The germination of seeds of celery (Apium graveolens L.) becomes progressively thermoinhibited on incubation in the dark at high temperatures, the inhibitory temperature being dependent on the cultivar used. In two high-dormancy cultivars of celery, the production of germination inhibitors in seeds incubated in the dark at 26°C gradually increased over a 7-day period. Inhibitor production was measured by incubating seeds of the low-dormancy cultivar Florida 683 in homogenates of the thermoinhibited seeds of the high-dormancy cultivars and recording germination either in the light or with the gibberellins A4 and A7 (GA4/7) in the dark. Most Florida 683 seeds which failed to germinate in the homogenates after 15 days were induced to germinate by addition of N6-benzyladenine (BA). The presence of BA in addition to GA4/7 throughout incubation in the dark completely overcame the inhibitory effects of homogenates. This indicates that thermoinhibition of celery seeds is associated with the accumulation of a germination inhibitor which interacts with cytokinins. The temperature-dependent primary dormancy of cv Florida 683 celery seeds in darkness was broken by GA4/7 (2 × 10-4 M) alone but other growth regulators such as BA, ethephon or daminozide were necessary to break dormancy of cv Lathom Blanching seeds in the presence of GA4/7 at this concentration. Although AgNO3 partially inhibited both the ethephon- and BA- induced germination of cv Lathom Blanching seeds in the presence of GA4/7 in the dark it did not affect the promotive action of daminozide. Ethephon did not overcome the inhibitory action of high concentrations of AgNO3 in the light. The ethylene synthesis inhibitor aminoethoxyvinylglycine (AVG) did not inhibit the germination of cv Lathom Blanching seeds induced by growth regulators in the dark or in the absence of growth regulators in the light. Fusicoccin (FC) did not break celery seed dormancy unless applied in the presence of GA4/7. Germination of cv Lathom Blanching celery seeds treated with GA4/7 at 16°C in the dark was inhibited by the K+ ionophore benzo-18-crown-C-6 (18-C-6) and in the presence of Ca2+ by the Ca2+ ionophore A23187; the 18-C-6 inhibition was reversed by BA.It is concluded that the involvement of gibberellin in celery seed dormancy is not dependent on endogenous ethylene and is directly or indirectly controlled through the action of other hormones on transmembrane ion fluxes.  相似文献

The effects of a plant-derived smoke extract, BA and GA3 on the thermodormancy of Grand Rapids lettuce seeds were studied. Thermodormant lettuce seeds treated either with BA, GA3 or smoke extract alone did not germinate. Combinations of BA with smoke extract and BA with GA3 were most effective in overcoming induced thermodormancy. GA3 plus smoke did not break the induced thermodormancy. The effects of the different treatments on germination were concentration dependent. Ethephon (Eth), gibberellin A3, A4 + 7 (GA3, GA4 + 7), and 6-benzyladenine (BA) removed secondary dormancy of Amaranthus caudatus seeds. The GAs and BA potentiated the effect of ethephon or 1-aminocyclopropane-1-carboxylic acid (ACC), an ethylene biosynthesis precursor, in terms of the rate or final percent of germination. Aminoethoxyvinylglycine (AVG), an ACC synthase activity inhibitor, was observed to simultaneously inhibit the release from dormancy effected by GA3 or BA as well as the ethylene production stimulated by these regulators. Breaking of secondary dormancy by GA3, GA4 + 7 or BA was prevented by 2,5-norbornadiene (NBD), an inhibitor of ethylene binding. Ethylene completely or markedly reversed the inhibitory effect of NBD. We thus conclude that the removal of secondary dormancy in Amaranthus caudatus seeds by gibberellin or benzyladenine involves ethylene biosynthesis and action.  相似文献

