Variation in nrlTS sequences among seven members of a seemingly natural group of Saxifraga (the S. pentadactylis complex; series Pentadactyles ) is phylogenetically analyzed. The two most parsimonious trees indicate that the S. pentadactylis complex is not a monophyletic group, because 5. intricata is excluded and is most closely related to S. exarata (series Cespitosae ). A well-defined lineage includes, however, six of the seven members of the S. pentadactylis complex. The tree topology suggests that this remaining group of six taxa may have diverged in the northeastern part of the Iberian Peninsula. Two sublineages can be identified from the ITS phylogenetic reconstruction: S. losae-S.moncayensis and the three subspecies of S. pentadactylis (subspp. pentadactylis, willkommiana , and almanzorii ). The relationships between S. losae and S. moncayensis provide an example of north-south speciation along the Sistema Ibérico mountain range. The three subspecies of S. pentadactylis form a monophyletic group resolved into one clade containing subspp. willkommiana and almanzorii , sister to subsp. pentadactylis. The molecular evidence agrees with chorological, cytotaxonomical, chemosystematic, and reproductive data indicating two differentiation patterns within S. pentadactylis. Transition to predominant autogamy in subsp. almanzorii probably resulted in rapid reproductive isolation from the outcrossing subsp. willkommiana in part of the Sistema Central mountain range, as suggested by their two distinct floral types but similar ITS sequences. Allopatric separation of populations of subsp. willkommiana from subsp. pentadactylis (the Pyrenees) has resulted in molecular divergence and some morphological differentiation.  相似文献

Adult roach, bream and their presumed F1 hybrid from an Anglian Water reservoir were identified on the basis of morphological and meristic characteristics. The hybrid was clearly intermediate. Four hybrid breeding crosses were induced to spawn by hypophysis. A bream × roach cross (female named first) failed to produce fertile eggs, whereas F1 hybrid × roach, roach × F1 hybrid and F1 hybrid × F1 hybrid all produced fry. Fertility (defined as survival of eggs to hatching) was high for the F1 hybrid × roach back-cross (56%) but low for the others (<2%), in comparison to the pure species controls (roach 69%, bream 76%). Progeny from these crosses were reared until anal fin rays could be counted. These counts indicated intermediacy between the parents and back-crossed individuals, and similarity between F1 hybrids and their F2 progeny.  相似文献

Abstract.— We analyzed the rate at which postzygotic incompatibilities accumulate in birds. Our purposes were to assess the role of intrinsic F1 hybrid infertility and inviability in the speciation process, and to compare rates of loss of fertility and viability between the sexes. Among our sample more than half the crosses between species in the same genus produce fertile hybrids. Complete loss of F1 hybrid fertility takes on the order of millions of years. Loss of F1 hybrid viability occurs over longer timescales than fertility: some viable hybrids have been produced between taxa that appear to have been separated for more than 55 my. There is strong support for Haldane's rule, with very few examples where the male has lower fitness than the female. However, in contrast to Drosophila , fertility of the homogametic sex in the F1 appears to be lost before viability of the heterogametic sex in the F1. We conclude that the time span of loss of intrinsic hybrid fertility and viability is often, but not always, longer than the time to speciation. Premating isolation is an important mechanism maintaining reproductive isolation in birds. In addition, other factors causing postzygotic reproductive isolation such as ecological causes of hybrid unfitness, reduced mating success of hybrids, and genetic incompatibilities in the F2s and backcrosses may often be involved in the speciation process.  相似文献

