Hemp (Cannabis sativa L.) is an emerging dioecious crop grown primarily for grain, fiber, and cannabinoids. There is good evidence for medicinal benefits of the most abundant cannabinoid in hemp, cannabidiol (CBD). For CBD production, female plants producing CBD but not tetrahydrocannabinol (THC) are desired. We developed and validated high‐throughput PACE (PCR Allele Competitive Extension) assays for C. sativa plant sex and cannabinoid chemotype. The sex assay was validated across a wide range of germplasm and resolved male plants from female and monoecious plants. The cannabinoid chemotype assay revealed segregation in hemp populations, and resolved plants producing predominantly THC, predominantly CBD, and roughly equal amounts of THC and CBD. Cultivar populations that were thought to be stabilized for CBD production were found to be segregating phenotypically and genotypically. Many plants predominantly producing CBD accumulated more than the current US legal limit of 0.3% THC by dry weight. These assays and data provide potentially useful tools for breeding and early selection of hemp.  相似文献

When shoots of young plants of hemp (Cannabis sativa L.) and spinach (Spinacea oleracea L.) were cultured as cuttings and allowed to regenerate advenitious roots, ca. 80–85% became female (formed pistillate flowers) regardless of whether the leaves were left on the plants or were cut off (except for the 2–3 uppermost ones) after the beginning of adventitious-root formation. But when the leaves were cut off and the cuttings treated with gibberellic acid (GA3, 25 mg/l) ca. 77–80% of the plants became male (formed staminate flowers). The result was quite similar when roots and leaves of young hemp plants were removed at the same time and the cuttings treated with GA3. It is suggested that the leaves play an essential role in sex expression in hemp and spinach and that this role is related to gibberellin synthesis in the leaves.  相似文献

When the shoots of young hemp (Cannabis sativa L.) plants were cut off the roots, cultured as cuttings, and regenerating (adventitious) roots were removed as soon as appearing, ca. 80–90% of the plants became male (had staminate flowers) whereas if the roots were allowed to develop a similar percentage became female (pistillate flowers). Treatment of de-rooted cuttings with 6-benzylaminopurine (15 mg/l) restored the percent of female plants to ca. 80. It is suggested that the root system plays an essential role in sex expression in hemp and that this role is related to cytokinin synthesis in the root.  相似文献

The influence of abscisic acid (ABA) on plastidial and cytosolic terpenoids and on two key enzymes for terpenoid biosynthesis was determined in vegetative stage of Cannabis sativa L. Low concentration of ABA (1 μM) increased 1-deoxy-D-xylulose 5-phosphate synthase (DXS) activity in treated plants in comparison to control plants. The amounts of chlorophyll a and carotenoids increased in response to ABA treatment but chlorophyll b content declined. The accumulation of α-tocopherol was stimulated only by 10 μM ABA. The ABA-treated plants showed a decline in 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) activity which was followed by a decrease in squalene and phytosterol content. ABA also decreased tetrahydrocannabinol (THC) and cannabidiol (CBD) contents. The essential oil had higher ratios of monoterpenes to sesquiterpenes as ABA-treated plants had less numbers of sesquiterpenes in comparison with control plants. Influence of ABA on the amounts of sesquiterpenes was different, some of them showed decrease of content and others increase of content.  相似文献

Here we demonstrate for the first time that cannabidiol (CBD) acts to protect synaptic plasticity in an in vitro model of Alzheimer’s disease (AD). The non-psycho active component of Cannabis sativa, CBD has previously been shown to protect against the neurotoxic effects of beta amyloid peptide (Aβ) in cell culture and cognitive behavioural models of neurodegeneration. Hippocampal long-term potentiation (LTP) is an activity dependent increase in synaptic efficacy often used to study cellular mechanisms related to memory. Here we show that acute application of soluble oligomeric beta amyloid peptide (Aβ1–42) associated with AD, attenuates LTP in the CA1 region of hippocampal slices from C57Bl/6 mice. Application of CBD alone did not alter LTP, however pre-treatment of slices with CBD rescued the Aβ1–42 mediated deficit in LTP. We found that the neuroprotective effects of CBD were not reversed by WAY100635, ZM241385 or AM251, demonstrating a lack of involvement of 5HT1A, adenosine (A2A) or Cannabinoid type 1 (CB1) receptors respectively. However in the presence of the PPARγ antagonist GW9662 the neuroprotective effect of CBD was prevented. Our data suggests that this major component of Cannabis sativa, which lacks psychoactivity may have therapeutic potential for the treatment of AD.

