Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Growth rate estimates () of phytoplankton populations that were sampled from nitrogen-limited continuous cultures and then incubated for short durations in batch culture with added14C-HCO3 were significantly different than steady-state growth rates () for 3 of 5 marine phytoplankton species. Two diatoms,Thalassiosira weissflogii andChaetoceros simplex, displayed virtually identical growth rates (=) over a wide range of, whereas for a third diatom,Phaeodactylum tricornutum, was overestimated by an average of 40% compared to. In contrast, was underestimated by the14C technique for the two remaining species: up to 40% at a steady-state of 1.0 day–1 for the chlorophyteDunaliella tertiolecta and up to 100% at of 1.4 day–1 for the haptophytePavlova lutheri. For the latter two species the divergence between and appeared to increase with increasing steady-state. A simple model of labeled and total carbon flow between the aqueous phase and cellular biomass was constructed to demonstrate that respiration was negligible when=, but was significant when>. In the cases in which<, a rapid physiological alteration presumably took place once the steady state was disturbed and cells were placed in the incubation chambers, which perhaps was related to the nutritional state of the cultures at the time of sampling. Questions thus are raised regarding our ability to measure accurately primary productivity from shipboard experiments with confined samples of phytoplankton from nutrient-impoverished waters that probably are less hardy than the laboratory cultures used in these studies.  相似文献

Cell suspension cultures of Agave amaniensiswere able to grow in media containing 10 – 240 M copper ions, and could remove more than 67% copper ions from the media. The cells accumulated up to 106 mg g–1 copper ions in the biomass. Copper ions at 240 M caused a decrease in growth index and packed cell volume of the cultures of 61.5 and 53.3%, respectively. The presence of copper ions caused the cell walls to thicken and to be more wrinkled. Certain amino acids were released in high concentration into the media. The hecogenin content in the biomass increased up to 157.9% at 20 M copper ions.  相似文献

Uptake kinetics of nitrogen derived from sewage–seawater mixtures (2.5–20% v/v effluent) were determined in the laboratory for Ulva rigida (Chlorophyceae) native from Bahía Nueva (Golfo Nuevo, Patagonia, Argentine). In terms of nitrogen concentration, experimental enrichment levels varied between 53.7 and 362.3M of ammonium and between 0.77 and 6.21M of nitrate+nitrite. Uptake rates were fitted to the Michaelis–Menten equation, with the following kinetic parameters: ammonium: Vmax = 591.2molg–1h–1, K s=262.3M, nitrate+nitrite: V max=12.9molg–1h–1, K s=3.5M). Both nutrients were taken up simultaneously, but ammonium incorporation was faster in all cases. The results show a high capability of Ulva rigida to remove sewage-derived nitrogen from culture media. In the field, most of the nitrogen provided by the effluent would be tied up in algal biomass, supporting low nitrogen levels found at a short distance away from the source.  相似文献

Development of an L6 myoblast in vitro model of moniliformin toxicosis   总被引:1,自引:0,他引:1  
L6 myoblasts were used as an in vitro model to investigate the role of moniliformin and its interaction with monensin in turkey knockdown syndrome and sudden death syndromes in poultry. Cell viability and microscopic and ultrastructural alterations noted in L6 myoblasts cultured in the presence of moniliformin (0.0–0.3 g/l) were compared to those observed in parallel cultures also containing one of the following compounds: selenium (0–0.004 ng/l), thiamine (0–0.3 g/l), or pyruvate (0–0.46 g/l). Marked dilation of the RER, membranous whorls, glycogen deposition, membrane-bound cytoplasmic inclusions and necrosis were observed in myoblasts exposed to 0.03/2-0.30 g moniliformin/l medium. Supplementation of medium with thiamine and pyruvate, or selenium, provided significant protection to cells exposed to 0.0–0.3 g/l or 0.0–0.15 g moniliformin/l, respectively. Dose-dependent differences in protein and ATP production were not detected. Myoblasts grown in medium containing 0–0.15 g moniliformin/l and 7.5–50.0 M A23187, beauvericin or monensin had degrees of cytotoxicity similar to parallel cultures receiving only an ionophore. L6 myoblasts were a useful model of moniliformin toxicosis. The findings of this study suggest cytotoxicity due to moniliformin in L6 myoblasts may be due in part to oxidative damage and altered pyruvate metabolism, and that moniliformin does not predispose myoblasts to ionophore toxicosis. This study supports the results of in vivo investigations in poultry that moniliformin and monensin do not act synergistically to induce knockdown or monensin toxicosis.  相似文献

