A study was conducted to determine the effect of broiler poultry litter (BL) supplementation to spent mushroom substrate (SMS) on its storage and feed-nutritional value improvement. In Exp. 1, the sawdust-based SMS from a king oyster mushroom (Pleurotus eryngii) farm was mixed with BL at 0%, 25%, 50%, 75% and 100% levels on a wet basis and deepstack stored for short-term (1-wk) and long-term (3-wk). At 1-wk of short-term deepstack storage, SMS with more than 50% BL levels showed favorable conservation. At 3-wk of long-term storage, all treatments except for BL 100% had a serious fungal problem. Based on chemical parameters, BL-blending to SMS practically improved the feed-nutritional value of the mixtures. Since the deepstacking method was not effective for long term storage, in Exp. 2 SMS ensiled with or without BL was attempted to improve long-term (3-wk) storage. All the ensiled treatments (SMS 100%, SMS 75%+BL 25% and SMS 50%+BL 50%) had desirable fermentation. As in deepstacking, BL-blending to SMS improved the nutritive value of the ensiled mixtures. The populations of total bacteria, lactic acid bacteria and yeast were highest (P<0.05) when 75% SMS was blended with 25% BL. In conclusion, blending 50% or more BL with SMS was effective for the short-term (1-wk) deepstack storage. For long-term (3-wk) storage of SMS, an ensiling method was effective with or without the addition of BL.  相似文献

The effect of carbon source addition on the operation of a sequencing batch reactor in order to remove nitrogen and COD of poultry wastewater was studied. The reactor was constructed with a glass tube having a volume of 7 l and a jacket for temperature control. The reactor bottom consisted of a conical porous stone in order to promote liquor aeration and agitation. Initial conditions and operation strategies were adjusted to improve the final effluent quality. According to the attained experimental results, it was verified that nitrification and denitrification can occur simultaneously in aerated culture, contrary the observation of some authors.  相似文献

Broiler litter was deep stacked at 30 and 40% moisture or ensiled at 40% moisture for 6 weeks in quadruplicate and subsequently evaluated for changes in chemical composition, pathogenic microbial load and in vitro dry matter degradability (IVDMD). A metabolism trial was conducted with lambs, in which a low-protein basal diet supplemented with litter, deep stacked at 30% and 40% moisture for 6 weeks or ensiled were compared with an isocaloric, isonitrogenous soybean meal supplemented diet. The maximum stack temperature of 65°C (irrespective of moisture level) was attained on day 7 at 45 cm depth and 60.5°C on day 10 at 80 cm depth, which eliminated coliforms. Increases in lactic and acetic acids were recorded with increase in moisture levels and advancement of stacking period. A small increase in crude protein (CP) content was associated with considerable declines in organic matter (OM), cellulose, neutral detergent fiber (NDF) and hemicellulose in deep-stacked litters. The apparent digestibility of nutrients and N-retention in the litter-supplemented diets were similar to those of the soybean meal supplemented diet. It is concluded that by deep stacking at 30 or 40% moisture, or ensiling, the litter was rendered free of pathogens and could be incorporated in the diet of livestock without adversely affecting digestibility of fiber and efficiency of N utilization.  相似文献

The effect of two abundant, easily available and very low-cost agro-industrial organic residues, i.e., filter cake from the sugar industry and poultry litter, on the composting stabilization time of coffee pulp and on the quality of the produced compost, was evaluated. Piles of one cubic meter were built and monitored within the facilities of a coffee processing plant in the Coatepec region of the State of Veracruz, Mexico. Manual aeration was carried out once a week. A longer thermophilic period (28 days) and a much lower C/N ratio (in the range of 6.9–9.1) were observed in the piles containing the amendments, as compared to the control pile containing only coffee pulp (14 days and a C/N ratio of 14.4, respectively). The maximum assimilation rate of the reducing sugars was 1.6 g kg-1 d-1 (from 7.5 to 5.3%) during the first two weeks when accelerators were present in the proportion of 20% filter cake plus 20% poultry litter, while they accumulated at a rate of 1.2 g kg-1 d-1 (from 7.4 to 9.13%) during the same period in the control pile. The best combination of amendments was 30% filter cake with 20% poultry litter, resulting in a final nitrogen content as high as 4.81%. The second best combination was 20% filter cake with 10% poultry litter, resulting in a compost which also contained a high level of total nitrogen (4.54%). It was concluded that the use of these two residues enhanced the composting process of coffee pulp, promoting a shorter stabilization period and yielding a higher quality of compost.  相似文献

