreover, cotton, tomato, melon and cucumber seedlings inoculated with several virulent F. oxysporum isolates from jojoba did not show any symptoms of wilt or defoliation. These results indicate a high degree of specificity of the Fusarium isolates from jojoba; therefore, it is suggested that this isolate be defined as F. oxysporum f. sp. simmondsia.  相似文献

A field soil, artificially infested with pathogenic isolates of Fusarium oxysporum f. sp. melonis was continuously used for screening resistant varieties of melon to Fusarium wilt. After 9–10 years of continuous cropping with resistant varieties, the soil had developed induced suppressiveness. Seven to 9 experimental replantings of the induced suppressive soil with the susceptible cultivar of melon, ‘Ein-Dor', nullified its suppressiveness. This was expressed by 90 % disease incidence. Only 2 replantings were required to obtain the same disease incidence in an adjacent field of a conducive soil. Nonpathogenic isolates of F. oxysporum, isolated from the rhizospheres of melon seedlings, induced various degrees of soil suppressiveness when added to soil at various ratios to the pathogenic isolate.  相似文献

Soil samples from both healthy and diseased paprika roots were tested to identify their mycoflora. Thirty-one species belonging to 16 genera were collected from rhizosphere and rhizoplane samples. The most frequently isolated fungi were Aspergillus flavus, A. niger, A. terreus, Fusarium oxysporum, Penicillium jensenii and Trichoderma harzianum. Fusarium oxysporum was the most common Fusarium species in the rhizoplane samples of diseased roots and identification was confirmed by RAPD-PCR technique. Trichoderma harzianum, T. pseudokoningii and Glioclaium roseum were chosen to study their biological control efficiency against Fusarium oxysporum. These fungal species reduced the percentage of seedling infection to 25, 40 and 50%, respectively. With the increasing of fungicide (Folicur and Ridomil) doses the dry weight of F. oxysporum decreased. Also, the increasing of fungicide dose lead to a slight decrease in the dry weight of T. harzianum, T. pseudokoningii and Glioclaium roseum.  相似文献

The potential of the biological control fungus Penicillium oxalicum to suppress wilt caused by Fusarium oxysporum f. sp. melonis and F. oxysporum f. sp. niveum on melon and watermelon, respectively, was tested under different growth conditions. The area under disease progress curve of F. oxysporum f. sp. melonis infected melon plants was significantly reduced in growth chamber and field experiments. In glasshouse experiments, it was necessary to apply P. oxalicum and dazomet in order to reduce Fusarium wilt severity in melons caused by F. oxysporum f. sp. melonis. For watermelons, we found that P. oxalicum alone reduced the area under the disease progress curve by 58% in the growth chamber experiments and 54% in the glasshouse experiments. From these results, we suggested that P. oxalicum may be effective for the management of Fusarium wilt in melon and watermelon plants.  相似文献

The purpose of this research was to determine whetherBacillus subtilis,nonpathogenicFusarium oxysporum,and/orTrichoderma harzianum,applied alone or in combination to chickpea (Cicer arietinumL.) cultivars ‘ICCV 4’ and ‘PV 61’ differing in their levels of resistance to Fusarium wilt, could effectively suppress disease caused by the highly virulent race 5 ofFusarium oxysporumf. sp.ciceris.Seeds of both cultivars were sown in soil amended with the three microbial antagonists, alone or in combination, and 7 days later seedlings were transplanted into soil infested with the pathogen. All three antagonistic microorganisms effectively colonized the roots of both chickpea cultivars, whether alone or in combination, and significantly suppressed Fusarium wilt development. In comparison with the control, the incubation period for the disease was delayed on average about 3 days and the final disease severity index and standardized area under the disease progress curve were reduced significantly between 14 and 33% and 16 and 42%, respectively, by all three microbial antagonists. Final disease incidence only was reduced byB. subtilis(18–25%) or nonpathogenicF. oxysporum(18%). The extent of disease suppression was higher and more consistent in ‘PV 61’ than in ‘ICCV 4’ whether colonized byB. subtilis,nonpathogenicF. oxysporum,orT. harzianum.The combination ofB. subtilis+T. harzianumwas effective in suppressing Fusarium wilt development but it did not differ significantly from treatments with either of these antagonists alone. In contrast, the combination ofB. subtilis+ nonpathogenicF. oxysporumtreatment was not effective but either antagonist alone significantly reduced disease development.  相似文献