Picoplankton (plankton 3 m) biomass was determined by flow cytometry in three European estuarine systems (Krka Estuary in Croatia, Rhône Delta in France, and Lena Delta and Laptev Sea in Russia). The size of natural phytoplankton groups was obtained by a calibration curve, with different picoplankton's strains (from 1.6 to 3.4 m), measured by a Coulter counter (size) and a flow cytometer (light-scattering). Two natural groups of picoplankton were identified by flow cytometry in the three systems: Synechococcus sp and picoeukaryotes. Picoplankton cells abundance ranged between: 2800 and 42000, 5000 and 37000, 1000 and 50000 cells ml–1 in the Krka estuary, in the Rhône delta and in the Lena-Laptev system, respectively. In the Krka estuary, picoplankton biomass ranges between 11 and 68 gC l–1. It can make up as much as 88% of the total photosynthetic plankton population and 50% of total organic particulate carbon. Picoplankton biomass was greater in the summer than in the autumn. At the halocline layer this biomass can attempt ca. 390 gC l–1during the summer cruise. In the Rhône delta, a lower picoplankton biomass (6–39 gC l–1) was observed at the end of the winter. These biomass represented between 0.4 and 22% of the particulate organic carbon, which could reach 71% of the total photosynthetic plankton biomass at the marine station. In the Lena-Laptev system, picoplankton biomass varied between 6 and 56 gC l–1 in surface waters. Picoplankton biomass decreased with depth, but picoeukaryotes were still observed in deep samples (20, 30 m) in the Laptev Sea, showing a considerable autotrophic activity in spite of low temperatures (0–1 °C). Although the widely dispersed estuary geographic distribution and their different estuarine characteristics, the data point out that these small organisms can also play an important role in the transfer of organic carbon from rivers to oceans and that flow cytometry can be able to detect these small cells in turbid systems.  相似文献

Seedlings ofAlnus incana (nodulated and non-nodulated) andBetula papyrifera were fertilized with varying amounts (0, 10, 25, 50, 100, 250 and 500 g N g–1 soil) of labelled ammonium-N or nitrate-N ( 5.2 A% excess15N as ammonium sulphate or potassium nitrate). After 4 months in the greenhouse,15N excess in the plants were determined and an isotope dilution equation was applied to determine the percent of biomass N fixed by theA. incana/Frankia system. When ammonium was used as the sole N source and birch as the non-fixing reference, N-fixation accounted for 95%, 87% and 60% of the plant nitrogen yields with 10, 25 and 50 g N g–1 rates, additions respectively. At the 100 g N g–1 fertilization and above N-fixation accounted for less than 10% of the N yield. Similar results were obtained when non-nodulatedA. incana was used as non-fixing reference. With nitrate as the sole N source, N-fixation accounted for 98%, 97%, 97%, 86%, 56% and 12% of N yield with 10, 25, 50, 100, 250 and 500 g N g–1 additions respectively. These values were similar for both types of reference plants. The direct isotope dilution method was compared to that of the total nitrogen difference method. There was good agreement between the two methods up to 50 g N g–1 for ammonium and up to 100 g N g–1 for nitrate. The difference method produced negative values at high concentrations of nitrogen fertilization. Again similar results were obtained by the two reference plants. The results indicate that birch can be used as a non-fixing control in isotope dilution studies but that care must be exercised in selecting the type and quantity of labelled nitrogen fertilizer.  相似文献