 A 400-bp RAPD marker generated by a primer of random decamer sequence has been found associated with the male sex phenotype in 14 dioecious cultivars and accessions of hemp (Cannabis sativa L.). The primer OPA8 generates a set of bands, most of which polymorphic among all the individual plants tested, and 1 of which, named OPA8400, present in all male plants and absent in female plants. A screening of 167 plants belonging to different genotypes for the association of the OPA8400 marker with the sex phenotype revealed that only in 3 cases was the 400-bp band was present in plants phenotypically female; on the contrary, in male plants the band was never missing, while in monoecious plants it was never present. Despite this sex-specific association, the sequences corresponding to OPA8400 were present in both staminate and carpellate plants, as revealed by Southern blotting and hybridization with the cloned RAPD band. The RAPD marker was sequenced, and specific primers were constructed. These primers generated, on the same genotypes used for RAPD analysis, a SCAR marker 390 bp in length and male-specific. This SCAR is suitable for a precise, early and rapid identification of male plants during breeding programs of dioecious and monoecious hemp. Application, through the root system, of growth regulators to hemp (Cannabis sativa L.) plants having 2–3 pairs of visible leaves caused pronounced shifts of sex expression in the adult individuals. Treatment with gibberellic acid (25 mg/l) resulted in more than 80% of the plants being male, i.e. having staminate flowers (controls, ca. 30%). Treatment with 6-benzylaminopurine and with indole-3-acetic acid (in either case, 15 mg/l) resulted in all plants being either female (pistillate flowers) or intersexes (bisexual flowers); treatment with abscisic acid (10 mg/l) had a similar but somewhat less pronounced effect.  相似文献

Conditioning of Heavy Metal-Polluted River Sediment by Cannabis sativa L.   总被引:1,自引:0,他引:1  
We are currently developing a process for the remediation of heavy metal-polluted sediment by solid-bed bioleaching. Unfortunately, freshly dredged sediment is impermeable to water, unsuitable for solid-bed leaching, and therefore needs preliminary conditioning to improve its structure. A pilot-scale conditioning plant consisting of several basins (each with a base of 6 m2 and a depth of 0.7 m) was filled with freshly dredged, slightly predewatered sediment from the Weisse Elster River. Hemp (Cannabis sativa L.) was chosen for conditioning because it develops quickly from seeds, produces large amounts of biomass, and forms commercially useful fibers. Irrespective of the plant cover, the sediment dried out, was oxidized and turned acidic. However, its structure and permeability only improved when the sediment was covered with plants and not when all the vegetation appearing was removed regularly. Although hemp germinated completely regularly on sediment, 1 week after seeding the leaves became partially discolored, deformed, and dry, and ultimately about 95% of all the hemp plants died. The surviving hemp produced shorter shoots and much lower amounts of biomass than hemp on unpolluted soil. Studies in pots and hydrocultures indicated that the inhibition of hemp growth is caused by the synergistic action of a low pH and several mobile toxic metals. Hemp on sediment incorporated much higher quantities of heavy metals (especially Zn, Cd, and Ni) than hemp on unpolluted soil, which calls the commercial use of the former seriously in question.  相似文献

Cannabis sativa is an interesting crop for several industrial uses, but the legislations in Europe and USA require a tight control of cannabinoid type and content for cultivation and subsidies release. Therefore, cannabinoid survey by gas chromatography of materials under selection is an important step in hemp breeding. In this paper, a number of Cannabis accessions were examined for their cannabinoid composition. Their absolute and relative content was examined, and results are discussed in the light of both the current genetic model for cannabinoid’s inheritance, and the legislation’s requirements. In addition, the effectiveness of two different types of markers associated to the locus determining the chemotype in Cannabis was evaluated and discussed, as possible tools in marker-assisted selection in hemp, but also for possible applications in the forensic and pharmaceutical fields.  相似文献