Whole cells of Bacillus sp., a bacitracin-producing bacteria, were immobilized in polyacrylamide gel. The continuous production of bacitracin by an immobilized whole-cell-containing air-bubbled reactor was examined with 0.5% peptone solution. The bacitracin productivity (28 units/ml/hr) obtained with this system was higher than that with a batch system. The effluent bacitracin concentration increased with increasing aeration rate and reached a steady-state maximum above the aeration rate of 3.0 liter/min. A high bacitracin productivity was retained for at least eight days when the gel was washed with sterilized saline at a flow rate of 250 ml/hr for 2 hr once a day. The half-life of the immobilized whole-cell system was about 10 days. Bacitracin productivity by the immobilized whole-cell reactor was higher than that by a conventional continuous fermentation process at high dilution rates.  相似文献

In this study, we utilized a unique strategy for fed-batch fermentation using ethanol-tolerant Saccharomyces cerevisiae to achieve a high-level of ethanol production that could be practically applied on an industrial scale. During this study, the aeration rate was controlled at 0.0, 0.13, 0.33, and 0.8 vvm to determine the optimal aeration conditions for the production of ethanol. Additionally, non-sterile glucose powder was fed during fed-batch ethanol fermentation and corn-steep liquor (CSL) in the medium was used as an organic N-source. When aeration was conducted, the ethanol production and productivity were superior to that when aeration was not conducted. Specifically, the maximum ethanol production reached approximately 160 g/L, when the fermentor was aerated at 0.13 vvm. These findings indicate that the use of a much less expensive C-source may enable the fermentation process to be directed towards the improvement of overall ethanol production and productivity in fermentors that are aerated at 0.13 vvm. Furthermore, if a repeated fed-batch process in which the withdrawal and fill is conducted prior to 36 h can be employed, aeration at a rate of 0.33 and/or 0.8 vvm may improve the overall ethanol productivity  相似文献

Static 90 day bioassays were made in outdoor cement tanks (300 l capacity) with fries of common carp (Cyprinus carpio) exposed to 1.0, 2.5 and 5.0 mg/l of cadmium (Cd) treated individually and with 250 mg/l poultry litter. Addition of poultry litter significantly reduced the concentrations of dissolved Cd in water and whole body Cd in fish and plankton. In contrast, concentrations of total Cd in sediment increased under the influence of poultry litter. Cadmium showed a distinct partitioning pattern in the sediment. Addition of poultry litter did not change the partitioning pattern. Growth of fish, in respect to length and weight, was reduced by all levels of Cd exposure. In contrast, maximum weight gains, increased lengths, increased specific growth rates and yields were recorded under the influence of poultry litter alone. Addition of poultry litter reduced the ill effects on growth of fish produced by Cd and proved to be a promising tool to counter Cd pollution in aquaculture.  相似文献

Native microbial populations can degrade poultry waste, but the process can be hastened by using feather-degrading bacteria. Strains of Bacillus licheniformis and a Streptomyces sp. isolated from the plumage of wild birds were grown in a liquid basal medium and used to inoculate feathers in compost bioreaction vessels. Control vessels had only basal medium added to the feathers, litter and straw. Temperature, ammonia, carbon and nitrogen were monitored for 4 weeks. Scanning electron microscopy of the feather samples showed more complete keratin-degradation, more structural damage, and earlier microbial biofilm formation on inoculated feathers than on uninoculated feathers. A diverse community of aerobic bacteria and fungi were cultured early, but declined rapidly. Thermophilic B. licheniformis and Streptomyces spp. were abundant throughout. Enteric gram-negative bacteria, (e.g., Salmonella, E. coli) originally found on waste feathers were not recovered after day 4. Vessel temperatures reached 64-71 degrees C within 36 h and stabilized at 50 degrees C. When tumble-mixed at day 14, renewed activity peaked at 59 degrees C and quickly dropped as available carbon was used. Feathers soaked in an inoculum of B. licheniformis and Streptomyces degraded more quickly and more completely than feathers that were not presoaked. Inoculation of feather waste could improve composting of the large volume of feather waste generated every year by poultry farms and processing plants.  相似文献

