Experimental induction of plant chemical defences with methyl jasmonate (MeJa) is a valuable tool for understanding the ecology of plant defensive responses. However, few studies have examined whether MeJa-induced defences in conifers are effective against insect herbivores. We studied, in 17 half-sib Pinus pinaster families, (i) the effect of MeJa application on plant growth and on the induction of diterpenoid resin in different sections of the stem; (ii) whether MeJa-induced defences increase the resistance of living pine juveniles against the large pine weevil Hylobius abietis in an in vivo bioassay and (iii) the induction of resin content after weevil wounding. Resin concentration was greater in the upper section of the stem compared with basal sections in both MeJa-induced and non-induced seedlings. Sixty days after MeJa application, treated plants showed 40% greater resin content all along the stem, but reduced height growth compared to control plants. MeJa-induction was effective against the pine weevil, as induced seedlings were 21% less damaged than control plants. Wounding activity by H. abietis produced a strong local defensive response after 48 h, where resin concentration was double that observed in the basal and apical sections not exposed to the insects.  相似文献

In this study we tested whether Pityogenes chalcographus L. beetles preferably infest their host tree, Picea abies (L.) Karst., through lenticels or not. In a second step the resin canal system, which is important for defence against bark beetles, was investigated under lenticels and under lenticel-free periderm. Beetle colonisation tests on breeding logs resulted in 93.2% successful invasions through lenticels, 4.2% through lenticel-free periderm, 1.2% through wounds, and 1.4% near branches (n=1,606). Three main reasons why beetles invade preferable sites are discussed, taking the function of lenticels to supply gas exchange of the interior tissues into consideration. First, attractive volatiles are more easily detectable over the lenticels, and this could be a stimulus for the bark beetles to invade their hosts through these structures. Secondly, the very loosely arranged tissues of the lenticel and bigger intercellular spaces in the cortex and secondary phloem under the lenticel permit the beetles easier movement towards the vascular cambium. Thirdly, the total resin canal area of both primary and secondary canals and the number of secondary resin canals were reduced under lenticels. A higher resin canal density beneath lenticels would constitute a barrier to gas movement.  相似文献

The aim of this study was to analyse the regeneration of Pinus pinaster after wildfire and the possible inter and intraspecific competition during the first 3 years after fire. The study area is located in a P. pinaster stand in León province (NW Spain). Three study sites (S1, S2 and S3) were established in an area burned in 1998. In each site, three permanent plots (20 × 1 m) were marked. A total of 20 quadrats of 1 m 2 were studied in each plot. The number and height of pine seedlings 1, 2 and 3 years after fire was recorded in each quadrat. The regeneration of understorey vegetation in the quadrats was analysed concurrently. The significance of linear correlations among the number and height of seedlings and understorey vegetation cover was tested by calculating Pearson correlation coefficients.Seed germination and seedling emergence took place massively during the first year after the fire and decreased through time. The height growth was constant over the 3 years at site S2, while a growth burst could be observed between years 2 and 3 at sites S1 and S2. Also, pines from site S2 reached shorter maximum heights in all years compared to pines from site S1 and S3. The understorey vegetation showed minimal regeneration during the first year but then increased greatly with time. Woody understorey cover and total vegetation cover were negatively correlated with pine seedling density in sites with a high number of seedlings (e.g. S1 and S3). When woody cover, total cover and pine seedling density were low (e.g. S2), there were no correlations. There was a positive correlation between vegetation cover and the maximum height of Pinus seedlings in all study sites.  相似文献

Rice gene Oryza sativa Drought Stress Response-1 (OsDSR-1) was one of the genes identified to be responsive to drought stress in the panicle of rice at booting and heading stages by both microarray and quantitative real-time PCR analyses. OsDSR-1 encodes a putative calcium-binding protein, and its overexpression in Arabidopsis rendered transgenic plants to produce much shorter lateral roots (LRs) than wild-type (WT) plants in the medium supplemented with abscisic acid (ABA), suggesting that OsDSR-1 may act as a positive regulator during the process of ABA inhibition of LR development. No significant difference was observed in the total LR length between WT and transgenic plants in the media with the increase of only osmotic stress caused by NaCl, LiCl, and mannitol, while transgenic Arabidopsis seedlings appeared to produce larger root systems with longer total LR lengths under high-potassium conditions than WT seedlings. Further analysis showed that external Ca2+ was required for the production of larger root systems, indicating that the promotion by OsDSR-1 of the LR development of transgenic Arabidopsis seemed to occur in a Ca2+-dependent manner under high-potassium conditions. We propose that OsDSR-1 may function as a calcium sensor of the signal transduction pathway controlling the LR development under high-potassium conditions.  相似文献

