The potato tuber moth (PTM) is an oligophagous herbivore and a severe pest of solanaceous crops in many countries of the world. Previously, we reported host expansion and damage potential of PTM on tomato, a congeneric crop of potato. Here we tested adult olfactory behaviour of PTM to leaves of five different cultivated tomato varieties including Moneymaker, Campari, Ailsa craig, LA3475 and E6203, and one wild species, Solanum pimpinellifolium. Tomato leaf hydro-distilled oils of Moneymaker, Campari, Ailsa craig, S. pimpinellifolium and E6203 showed strong attractiveness and LA3475 exhibited repulsiveness for adult PTM of both sexes in two-armed bioassays. Volatiles of Moneymaker, Campari, Ailsa Craig, S. pimpinellifolium and E6203 showed attractiveness for mated adult PTM of both sexes (> 70%). The extracted oil of all tomato leaves contains monoterpenes, sesquiterpenes, diterpenes and C6 alcohols as well as other components. Phytol is one of the common and major compounds in all tomato varieties. Phytol showed weak attractiveness: 60.8–63.6% and 57.6–60.6% for male and female PTM, respectively when tested at concentrations of 0.1–10 mg/mL. Therefore, the attractiveness of tomato leaf volatiles might be due to synergistic effects of plant volatile mixtures. Identification PTM attractant volatile in tomato leaf is important in its control in environmentally friendly manner.  相似文献

Potato tuber moth (PTM) is a primary pest of potato which had invaded into Korea in 1968 and its distribution was restricted in the southern part of the peninsula. Possibly due to the climate change, its distribution expanded toward the Northern part of South Korea including Gyounggi and Gangwon province where tomato production is high. Host expansion and damage potential of PTM was evaluated on tomato, a congeneric crop of potato. Three commercial varieties of tomato (Dafnis, Bangul and Jujube) and a potato (Superior-Sumi) were subjected to PTM larvae. The results showed that host suitability of tomato to PTM was lower than potato, but PTM could completely develop and reproduce on tomato leaves or fruits. Development time was varied relative to tomato varieties and plant age. PTM showed lower performance on the leaves and fruits of cherry type tomatoes. Higher survival was observed in the large sized fruit of tomato varieties. This study showed substantial evidence that the invaded PTM could damage the tomato cultivation.  相似文献

The potato tuber moth (PTM),Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), is a major pest of processing tomatoes,Lycopersicon esculentum Mill. (Solanaceae), in Israel. The larvae penetrate the tomato fruit through the stem end and present a serious threat to crop quality. Foliage and fruit samples were taken in nine commercial tomato fields located in Israel's three main tomato growing areas, two of which are potato growing areas as well. PTM was not found where potatoes were absent. Potato harvest in nearby fields was found to be the most significant factor affecting seasonal trends in PTM population density in tomatoes. All four larval instars were found in foliage on all sampling dates. Significantly higher proportions of first instars were found during the population density increase which followed potato harvest. Damaged fruits did not contain first instar larvae, indicating that PTM never undergoes complete development within tomato fruit. Fruit damage levels at harvest were positively correlated to the peak mean population densities on foliage and the date they were observed. In tomato fields not adjacent to potatoes, infestation was first observed at the edge of the field. Both before and after the potato harvest in nearby fields, population density at the edge of the field was significantly higher than at the center. In tomato fields adjacent to potatoes, no significant differences were found between population densities at the edge and center before the potatoes were harvested. After the potato harvest, population density at the center of tomato fields was higher than at the edge. Deceased, October 1988  相似文献

