Rice tillering is one of the most important agronomic traits that determine grain yields.Our previous study has demonstrated that the MONOCULM1(MOC1)gene is a key component that controls the formation of rice tiller buds.To further elucidate the molecular mechanism of MOC1 involved in the regulation of rice tillering.we performed a yeast-two-hybrid screening to identify MOC1 interacting proteins(MIPs).Here we reported that MIP1 interacted with MOC1 both in vitro and in vivo.The overexpression of MIP1 resulted in enhanced tillering and reduced plant height.In-depth characterization of the context of MIP1 and MOC1 would further our understanding of molecular regulatory mechanisms of rice tillering. 相似文献

ARGONAUTE (AGO) proteins play crucial roles in plant defence against virus invasion. To date, the role of OsAGO2 in rice antiviral defence remains largely unknown. In this study, we determined that the expression of OsAGO2 in rice was induced upon rice black-streaked dwarf virus (RBSDV) infection. Using transgenic rice plants overexpressing OsAGO2 and Osago2 mutants generated through transposon-insertion or CRISPR/Cas9 technology, we found that overexpression of OsAGO2 enhanced rice susceptibility to RBSDV infection. Osago2 mutant lines exhibited strong resistance to RBSDV infection through the elicitation of an early defence response, including reprogramming defence gene expression and production of reactive oxygen species (ROS). Compared to Nipponbare control, the expression level of OsHXK1 (HEXOKINASE 1) increased significantly, and the methylation levels of its promoter decreased in the Osago2 mutant on RBSDV infection. The expression profile of OsHXK1 was the opposite to that of OsAGO2 during RBSDV infection. Overexpression of OsHXK1 in rice also induced ROS production and enhanced rice resistance to RBSDV infection. These results indicate that OsHXK1 controls ROS accumulation and is regulated by OsAGO2 through epigenetic regulation. It is noteworthy that the Osago2 mutant plants are also resistant to southern rice black-streaked dwarf virus infection, another member of the genus Fijivirus. Based on the results presented in this paper, we conclude that OsAGO2 modulates rice susceptibility to fijivirus infection by suppressing OsHXK1 expression, leading to the onset of ROS-mediated resistance. Rice dwarf virus crystals belong to space group I222 with cell parameters a = 770 (2), b = 795 (5), c = 814 (5) Å and α = β = γ = 90°. The unit cell of the crystal contains two viruses at the origin and body-centred positions. Using data synthesized from a rice dwarf virus model crystal in the space group I222, the possibility of ab initio phasing was thoroughly examined. The centric nature of the initial phases was unexpectedly broken by extensive iteration of the non-crystallographic symmetry averaging. The structure of rice dwarf virus was then solved with ab initio phasing up to 20 Å resolution. The triangulation number determined by the present study is T = 13, which is different from the triangulation number, T = 9, previously determined by electron microscopy [Uyeda & Shikata (1982). Ann. Four gibberellin (GA1, GA3, GA4 and GA37) glucosyl esters were synthesized and found to be as active as their respective free acids in the rice seedling bioassay. 利用micro array 技术对水稻幼苗在营养胁迫条件下根部基因表达的研究中发现: 一个与豌豆Pra2(小G蛋白)基因有同源性的基因的RNA水平在营养胁迫后再补充营养时, 表达量下降。用RT-PCR和PCR方法分别获得该基因的cDNA克隆—OsPra2和该基因翻译起始位点上游1 kb的启动子序列。OsPra2基因编码的蛋白质具有结合GTP/GDP的4个保守结构域和构成小G蛋白Rab家族的特有的结构域。该基因cDNA与GST蛋白基因融合表达载体在洋葱表皮细胞中的瞬间表达结果显示该蛋白定位在在细胞膜和细胞核上, OsPra2基因启动子与GUS报告基因融合表达转基因水稻显示该基因启动子驱动GUS在胚芽鞘和根中表达, 35S启动子驱动OsPra2基因过表达转基因水稻与野生水稻株型相比明显矮化, 类似BR缺陷型植物株型。本实验还对OsPra2和P450蛋白的相互作用及在BR代谢途径中的可能作用进行了分析。 相似文献

The properties of various mono- and di-substituted analogues of 2-phenoxypropionic acid as inhibitors of tillering were investigated on wheat, barley and rice. Highest levels of activity were shown by (R)2-Cl,5-Cl, (R)2-Cl,5-F, and (R)2-Cl,5-methyl analogues. Few or no signs of phytotoxic effects (leaf chlorosis or necrosis) were evident on wheat or barley following spray application of these compounds. Rice was both more susceptible to inhibition of tillering and phytotoxic effects. However, almost complete inhibition of tillering was achieved by application of some compounds to rice with little or no phytotoxicity. Comparisons were made between the properties of these compounds and commercially used phenoxyacetic and phenoxypropionic herbicides and plant growth regulators. Dichlorprop inhibited tillering in rice, fenoprop in wheat and rice, and fluroxypyr in wheat, all without phytotoxic effects. 相似文献

水稻(Oryza sativa L.)是重要的粮食作物,其植株光合作用与肥料利用的效率是决定水稻产量的重要因素。本研究在水稻‘科辐粳7号’的辐射诱变群体中筛选到一个叶片黄化且分蘖减少的突变体,并将其命名为yllt10(yellow leaf and less tillering 10)。与野生型对照相比,该突变体叶片的叶绿素含量降低、叶绿体的结构发育异常、光合作用速率显著下降。遗传分析表明yllt10突变体的表型受单隐性核基因控制,利用图位克隆技术将YLLT10基因定位于水稻第10号染色体J4与J5两个分子标记之间,通过对该区间内的注释基因进行PCR测序,发现在yllt10突变体中CAO1/PGL基因的第1个外显子发生单碱基缺失,从而导致该基因的移码突变,是CAO1/PGL基因的一个新等位变异。此外,在不同氮素浓度种植条件下,yllt10突变体均表现出对氮素不敏感的表型。本研究表明YLLT10基因调控水稻的叶色和分蘖数,影响水稻的光合作用和产量,对该基因的功能机制研究能够为水稻高产育种提供一定的理论依据。 相似文献