Two populations of Sagittaria brevirostra from the same lake were sampled 10 years apart and yielded similar data on inflorescence structure and on numbers and ratios of male and female flowers. Larger inflorescences have relatively more male than female flowers than do smaller inflorescences. Experiments in semi-natural conditions were undertaken to assess hemp metal tolerance and its ability to accumulate cadmium, nickel and chromium. Cannabis sativa was grown in two soils, S1 and S2, containing 27, 74, 126 and 82, 115, 139 g g–1 of Cd, Ni and Cr, respectively. After two months from germination and at ripeness, no significant alteration in plant growth or morphology was detected. On the contrary, a high hemp reactivity to heavy metal stress with an increase in phytochelatin and DNA content was observed during development, suggesting the Cannabis sativa ability to avoid cell damage by activating different molecular mechanisms. Metals were preferentially accumulated in the roots and only partially translocated to the above-ground tissues. The mean shoot Cd content was 14 and 66 g g–1 for S1 and S2 soil, respectively. Although not negligible concentrations they were about 100 times lower than those calculated for the hyperaccumulator Thlaspi caerulescens. Similarly Ni uptake was limited if compared with that of the Ni-hyperaccumulator Alyssum murale. Chromium uptake was negligible. As expected on the base of the metal concentration detected in ripe plants, no statistically significant variation in soil metal content was detected after one crop of hemp. Nevertheless, a consistent amount (g) of Cd and Ni is expected to be extracted by 1 ha biomass of hemp (about 10 t) per year and along the time a slow restoration of deeper soil portions can be obtained by its wide root system (at least 0,5 m deep). In addition, the possibilities of growing hemp easily in different climates and using its biomass in non-food industries can make heavy metal contaminated soils productive. Selective breeding has produced cannabis plants for specific uses, including high-potency marijuana strains and hemp cultivars for fiber and seed production. To assess variation in the proportion of self‐fertilized seeds among flowers within inflorescences and the relationship between floral traits and the rate of self‐fertilization, the proportion of self‐fertilized seeds among individual flowers was estimated using ten microsatellite markers in self‐compatible plants of Aquilegia buergeriana var. oxysepala. Within‐inflorescence variation in floral traits, such as the duration of the male and female phases, flower size, herkogamy and the number of pollen grains and ovules in two natural populations, were investigated. The first flower in an inflorescence produced more seeds and a higher proportion of self‐fertilized seeds than the second flower. The higher proportion of self‐fertilized seeds in the first flowers was accompanied by a higher number of pollen grains and ovules in the bud stage and the female phase. These results indicate that the high proportion of self‐fertilized seeds in the first flowers in an inflorescence may be due to the high number of remaining pollen grains in the female phase. This suggests that variation in floral traits within inflorescences affects seed quality and quantity among flowers within inflorescences.  相似文献

PurposeColon cancer is a major public health problem. Cannabis-based medicines are useful adjunctive treatments in cancer patients. Here, we have investigated the effect of a standardized Cannabis sativa extract with high content of cannabidiol (CBD), here named CBD BDS, i.e. CBD botanical drug substance, on colorectal cancer cell proliferation and in experimental models of colon cancer in vivo.MethodsProliferation was evaluated in colorectal carcinoma (DLD-1 and HCT116) as well as in healthy colonic cells using the MTT assay. CBD BDS binding was evaluated by its ability to displace [3H]CP55940 from human cannabinoid CB1 and CB2 receptors. In vivo, the effect of CBD BDS was examined on the preneoplastic lesions (aberrant crypt foci), polyps and tumours induced by the carcinogenic agent azoxymethane (AOM) as well as in a xenograft model of colon cancer in mice.ResultsCBD BDS and CBD reduced cell proliferation in tumoral, but not in healthy, cells. The effect of CBD BDS was counteracted by selective CB1 and CB2 receptor antagonists. Pure CBD reduced cell proliferation in a CB1-sensitive antagonist manner only. In binding assays, CBD BDS showed greater affinity than pure CBD for both CB1 and CB2 receptors, with pure CBD having very little affinity. In vivo, CBD BDS reduced AOM-induced preneoplastic lesions and polyps as well as tumour growth in the xenograft model of colon cancer.ConclusionsCBD BDS attenuates colon carcinogenesis and inhibits colorectal cancer cell proliferation via CB1 and CB2 receptor activation. The results may have some clinical relevance for the use of Cannabis-based medicines in cancer patients.  相似文献

The chronology of Phomopsis ganjae conidia germination and infection of Cannabis sativa leaves was observed with the scanning electron microscope. A-conidia germination approached 100% after 24 h, appresoria initiation began after 36 h; B-conidia germinated by 52 h but were not infective. Four-week-old C. sativa seedlings were more susceptible than 16-week-old plants, males more than females. THC and CBD, extracted and separated via TLC, inhibited P. ganjae conidia germination and hyphal growth.  相似文献

Marijuana (Cannabis sativa L.) was sampled at nine progressive growth stages in Riley County, Kansas, and analyzed for four major cannabinoids: cannabidiol (CBD), della-8-tetrahydrocannabinol (delta-8-THC), delta-9-tetrahydrocannabinol (delta-9-THC), and cannabinol (CBN). Seasonal fluctuation in cannabinoids were related to stage of plant development. Cannabinoids were lowest in seedlings, highest prior to flowering and at an intermediate level thereafter until physiological maturity. Cannabinoids were highest in flowers and progressively lower in leaves, petioles, stems, seeds, and roots. Cannabinoid content of male and female flowers was not significantly different. Cannabidiol occurred in the highest concentrations (0.01 to 0.94% of dry matter) in all plant parts; delta-9-THC, the next highest (0.0001 to 0.06%) in the study over time. Cannabidiol content of leaf tissue of plants sampled from ten locations at flowering, ranged from 0.12 to 1.7%; delta-9-THC, from 0.01 to 0.49%. Some variation was attributed to environmental factors. Results indicate transformation of CBD to delta-9-THC to CBN. Environmental stress apparently increased delta-9-THC concentration, and bivalent ions: Mg, Mn, and Fe of leaf tissue could have regulated enzyme systems responsible for cannabinoid synthesis.  相似文献