Wild‐type and ACC‐oxidase antisense tomato plants ( Lycopersicon esculentum Mill. cv. Ailsa Craig and pTOM13) were grown in environment‐controlled rooms for 21 days under photoperiods of 8, 16 or 23.5 h at an irradiance of 300 µmol m−2 s−1. Photosynthetic pigments, photosynthesis, soluble carbohydrates, starch and ethylene were measured on the last fully expanded leaf. Increasing the photoperiod from 8 to 16 h stimulated all measured growth parameters in both cultivars. However, when the photoperiod was increased to 23.5 h, foliar yellowing and deformation were observed in the wild‐type Ailsa Craig whereas no change was observed in pTOM13. It was not possible to relate these foliar changes in Ailsa Craig to destruction of the photosynthetic apparatus by excess carbohydrate levels in the leaves. Because pTOM13 was antisense to ACC‐oxidase. it is proposed that yellowing and deformation in leaves of wild‐type tomato plants grown under long photoperiods may be caused by stress ethylene induced by a long photoperiod.  相似文献

The effect of plant age and daylength on glandular pubescence was determined for two lines of tomato derived from Lycopersicon hirsutum (BTN 979 and LA 1777A) and a variety of L. esculentum (N 91‐1‐1‐1‐1). Densities of type I, IV, VI and VII glandular trichomes were lowest in N 91‐1‐1‐1‐1 and, over all varieties, were more dense on plants aged greater than 6 wk. Daylength interacted with variety to significantly affect densities of type VII trichomes only. Host‐plant resistance to Phthorimaea operculella was determined in preliminary tests using insects cultured from founders from a potato crop and in confirmatory tests using (less readily available) insects recovered from foliage of a tomato crop. Mortality of ex‐potato neonates on LA 1777 A and BTN 979 foliage was higher 18 h after placement than for N 91‐1‐1‐1‐1, with no effect of day length or plant age. Mortality for ex‐tomato neonates followed a similar trend. Ten days later, two‐thirds of ex‐tomato larvae had established mines on N 91‐1‐1‐1‐1 but fewer (16.7%) were live on other varieties. Stepwise multiple regression using variety as the sole factor was significant (P<0.001) in accounting for 61.4% of the variation in ex‐tomato larval survival but addition of other factors to the regression model was not significant. BTN 979 supported fewer, smaller adults to develop than did N 91‐1‐1‐1‐1, whilst no adults developed on LA 1777 A. In a non‐choice test using ex‐potato adults, significantly more eggs were laid on N 91‐1‐1‐1‐1 than on L. hirsutum varieties and 9‐wk‐old plants were preferred over plants three weeks older or younger. The same variety and plant age trends were evident in a free‐choice test using ex‐tomato adults.  相似文献

Abstract

A series of laboratory experiments was conducted to investigate the nature of the factors which determine acceptance or rejection of plants for oviposition by potato tuber moth. Bare leaves of only two plant species (potato and egg-plant) were accepted for oviposition, but when covered with a layer of muslin, leaves of all plants examined—including several known non-host plants—were rendered acceptable. Thus, volatile repellent factors are not possessed by unacceptable plants; rather, they must be rejected either because of chemical deterrents detectable only on contact or because of unsuitably textured surfaces. Plant juices expressed on to a covering layer of muslin increased the acceptance of potato and egg-plant, had no effect with tobacco, and decreased acceptance of all known non-host plants and of tomato. Brushing the leaf surface without releasing plant juices induced a detectable deterrent effect with some plant species. There was no stimulation or deterrence when the oviposition surface was separated from expressed plant juices by about 2 mm. Tomato reacted like known non-host plants in all tests. Potato and egg-plant stimulated greater total egg deposition in most experiments, but strongly deterrent treatments did not reduce the total egg lay. Antennectomy reduced but did not eliminate the stimulatory effect of potato and the deterrent effect of bean plant juices. Virgin moths were not stimulated to oviposit by potato peel. It is concluded that selection of plants for oviposition is determined both by the physical nature of their surfaces and by chemical factors which are detected only on contact. The effects of stimulative chemical factors in host plants and deterrent factors in non-host plants reinforce each other in the plant selection process.  相似文献