Quantitative trait loci (QTL) influencing the weight of abdominal fat (AF) and of breast muscle (BM) were detected on chicken chromosome 5 (GGA5) using two successive F2 crosses between two divergently selected 'Fat' and 'Lean' INRA broiler lines. Based on these results, the aim of the present study was to identify the number, location and effects of these putative QTL by performing multitrait and multi-QTL analyses of the whole available data set. Data concerned 1186 F2 offspring produced by 10 F1 sires and 85 F1 dams. AF and BM traits were measured on F2 animals at slaughter, at 8 (first cross) or 9 (second cross) weeks of age. The F0, F1 and F2 birds were genotyped for 11 microsatellite markers evenly spaced along GGA5. Before QTL detection, phenotypes were adjusted for the fixed effects of sex, F2 design, hatching group within the design, and for body weight as a covariable. Univariate analyses confirmed the QTL segregation for AF and BM on GGA5 in male offspring, but not in female offspring. Analyses of male offspring data using multitrait and linked-QTL models led us to conclude the presence of two QTL on the distal part of GGA5, each controlling one trait. Linked QTL models were applied after correction of phenotypic values for the effects of these distal QTL. Several QTL for AF and BM were then discovered in the central region of GGA5, splitting one large QTL region for AF into several distinct QTL. Neither the 'Fat' nor the 'Lean' line appeared to be fixed for any QTL genotype. These results have important implications for prospective fine mapping studies and for the identification of underlying genes and causal mutations.  相似文献

Abstract. The chloroplasts of two species of the Crassulaceae and their F1 hybrid were compared by electron microscopy. The two species had contrasting leaf tissue δ13C values of −25°/ ( Sedum greggii ) and −13°/ ( Cremnophila linguifolia ), and the F1 hybrid had a value of − 18°/. S. greggii had a mean of 8.9 thylakoids per granum in contrast to C. linguifolia which had a mean of only 3.8 thylakoids per granum. The F1 hybrid had a mean of 6.4 thylakoids per granum. Reproductive isolation between species may often be attained by multiple isolating barriers, but the components are rarely studied in animal taxa. To elucidate the nature of multiple isolating barriers, we quantified the strength of three premating barriers, including ecologically based ones (seasonal, habitat, and sexual), two postmating–prehatching barriers (reduced egg hatchability and conspecific sperm precedence [CSP]), and one posthatching barrier, including four components of F1 hybrid reduced fitness, between two phytophagous ladybird beetles, Henosepilachna vigintioctomaculata and H. pustulosa . We detected five positive barriers (habitat isolation, sexual isolation, reduced egg hatchability, CSP, and reduced egg hatchability in backcrosses of F1 hybrids). None of these barriers entirely prevents gene exchange when it acts alone, but jointly they generate nearly complete reproductive isolation even between sympatric populations. Host fidelity contributed most strongly to reproductive isolation by reducing interspecific hybridization through several important types of ecological isolation, including microspatial, habitat, and seasonal isolation. The existence of multiple isolating barriers likely helps keep reproductive isolation stable and robust, by complementing changes in the strength of leaky barriers. This complementarity of multiple isolating barriers yields the concept of robustness of reproductive isolation, which is important when considering the long-term maintenance of species boundaries in coexisting species pairs.  相似文献

The effectiveness of geographical isolation, ecological isolation, temporal isolation, mechanical isolation, ethological isolation, cross-incompatibility, hybrid inviability, hybrid sterility and hybrid breakdown as practical barriers to gene flow in the field between Conospermum taxifolium, C. ericifolium, C. ellipticum and C. longifolium has been quantified. The barriers to gene flow between C. ericifolium and C. ellipticum are completely effective, owing to their allopatric distributions. The barriers to gene flow between C. taxifolium and these two species are only partially effective, as their ecological separation breaks down in intermediate habitats, and partially-fertile F1 plants can grow in the areas of overlap. The barriers to gene flow between C. longifolium and the other three species are almost completely effective, as cross-incompatibility is very high and the F1 plants are female-sterile.  相似文献

Transgenic oilseed rape ( Brassica napus ) plants can interbreed with nearby weedy Brassica rapa , potentially enhancing the weediness and/or invasiveness of subsequent hybrid offspring. We have previously demonstrated that transgenic mitigation effectively reduces the fitness of the transgenic dwarf and herbicide-resistant B. napus volunteers. We now report the efficacy of such a tandem construct, including a primary herbicide-resistant gene and a dwarfing mitigator gene, to preclude the risks of gene establishment in the related weed B. rapa and its backcross progeny. The transgenically mitigated and non-transgenic B. rapa  ×  B. napus interspecific hybrids and the backcrosses (BC1) with B. rapa were grown alone and in competition with B. rapa weed. The reproductive fitness of hybrid offspring progressively decreased with increased B. rapa genes in the offspring, illustrating the efficacy of the concept. The fitness of F2 interspecific non-transgenic hybrids was between 50% and 80% of the competing weedy B. rapa , whereas the fitness of the comparable T2 interspecific transgenic hybrids was never more than 2%. The reproductive fitness of the transgenic T2 BC1 mixed with B. rapa was further severely suppressed to 0.9% of that of the competing weed due to dwarfism. Clearly, the mitigation technology works efficiently in a rapeseed crop–weed system under biocontainment-controlled environments, but field studies should further validate its utility for minimizing the risks of gene flow.  相似文献

