Skotodormant seeds of Lactuca sativa Grand Rapids imbibed in darkness for 10 days (10-day DS) germinated poorly upon terminal treatment with red light (R) or gibberellin A3 (GA3). Soluble sugars in the imbibition solutions influenced the depth of skotodormancy. Ten-day DS seeds, imbibed in 50–500 mm sucrose or 100–500 mm glucose and given terminal GA3 germinated completely and germinated about 80% when imbibed in 100 mm galactose, mannose, lactose, or maltose. In contrast, terminal R applied to 10-day DS seeds caused only 20–50% germination. If given R at day 0 and imbibed for 10 days in darkness in 500 mm sucrose or glucose, seeds washed free of exogenous glucose or sucrose then germinated about 50% in darkness in water. These seeds responded to terminal R or GA3 with complete germination. When seeds were given FR at day 0, germination responses following terminal R or GA3 were significantly lower when the duration of DS was increased from 7–10 day DS to 15 days. In 10-day DS seeds given initial FR and imbibed in either solutions of 50 or 100 mm sucrose and KNO3, either terminal R or GA3 treatment gave complete or near complete germination. It is concluded that seed exposure to certain soluble sugars and/or nitrate during a 10-day DS protected certain substrates and thereby extended the sensitivity of the seeds to terminal R or GA3 treatment. The study provides substantial evidence for nonhormonal factors associated with light and GA action in the control of seed skotodormancy. Received October 30, 1996; accepted April 22, 1997  相似文献

Induction of inhibited growth halo, around filter paper discs or groundnut seeds dusted with aldrex T, diameter of which consistently increased with dressing application rate, was exhibited by the root nodule bacteriumRhizobium leguminosarum. Some cells sensitive to aldrex T gained recovery upon transference to fresh media denoting a bacteriostatic effect of the anti-fungal dressing.The viable bacterial cells steadily declined with fungicide concentrations between 100 and 500 p.p.m. An increase of 130% in viable colony count was recorded when the dose was increased to 700 p.p.m., and a relatively similar increase at 900 p.p.m., thus exhibiting a zone of inverted growth at 500 p.p.m. which was also confirmed by the optical density test. The latter was reliable after 24h incubation when the turbidity uniformly deminished with the fungicide dose to a maximal decline at 500 p.p.m. Aldrex T decreased the log and increased the stationary phases ofR. leguminosarum growth at all test doses.In vitro treatment ofR. leguminosarum with the fungicide exhibited some changes in bacterial morphology, cell lysis and shrinkage of cell membrane as revealed by the electron microscope.  相似文献

Seeds ofBromus sterilis L. germinated between 80–100% in darkness at 15° C but were inhibited by exposure to white or red light for 8 h per day. Exposure to far-red light resulted in germination similar to, or less than, that of seeds maintained in darkness. Germination is not permanently inhibited by light as seeds attain maximal germination when transferred back to darkness. Germination can be markedly delayed by exposure to a single pulse of red light following 4 h inhibition in darkness. The effect of the red light can be reversed by a single pulse of far-red light indicating that the photoreversible pigment phytochrome is involved in the response. Aims: To assess the effectiveness of sequential treatments of radish seeds with aqueous chlorine dioxide (ClO2) and dry heat in reducing the number of Escherichia coli O157:H7. Methods and Results: Radish seeds containing E. coli O157:H7 at 5·5 log CFU g?1 were treated with 500 μg ml?1 ClO2 for 5 min and subsequently heated at 60°C and 23% relative humidity for up to 48 h. Escherichia coli O157:H7 decreased by more than 4·8 log CFU g?1 after 12 h dry‐heat treatment. The pathogen was inactivated after 48 h dry‐heat treatment, but the germination rate of treated seeds was substantially reduced from 91·2 ± 5·0% to 68·7 ± 12·3%. Conclusions: Escherichia coli O157:H7 on radish seeds can be effectively reduced by sequential treatments with ClO2 and dry heat. To eliminate E. coli O157:H7 on radish seeds without decreasing the germination rate, partial drying of seeds at ambient temperature before dry‐heat treatment should be investigated, and conditions for drying and dry‐heat treatment should be optimized. Significance and Impact of the study: This study showed that sequential treatment with ClO2 and dry‐heat was effective in inactivating large numbers of E. coli O157:H7 on radish seeds. These findings will be useful when developing sanitizing strategies for seeds without compromising germination rates.  相似文献