This study tested the hypothesis that antagonistic interactions between abscisic acid (ABA) and ethylene mediate the effects of soil compaction on shoot growth. Isogenic wild‐type (Ailsa Craig), ABA‐deficient (notabilis) and a transgenic (ACO1AS) tomato genotype with a reduced capacity to synthesize ethylene were examined. Exogenous ABA was also applied. Leaf area was comparable when Ailsa Craig and ACO1AS were grown in uncompacted (1·1 g cm?3) or compacted (1·5 g cm?3) soil, but was lower in notabilis. However, a 1·1/1·5 g cm?3 split‐pot treatment invoked marked genotypic differences, whereby leaf area was comparable to 1·1 g cm?3 control plants in ACO1AS but was intermediate between the 1·1 and 1·5 g cm?3 treatments in Ailsa Craig and notabilis. ABA may be discounted as the root‐sourced signal responsible for reducing leaf area when the roots encountered compacted soil as Ailsa Craig and ACO1AS showed differing responses despite similar increases in xylem sap ABA concentration; leaf area was invariably lower in notabilis. These genotypic differences were correlated with ethylene evolution; thus the greater leaf area in ACO1AS was associated with its reduced ability to synthesize ethylene, whereas the reductions in leaf expansion observed when Ailsa Craig and notabilis encountered compacted soil were accompanied by increased ethylene production. Application of ABA had little effect on ACO1AS, but promoted a recovery of leaf expansion in notabilis, and more surprisingly in Ailsa Craig. These results suggest that antagonistic interactions between ABA and ethylene may regulate leaf expansion when the root system simultaneously encounters uncompacted and compacted soil.  相似文献

The tomato leafminer, Tuta absoluta (Lepidoptera: Gelechiidae), is considered to be a major pest that damages tomato (Solanum lycopersicum L; Solanaceae) crops in South American, European, and Mediterranean countries. This insect species is polyphagous (i.e., feeds on many types of food); hence, it could also develop on other cultivated host plants, principally solanaceous plants, such as potato (S. tuberosum L.; Solanaceae) and eggplant (S. melongena L.; Solanaceae). Therefore, we tested the hypothesis that host plant choice by adult T. absoluta is influenced by plant volatile organic compounds and larval host plant experience. One tomato cultivar (cv.) ‘Moneymaker’ and three potato cv. ‘Charlotte’ ‘Bintje,’ and ‘Nicola’ were tested. Using a flying tunnel, we observed that females reared on tomato preferred flying toward tomato and, to a lesser extent, potato cv. ‘Charlotte.’ These preferences might be explained by the high release of terpenes by these two plants. When conducting oviposition choice assays, we found no preference between tomato and potato in the number of eggs laid by females that had been previously reared on either host plant. This study indicates that the host finding behavior of T. absoluta is mediated by solanaceous volatiles, while oviposition behavior appears to depend on additional stimuli. These results provide baseline information for use in the development of new control strategies against T. absoluta using semiochemicals and plant breeding.  相似文献

Accessions of wild Lycopersicon spp. and selected Fl hybrid tomato cultivars were compared for their resistance to three whitefly-transmissible geminiviruses: Indian tomato leaf curl virus (ITmLCV) and tomato yellow leaf curl viruses from Sardinia (TYLCV-Sar) and Senegal (TYLCV-Sen). The resistance of different plant lines was expressed in different ways but in most instances a given line reacted similarly to graft inoculation with the three viruses. L. pimpinellifolium LA1478 produced as much virus antigen, assessed by triple antibody sandwich-ELISA, as the susceptible cv. Moneymaker but developed only very mild symptoms and is therefore tolerant of infection. In L. hirsutum LA1777 and L. peruvianum CMV-INRA, very mild or no symptoms developed but antigen concentrations were substantially less than in Moneymaker. L. chilense LA1969 remained symptomless and its antigen concentration was < 1% of that in Moneymaker. Symptoms were mild or barely evident in the Fl hybrid cultivars. Cultivars Tyking and Fiona had antigen concentrations about 5–10% of those of Moneymaker, whereas TY20, Top 21 and Tyger had intermediate antigen concentrations. In a few instances, the extent to which virus accumulation was restricted depended on the challenge virus. Accumulation of TYLCV-Sen in TY20, Top 21 and Tyger was less affected than that of the other two viruses, and accumulation of TYLCV-Sar in accessions LA1777 and CMV-INRA was less affected than that of TYLCV-Sen or ITmLCV. Tissue-printing tests showed that ITmLCV and TYLCV-Sen antigens were confined to phloem tissue. In Tyking, the number of virus antigen-containing phloem traces and the antigen content of individual traces were less than in Moneymaker but the partitioning of antigen between internal and external phloem was unaffected.  相似文献