The inability to induce rooting of in vitro-established Protea cynaroides microshoots has prevented the production of complete plantlets. A successful shoot-tip micrografting technique was developed using in vitro-germinated P. cynaroides seedlings as rootstocks and axenic microshoots established from pot plants as microscions. Thirty-day old seedlings, germinated on growth-regulator-free, half-strength Murashige and Skoog medium, were decapitated and a vertical incision made from the top end. The bottom ends of microshoots established on modified Murashige and Skoog medium were cut into a wedge (‘V’) shape, and placed into the incision. The micrografted explants were cultured in a growth chamber with the temperature adjusted to 25 ± 2°C, with a 12-h photoperiod. Best results were obtained by placing the microscions directly onto the rootstock without any pre-treatments. Dipping the explants in anti-oxidant solution or placing a layer of medium around the graft area led to the blackening of the microscion.  相似文献

Little is known about the molecular processes involved in the phloem response to aphid feeding. We investigated molecular responses to aphid feeding on celery (Apium graveolenscv. Dulce) plants infested with the aphid Myzus persicae, as a means of identifying changes in phloem function. We used celery as our model species as it is easy to separate the phloem from the surrounding tissues in the petioles of mature leaves of this species. We generated a total of 1187 expressed sequence tags (ESTs), corresponding to 891 non-redundant genes. We analysed these ESTs in silico after cDNA macroarray hybridisation. Aphid feeding led to significant increase in RNA accumulation for 126 different genes. Different patterns of deregulation were observed, including transitory or stable induction 3 or 7days after infestation. The genes affected belonged to various functional categories and were induced systemically in the phloem after infestation. In particular, genes involved in cell wall modification, water transport, vitamin biosynthesis, photosynthesis, carbon assimilation and nitrogen and carbon mobilisation were up-regulated in the phloem. Further analysis of the response in the phloem or xylem suggested that a component of the response was developed more specifically in the phloem. However, this component was different from the stress responses in the phloem driven by pathogen infection. Our results indicate that the phloem is actively involved in multiple adjustments, recruiting metabolic pathways and in structural changes far from aphid feeding sites. A transfer DNA (T-DNA) carrying the marker gene nptII was detected in the genomes of diploid and haploid maize plants obtained after the treatment of pistil filaments with a suspension of Agrobacterium during artificial pollination. PCR analysis of total DNA isolated from 155 canamycin-resistant diploid F1 seedlings revealed T-DNA insertions in the genomes of 111 plants (32.7% of the total number of analyzed seeds). The example of matroclinal haploids was used to demonstrate that T-DNA may be transported to the egg cell by the growing pollen tube (PT). Twelve out of 16 analyzed haploid plants contained the T-DNA insertion. The possible mechanism of the transfer of the Agrobacterium T-DNA to the maize genome during pollination is discussed.  相似文献

Miconia calvescens (Melastomataceae) is an invasive alien tree in native forests on some Pacific islands and a potentially invasive species in Australia. Searches for potential classical biocontrol agents have been undertaken for over a decade in the centre of origin (Central and South America). Salbia lotanalis (Lepidoptera: Pyralidae) is a leaf roller which has been recognized as a promising classical biocontrol agent for M. calvescens. This paper presents the biology and an impact study of S. lotanalis on M. calvescens. Life table parameters showed that S. lotanalis has a high reproductive capacity, with up to six generations a year. Miconia calvescens seedlings attacked by S. lotanalis caterpillars had their growth rate significantly reduced. Seedlings subjected initially to a high level of defoliation (80%) caused by caterpillars had leaf fall and a lower leaf area than controls after 210 days. Considering the high population growth rate and significant impact on young plants, S. lotanalis appears to have a high potential for use as a classical biological control agent to be used against M. calvescens.  相似文献