ABSTRACT

An infusion of Ruta graveolens L. was tested for its inhibitory effects upon radish germination at the cyto-physiological level. Radish seeds were placed under optimum conditions for germination either in water (control) or in the presence of rue infusion (treated seeds). Morpho-physiological observations indicate that in treated radish seeds the inhibition of germination is accompanied by reduced water uptake and delayed reactivation of the outermost living layer, i.e. the aleurone cells. Compared to the control, aleurone cells of treated seeds present many large lipid droplets and protein bodies, without differentiated organelles. Moreover, chemical and biochemical analyses show that the treatment impairs the metabolic pathways of germination, such as the mobilization and utilization of seed reserves, and the loosening of cell walls. In fact, in treated seeds we found i) increased contents of glucose and galactose, ii) higher concentration of malic acid and iii) lower activities of some glycosidases compared to the control. Results suggest that aleurone cells may play an active part in controlling the embryo's metabolism.  相似文献

Summary The effect of culture filtrates of some common rhizosphere fungi on the seed germination ofDichanthium annulatum, Bothriochloa pertusa andSetaria glauca was studied. Surface sterilized seeds were soaked in the fungal culture filtrates for 24 hours and after several washings in distilled water transferred to moist filter paper in Petri dishes and the percentage germination recorded. No significant inhibition or stimulation of germination was noticed in any of the treatments. The only exception wasTrichoderma viride which inhibited the germination ofD. annulatum seeds to some extent.  相似文献

The effect of iprodione on the seed-borne phase of Alternaria brassicicola   总被引:2,自引:0,他引:2  
Alternaria brassicicola infection of Brassica oleracea seeds was effectively controlled by a dust application of iprodione (Rovral 50% w.P.). At 2.5 g a.i./kg the seed-borne fungus was usually eliminated from samples with up to 61.5% affected seeds (35.5% internally diseased) but higher levels of infection required increased doses for complete eradication of the fungus. The germination of healthy seeds, including samples from 7–yr-old stocks, on filter paper was unaffected by the treatment. However, the germination of diseased samples, particularly those internally infected with A. brassicicola, was improved. More seedlings emerged from iprodione treated than from untreated seeds in glasshouse soil but the differences were not significant. The application of gamma-hexachlorocyclohexane to iprodione treated seeds sown in soil did not adversely affect subsequent emergence or disease control. Disease control was maintained and germination was not affected by the treatment when treated infected seeds were stored for 2 yr at 10 °C, 50% r.h. In a field trial iprodione seed treatment reduced seedling infection in a cabbage crop grown from naturally diseased seeds (100% contaminated, 45.5% internally infected) from 5.6 to 0.04%.  相似文献

The effects of fusicoccin on the germination of dormant, light-requiring or abscisic acid-inhibited seeds has been investigated. (1) Fusicoccin (10?6M) induces germination in dormant wheat seeds (Triticum durum cv. Cappelli; 1972 crop) and stimulates it in seeds already relieved from dormancy (1971 crop), with an effect similar to that of gibberellic acid. (2) Fusicoccin (1.5 × 10?6M) is more active than the two phytohormones gibberellic acid and benzyladenine and than white light in stimulating light-requiring lettuce seeds (Lactuca sativa cv. Grand Rapids) to germinate. Germination of radish seeds (Raphanus sativus) is also accelerated by fusicoccin, while benzyladenine and gibberellic acid are less active in this material. (3) Fusicoccin (1.5 × 10?5M) removes almost completely the inhibitory effect of abscisic acid on germination of radish and lettuce seeds, whereas benzyladenine (10?4M) and gibberellic acid (3 × 10?4M) remove the inhibition only partially. The possible relationship between these results and previous information on growth by cell enlargement is discussed in terms of the mechanism of action of fusicoccin as compared with natural hormones.  相似文献