An important strategy for obtaining a safer transgenic plant may be the use of a spatial- or tissue-specific promoter, instead of a constitutive one. In this study, we have used a light-inducible maize PEPC promoter to regulate the cry1Ab gene, aiming to produce transgenic potatoes that are resistant to potato tuber moth (PTM) (Phthorimaea operculella, Zeller). Out of 60 regenerated lines having normal phenotypes, 55 lines were PCR-positive for both the cry1Ab and nptII genes. Southern analysis on three selected putative transgenic lines revealed that they have only a single intact copy of the cry1Ab gene. An investigation of the Cry1Ab protein in the leaves and light-exposed (LE) tubers of the transgenic lines demonstrated the presence of the protein in the foliage and green tubers but not in the light-not exposed (LNE) tubers. A bioassay analysis of excised leaves of nine randomly selected lines showed that eight lines had 100% PTM larval mortality. Confirming results were obtained in six selected lines using the whole plant bioassay in the greenhouse. LE transgenic tubers also exhibited 100% larval mortality; however, the levels of damage to the LNE transgenic tubers were high and statistically the same as those incurred by the non-transgenic ones. Based on the results, we believe that this spatial expression of Cry1Ab using the light-inducible PEPC promoter can control PTM infestation in the field and significantly reduce pollution transmission to storage potatoes.  相似文献

In this study a Brazilian granulovirus strain, PhopGV, isolated from the potato tuber moth (PTM) Phthorimaea operculella, was investigated regarding its potential for biological control and in vivo production. The relationship between mortality of P. operculella larvae and virus concentration was determined at different temperatures on potato tubers and susceptibility of P. operculella to PhopGV was also determined on potato leaves. Virulence of PhopGV to P. operculella was not affected by temperatures from 18 to 30°C. The median lethal concentration (LC50) of larvae fed on potato foliage treated with PhopGV was not higher than that verified with larvae fed on treated tubers. Optimal conditions for production of virus-infected larvae were obtained by using the virus suspensions of 41 × 105, 6.3 × 105 and 62 × 105 OBs ml−1 at 18, 24 and 30°C, which resulted in 32.0, 31.4 and 34.8% of infected larvae collected, respectively. The maximum percentage of infected larvae recovered from tubers was not affected by temperature. However, time for production of virus-infected larvae was longer at 18°C and shorter at 30°C. Persistence of PhopGV was determined on stored tubers and we observed that the virus remained effective for at least two months, causing up to 84.2% mortality of P. operculella at 1 × 107 OBs ml−1. The pathogen was also highly virulent to tomato pinworm, Tuta absoluta, inflicting high percentage of mortality, delaying larval growth and inhibiting pupation. This Brazilian PhopGV strain has potential to control PTM larvae on potato tubers at a broad range of temperature and can be produced in vivo using virus-treated tubers.  相似文献

Levels of endogenous abscisic acid (ABA) in wild type were not required for the synthesis of heat shock proteins in detached leaves of tomato (Lycopersicon esculentum Mill., cv Ailsa Craig). Heat-induced alterations in gene expression were the same in the ABA-deficient mutant of tomato, flacca, and the wild type. Heat tolerance of the mutant was marginally less that the wild type, and in contrast, ABA applications significantly reduced the heat tolerance of wild-type leaves. It was concluded that elevated levels of endogenous ABA are not involved in the tomato heat shock response.  相似文献