Mature seed-derived embryogenic calli of indica rice (Oryza sativa L. cv. PAU201) were induced on semisolid Murashige and Skoog medium supplemented with 2.5 mg dm−3 2,4-dichlorophenoxyacetic acid + 0.5 mg dm−3 kinetin + 560 mg dm−3 proline + 30 g dm−3 sucrose + 8 g dm−3 agar. Using OsglyII gene, out of 3180 calli bombarded, 32 plants were regenerated on medium containing hygromycin (30 mg dm−3). Histochemical GUS assay of the hygromycin selected calli revealed GUS expression in 50 % calli. Among the regenerants, 46.87 % were GUS positive. PCR analysis confirmed the presence of the transgene of 1 kb in 60 % of independent plants. Further, these plants have been grown to maturity in glasshouse. In vitro screening for salt tolerance showed increase in fresh mass of OsglyII putative transgenic calli (185.4 mg) as compared to control calli (84.2 mg) on 90 mM NaCl after 15 d. When exposed to 150 mM NaCl, OsglyII putative transgenic plantlets showed normal growth while the non-transgenic control plantlets turned yellow and finally did not survive.  相似文献

We present data on the habitat, density, and spatial distribution of Rivulus giarettai, and discuss some biotic and abiotic variables related to its abundance in Free Flowing Waters (FFW) and Dam Reservoirs (DR) in palm grove (Mauritia flexuosa) marshes (Veredas) in Uberlandia, Minas Gerais, southeastern Brazil. The mean density (individuals/plot) of R. giarettai was about 13 times higher in FFW than in DR. In FFW, the density of R. giarettai was highest at intermediate amounts of substrate (plant mass) and it was positively rank-correlated with the depth, and the number of arthropods. Individuals occurred in an aggregated distribution. The aggregated pattern could be related to a concentration of individuals in microhabitats neither too much exposed nor completely saturated by plants. R. giarettai was relatively abundant and tolerant to slight man-made habitat modifications. Damming appeared to be especially problematic by negatively affecting its density.  相似文献

Survey of seven strains determined as Septonema ochraceum (Dothideomycetes, inc. sed.) isolated from pine litter or obtained from public collections revealed three new species, Fusicladium cordae, F. sicilianum (Venturiaceae), Cladophialophora matsushimae (Herpotrichiellaceae) and a cryptic species morphologically identical to Devriesia americana (Teratosphaeriaceae), but phylogenetically distinct. Morphological survey and phylogenetic analysis using nucleotide sequence data from the nuclear ribosomal subunit genes indicate a close relationship within three species colonising pine litter needles, F. cordae, F. pini and F. ramoconidii. F. sicilianum is most related to F. rhodense. C. matsushimae represents a species belonging to one of the lineages of the polyphyletic genus Cladophialophora. None of the strains observed can be classified morphologically as S. ochraceum, of which the type material does not exist.  相似文献

The wolf spider Pardosa cribata Simon is the most abundant ground-dwelling spider inhabiting citrus orchards in eastern Spain. However, little is known about its activity-density and its predatory role in the citrus agrosystem. Here we report on the activity-density of P. cribata monitored by pitfall traps, and on its capacity to prey on two citrus pests that appear both in the citrus canopy and the ground cover, Ceratitis capitata (Wiedemman) and Myzus persicae (Sulzer), respectively. Pardosa cribata was present in citrus orchards throughout the year, with a peak in spring and a higher peak in summer. Pardosa cribata preyed on adults and third-instar larvae but not on pupae of C. capitata. A type II functional response was obtained for teneral-like adults, with an estimated attack rate (a′) of 0.771 ± 0.213 days−1 and a handling time (T h) of 0.051 ± 0.013 days. Pardosa cribata also preyed efficiently on M. persicae, giving a type II functional response with an estimated attack rate and handling time of 2.833 ± 0.578 days−1 and 0.031 ± 0.001 days, respectively. The data reported here indicate that this wolf spider could play an important role in regulating both these pests, and therefore might contribute to developing conservation biological control strategies for citrus pests. The influence of sugars and growth regulators on shoot and root growth of Dactylorhiza species was studied under in vitro conditions. The seedling development was stimulated with the application of glucose and sucrose at concentration of 10 g dm−3 each. The improvement of shoot growth rate and shoot length was enhanced by cytokinins N 6-(2-isopentenyl)adenine or N 6-benzyladenine and their combination with auxin indolebutyric acid (IBA). The root growth rate and root length of seedlings increased in the presence of IBA and α-naphthaleneacetic acid. Individual Dactylorhiza species showed statistically significant differences in shoot and root development depending on sugar and growth regulator combinations.  相似文献