Supernatant malate dehydrogenase (MDH) isozymes (as visualized by starch gel electrophoresis) are encoded by two distinct gene loci in both the largemouth and smallmouth bass. When an interspecific F1 hybrid is formed between these two fish, a unique MDH isozyme is generated. The results of freeze-thaw molecular hybridization (which is the first application of this technique to MDH) indicate that this unique isozyme in the F1 hybrid is a heterodimer composed of one subunit of each parental type. The F1 hybrids produced F2 hybrids which in turn formed the F3 hybrid population. The inheritance of alleles at the MDH-B locus is consistent with a single Mendelian autosomal locus. Furthermore, there is no evidence of linkage between the lactate dehydrogenase-E locus and the MDH-B locus.  相似文献

Based on the observation of a grey phenotype in the F1 generation from a cross between two white plumage duck varieties, the white Kaiya and the white Liancheng , we hypothesized a possible interaction between two autosomal loci that determine grey plumage. Using the parental and F1 individuals, seven testing combinations including five different F1 intercrosses (F2) and two different backcrosses (BC1 and BC2) were designed to test our hypothesis. It was demonstrated by chi-squared analysis that six test matings produced offspring in the expected ratios between the grey and white, with P- values ranging from 0.50 to 0.99. Another mating, where all white offspring were expected, produced 33 white individuals. These results verified that the interaction between two loci produced the grey phenotype. The C locus, which carries the recessive allele ( c ), was previously thought to be the only gene responsible for white plumage in the duck. This is the first report that an allele ( t ), carried by the white Liancheng at a different autosomal locus, also determines white plumage in ducks. Furthermore, the dominant alleles at both loci can interact with each other to produce the grey phenotype, and a new dark phenotype, observed in some F2 individuals, can be attributed to the dosage effect of the T allele.  相似文献

Abstract.  1. In insects, larval diet can have a major impact on development, survival, and reproductive success. However, resource availability at the adult phase of the life cycle is also likely to have strong effects in species where there is an extended period of sexual maturation following adult eclosion.
2. The effect of diet on the survival and reproductive success of the lekking Hawaiian fruit fly, Drosophila grimshawi , was explored. Two generations of emerging adults were exposed to one of two feeding regimes: 'constant' and 'varied' (corresponding to food 'each day' or 'every other day' respectively). The impact of resource availability on survival and reproductive success in each generation was then investigated.
3. The probability of survival to 5 weeks old was higher for individuals fed a constant diet than individuals fed a varied diet, but was comparable for males and females.
4. There was a significant maternal effect on offspring survival. Offspring whose mothers were reared on a constant feeding regime had higher survival than offspring whose mothers were reared on a varied diet.
5. There was no relationship between feeding regime and the quantity of pheromones deposited by males (a measure of male reproductive investment); however F2 sons were more likely to deposit pheromones and deposited a larger quantity of pheromone than their F1 sires. The number and sex ratio of offspring (a measure of female reproductive effort) emerging from the F1 generation was unrelated to maternal or paternal feeding regime.