Ditylenchus destructor and Ditylenchus dipsaci are economically important plant‐parasitic nematodes, affecting potato production mostly in temperate climates. Management through crop rotation is not feasible because of their wide host range. These nematodes are listed as quarantine pests in many countries. Limited information exists on the resistance and tolerance of currently cultivated potatoes to D. destructor and D. dipsaci. Two greenhouse experiments were conducted to screen 25 potato varieties for resistance to and tolerance for D. destructor and D. dipsaci infections. Reproduction factor (RF) and relative susceptibility (RS) were used to evaluate resistance, while potato tuber damage and tuber weight reduction was used to evaluate tolerance. Based on the RF, 16 varieties were evaluated as susceptible (S) while 5 varieties were evaluated as resistant (R) to D. destructor; varieties ‘Innovator’, ‘Aveka’ and ‘Spunta’ were resistant to D. dipsaci based on RF. ‘Désirée’ was observed to be highly susceptible to D. destructor and D. dipsaci in both experiments and was used as the standard susceptible control variety for the calculation of RS. A scale of 1–9 was used to classify RS of the potato varieties to D. destructor and D. dipsaci, where 9 indicated the highest level of resistance. All classes of resistance to D. destructor and D. dipsaci were observed in the potato varieties tested in the experiments. Six varieties had significantly lower RS to D. dipsaci than the standard susceptible control variety. Tolerant to highly sensitive potato varieties to both nematodes were also observed. RS and external potato tuber damage were identified as suitable methods for resistance and tolerance determination, respectively. This study provides essential information on the status of resistance and tolerance in potato varieties against D. destructor and D. dipsaci but needs to be confirmed under field conditions.

To probe the role of xanthophylls in non-photochemical quenching (NPQ) and the compensatory acclimated photoprotection mechanisms, a tomato (Lycopersicon esculentum Mill. cv. Ailsa Craig) Xa mutant with deficit in lutein (L) and neoxanthin (N) contents was used. The Xa mutant showed lowered NPQ, an increased degree of de-epoxidation state [(A+Z)/(V+A+Z)], and decreases of photosystem 2 (PS2) antenna size. Although the Xa mutant had a CO2 assimilation rate similar to that of Ailsa Craig, it exhibited a much larger stomatal conductance (g s) than Ailsa Craig. Decreased electron flux in PS2 (J PS2) for the Xa mutant was associated with electron flux for photorespiratory carbon oxidation (J o) and alternative electron flux in PS2 (J a) while electron flux for photosynthetic carbon reduction (J c) was not different from Ailsa Craig. Moreover, the Xa mutant also exhibited higher activities of antioxidant enzymes, higher contents of ascorbate and glutathione, and lower contents of reactive oxygen species. Hence some compensatory acclimated mechanisms of photoprotection operated properly in the lack of NPQ and xanthophylls.  相似文献

The influence of host plant on population dynamics of an invasive pest, Tuta absoluta was studied on three economically important solanaceous crops. Experiments were conducted in laboratory (29 ± 0.5°C, 75 ± 5% RH and a photoperiod of 14:10 hr [L:D]) using tomato (Solanum lycopersicum L.), potato (Solanum tuberosum L.) and eggplant (Solanum melongena L.). Results indicated that intrinsic rate of increase (r), finite rate of increase (λ) and net reproductive rate (R0) were higher, and mean generation time (T) was the shortest on tomato. Results suggested that T. absoluta developed on all the three plants, and tomato plant was most preferred one. Results suggested that T. absoluta has a potential to become a serious pest on potato and even on eggplant under favourable conditions. We used the life tables of 0.025th and 0.975th percentiles of bootstraps to project the uncertainty of population growth, a new concept.  相似文献