This study addresses the temporal distribution of forms of phosphorus in the soil of a temporarily flooded riparian forest of the valley of the river Garonne (Southwest of France). A sequential extraction for forms of phosphorus of increasing chemical stability was used. During the study period (13 months), the forest was flooded a few days during March and May. In winter, resin-Pi concentration was high (26 g g–1) in comparison to spring values (<9 g g–1). NaHCO3-Po, NaHCO3-Pi or NaOH-Pi concentrations increased during winter (up to 74, 124 and 78 g g–1 respectively) and decreased significantly during spring (32, 44 and 32 g g–1 respectively). This pattern was attributed to simultaneous mineralization and plant uptake during the growing season and to the flood events (erosional processes and P-release). During summer and fall, resin-Pi concentration increased significantly (up to 26 g g–1 in October). NaHCO3-Po concentrations remained low during spring and summer (<33 g g–1), and increased significantly in fall (>45 g g–1 NaHCO3-Pi or NaOH-Pi increased in late spring or summer (90 g g–1 and 68 g g–1 respectively). Increasing concentrations of the labile forms during late spring or summer were ascribed to the warm temperature and soil dryness that limited plant growth. HCl-Pi increased regularly after the floods (174 g g–1 before the flood events to 254 g g–1 after the floods). Residual P presented a similar pattern i.e. 214 g g–1 and 279 g g–1 respectively before and after the flood events. This pattern was attributed to a progressive incorporation of flood deposits to the soil.  相似文献

Despite reports that boron (B) requirements differ among plant species there is a shortage of critical evidence to demonstrate unequivocally whether species differ in internal or external B requirements or both. The present research was conducted to establish the external and internal B requirements of three contrasting species, a woody dicot (marri), an herbaceous dicot (sunflower) and a monocot (wheat) using B-buffered solution culture. Boron-buffered solution culture provided satisfactory control of external B concentrations ranging from 0.04 to 30 M throughout the 20- (sunflower and wheat) or 40-day (marri) growth period. At low external B concentrations ( 0.13 M), the growth of marri and sunflower was severely depressed but by contrast the vegetative growth of wheat plants was satisfactory and free of B deficiency symptoms. Marri and sunflower plants achieved total maximum shoot growth at 1.2 M B in solutions while wheat plants did so at 0.6 M B. The critical B concentrations (mg kg–1 dry matter) in the youngest open leaf blades of marri, sunflower and wheat plants were 17.9, 19.7 and 1.2 on 20, 10 and 10 days after transplanting (DAT), respectively. Lower internal and external B requirements of wheat were matched by a lower uptake rate of B compared to marri and sunflower.  相似文献

Atriplex gmelini plants were regenerated via organogensis from hypocotyl explants. Callus lines were induced from the hypocotyl explants on Linsmaier and Skoog (LS) medium supplemented with 1 M benzyladenine and 5 M -naphthaleneacetic acid in the dark. Shoots were regenerated from the callus lines on LS medium supplemented with 20 M thidiazuron and 0.1 M -naphthaleneacetic acid under a high-intensity light condition (450 mol m–2 s–1). The regenerated shoots were rooted on LS medium without growth regulators to obtain fully developed plants. We succeeded in transforming Atriplex gmelini from callus lines using Agrobacterium tumefaciens.  相似文献

Glycerol/methanol and sorbitol/methanol mixed-feed fermentation strategies for the production of recombinant proteins by Pichia pastoris were compared in order to examine sorbitol's potential as a carbon source. Although P. pastoris does have a lower cell yield on sorbitol than on glycerol, the specific rate of product formation is higher (60 g protein g–1 dry wth for sorbitol/methanol, vs 45 g protein g–1 dry wth for glycerol/methanol), resulting in comparable final recombinant expression levels. Importantly, the presence of residual sorbitol in the growth medium appears to be less repressive to the alcohol oxidase promoter in this organism, providing a more forgiving means of operating mixed-feed fed-batch recombinant P. pastoris fermentations.  相似文献