We assessed the fertility (reproductive success, litter size, testis weight, spermatocyte-to-spermatid ratio) of F1s and backcrosses between different wild-derived outbred and inbred strains of two mouse subspecies, Mus musculus domesticus and M. m. musculus . A significant proportion of the F1 females between the outbred crosses did not reproduce, suggesting that female infertility was present. As the spermatocyte-to-spermatid ratio was correlated with testis weight, the latter was used to attribute a sterile vs. fertile phenotype to all males. Segregation proportions in the backcrosses of F1 females yielded 11 (inbred) to 17% (outbred) sterile males, suggesting the contribution of two to three major genetic factors to hybrid male sterility. Only one direction of cross between the inbred strains produced sterile F1 males, indicating that one factor was borne by the musculus X-chromosome. No such differences were observed between reciprocal crosses in the outbred strains. The involvement of the X chromosome in male sterility thus could not be assessed, but its contribution appears likely given the limited introgression of X-linked markers through the hybrid zone between the subspecies. The divergence in reproductive features and hybrid fertility patterns between two chromosomal races (2 n  = 40, 40St, and 2 n  = 22, 22Rb) of the house mouse in Tunisia were re-assessed on a larger sample of wild and laboratory-bred individuals than studied hitherto. Results showed that litter sizes were significantly smaller in 40St than in 22Rb mice, contrary to previous analyses. This suggests that variation in litter size between the two chromosomal races is more likely related to selective and/or environmental factors acting locally than to interracial reproductive trait divergence. However, the significantly reduced litter size of F1 hybrids compared with parental individuals was confirmed, and further highlighted a sex difference in hybrid infertility, as F1 females produced fewer litters and of smaller size than males. Histological analyses of F1 and backcrosses showed a breakdown of spermatogenesis in males and a significantly reduced primordial follicle pool in females. The degree of gametogenic dysfunction was not related to the level of chromosomal heterozygosity per se , but a significant effect of two Rb fusions on follicle number was observed in hybrid females. Hybridization between divergent lineages often results in reduced hybrid viability. Here we report findings from a series of independent molecular analyses over several seasons on four life stages of F1 hybrids between the newts Triturus cristatus and T. marmoratus . These two species form a bimodal hybrid zone of broad overlap in France, with F1 hybrids making up about 4% of the adult population. We demonstrate strong asymmetry in the direction of the cross, with one class ( cristatus -mothered) making up about 90% of F1 hybrids. By analyzing embryos and hatchlings, we show that this asymmetry is not due to prezygotic effects, as both classes of hybrid embryos are present at similar frequencies, implicating differential selection on the two hybrid classes after hatching. Adult F1 hybrids show a weak Haldane effect overall, with a 72% excess of females. The rarer marmoratus -mothered class, however, consists entirely of males. The absence of females from this class of adult F1 hybrids is best explained by an incompatibility between the cristatus X chromosome and marmoratus cytoplasm. It is thus important to distinguish the two classes of reciprocal-cross hybrids before making general statements about whether Haldane's rule is observed.  相似文献

Three species of Darwin's Finches hybridize on the Galapagos island of Daphne Major. We examined mating patterns to determine if hybrids exhibit mate preferences. Geospiza fortis x G. scandens F1 hybrids backcrossed to both of the parental species, whereas all backcrosses bred with the parental species to which they were most related, or with hybrids. Paternal song was shown to be the crucial factor determining the mating pattern of G. fortis x G. scandens F1 hybrids and their offspring. Song is culturally inherited, transmitted faithfully from father to son (with few exceptions) as a result of an imprinting-like process. Size also contributes to the choice of mates. G. fortis x G. scandens F1 hybrid females paired with large G. scandens -like G. fortis males. G. fortis x G. fuliginosa F1 hybrids paired negatively assortatively with respect to the size of their G. fortis mates. Non-random mating of hybrids based on song, a non-generic trait, has interesting evolutionary consequences. Song characteristics and nuclear and mitochondrial genes flow from G. fuliginosa into the G. fortis population, whereas the direction of transfer of genetic and song information between G. fortis and G. scandens depends on which song was sung by the father of the hybrids.  相似文献

Abstract.  1. Changes in the frequency of Cry1Ac resistance genes and shifts in tolerance of cotton bollworm, Helicoverpa armigera , to the Cry1Ac toxin were assessed using bioassays of F1 and F2 offspring of isofemale lines from Anci County of Hebei Province (a multiple-crop system including corn, soybean, peanut, and Bt cotton) and Xiajin County of Shandong Province (an intensive Bt cotton planting area) in Northern China during 2002–2005.
2. A conservative analysis of the overall results indicated that there was a small increase in the frequency of major, non-recessive resistance genes over time.
3. The relative average development ratings [RADR – growth rate of a line on a Bt diet in proportion to the growth rate on a non-Bt (NBT) diet] of the bollworm larvae in F1 tests increased significantly from year to year, indicating a gradual trend towards higher tolerance to Cry1Ac in the field populations.
4. There were also significant positive correlations between RADR of the lines in the F1 generation and the RADR of their F2 offspring, indicating that the tolerance was genetically based.
5. Quantitative genetic simulation analysis showed that resistance of H . armigera to Bt cotton in Xiajin could evolve to a high level in 11–15 years if no effective resistance management measures are carried out.  相似文献