The percentage germination of seeds of parsley cv. Imperial Curled was higher in the light than in the dark, the high temperature limits for germination being 30 and 28°C for light and dark respectively. At the higher temperatures, the germination rate was slower in the dark. At 30°C, treatment with a gibberellin A4/7 mixture at 2 × 10–4 M partially alleviated the inhibiting effect of darkness on the germination percentage. Pre-incubation of parsley seeds at 35°C in the dark for 30 h increased the rate, but decreased the percentage, of germination of seeds incubated at 15°C in the light. Germination and seedling emergence studies were made on seed harvested from four different umbel positions. Although heavier seeds were produced from primary umbels than from other umbel orders, they were less viable as measured by seedling emergence in the glasshouse. The rate of emergence was decreased with increasing umbel order i.e. with later seed development: this was reflected in subsequent seedling weights, with seedlings from quarternary umbel seeds being about half the weight of those from primary umbel seeds. Imbibition of celery (Apium graveolens L.) seeds at 32°C for up to 96 h lowered the upper temperature limit for germination. If this high temperature treatment was given in the light, these seeds germinated slightly earlier than those treated in the dark although the final percentage germination was similar for both treatments. The inhibitory effect of the high temperature treatment was completely removed by allowing the seeds to imbibe in a mixture of the gibberellins A4 and A7 (GA4/7) and partially removed by the cytokinin N6-benzylaminopurine (BA). GA4/7 was less effective when added before rather than after the high temperature treatment, whereas the opposite was true of BA. At constant temperatures more GA4/7 was required to promote germination as the temperature was raised but addition of BA reduced the concentration of GA4/7 required. A model is proposed for the control of celery seed germination by light and temperature through the action of endogenous cytokinins and gibberellins.  相似文献

Effects of the gibberellins A4+7(GA4+7) and A3(GA3), benzyladenine (BA) and forchlorfenuron (CPPU) on deposition of the cuticular membrane (CM) in developing tomato (Lycopersicon esculentum L.) fruit were investigated. Growth regulators were applied when fruit development within trusses ranged from the flower to the mature stage. Developmental stage of fruit at the time of application was indexed by fruit diameter. Fruit were harvested at maturity, the CM isolated enzymatically on an individual fruit basis and mass of CM per unit fruit surface area calculated. In mature fruit, mass of CM per fruit increased with fruit size, but mass of CM per unit surface area was independent of fruit size, position within a truss and position of the truss on the plant. GA4+7 and GA3 increased CM mass per unit fruit surface area at concentrations up to 300 mg l−1. Young fruit (5–10 mm diam. at time of application) was most responsive. Responsiveness decreased as fruit development at application progressed towards maturity. There was no consistent effect of GA4+7 or GA3 on fruit mass. BA (up to 100 mg l−1) or CPPU (up to 3 mg l−1) had no significant effect on CM mass per unit surface area regardless of developmental stage. Higher concentrations of BA or CPPU decreased CM mass per unit surface area. There was no effect of BA or CPPU on fruit mass. Potential mechanisms and benefits of a gibberellin induced increase in CM deposition are discussed.  相似文献

The white-light-(WL) induced enlargement of dicotyledonous leaf cells is known to occur via an acid-growth mechanism; i.e., WL causes leaf cells to excrete protons which lead to an increase in wall extensibility and thus cell enlargement. Gibberellic acid (GA3) and N6-benzyladenine (BA) also induce leaf cell enlargement. To see if they also act via acid-induced cell wall loosening, a comparison has been made of WL-, GA3-and BA-induced growth of strips, taken from primary leaves of bean (Phaseolus vulgaris L.) plants raised in continuous red light for 10 d. White light, GA3 and BA all increased wall extensibility as measured by the Instron technique, and this change preceded the increase in growth rate. However, whereas WL induced significant proton excretion, neither GA3 nor BA caused any acidification of the apoplast. Furthermore, neutral buffers, which effectively inhibited the growth induced by WL, were without effect on growth promoted by either GA3 or BA. These results indicate that while WL, GA3 and BA all initiate growth in bean leaves by altering cell-wall properties, GA3 and BA do so through some wall loosening mechanism other than wall acidification. Quantitative estimates of gibberellin A9 in Norway spruce extracts obtained by gas chromatography-mass spectrometry, radioimmunoassay (RIA_ and bioassay were compared after successive purifications of the extracts. The extracts were assayed in several dilutions with and without the addition of standard gibberellin A9, thus showing the effect of extract components on the response of the