Six species of Fusarium were identified among approximately 7,000 cultures isolated from roots and soils collected from prairies and cornfields in southern Minnesota. In both soil sources, F. oxysporum and F. solani predominated, followed in order by F. roseum, F. episphaeria, F. tricinctum, and F. moniliforme. The same order was obtained for the 28 species of prairie grasses and forbs as for corn. From prairie plant and corn roots, respectively, F. oxysporum was isolated from 64 and 90%; F. solani, 26 and 80%; F. roseum, 8 and 35%; and F. tricinctum, 6 % each. In 10 of 17 companion soil pairs, populations of Fusarium species were higher in cornfield than in prairie soils. Populations of F. roseum and F. moniliforme especially, were higher in cornfield soils of sample pairs, but populations of F. solani were higher in prairie soils. Fusarium roseum ‘Equiseti’ was the predominant cultivar of this species in both prairie and cornfield soils. Thus all six species of Fusarium appeared to be indigenous to the prairie, and some, especially F. roseum, apparently had increased in prevalence in soil and roots by corn culture.  相似文献

Okra was grown in field plots of Tifton loamy sand naturally infested with the nematodes Meloidogyne incognita and Criconemoides ornalus and the pathogenic fungi Fusarium oxysporum, F. solani, F. roseum, and Pythium spp. Plots were treated with various soil pesticides and left exposed or covered with biodegradable paper film mulch under trickle irrigation. Soil was assayed for nematodes and fungi, and plant roots were examined for root-rot and insect damage. Fewer nematodes and fungi generally were recovered from soil treated with DD-MENCS (with and without film mulch) or methyl bromide-chloropicrin (2:1) (MBC) and film mulch than from nontreated soil. Funfigation with DD-MENCS or MBC suppressed populations of M. incognita, C. ornatus, F. oxysporum, F. solani, F. roseum, and Pythium spp. Ethoprop (alone or combined with other pesticides), sodium azide, and chloroneb were less effective than DD-MENCS and MBC. Plant growth anti yield were greatest when nematodes and pathogenic fungi were controlled. Yield was increased 3-fold by DD-MENCS + film mulch or MBC + film mulch in comparison with the average yield of okra produced in Georgia. The root-knot nematode-Fusarium wilt complex was most severe in nonfuntigated soil.  相似文献

Sesame (Sesamum indicum L.) is one of the most important oilseed crops in Egypt and worldwide. It is being infected with many pathogens, among these pathogens Fusarium oxysporum f.sp. sesami (Zap.) Cast is causing severe economic losses on sesame. In this study, antagonistic capability of 24 isolates of Trichoderma spp. was assessed in vitro against F. oxysporum f.sp. sesami. Two strains; T. harzianum (T9) and T. viride (T21) were revealed to have high antagonistic effect against F. oxysporum f.sp. sesami in vitro with inhibition percentage about 70 and 67%, respectively. These two isolates proved to have high ability to control Fusarium wilt disease under greenhouse conditions. The highest reduction in disease severity was achieved with T. viride followed by T. harzianum with reduction in disease severity about 77 and 74%, respectively. This study revealed that the time of application of bioagents is a decisive factor in determining the efficacy of Trichoderma isolates to control Fusarium wilt of sesame. It was revealed that the highest reduction in the disease severity was achieved when either Trichoderma viride or T. harzianum were applied 7 days before challenging with the F. oxysporum f.sp. sesami.  相似文献

Abstract

Biological and nutrient management of soil borne disease is increasingly gaining stature as a possible practical and safe approach. Inhibitory effects of fungal and bacterial antagonists were tested under in vitro conditions against the wilt pathogen of alfalfa Fusarium oxysporum f. sp. medicaginis. Trichoderma harzianum and Pseudomonas fluorescens (PI 5) were found to be effective against the alfalfa wilt pathogen. Manganese sulphate at 500 and 750 ppm inhibited the mycelial growth of F. oxysporumf. sp. medicaginis under in vitro conditions. In pot culture studies, manganese sulphate at 12.5 mg/kg reduced the wilt incidence (23.33%). Combined application of manganese sulphate 12.5 mg/kg + T. harzianum 1.25 mg/kg of soil significantly reduced the wilt incidence accompanied by improved plant growth and yield in pot culture. The mixture of manganese sulphate (25 kg/ha) + T. harzianum (2.5 kg/ha) significantly reduced the wilt incidence when applied as a basal dose in the field conditions. The average mean of disease reduction was 62.42% over control.  相似文献