Seed dispersal by animals is one of the most important plant-animal mutualisms, but saurochory, the dispersal of seeds by reptiles, has received little attention. We investigated the role of the Florida box turtle (Terrapene carolina bauri) as a seed dispersal agent in pine rockland forests of the lower Florida Keys and examined the effect of turtle digestion on seed germination. We obtained seeds of 11 species with fleshy fruits and 2 species with non-fleshy fruits (a grass and legume) from the feces of 145 box turtles collected on Key Deer National Wildlife Refuge from 1999 to 2000. We planted the seeds of nine species and germination percentage (percentage of seeds that germinated during the experiment) varied from 10% to 80%. Comparative germination experiments were conducted with Thrinax morrissii, Serenoa repens, and Byrsonima lucida. We compared the germination percentage and germination rate (number of days from planting to seedling emergence) of seeds from three treatments (seeds recovered from feces, control seeds with pulp, and control seeds without pulp) and continued these experiments for up to 2 years. Passage through the box turtle digestive tract greatly enhanced the germination percentage and germination rate of S. repens, but decreased the germination percentage of B. lucida and T. morrissii, and decreased germination rate for T. morrissii. Subsequent destructive seed viability tests revealed that many ungerminated T. morrissii seeds remained viable, suggesting long-term seed dormancy may occur, even after passage through the turtle digestive system. In addition, the proportion of ungerminated seeds which remained viable was greater for seeds recovered from turtle feces than from control seeds with pulp. Furthermore, removal of fleshy pulp either manually or by the turtle digestive system may allow T. morrissii to escape insect predation.  相似文献

Abstract Calmodulin (Cam), the heat-stable, ubiquitous, Ca2+-dependent regulator protein, has been purified to apparent homogeneity from germinating radish seeds (Raphanus sativus). The characteristics of radish Cam-molecular weight, absorption spectrum, Ca2+-dependent activation of brain phosphodiesterase (PDE)-are very similar to those described for Cam from other plant materials. Radish Cam, like other plant Cam, shows some differences to Cam of calf brain. The total amount of Cam in radish embryos at 24 h of germination is ca. 37 μg g−1 fresh weight. Approximately 95% of the total amount of Cam is present in the soluble fraction (supernatant at 100,000 g). The level in the embryo axis strongly increases in the first 24 h of germination (+540%); this increase is strongly reduced when the germination is inhibited by abscisic acid (ABA). In the presence of Ca2+, no ‘free’ Cam (i.e. not bound to other structures) is present in the soluble fraction, suggesting that, during early germination, Cam level is a limiting factor for the activities of Ca2+ -Cam-dependent systems. These studies suggest that Cam plays an important role in the early phases of seed germination. An inhibitor of the Ca2+-Cam-dependent phosphodiesterase is present in the soluble fraction from radish embryos; this substance decreases during germination. A possible role of this inhibitor during the early germination phases is hypothesized.  相似文献

Both dark and red light germination of lettuce seeds (cv. “Maikönig”) as well as their root and hypocotol elongation were inhibited when the seeds were sown in petri dishes together with a few seeds of Heracleum laciniatum Horn. This inhibition was not significantly counteracted by the presence of gibberellic acid (GA3) or/and 6-benzylaminopurine (BA). However, a large proportion of the lettuce seeds germinated abnormally (only cotyledons emerged) when treated with BA in the presence of Heracleum seeds. GA3 had alone no significant effect on abnormal germination, but it counteracted the effect of BA to some extent. The inhibitory effect of Heracleum seeds gradually disappeared during a moist incubation period of one to seven days in darkness at 25°C. When lettuce seeds were pre-incubated together with Heracleum seeds for one to five days the remaining, non-germinated lettuce seeds had lost their ability for subsequent germination in darkness in distilled water. This induced dark dormancy was to a great extent broken by red light, but not by GA3 or/and BA. H. laciniatum seeds inhibited the germination of Salix pentandra seeds and to some extent also the germination of radish but had no effect on the germination of spruce.  相似文献