assays. Radioimmunoassay produced estimates comparable to gas chromatography-mass spectrometry after one purification step by high-performance liquid chromatography. Extracts purified by polyvinylpyrrolidone-column chromatography and solvent partitioning but not high-performance liquid chromatography resulted in inaccurate RIA estimates. The performance of the RIA could be monitored by logit-log transformations of the standard curve and extract dilution curve and by calculating the slope of the standard addition curve. It was, however, not possible to correct for the interference caused by extract components by the standard addition procedure. Growth Regulators and the Phytochrome-Mediated Dormancy of Celery Seeds   总被引:2,自引:0,他引:2  
Seeds of five celery (Apium graveolens L.) cultivars germinated at 15°C in the light or dark but at 22°C only in the light. This light requirement was overcome by treatment with a mixture of the gibberellins GA4 and GA7 (GA4/7) but interactions of cytokinins, daminozide, ethephon, EDTA and N-phenyl-N′-4-pyridylurea (NC5392) with GA4/7 were observed. Varietal differences in response to GA4/7 concentration and the requirement for cytokinins were related to the upper temperature limits for germination of the different cultivars. Seeds of cultivars responding to low concentrations of GA4/7 appeared to contain less natural inhibitor than those requiring either high concentrations of GA4/7 or cytokinin in addition to low GA4/7. The cytokinin requirement for germination was partially removed by leaching the seeds with water. Interaction studies with applied hormones indicated that in seeds incubated in the light inhibition by abscisic acid was partially alleviated by N6-benzyladenine but not by GA4/7 application. The implications of these results are discussed in relation to the involvement of natural plant hormones in the dormancy mechanism of celery seeds.  相似文献

The flowering ofSalvia splendens Sellow under noninductive short days is promoted by exogenous application of estrogen fraction isolated from flowering sage plants, gibberellin GA4+7 and to some extend N6-benzyladenine and estradiol. The persistence of gibberellin A3 on plant surfaces was examined using fruit of Marsh seedless grapefruit (Citrus paradisi Macf.) and an inert glass model system. 14C-gibberellin A3 was applied to surfaces in aqueous treatment solutions or in waxing solutions. Dried-out treatment residues were removed by washing and analyzed for total and GA3-like radioactivity. Gibberellin A3 persisted without significant loss for at least 7 d in aqueous treatment solutions (pH 4.0 or 6.2) but was less persistent in the pH 10.4 waxing solution (t1/2=7 d).Loss of total peel surface radioactivity was fast during the first 3 days, slowing down afterwards. After 14 days 73% of the initial radioactivity could still be recovered from fruit peel surface and 70% of the recovered radioactivity was still in the form of gibberellin A3. Gibberellin A3 was somewhat more persistent in residues from pH 4 than pH 7 treatment solutions. Light had a slight enhancing effect on gibberellin A3 decomposition on fruit peel under growth chamber conditions. After 12 d at 100% relative humidity, 88% of the radioactivity on glass surfaces was still in the form of gibberellin A3, as against 45% at a relative humidity of 50%. Simulated field conditions, combining daily fluctuations in light, temperature and relative humidity, markedly enhanced gibberellin A3 decomposition on glass surfaces (t1/2=2 d). A cytokinin photoaffinity reagent, 8-azido-N 6-benzyladenine (8N3BA), was synthesized from 8-bromoadenosine via azide replacement, benzylation at N–1, rearrangement to the N-6-benzyl derivative and acid hydrolysis. The compound thus obtained was found to have full cytokinin activity in the moss and tobacco cell-suspension bioassays. Photolysis of 8N3BA was accomplished with long and short-wavelength ultraviolet light and produced compounds which had very little or no biological activity in the two bioassays. In-vivo photolysis of 8N3BA caused loss of the cytokinin activity of this compound in moss protonemata. Kinetin at 10 mg l–1 increased the number of flowers produced on Rosa damascena plants while GA3 inhibited flowering. In the leaves of non-flowering plants GA-like activity was high while specific cytokinin activity (fraction-II) was significantly higher in flowering plants. A cell-free system prepared from developing seed of runner bean (Phaseolus