A biomass adapted to degrade toluene and xylenes in mixture was grown in a batch reactor with substrates supplied by pulses. The inhibition of biomass growth in the course of substrate degradation was investigated. The maximal biomass concentration of 7 g l–1 was obtained using 150 l of toluene and 15 l of a mixture of xylenes in one litre of liquid medium, and the maximal biomass productivity and yield were 53 mg l–1 h–1 and 0.32 gDW g s –1 , respectively. Higher quantities of substrate added by pulses, that is 200 l of toluene with 20 l of xylenes and 300 l of toluene with 30 l of xylenes, caused an accumulation of metabolites. These higher quantities of substrates caused inhibition of microbial growth. Among the metabolites produced, 4-methyl catechol was found in large quantities in the culture medium and in the cells.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献

A micromanipulation method is described for measuring the bursting forces of bacteria and relating them to cell size. At a compression speed of 6.2 m s–1, bursting forces of three samples of rapidly growing Staphylococcus epidermis from a batch culture varied from 3 to 34 N with an average value of 13.8 N (standard error 0.8 N). Escherichia coli grown in continuous culture at a specific growth rate of 0.5 h–1 had bursting forces varying from 1 to 9 N with an average value of 3.6 N (standard error 0.4 N). In squeeze-hold experiments, force relaxation was observed, which was attributed to water loss from the cells, or viscoelasticity, or both. At high compression speed, such as 6.2 m s–1, this relaxation could be neglected. Micromanipulation strength measurements might be used in studies of cell mechanical disruption and of the dependence of cell strength on cell physiology.  相似文献

Lyophilized aqueous extract of Maytenus ilicifolia leaves (LAEMIL) is commonly used in Brazilian folk medicine in the treatment of dyspepsia as well as gastric ulcers. We have investigated the effect and the possible mechanism of action of the LAEMIL on acid secretion in isolated frog gastric mucosa incubated in an Ussing chamber. It was observed that LAEMIL (7–28 mg%) as well as cimetidine (125–4,000 M), a well-known histamine H2 receptor antagonist, decreased basal acid secretion in a concentration-dependent manner. Similarly to cimetidine (190 M), LAEMIL (21 mg%) also inhibited gastric acid secretion induced by increasing concentrations of histamine (50–800 M). The EC50 values for histamine alone and histamine in the presence of LAEMIL or cimetidine were 94.6 M (71.1–125.9 M), 244.9 M (209.4–286.4 M) and 142.2 M (23.6–855.0 M), respectively. LAEMIL, histamine and cimetidine were effective on acid secretion only when added to the serosal surface of the mucosa. Furthermore, simultaneous addition of LAEMIL and cimetidine at concentrations, per se, ineffective, caused a 16% reduction in the basal acid secretion [from 8.3±0.3 to 6.9±0.2 Eq g–1 (15 min)–1, n=4]. Although effects such as inhibition of histamine biosynthesis and/or histamine release can not be ruled out, our data suggest that LAEMIL, like cimetidine, reduces acid secretion in the isolated frog gastric mucosa by antagonising histamine H2 receptors.Abbreviations LAEMIL Lyophilized aqueous extract of Maytenus ilicifolia leaves - Hist Histamine - Cim Cimetidine  相似文献

Summary The architecture of the media of arterial vessels in dog brain was investigated using scanning electron microscopy. The arrangement and shape of the circularly-oriented smooth muscle cells varied with vessel diameter: The arteries (>100 m in diameter) had 4–10 layers of spindle-shaped smooth muscle cells; the muscular arterioles (30–100 m), 2–3 layers of spindle-shaped smooth muscle cells; the terminal arterioles (10–30 m), a compact layer of spindle-shaped smooth muscle cells with more dominant nodular or rod-like processes and thin lateral processes; and the precapillary arterioles (5–15 m), a less compact layer of branched smooth muscle cells.Longitudinally-oriented muscles were observed in the medio-adventitial border. The distribution and arrangement of these muscles varied with vessel size: in the large arteries (> 300 m in diameter), at the branching sites only; in the small arteries (100–300 m), at both the branching and non-branching sites; in the muscular arterioles, at both the branching and non-branching sites in a reticular arrangement with some muscle cells having an asteroid appearance; in the terminal aterioles, only asteroid-like muscle cells were found at the branching and non-branching sites.  相似文献