We developed and validated a treatment to inactivate Escherichia coli O157:H7 on radish seeds without decreasing seed viability. Treatments with aqueous ClO(2) followed by drying and dry-heat treatments were evaluated for efficacy to inactivate the pathogen. Conditions to dry radish seeds after treatment with water (control) or ClO(2) were established. When treated seeds with high water activity (a(w)) (>0.99) were stored at 45°C and 23% relative humidity (RH), the a(w) decreased to <0.30 within 24 h. Drying high-a(w) seeds before exposing them to dry-heat treatment (≥60°C) was essential to preserve seed viability. The germination rate of radish seeds which had been immersed in water for 5 min, dried at 45°C and 23% RH for 24 h, and heated at 70°C for 48 h or at 80°C for 24 h was not significantly decreased (P ≤ 0.05) compared to that of untreated radish seeds. Sequential treatments with ClO(2) (500 μg/ml, 5 min), drying (45°C, 23% RH, 24 h), and dry heating (70°C, 23% RH, 48 h) eliminated E. coli O157:H7 (5.9 log CFU/g) on radish seeds and, consequently, sprouts produced from them without decreasing the germination rate. These sequential treatments are recommended for application to radish seeds intended for sprout production.  相似文献

Alcorn , Stanley M. (U. S. Dept. of Agric., Tucson, Ariz.), and Edwin B. Kurtz , Jr . Some factors affecting the germination of seed of the saguaro cactus (Carnegiea gigantea). Amer. Jour. Bot. 46(7): 526–529. 1959.—Germination of saguaro cactus seeds is stimulated by red light (approx. 6550 A) or daylight and far-red light (approx. 7350 A) counteracts this effect. About 0.1% germinate in continuous darkness. A single exposure to red light was most effective when the seeds were imbibed 24 hr., but maximum germination resulted from multiple exposures to red light during a 72-hr. imbibition period. The optimum temperature for germination was 25°C.; no germination occurred at 15°C. and only slight germination at 35°C. Imbibition of light-treated seeds in 0.05 to 0.2% KNO3 increased germination. Germination of seeds in either light or dark was increased by imbibing the seeds in 500 to 1000 p.p.m. gibberellic acid.  相似文献

Summary We have developed an improved artificial seed system by using a hot-water extract from a marine cyanobacterium, Synechococcus sp. NKBG 042902. Carrot somatic embryos (Daucus carota L.) were divided into two size categories (> 800 m and 425–800 m). High frequency germination (91%) was obtained using the large somatic embryos encapsulated in calcium alginate gel containing 400 mg 1–1 of extract. This compares to 35% without addition of the extract. A non-dialysate fraction of the extract showed strong germination-promoting activity compared with a dialysate fraction. The germination frequency of artificial seeds containing 100 mg 1–1 of non-dialysate fraction was more than 90%. Almost all germinating artificial seeds developed into plantlets within 4 days. We also achieved high frequency germination (60%) of artificial seeds encapsulating small somatic embryos (425–800 m) that contained 100 mg 1–1 of non-dialysate (control 9%). Although the small somatic embryos showed a lower germination frequency than the large embryos, the plantlet development process in these seeds was far more vigorous. Such a high germination frequency has not previously been reported for a carrot artificial seed system.  相似文献