尖孢镰孢菌古巴专化型(Fusarium oxysporum f.sp.cubense)是香蕉枯萎病的病原菌,该菌是一种土壤习居菌,了解香蕉根区土壤中真菌多样性及镰孢菌属(Fusarium)真菌所占比例,对如何减少土壤中的病原菌、预防香蕉枯萎病的发生有重要的指导意义。该文通过采集不同宿根年限的香蕉健康植株和枯萎病植株的根区土壤,利用高通量测序技术测定土壤样品中的真菌种群。结果表明:(1)同一宿根年限的香蕉植株中,健康植株根区土壤中所获的reads及OTUs数量均高于枯萎病植株,说明健康植株根区土壤的真菌多样性丰富于枯萎病植株。(2)除了一年生香蕉枯萎病植株以担子菌门(Basidiomycota)为主外,其他土壤样品中均以子囊菌门(Ascomycota)为主,其中的丛赤壳科最高相对丰度来自三年生健康植株的根区土壤(26.02%),其次是五年生的枯萎病植株根区土壤(15.56%)。(3)在丛赤壳科中,镰孢菌属在三年生健康植株土壤中的相对丰度最高(2.54%),在其他样品中的相对丰度在0.1%~0.65%之间;在镰孢菌属中,腐皮镰孢菌(Fusarium solani)的相对丰度(0~1.59%之间)高于尖孢镰孢菌(F.oxysporum),尖孢镰孢菌仅占很小的比例(相对丰度0~0.08%之间)。可见,在不同香蕉植株的根区土壤中,健康植株的根区土壤真菌多样性高于枯萎病植株,无论是健康植株还是枯萎病植株的根区土壤中,作为香蕉枯萎病病原菌的镰孢菌属或尖孢镰孢菌的群体均不占主导地位。  相似文献

The interaction between Fusarium oxysporum (cause of cotton wilt) and Cephalosporium maydis (cause of maize late-wilt) on cotton roots is associated with an appreciable decrease in the severity of the cotton wilt disease. Reduction in infection is more pronounced when the latter fungus precedes the former in the soil than when they are inoculated simultaneously. C. maydis exerts little or no such effect when it follows F. oxysporum in the soil. C. maydis grows on the surface of cotton roots near growing points as a root-surface inhabitant. Dark red lesions are produced but these disappear, as does the fungus, when the root becomes hardened either naturally or in response to the growth of the fungus on the surface. The presence of the fungus is associated with increased production of root laterals. Cotton plants, including those which may appear healthy, show only mild internal symptoms of Fusarium infection when grown in soil inoculated with the two fungi, suggesting that the decreased severity of wilt is largely due to increased tolerance of the plants to infection with the disease as a result of increased number of root laterals. It is also possible that cotton roots with C. maydis on their surface become less suitable for the progress of F. oxysporum. F. oxysporum produces in culture a metabolite inhibitory to C. maydis. This may partly account for the little effect that the latter fungus exerts on the severity of wilt when it follows F. oxysporum in the soil. It appears that the interaction between F. oxysporum and C. maydis does not affect the pathogenicity of the latter fungus to maize.  相似文献

Fusarium wilt is caused by F. oxysporum Schlecht end. Fr. f. sp. ciceris (FOC) is a devastating disease of chickpea in Algeria. In this study, antagonistic effects of B. subtilis MF352017 (Bs1) and Trichoderma harzianum KX523899 (T5) isolated from the rhizosphere of chickpea were investigated separately and in combination for their efficacy in controlling the disease in vivo. The efficacy of the antagonistic biocontrol agents on Fusarium wilt was evaluated based on vegetative and root growth parameters of chickpea. Seed bacterisation with B. subtilis MF352017 (Bs1) and seed treatment with T. harzianum (T5) significantly protected chickpea seedlings from FOC as compared to untreated plants. Plant protection was more pronounced in T. harzianum-treated plants than in bacterised plants. The application of both antagonists effectively suppressed 93.67% of the disease and also enhanced plant growth leading to increased plant height, root length, fresh and dry weights of shoot and root. The mixture of antagonists increased the effectiveness of B. subtilis MF352017 (Bs1) isolate on Fusarium wilt and improved chickpea growth.  相似文献

The rhizospheric soil microfungi from a native forest (undisturbed and disturbed) were studied using soil dilution plate and soil washing methods. Fungi were isolated using slightly acid and alkaline culture media. 54 taxa were isolated: 49 from undisturbed forest soil and 37 from disturbed forest soil. Acremonium sp., Aspergillus ustus, Coemansia pectinata, Doratomyces stemonitis, Fusarium solani, F. oxysporum, Gliocladium roseum, Humicola fusco-atra, Mortierella sp., Penicillium lilacinum, Trichoderma harzianum, and T. koningii, showed the highest frequency, in both, undisturbed and disturbed forests. In undisturbed soil forest the biodiversity index was 3.97 whereas in disturbed ones was 3.89.  相似文献