For composting, passive aeration can save energy costs while being just as efficient as forced or active aeration. Passive aeration requires the proper design of aeration ducts, and thus, the proper prediction of the convective airflow rates created by the temperature differential between the compost and the ambient air. To establish such relationship, the temperature and convective air flow regimes of composts were investigated using three bulking agents (wood shavings, hay and straw), each at three moisture contents (MC-60%, 65% and 70%) spanning the normal values. All bulking agent and aeration treatments were aerated in duplicate under passive and active regimes. Laboratory vessels of 105 L were used for all treatments. Passive aeration treatments produced temperatures above 57 degrees C, as did the treatments actively aerated at 4 mg of air s(-1) kg(-1) of initial dry compost material. Compost MC had an effect only on the peak compost temperature, occurring between day 2 and 6. After 6 days of composting, MC no longer had any effect on temperature regime because of the loss of moisture by each mixture. A relationship was established between the Grasholf number (Gr-ratio of buoyancy to viscous forces) and the convective airflow rates, to size the aeration ducts for passive aeration. In general, convective airflow rates ranged from 1.5 to 0.7 mg of dry air s(-1) kg(-1) of initial compost dry matter, from day 0 to day 20, respectively, and for all compost treatments. This airflow rate sizes the aeration ducts installed under compost piles for passive aeration. As compared to straw where airflow rate dropped over a given level of Gr, wood shavings and hay were found to be more effective as bulking agents, as their airflow rate increased constantly with Gr.  相似文献

Broiler chicken litter was kept as a stacked heap on a poultry farm, and samples were collected up to 9 months of storage. Chicken litter inoculated with desiccation-adapted Salmonella cells was heat-treated at 75, 80, 85, and 150°C. Salmonella populations decreased in all these samples during heat treatment, and the inactivation rates became lower in chicken litter when storage time was extended from 0 to 6 months. There was no significant difference (P > 0.05) in thermal resistance of Salmonella in 6- and 9-month litter samples, indicating that a threshold for thermal resistance was reached after 6 months. Overall, the thermal resistance of Salmonella in chicken litter was affected by the storage time of the litter. The changes in some chemical, physical, and microbiological properties during storage could possibly contribute to this difference. Moisture and ammonia could be two of the most significant factors influencing the thermal resistance of Salmonella cells in chicken litter. Our results emphasize the importance of adjusting time and temperature conditions for heat processing chicken litter when it is removed from the chicken house at different time intervals.  相似文献

The effect of yeast propagated at different aeration conditions on yeast physiology, fermentation ability, and beer quality was investigated using three strains of Saccharomyces cerevisiae. It was shown that yeast cells grown under continuous aeration conditions during propagation were almost two times higher as compared with discontinuous aeration conditions. The maximum of cell growth of all samples reached between 36 h and 48 h. The concentration of trehalose was increased under continuous aerated yeasts, whereas glycogen was decreased. It was also observed that the concentration of glycogen and trehalose in yeast cells had no direct effect on subsequent fermentation ability. The effect of yeast propagated under different aeration conditions on subsequent fermentation ability was different from yeast strains, in which the influence will be most pronounced at the first fermentation. Later, the yeasts might regain its original characteristics in the following fermentations. Generally, continuously propagated yeast had a positive effect on beer quality in subsequent fermentation. Hence, the concentration of aroma compounds obtained with yeast propagated under 6 1/h for 48 h aeration was lower than those grown under other aeration conditions in the bottom yeasts; in particular, the amounts of phenylethyl alcohol, ester, and fatty acids were decreased.  相似文献

Very little is known about the microbial composition of animal bedding wastes, including poultry litter, and what is known has been deduced from standard culture methods, by which some fastidious organisms that exist in the environment may not be detected. We evaluated the bacterial composition of poultry litter by using a combination of culture and molecular detection. Total aerobic bacteria in poultry litter were detected by culture at 10(9) CFU/g of material. Enteric bacteria such as Enterococcus spp. and coliforms composed 0.1 and 0.01%, respectively, of the total aerobic cultivatable bacteria in poultry litter; no Salmonella strains were detected by culture. In order to characterize the most abundant bacterial groups, we sequenced 16S ribosomal DNA (rDNA) genes amplified by PCR with microbial community DNA isolated from poultry litter as the template. From the 16S rDNA library, 31 genera were identified. Twelve families or groups were identified with lactobacilli and Salinococcus spp. forming the most abundant groups. In fact, 82% of the total sequences were identified as gram-positive bacteria with 62% of total belonging to low G+C gram-positive groups. In addition to detection of 16S rDNA sequences associated with the expected fecal bacteria present in manure, we detected many bacterial sequences for organisms, such as Globicatella sulfidofaciens, Corynebacterium ammoniagenes, Corynebacterium urealyticum, Clostridium aminovalericum, Arthrobacter sp., and Denitrobacter permanens, that may be involved in the degradation of wood and cycling of nitrogen and sulfur. Several sequences were identified in the library for bacteria associated with disease in humans and poultry such as clostridia, staphylococci, and Bordetella spp. However, specific PCR targeting other human and veterinary pathogens did not detect the presence of Salmonella, pathogenic Escherichia coli, Campylobacter spp., Yersinia spp., Listeria spp., or toxigenic staphylococci. PCR and DNA hybridization revealed the presence of class 1 integrons with gene cassettes that specify resistance to aminoglycosides and chloramphenicol. Only from understanding the microbial community of animal wastes such as poultry litter can we manage animal disease and limit the impact of animal waste on the environment and human and animal health.  相似文献