以大白菜、萝卜、番茄和黄瓜种子为受体,采用实验室培养皿种子发芽生物测试法研究了黄瓜种子浸提液、种子萌发、胚根和芽苗分泌物、芽苗腐解物和芽苗浸提液的化感效应。结果表明:(1)黄瓜种子浸提液对大白菜、萝卜、番茄和黄瓜种子萌发均有化感抑制作用,即黄瓜种子内含有某些化感抑制物质。(2)在水浸提过的黄瓜种子萌发过程中,它不仅对其近邻套种的大白菜、萝卜和番茄种子萌发产生化感抑制作用,而且其胚根和芽苗分泌物对后茬播种的4种蔬菜种子发芽也表现出不同程度的化感抑制作用;黄瓜芽苗腐解物和芽苗水浸提液也对各受体蔬菜种子发芽与生长产生不同程度的化感抑制作用,且随着腐解芽苗量的增加或浸提液浓度的升高,各受体蔬菜种子的发芽指标值、化感效应指数值和综合效应值随之降低。(3)黄瓜种子浸提液及芽苗各器官的化感物质对黄瓜种子的萌发与生长产生了自毒作用,且黄瓜芽苗腐解物、芽苗浸提液、胚根及芽苗分泌物对受体黄瓜的自毒作用均为最大。研究发现,黄瓜种子浸提液、种子萌发时期以及芽苗各器官的化感物质主要是通过抑制受体胚根的生长而起化感抑制作用,即受体蔬菜种子胚根对化感效应最为敏感;因黄瓜种子及萌发期释放化感物质的途径有所不同,导致受体大白菜、萝卜、黄瓜和番茄的化感响应也不相同;在黄瓜种子萌发和芽苗生长的早期,化感物质即开始在芽苗体内进行合成与积累,一部分可通过胚根和芽苗分泌途径释放到环境中,另一部分可通过芽苗腐解途径释放化感物质,并对受体蔬菜种子萌发与生长表现出较强的化感抑制作用。  相似文献

Analysis of the rate-limiting dehydrogenases of the oxidative pentosephosphate pathway (OPPP), as well as changes in the contributions of14C1- and 14C6-labelled glucose torespired CO2 during germination, indicated activation of the OPPPduring Arabidopsis seed germination. An approximatelyfour-fold increase in free proline (Pro) was also observed prior to radicleemergence in most seeds. Delayed radicle emergence in transgenicArabidopsis seeds which expressed an antisense copy of thegene encoding the Pro biosynthetic enzyme1-pyrroline-5-carboxylate synthetase correlated with anapproximately 35% reduction in the maximal concentration of Pro accumulated bythe antisense lines during germination. A dose-dependent inhibition ofArabidopsis seed germination by millimolar concentrationsof Pro capable of feedback inhibition of Pro synthesis reinforced the role ofPro synthesis in promoting germination. The ability of the artificial oxidantsmethylene blue and phenazine ethosulphate to overcome the inhibitory effects ofPro suggests a functional link between elevated Pro synthesis and increasedOPPPactivity and the importance of coupling of both pathways in stimulatinggermination.  相似文献

A new method for germination bioassays in allelopathy was evaluated. Inspired by pharmacology, allelochemical quantity to test on different target seeds was calculated according to seeds biometry. The first step consisted in measuring volume, mass, surface and shape of Lactuca sativa and Raphanus sativus seeds. The radish seed mass, surface, volume and contact area biometric parameters were respectively 10, 2, 6 and 1.7 that of lettuce. Two germination bioassay sets were compared: (i) a conventional one, testing the same concentration of allelochemical (2-benzoxazolinone termed as “BOA”) on the two species and, (ii) a biometrics enhanced seed test (“BEST” method), employing quantities of the chemical that were calculated in proportion to seed biometry parameters. The conventional method indicated that 1 mM BOA slowed and decreased germination rate (radish 50 %, lettuce 10 %) whereas 10 and 0.1 μM did not induce any effect. The BEST method offered more differentiated results: applied BOA according to seed volume induced significant inhibition of radish germination (both quantities), while lettuce was only affected by the highest dose. The same occurred when considering grain surface and contact area. Thus, the BEST assay showed more clearly that radish was more sensitive to BOA than lettuce. Compared to a setup with identical test compound concentrations for all tested species, the BEST method provides more differentiated results contributing to a more realistic comparative susceptibility assessment.  相似文献