coccineus L.) converted [14C]gibberellin A12-aldehyde to several products. Thirteen of these were identified by capillary gas chromatography-mass spectrometry as gibberellin A1 (GA1), GA4, GA5, GA6, GA15, GA17, GA19, GA20, GA24, GA37, GA38, GA44 and GA53-aldehyde, all giving mass spectra with 14C-isotope peaks. GA8 and GA28 were also identified but contained no 14C. Protoplasts isolated from embryogenic callus of Citropsis schweinfurthii (Engl.) Swing. & M. Kell were cultured in MT (Murashige and Tucker 1969) basal medium containing 5% sucrose supplemented with 0.0, 0.001, 0.01, 0.1 or 1.0 mg l–1 BA, 0, 300, 600 or 900 mg l–1 malt extract and 0.6 M sorbitol. The highest plating efficiency was obtained on MT basal medium containing 5% sucrose supplemented with 0.01 mg l–1 BA and 600 mg l–1 malt extract. MT basal medium containing 5% sucrose and supplemented with 0.01 mg l–1 kinetin was found to be a medium suitable for the development of globular somatic embryos derived from protoplasts into heart-shaped somatic embryos with cotyledon-like structures. The highest percentage of shoot formation was obtained using 0.1 mg l–1 GA3. Elongation growth and gibberellin (GA9) metabolism in excised hypocotyls of lettuce (Lactuca sativa L. cv. Arctic) were investigated. Exogenously supplied GA9 stimulates elongation of hypocotyl sections and this response is intermediate between that elicited by GA1 or GA20 and GA4/7 mixture. Although uptake of radioactivity from [3H]GA9 increases with time, this gibberellin does not accumulate in the tissue but is rapidly converted to a compound with HPLC properties resembling those of [3H]GA20. After 2 h incubation in [3H]GA9, the presumptive GA20 represents 90% of the acidic ethyl acetate-soluble radioactivity in the tissue. Radioactivity is also associated with an acidic butanol-soluble fraction containing two components resolvable by HVE. The major component is similar in electrophoretic properties to a GA-glucosyl ether while the other compares to a GA-glucosyl ester. Conversion of [3H]GA9 to its [3H]GA20-like metabolite is reduced by addition of carrier GA9 or GA4/7 at concentrations as low as 1 M, while GA1, GA3 and L-proline are without effect. Formation of the GA20-like compound can be blocked by the addition of 2,2-dipyridyl, and this inhibitory effect of dipyridyl can be reversed by addition of Fe2+. At 200 M dipyridyl, elongation growth as well as [3H]GA9 metabolism are reduced by 80%. The fate of [14C] gibberellin A3 and [3H] gibberellin A1 was examined in senescing fruit of Shamouti orange (Citrus sinensis L. Osbeck) and tomato (Lycopersicon esculentum Mill.). Gibberellin A3 was highly persistent in Citrus peel (t 1/2=18 days) and to a lesser degree in tomato (t 1/2=5.5 days). Ethylene and ethephon caused a slight enhancement of gibberellin A3 metabolism in Citrus and tomato fruit, respectively. Gibberellin A1 was metabolized by Citrus peel at a relatively high rate (t 1/2 < 24 h) and ethylene slightly reduced this rate. BA at 10–5 M, GA3 at 3×10–4 M or GA4+7 at 3×10–5 M partially or largely reversed the inhibition of Amaranthus caudatus seed germination due to JA-Me. BA or GA3 did not affect ethylene production and ACC oxidase activity in vivo in the presence of JA-Me before radicle protrusion. However, both increased ethylene production after 72 h of incubation, when the reversal of the JA-Me inhibition of seed germination was observed. AVG at 3×10–4 M decreased ethylene production when it was applied simultaneously with BA and JA-Me or GA3 and JA-Me, but it had no effect on seed germination. NBD almost completely reversed the stimulatory effect of BA, GA3 or GA4+7 on the germination of seeds in the presence of JA-Me. Exogenous ethylene reversed the inhibitory effect of NBD. The results indicate that action of endogenous ethylene is involved in the response of JA-Me inhibited seeds to BA or GAs.  相似文献

An antiserum against gibberellin A4 (GA4) raised in rabbits and its partially purified antibodies were used to develop radioimmunoassay (RIA) and indirect enzyme-linked immunosorbent assays (ELISAs) for GA4. Of three immunoassays tested, an ELISA based on the NAD-dependent redox cycle (enzyme-amplified ELISA) had highest sensitivity. Levels of methylated GA4 detected by this most sensitive method ranged from 0.1 fmol/assay (3.5 fg/assay) to 0.1 pmol/assay (3.5 pg/assay) suggesting applicability of this method to the detection of gibberellins in purified plant extracts.  相似文献