The numbers of viable bacteria in built up poultry litter were found to be 1010-1011/g fresh weight and appeared to be little affected by factors such as age, temperature, moisture content and pH. Counts for unused litter and poultry droppings were lower. In built up litter of high alkalinity coryneform bacteria were predominant; micrococci occurred sporadically and small numbers of nocardias, streptomycetes, aerobic spore formers and streptococci were encountered. A variety of Gram negative bacteria also occurred, the numbers of which appeared to be controlled by alkalinity; they were less abundant in litters where the pH and buffering capacity were high. Strongly alkaline conditions also tended to lower the fungal counts but had no effect on the count of enterococci.  相似文献

Utilization of cellulose from waste paper by Myrothecium verrucaria   总被引:4,自引:0,他引:4  
Extensive screening studies on cellulolytic bacteria and fungi led to the selection of Myrothecium verrucaria as the organism producing the maximum rate of protein biosynthesis from ball-milled newspaper. Studies in aerated stirred-jar fermentors were carried out to determine the conditions for maximum protein synthesis rate and maximum final protein concentration. The optimum aeration rate was 250 to 374 mM of oxygen at 300 to 400 rpm stirring rate. The pH optimum was broad, from 3.9 to 6.5. Urea at 0.03% and yeast autolysate at 0.1% stimulated growth rate and protein production. The maximum rate of protein biosynthesis and the maximum protein yield were 0.3 g/liter/day and 1.42 g/liter, respectively, from medium G3 with 4% ball-milled newspaper. The final product, obtained by evaporation of the total culture, was 33.7 g from one liter of medium which originally contained 40 g of ball-milled newspaper and 11.3 g of other dissolved materials. The protein content of this final product was 3.3 g, calculated from total organic N × 6.25 or 1.42 g calculated from the biuret method. Both the synthesis rate and the final cell yield are below those obtainable by growing Fungi Imperfecti, yeasts or bacteria on soluble materials such as glucose.  相似文献

The lower leaves of tobacco plants were inoculated with leaf mosaic virus and the activities of oxygenase, peroxidase, catalase, and invertase were followed in leaves of comparable age at intervals of 2 or 3 days over a period of 21 days. The inoculated leaves exhibited a great decrease relative to normal tissue in the activity of oxygenase and peroxidase on the 6th day. Younger leaves showed this minimum at a progressively later date. A great decrease in the activities of these enzymes was attained by the 14th to the 18th day. This maximum was followed by a decrease. Catalase exhibited an increased activity which reached a maximum at about the 8th day. A second maximum was observed on the 16th to the 18th day. Invertase reached a minimum, relative to normal plants, on about the 8th day. A second minimum was approached on the 16th to the 18th day. These data show that profound disturbances in the physiology of infected plants occur many days before the leaf juice attains an infectious concentration of virus. The observed activities could not be due therefore to metabolic activities of the virus particles themselves. Since infectivity is attained only after a period of profound physiological disturbance, it seems possible that the virus protein develops as a product of abnormal metabolism.  相似文献

Knowledge of the population fluctuation and spatial distribution of pests is fundamental for establishing an appropriate control method. The population fluctuation and spatial distribution of the Alphitobius diaperinus in a poultry house in Cascavel, in the state of Parana, Brazil, was studied between October, 2001 and October 2002. Larvae and adults of the lesser mealworm were sampled weekly using Arends tube traps (n = 22) for six consecutive flock grow-outs. The temperature of the litter and of the poultry house was measured at the same locations of the tube traps. Beetle numbers increased continuously throughout all the sampling dates (average 5,137 in the first week and 18,494 insects on the sixth week). Significantly greater numbers of larvae were collected than adults (1 to 20 times in 95% of the sampling points). There was no correlation between temperature and the number of larvae and adults collected, therefore no fluctuation was observed during the sampling period. The population growth was correlated to litter re-use. The highest temperatures were observed in deep litter. The spatial distribution of larvae and adults in the poultry house was heterogeneous during the whole period of evaluation. Results suggest that monitoring in poultry houses is necessary prior to adopting and evaluating control measures due to the great variability of the insect distribution in the poultry house.  相似文献