The effects of three salinities (0, 100 and 500 mM NaCl) and four constant temperatures (10, 20, 30 and 35 °C) on seed germination of Halostachys caspica (M. B.) C. A. Mey., Kalidium foliatum (Pall.) Mop. and Halocnemum strobilaceum (Pall.) Bieb. were investigated. After seeds were treated with different concentrations of NaCl at constant temperatures of 10–35 °C for 16 days, ungerminated seeds were transferred to distilled water for 10 days to investigate the total germination; after this time, the ungerminated seeds from the 10 and 20 °C treatments were then moved to 35 °C for another 5 days to determine the final germination. The three plant species in the present experiment are salt-resistant euhalophytes growing in high saline soils in the Zhungur Basin in Xinjiang, a northwest province of China.Compared with germination under control conditions, germination percentages of all three species were not affected by 100 mM NaCl at 10–35 °C, while severely inhibited by 500 mM NaCl; germination percentages were very low at 10 °C up to 100 mM NaCl for all species; the optimum temperature for germination of H. caspica and K. foliatum was 20–30 °C, while 35 °C for H. strobilaceum, up to 100 mM NaCl; seeds did not suffer ion toxicity for all species, as evidenced by the high total germination after ungerminated seeds pretreated with 500 mM NaCl were transferred to distilled water at constant temperatures of 10–35 °C for 10 days, and the high final germination after the ungerminated seeds from the 10 and 20 °C treatments were subsequently moved to 35 °C for another 5 days; Halostachys caspica had greater sensitivity to increasing temperatures from 10 and 20 °C to 35 °C compared with the other two species.  相似文献

Summary The lengthy dormancy requirement of yew seeds can be overcome with a simple in vitro method. Viable embryos were excised from seeds ofTaxus brevifolia and four cultivars ofT. media over a range of developmental stages. Embryos were cultured in several basal media formulations (Whites’, Gamborg’s B5 and Murashige and Skoog’s) under dark or light. After a lag period of 1 to 2 wk, embryos of both species germinated precociously. Germination rates of up to 70% were obtained withT. media cv. Hicksi embryos. The highest rates of germination were obtained in White’s and MS media. Embryos excised from green seeds with undeveloped arils showed the highest germination rates. As the seeds approached maturity, in vitro germination rates of the excised embryos declined dramatically. Green seeds and seeds with developing arils could be stored at 5° C without large loss in embryo germination. Seeds with fully developed arils could be stored frozen at −20° C for 1 wk while still allowing about 50% of embryo germination. At least 30% of the precociously germinated embryos of both species were able to develop into full seedlings. Klebsiella was found to multiply and colonize growing radish root bulb surfaces following the inoculation of seeds with 101–104 cells. All 29 cultures ofKlebsiella originally isolated from 5 different sources were capable of growth to 106–107 colony-forming units/g of root bulb within 1 week after seed germination. Linear regression analysis illustrated differences inKlebsiella survival rates over 4 weeks of radish plant development. Analysis of covariance showed the survival ability wasKlebsiella from vegetables>mastitis>human, water, and pulp mill isolates. It was also shown thatKlebsiella species 2 had a significantly higher survival rate than the otherKlebsiella species. This finding correlates well with the observation thatKlebsiella species 2 is the most commonKlebsiella species isolated from vegetables. Average densities for allKlebsiella groups at plant harvest (5 weeks) ranged from 103–105 colony-forming units/g of radish plant. The possible health significance of these densities ofKlebsiella on vegetables consumed raw by humans is discussed.  相似文献