Previous studies have demonstrated that the obligate myrmecophytism between Macaranga ant-plants and Crematogaster plant-ants is highly species specific, although multiple Macaranga species can coexist in a microhabitat. However, the species specificity has been described based on the study of trees with established plant-ant colonies. We studied how the process of settling into the partner Macaranga seedlings by single foundress Crematogaster queens contributes to species specificity. By sampling seedlings of three sympatric Macaranga myrmecophytes species in the field, we tested two hypotheses. The first is that foundresses correctly select their specific partner plant species when they settle into seedlings. The second hypothesis is that the seasons in which seedlings available for settling by foundresses appear are segregated among the Macaranga species, and the seasons in which foundress queens settle are synchronized to the appearance of seedlings of specific partner species; thus species specificity is consequently generated. Our results support the former hypothesis but not the latter: we always observed foundresses settling species-specific host plants, and seedlings suitable for settling were always available in each Macaranga species. We studied the oviposition performance of Fopius arisanus (Sonan) (Hymenoptera: Braconidae) attacking eggs of four fruit flies of the genus Anastrepha Schiner (Diptera: Tephritidae) under laboratory conditions. The complete process of oviposition on an individual egg of Anastrepha ludens (Loew) lasts in average 85.4 ± 2.9 s, including a tremor (25.8 ± 1.03 s) observed in the middle of this process related to the egg’s descent. The average parasitism of A. ludens egg was 60.9 ± 7.5%, with only 1.2% of superparasitized eggs. During individual acts of oviposition, we noted that F. arisanus possesses a highly flexible ovipositor that curves easily as it searches for additional suitable eggs, which may be of particular benefit when a female finds large clutches of eggs. The individual oviposition of F. arisanus in host fruits attacked by Anastrepha spp. varies with the egg clutch size of each fruit fly species: A. serpentina laid the biggest egg clutches (21.3 ± 1.4), followed by A. ludens (14.2 ± 0.9), and A. striata (1.0 ± 0.0) (=A. obliqua). The time spent by F. arisanus in individual ovipositions was parallel to these findings, reinforcing the idea that F. arisanus attacks several eggs in each individual insertion of its ovipositor. Neither formal oviposition acts, nor adult emergences of F. arisanus were registered in A. obliqua. We discuss the potential of F. arisanus as natural enemy of fruit flies of the genus Anastrepha, and explore the eventual developing of its mass rearing. Handling Editor: Torsten Meiners.  相似文献

Ophiostomatoid fungi colonize the conducting tissues of conifer stems, the phloem and the xylem. These pathogenic fungi penetrate into the stem through injuries made by xylophagous insects vectoring these pathogens. In this study the response of the phloem of Scotch pine (Pinus sylvestris L.) to wounding (treatment 1) was compared with the response to wounding combined with application of high-molecular-weight compounds isolated from the mycelium of the ophiostomatoid fungus Ceratocystis laricicola Redfern & Minter (treatment 2). Both treatments induced the appearance of necrosis in the inner bark, the formation of periderm separating living and dead tissues, and formation of the callus alongside the wound perimeter. In addition, the bark accumulated lignin, bound proanthocyanidins, and resins, with a parallel decrease in the content of free proanthocyanidins, low-molecular-weight carbohydrates, and non-lignin components of the cell wall (P > 0.95). The size of necrotic spots, as well as changes in the content of most substances, were significantly higher in the treatment 2 than in the treatment 1 (P > 0.95). The accumulation of lignin in cell walls of phloem sieve cells was delayed in the treatment 2 as compared with that in the treatment 1. This suggested that the mycelial extract temporarily inhibited lignification at the early stage of the wound response. This disturbance of the cell wall protective transformation led to the hypothesis that the fungal suppressors retard the repair of inner bark injured by insects, thereby favoring the invasion of conifer tissues by ophiostomatoid fungi.  相似文献