The leaf temperature of six annual and six perennial plant species was monitored during spring and summer on a sand dune ecosystem in the delta Mediterranean coast of Egypt. During winter, leaves of all tested perennial species attained temperatures higher than the air temperature at night and shortly after sunrise, with maximum leaf–air temperature differences reaching up to 8°C. The lowest differences were less than 1°C. Around noon, the leaves of several species attained temperatures lower than that of the air whereas others showed higher temperatures. The opposite was true during summer, when leaf temperatures were lower than air temperature. The maximum leaf–air temperature differences occurred after midnight towards sunrise and reached up to 10°C. The lowest differences were found around noon and were of less than 5°C. The annual plant species have more pronounced variations than perennials in their leaf temperatures during the night and for most of the day. The leaves were heated or cooled a few degrees above or below the air temperature. The results are discussed in relation to the morphological characters of the leaves. The variation in leaf temperature at different times of the day was significantly related to leaf morphology, specific leaf area, thickness, volume, leaf area index and the surrounding environment.  相似文献

This study aimed to find optimal operation conditions for nitrogen removal from high strength slaughterhouse wastewater at 11 °C using the intermittently aerated sequencing batch reactors (IASBRs) so as to provide an engineering control strategy for the IASBR technology. Two operational parameters were examined: (1) loading rates and (2) aeration rates. Both the two parameters affected variation of DO concentrations in the IASBR operation cycles. It was found that to achieve efficient nitrogen removal via partial nitrification–denitrification (PND), “DO elbow” point must appear at the end of the last aeration period. There was a correlation between the ammonium oxidizing bacteria (AOB)/nitrite oxidizing bacteria (NOB) ratio and the average DO concentrations in the last aeration periods; when the average DO concentrations in the last aeration periods were lower than 4.86 mg/L, AOB became the dominant nitrifier population, which benefited nitrogen removal via PND. Both the nitrogen loading rate and the aeration rate influenced the population sizes of AOB and NOB. To accomplish efficient nitrogen removal via PND, the optimum aeration rate (A, L air/min) applied can be predicted according to the average organic loading rates based on mathematical equations developed in this study. The research shows that the amount of N2O generation in the aeration period was reduced with increasing the aeration rate; however, the highest N2O generation in the non-aeration period was observed at the optimum aeration rates.  相似文献

Aerosolized bacteria were recovered up to 930 m downwind of three sewage treatment plants in Jefferson County, Ky. This distance includes homes in the proximity of several hundred such plants in that county. Bacterial counts were elevated on foliage near activated sludge tanks; although these counts decreased rapidly, at 48 h after exposure they were significantly higher than the counts on unexposed leaves. The 50% lethal dose of aerosolized Klebsiella pneumoniae was comparable to the 50% lethal dose of a virulent clinical isolate, and enteric bacteria were recovered from the respiratory organs of mice after forced inhalation adjacent to an aerated sludge tank. The coliform density in the effluents of the plants tested was inversely related to the airborne bacterial load at those plants. This relationship was attributed to the correlation between effluent quality and extent of aeration of activated sludge. Wind direction and distance influenced the airborne counts, but the extreme variation in counts indicates that it is not possible to predict emission rates accurately in an open ecosystem. Airborne enteric bacteria also were isolated near a decorative fountain used by humans for wading. The discovery of these sources of aerosolized microorganisms from polluted waters in densely populated areas suggests that a potential health hazard may be created by the increased probability of inhaling and ingesting microorganisms of fecal origin.  相似文献

Studies have been conducted over a 2-year period to determine the extent to which a poultry processing plant served as a reservoir of salmonellae reaching the external environment, to examine the question of the importance of salmonellae in the environment, and to consider how best to control the spread of the organisms. The studies have been undertaken at a chicken processing plant handling between 75,000 and 80,000 birds per day. Populations of salmonellae and indicator bacteria were estimated in the raw wastes, through the waste treatment plant, and in the receiving stream waters. The results demonstrate that salmonellae are present in poultry processing wastes in a surprisingly constant relation to fecal coliforms (in excess of 1 Salmonella per 500 fecal coliforms), that serotypes in the environment are constantly changing, and that they may reflect unusual conditions in the processing plant, or a possible source of infection among human and animal residents of the environment. Disinfection of poultry processing wastes is recommended.  相似文献