Fusarium wilt disease is a growing concern in cucurbit crops in the Philippines. Most often than not, farmers highly depend on commercial fungicides for control but these chemicals are very expensive and not environment-friendly. Biofumigation and green manuring using Brassica plants is a potential alternative for sustainable management of this destructive disease. A study was conducted to evaluate the efficacy of mustard var. Montevede as a biofumigant and green manure to control Fusarium wilt disease of bittergourd (Momordica charantia L.) and bottlegourd (Lagenaria siceraria (Mol.) Standl.). In vitro assay of mustard slurry resulted in 100% suppression of the mycelial growth of F. oxysporum f. sp. momordicae and F. oxysporum f. sp. lagenariae isolates after exposure to 5, 10 and 15?g of mustard slurry compared with the control. Similarly, incorporation of the macerated mustard leaves in the infested soil reduced Fusarium wilt incidence by 100% in bittergourd and bottlegourd. The effect of mustard was comparable to Bavistin® fungicide both in vitro and in vivo.  相似文献

In Egypt, sesame cultivation is subject to attack by wilt and root-rot diseases caused by Fusarium oxysporum f.sp. sesami (Zap) Cast. and Macrophomina phaseolina (Maubl) Ashby causing losses in quality and quantity of sesame seed yield. Bacillus subtilis and Trichoderma viride isolates which were isolated from sesame rhizosphere were the most effective to antagonise fungal pathogens, causing high reduction of hyphal fungal growth. Trichoderma viride was found to be mycoparasitic on Fusarium oxysporum f.sp. sesami and M. phaseolina causing morphological atternation of fungal cells and sclerotial formation. In general, Bacillus subtilis, T. viride, avirulent Fusarium oxysporum isolate and Glomus spp. (Amycorrhizae) significantly reduced wilt and root-rot incidence of sesame plants at artificially infested potted soil by each one or two pathogens. Data obtained indicate that Glomus spp significantly reduced wilt and disease severity development on sesame plants followed by T. viride. Meanwhile, avirulent Fusarium oxysporum isolate followed by Glomus spp. were effective against root-rot disease incidence caused by M. phaseolina. Glomus spp. followed by B. subtilis significantly reduced wilt and root-rot disease of sesame plants. All biotic agents significantly reduced F. oxysporum f.sp. sesami and M. phaseolina counts in sesame rhizosphere at the lowest level. Glomus spp. and the avirulent isolate of F. oxysporum eliminated M. phaseolina in sesame rhizosphere. Meanwhile T. viride was the best agent at reducing F. oxysporum at a lower level than other treatments. Application of VA mycorrhizae (Glomus spp.) in fields naturally infested by pathogens significantly reduced wilt and root-rot incidence and it significantly colonised sesame root systems and rhizospheres compared to untreated sesame transplantings.  相似文献

During years 2001, 2002 and 2003 the gravity of the Fusarium wilt in 1000 hectares of melon culture was evaluated in Colima (Mexico). In spite of the soil disinfections with methyl bromide, the losses could reach 25% of the final production. The analysis of 4 soil samples from the fields with ill plants, in a selective medium for Fusarium, allowed to detect the presence of F. oxysporum. By means of the presented technique “soil phytopathometry”, 31 isolates of F. oxysporum f. sp. melonis were obtained from the soil samples. The isolates were inoculated on melon plants to evaluate their pathogenicity. The 31 isolates inoculated, produced the symptoms of chlorosis and wilting, in melon cultivars that allowed us to affirm that all isolates were race 1 of F. oxysporum f. sp. melonis. Being this the first news of the presence of F. oxysporum f. sp. melonis in the state of Colima (Mexico).  相似文献

Summary Thirty two bacteria antagonistic to a number of phytopathogenic fungi were isolated from soil samples. One bacterial strain, designated as M 51, appeared to be particularly active towardsF. oxysporum f. sp.dianthii, in vitro andin vivo and it was inhibitoryin vitro to three otherFusarium spp. used. Tests to find if there was protection against fusarium wilt were carried out by three different methods of inoculation of the cuttings: a) dipping of cuttings for ten minutes in bacterial suspension; b) spraying of suspension on perlite where the rooted cuttings were planted; c) spraying the greenhouse bench rooting boxes, where the non-rooted cuttings were planted, with bacterial suspension. Following this all the cuttings were transplanted into soil naturally highly infested withFusarium oxysporum f. sp.dianthii (3000 units/g). Good protection against fusarium wilt was obtained for cuttings inoculated by method (b). However protection decreased gradually about 60 days after they were transplanted; both control and inoculated cuttings showed a comparable mortality rate. Method of inoculation and the development of the protective effect are discussed.  相似文献

Soil fusaria from various areas in and around Surrey were estimated on medium selective forFusarium. 4 species ofFusarium, F. episphaeria. F. oxysporum, F. roseum andF. solani were detected.F. roseum was the most widely distributed species and present in highest numbers. Among 3 cultivars of this species obtained, non-pathogenic clones of “Culmorum” was always present. The other 3 species ofFusarium were less widely distributed and occurred in lower numbers, withF. solani being the most restricted in distribution and present in very small populations. NoFusarium was recorded for soils from natural field vegetation (except grass areas) and forests.  相似文献