Abstract. The presence or absence of epistasis, or gene interaction, is explicitly assumed in many evolutionary models. Although many empirical studies have documented a role of epistasis in population divergence under laboratory conditions, there have been very few attempts at quantifying epistasis in the native environment where natural selection is expected to act. In addition, we have little understanding of the frequency with which epistasis contributes to the evolution of natural populations. In this study we used a quantitative genetic design to quantify the contribution of epistasis to population divergence for fitness components of a native annual legume, Chamaecrista fasciculata . The design incorporated the contrast of performance of F2 and F3 segregating progeny of 18 interpopulation crosses with the F1 and their parents. Crosses were conducted between populations from 100 m to 2000 km apart. All generations were grown for two seasons in the natural environment of one of the parents. The F1 often outperformed the parents. This F1 heterosis reveals population structure and suggests that drift is a major contributor to population differentiation. The F2 generation demonstrated that combining genes from different populations can sometimes have unexpected positive effects. However, the F3 performance indicated that combining genes from different populations decreased vigor beyond that due to the expected loss of heterozygosity. Combined with previous data, our results suggest that both selection and drift contribute to population differentiation that is based on epistatic genetic divergence. Because only the F3 consistently expressed hybrid breakdown, we conclude that the epistasis documented in our study reflects interactions among linked loci.  相似文献

Twenty‐one allozyme loci in samples of wild‐caught and hatchery‐reared Indian major carps from Bangladesh were analysed. Bayesian model‐based clustering analysis revealed the presence of four taxa, corresponding to the three known species along with a fourth unknown taxon present in two hatchery samples. Individual admixture coefficients showed that 24% of all hatchery‐reared fishes were hybrids, whereas a single hybrid was observed in the wild‐caught samples. Only catla Catla catla × rohu Labeo rohita and mrigal Cirrhinus cirrhosus × rohu hybrids were observed, the vast majority of which were F1 hybrids, though five individuals represented putative backcrosses. Mitochondrial DNA analysis revealed that catla × rohu hybridization primarily involved catla males and rohu females, whereas mrigal × rohu hybrids primarily resulted from rohu males and mrigal females. Despite the high percentage of F1‐hybrids in hatchery samples, reproductive barriers among species have so far precluded widespread introgression. Continued hybridization may eventually lead to a breakdown of species barriers, thereby compromising the genetic integrity of the species in the wild, and leading to production losses in aquaculture.  相似文献

To characterize the genetic basis of voluntary calcium consumption, we tested C57BL/6J mice (B6; with low avidity for calcium), PWK/PhJ mice (PWK; with high avidity for calcium) and their F1 and F2 hybrids. All mice received a series of 96-h two-bottle preference tests with a choice between water and the following: 50 m m CaCl2, 50 m m calcium lactate, 50 m m MgCl2, 100 m m KCl, 100 m m NH4Cl, 100 m m NaCl, 5 m m citric acid, 30 μ m quinine hydrochloride and 2 m m saccharin. Most frequency distributions of the parental and F1 but not F2 groups were normally distributed, and there were few sex differences. Reciprocal cross analysis showed that B6 × PWK F1 mice had a non-specific elevation of fluid intake relative to PWK × B6 F1 mice. In the F2 mice, trait correlations were clustered among the divalent salts and the monovalent chlorides. A genome screen involving 116 markers showed 30 quantitative trait loci (QTLs), of which six involved consumption of calcium chloride or lactate. The results show pleiotropic controls of calcium and magnesium consumption that are distinct from those controlling consumption of monovalent chlorides or exemplars of the primary taste qualities.  相似文献