A dwarf mutant of rice(Oryza.sativa L.)by mutagenesis of ethylene methylsulfonate(EMS)treatment from Nipponbare was identified.The mutant exhibited phenotypes of dwarfism and withered leaf tip(dwll).Based on the internode length of dwl1,this mutant be longs to the dm type of dwarfing.Analysis of elongation of the second sheath and α-amylase activity in endosperm showed that the phenotype caused by dwll was insensitive to gibberellin acid treatment.Using a large F2 population derived from a cross between the dwll and an indica rice variety,TN1,the DWLl gene was mapped to the terminal region of the long arm of chromosome 3.Fine-mapping de-limited it into a 46 kb physical distance between two STS markers,HL921 and HL944,where 6 open reading frames were predicted.Cloning of DWL1 will contribute to dissecting molecular mechanism that regulates plant height in rice,which will be beneficial to molecular assisted selection of this important trait. 相似文献

近年来,水稻黑条矮缩病严重影响我国水稻生产,培育高抗黑条矮缩病的水稻新品种迫在眉睫。获得抗性好的水稻资源是育种的基础。本研究通过对保存的2000份水稻地方品种连续2年设置2个试验点的黑条矮缩病抗性鉴定,初步筛选出连续2年没有发病的资源38份。对这38份中农艺性状较好的6份粳稻资源进行人工室内接虫鉴定,获得高抗水稻黑条矮缩病的水稻资源1份。与感病对照相比,水稻黑条矮缩病毒(Rice black-streaked dwarf virus, RBSDV)在抗病品种体内的表达量下降71.5倍,表明抗病品种对RBSDV在体内的复制有明显的抑制作用。本研究为水稻黑条矮缩病抗病育种和抗性基因的定位、克隆奠定了材料基础。 相似文献

Rice dwarf virus (RDV) is a double-shelled icosahedral virus.Using electron cryomicroscopy and computer reconstruction techniques, we have determined a 3.3 nm resolution three-dimensional (3D) structure of the inner shell capsid without the outer shell and viral RNA. The results show that the inner shell is a thin, densely packed, smooth structure, which provides a scaffold for the full virus. A total of 120 copies of the major inner shell capsid protein P3 forms 60 dimers arranged in a T=1 icosahedral lattice. A close examination on the subunit packing of the T=1 inner core P3 with that of the T=13l outer shell P8 indicated that P8 trimers connect with P3 through completely non-equivalent, yet highly specific, intermolecular interactions. 相似文献

具有发夹结构的双链RNA(hairpin RNA,hpRNA)能高效诱导转录后基因沉默的发生.以水稻(Oryza sativaL.)矮缩病毒(RDV)基因组中第八片段编码区128~754 bp的序列为臂构建hpRNA,并克隆到植物表达载体pROK-2上.通过农杆菌介导的方法转化水稻\"中花11\".Southern blot分析表明,共获得12株阳性转化体.用带有RDV的叶蝉(Nephotettix cincticeps)接种Tl代转hpRNA水稻,结果表明转基因水稻对RDV具有高抗性或表现为症状延迟.而相同序列的有义链的转基因水稻和空载体的转基因水稻表现为典型的RDV侵染症状.HpRNA在转基因水稻中对RDV高抗性发挥重要作用. 相似文献

半矮化多分蘖突变体f2-132由60Co-γ辐射诱变粳稻品种F2-285A获得。遗传分析表明该性状受1对隐性基因控制,已将突变基因定位在第4染色体长臂上2个Indel标记C4-Z3和C4-Z4之间,物理距离为46 kb。该区间内包含一个已报道的多分蘖基因D17/HTD1,对f2-132中的D17基因测序发现编码区第395位的碱基由T突变为C,导致第132位的氨基酸由苯丙氨酸变成丝氨酸。D17/HTD1编码类胡萝卜素裂解双加氧酶7(CCD7,carotenoid cleavage dioxygenase 7),是植物激素独脚金内酯(SLs,strigolactones)合成途径中的重要酶之一。利用SLs的人工合成类似物GR24处理f2-132,其多分蘖表型受到抑制。 相似文献

A spontaneous rice mutant, erect leaf1 (elf1–1), produced a dwarf phenotype with erect leaves and short grains. Physiological analyses suggested that elf1–1 is brassinosteroid-insensitive, so we hypothesized that ELF1 encodes a positive regulator of brassinosteroid signaling. ELF1, identified by means of positional cloning, encodes a protein with both a U-box domain and ARMADILLO (ARM) repeats. U-box proteins have been shown to function as E3 ubiquitin ligases; in fact, ELF1 possessed E3 ubiquitin ligase activity in vitro. However, ELF1 itself does not appear to be polyubiquitinated. Mutant phenotypes of 2 more elf1 alleles indicate that the entire ARM repeats is indispensable for ELF1 activity. These results suggest that ELF1 ubiquitinates target proteins through an interaction mediated by ARM repeats. Similarities in the phenotypes of elf1 and d61 mutants (mutants of brassinosteroid receptor gene OsBRI1), and in the regulation of ELF1 and OsBRI1 expression, imply that ELF1 functions as a positive regulator of brassinosteroid signaling in rice. 相似文献