The role of ethylene in regulating growth in tomato (Lycopersicon esculentum Mill.) during compaction stress was examined using wild-type (cv Ailsa Craig) and transgenic (ACO1AS) genotypes; the latter has a reduced capacity to produce ethylene. Ethephon or silver ions were applied to increase ethylene production or block its action. Shoot growth in both genotypes was comparable in uncompacted (1.1 g cm−3) and uniformly compacted soil (1.5 g cm−3). However, a 1.1/1.5-g cm−3 split-pot treatment invoked marked genotypic differences: growth was reduced in cv Ailsa Craig but was comparable to uncompacted control plants in ACO1AS. As xylem sap abscisic acid levels were similar, abscisic acid was not responsible for inhibiting growth in cv Ailsa Craig. These genotypic differences in growth were accompanied by increased ethylene evolution in cv Ailsa Craig, suggesting that the ability of ACO1AS to maintain growth in the split-pot treatment reflected its lower ethylene levels, a view supported by the observation that excising the roots in the compacted compartment reduced ethylene evolution and restored shoot growth in cv Ailsa Craig. Treatment with silver restored shoot growth in cv Ailsa Craig, whereas treatment with ethephon reduced growth in ACO1AS. Thus, ethylene apparently has a key role in determining growth when tomato plants encounter differential soil compaction.  相似文献

A cDNA library produced from mRNA isolated from the pericarp of wild-type tomato fruit (Lycopersicon esculentum Mill. cv Ailsa Craig) at the first visible sign of fruit ripening was differentially screened to identify clones whose homologous mRNAs were present at reduced levels in fruit of the tomato ripening mutant, ripening inhibitor, rin. Five clones were isolated (pERT 1, 10, 13, 14, 15). Accumulation of mRNA homologous to each of these clones increased during the ripening of wild-type fruit and showed reduced accumulation in ripening rin fruit. The levels of three of them (homologous to ERT 1, 13 and 14) were increased by ethylene treatment of the mutant fruit. A further clone, ERT 16 was identified for a mRNA present at a high level in both normal and mutant fruit at early stages of ripening. Database searches revealed no significant homology to the DNA sequence of ERT 14 and 15; however, DNA and derived amino acid sequence of ERT 1 both contain regions of homology with several reported UDP-glucosyl and glucuronosyl transferases (UDPGT) and with a conserved UDPGT motif. A derived amino acid sequence from the ERT 10 cDNA contains a perfect match to a consensus sequence present in a number of dehydrogenases. The ERT 13 DNA sequence has homology with an mRNA present during potato tuberisation. The presence of these mRNAs in tomato fruit is unreported and their role in ripening is unknown. The ERT 16 DNA sequence has homology with a ripening/stress-related cDNA isolated from tomato fruit pericarp.  相似文献

Strong viroid-caused pathogenesis was achieved in tomato cv. Rutgers by biolistic transfer of severe or lethal potato spindle tuber viroid (PSTVd) strains, while other tomato genotypes (e.g., Moneymaker) were tolerant. With reciprocal hybrids between sensitive and tolerant genotypes, we show that plant depression dominates over tolerance. Biolistic transfer of the most pathogenic PSTVd strain AS1 to Nicotiana benthamiana, which is considered to be a symptomless PSTVd host, led to a strong pathogenesis reaction and stunting, suggesting the presence of specific viroid pathogenesis-promoting target(s) in this plant species. Total levels of small siRNA-like PSTVd-specific RNAs were enhanced in strongly symptomatic tomato and N. benthamiana plants after biolistic infection with AS1 in comparison to the mild QFA strain. This indicates association of elevated levels of viroid-specific small RNA with production of strong symptoms. In symptom-bearing tomato leaves in comparison to controls, an RNase of approximately 18 kDa was induced and the activity of a nuclease of 34 kDa was elevated by a factor of seven in the vascular system. Sequence analysis of the nuclease cDNA designated TBN1 showed high homology with plant apoptotic endonucleases. The vascular-specific pathogenesis action is supported by light microscopic observations demonstrating a certain lack of xylem tissue and an arrest of the establishment of new vascular bundles in collapsed plants.  相似文献