In vitro regeneration of black nightshade (Solanum nigrum L.) plants was achieved through callus-mediated shoot organogenesis followed by 30 d indoor ex vitro adaptation to nutritional stress under environmental ambience and thereafter 6-d outdoor acclimatization in pots prior to field establishment. Relevant physiological parameters including pigment content, chlorophyll a fluorescence, net photosynthetic rate (P N), transpiration rate (E), and stomatal conductance (g s) of in vitro-regenerated plants were investigated during the course of ex vitro adaptation. During the first 4 d of indoor transplantation to potting substrate, there was a marginal reduction in the leaf chlorophyll and carotenoid contents but P N and E were strongly reduced. The stomatal conductance and E/P N ratio were significantly higher in plants up to 20 d of indoor adaptation than those of comparable age grown naturally from seeds. The shape of the OJIP fluorescence transient varied significantly with acclimatization, and the maximum change was observed at 2.0 ms. The 2.0 ms variable fluorescence (V j), 30 ms relative fluorescence (M 0), photon trapping probability (TR0/Abs), and photosystem II (PSII) trapping rate (TR0/RC) showed initial disturbance and subsequent stabilization during 30 d of indoor acclimatization. Energy dissipation (DI0/RC) and electron transport probability (ET0/TR0) showed an initial phase of increase during the 4 d after plants were transplanted outdoors. During the 6-d outdoor acclimatization after transfer of plants to soil, no significant change in total chlorophylls and carotenoids, E, and g s were observed, but P N improved after reduction on the first d. The OJIP-derived parameters experienced change on the first d but were stabilized quickly thereafter. There was no significant difference between outdoor acclimatized plants and those of the seed-grown plants of comparable age with respect to photosynthetic and fluorescence parameters. Direct transfer of plants without indoor acclimatization, however, showed a completely different trend with respect to P N, E, and OJIP fluorescence transients. The bearing of this study on optimizing micropropagation is discussed.  相似文献

The potential role of the resin system in the response of Austrian pine (Pinus nigra Arn.) seedlings to mechanical injury and fungal infection was studied in greenhouse experiments. Anatomical observations were performed on 2-year-old plants wounded at collar level and inoculated with Sphaeropsis sapinea (Fr.: Fr.) Dyko & Sutton in Sutton or Diplodia scrobiculata (J. de Wet, B. Slippers & M. J. Wingfield, sp. nov.; sensu de Wet et al. 2003), two fungal pathogens that cause shoot blight and canker on conifers, and that are characterized by different levels of aggressiveness. Histological examination of host tissue taken from the stem at 0, 8, and 12 cm above the treatment site revealed significant treatment- and time-dependent effects on the course of locally and systemically induced traumatic resin duct (TRD) development. Occurrence of TRDs was observed after 4 days only in seedlings inoculated with D. scrobiculata. At 12 days, TRDs were present also in mock-inoculated controls. No TRDs appeared in seedlings inoculated with S. sapinea. However, S. sapinea caused loss of vacuolar phenolics, severe disruption of cambial tissue and invaded the host xylem quickly and apparently unimpeded, whereas D. scrobiculata was never detected in the host xylem. Five-year-old Austrian pines subjected to the same stem base treatments were used to determine the resin mass flowing from the stem 30 cm above the treatment sites. Wounding and/or inoculation induced a significant, 8.3-fold average increase in systemic resin flow over the untreated trees 3 weeks after basal treatment, suggesting that wounding is the sole prerequisite for systemic induction of resin flow. The results are discussed in the context of current disease resistance models.  相似文献

Esteya vermicola (Ophiostomataceae) is the first reported endoparasitic fungus of the pinewood nematode (PWN), Bursaphelenchus xylophilus (Nematoda: Aphelenchoidoidea). It has high in vitro infectivity. In this study, the nematocidal effect of E. vermicola in logs was investigated and evaluated. Two months after inoculation of pine wilt-killed Pinus densiflora logs with E. vermicola conidia suspensions of 3 × 108 and 3 × 106 ml−1, the density of nematodes decreased by approximately 79% and 47%, respectively. When the fungus was sprayed on to four-year-old pine seedlings one month before PWN inoculation, the survival index of seedlings reached 0.67 compared with only 0.067 for control seedlings without fungal spraying. These results suggest that conidia spraying of Evermicola can, to some extent, protect pine trees from wilt disease. Moreover, infected nematodes and hyphae of Evermicola were observed in the treated wood sections.  相似文献

Although modern forestry takes into consideration the analysis of the effects of forest management on plant structure, diversity and seedlings, little is known about how those parameters respond to harvest techniques in the Mediterranean region. We investigated the effect of three different harvest intensities, respect to uncut controls, on understory plant species functional groups, richness, diversity and pine seedlings in a natural Maritime pine stand in Spain, three years after harvesting. The harvest treatments produced a reduction of the number of Pinus pinaster seedlings and woody species cover, and an increase of species richness (total and of annual species) and plant cover of annual species respect to control plots (CO). The Shannon diversity values showed no differences between treatments. These results emphasize that the tree harvest treatments analyzed are not suitable for the management of this P. pinaster stand. Otherwise, the reduction of pine seedling density by harvest treatments and the changes in richness and cover of functional groups would not induce the natural regeneration of this stand maintaining the understory plant layer.  相似文献