We tested the hypothesis that plants only stimulate net mineralization of N when intense competition for N exists between plants and heterotrophs. Nitrogen mineralization in the soil used was insensitive to the range of moisture fluctuations that were inevitable during plant growth. Pots were planted to wheat (Triticum aestivum L.) or left unplanted and received no straw, straw added in one central layer, or straw added uniformly through the whole soil volume. Through the addition of15 N-labelled nitrate, initial soil inorganic N was increased to 17 g g–1 in unplanted treatments and to 17 g g–1 and 72 g g–1 in planted treatments. Straw addition increased microbial immobilization of labelled N (soil inorganic N at planting), but did not reduce net mineralization of unlabelled soil N (soil organic N at planting), indicating that straw decomposers immobilized N early in the growth period. Plant growth did not reduce immobilization of N by straw decomposers. Net mineralization of N was not affected by plant growth at the low rate of N addition, but was reduced at the high rate of N addition. We conclude that the influence of wheat growth on net mineralization of N depends on soil N availability, with reductions in net mineralization at high N levels due to increased immobilization.  相似文献

Embryogenic culture was induced from the immature embryos of Quercus serrata using Marashige and Skoog's medium (MS) containing 0.1 M each of 2,4-d and BAP, and subcultured for seven months before isolation of protoplasts by using 1% Cellulase RS in 0.6 M mannitol solution. Efficient colony formation was obtained when protoplasts were cultured in a liquid MS medium containing 0.6 M mannitol, 3% sucrose and combination of 0.1 M or 1 M each of 2,4-d and BAP. Excluding ammonium nitrate from the MS medium resulted in the decrease of the percentage of colony formation. Total mercury was measured in different compartments of Lake Xolotlán's (Managua) ecosystemviz., sediments, water, fish and men. Sediments from 18 localities at 5 depths inside the sediment (5, 10, 15, 20 and 25 cm) contained an average concentration of 0.62 g Hg.g–1±0.46 at the surface, with extreme values of 0.16 and 1.8 g.g–1. The highest concentration was observed at 25 cm depth in front of the chlor-alkaly factory (ELPESA). This maximum is associated with the period of highest production of this factory. The highest mercury concentrations in water were also measured close to the discharge of ELPESA,viz. 787 g.Hg–1 in January and 506 g.g–1 in April. The mean mercury concentrations measured in the muscles of the most consumed fish were 0.63 g.g–1±0.22 (extreme values 0.22 and 1.45) inCichlasoma managuense, and 0.07 g.g–1±0.14 (extreme values 0.004 and 0.63) inC. citrinellum. The concentration in the liver was 0.79 g.g–1±1.29 inC. managuense and 0.62 g.g–1±0.44 inC. citrinellum. Human hairs (n=98) of fishermen and their families contained 5.03 g.g–1±6.2 (extreme values 0.02 and 38.22). The mean concentration measured in men was 6.22 g.g–1±6.34 (n=58), and in women 3.39 g.g–1±5.7 (n=40). The average mercury concentration of hairs of workers of ELPESA was 91.24 g.g–1±156.9 (extreme values 0.46 and 724.53; n=32). We conclude that total mercury levels in the various ecosystem compartments are very high and mercury contamination in the lake may be considered as dangerous for human health.  相似文献

A 20–40 m pellicular high density (3.7 g cm–3) expanded bed material has been designed for the capture of DNA and other large macromolecules. Anion exchangers fashioned out of these supports exhibited dramatically enhanced DNA binding capacities over commercial anion exchange adsorbents (6 mg ml–1, c.f. 50 g ml–1 at 10% breakthrough), due to a combination of small particle and fuzzy surface architecture created through the coupling of polyethylene imine chains.  相似文献
