The aim of the present work was to evaluate non-enzymic antioxidants during natural and artificially modulated senescence. Senescence of bean (Phaseolus vulgaris L. cv. Jantar) cotyledons was modulated by UV C irradiation or by the decapitation of plants apices. The content of beta-carotene and zeaxanthin decreased in control and decapitated plants but in UV C irradiated plants these contents increased. The degree of de-epoxidation increased in all cultivations with age. The content of total glutathione (sum of reduced and oxidized) sharply decreased in bean cotyledons grown in all conditions. Interestingly, the content of total ascorbate increased at the end of cotyledon life span of control and decapitated plants but decreased in UV plants. Decrease of reduced/oxidized ratio of ascorbate and glutathione during cotyledon ageing confirmed increasing oxidative stress during senescence in all cultivations.  相似文献

During ageing of bean (Phaseolus vulgaris L.) cotyledons in plants with modified life span the time-course of four cytokinins, ethylene, and the end products of free radical attack, lipofuscin-like pigments (LFP), were studied. UV irradiation shortened cotyledon life span, while epicotyl decapitation prolonged it. In controls, LFP increased at the senescence onset but at the end of life span it returned to the initial level. Ethylene increased more than 3-fold at the time of abscission. The content of individual cytokinins (zeatin, zeatin riboside, isopentenyl adenine, isopentenyl adenine riboside) varied differently during ageing but they did not decreased in any case under level observed in young cotyledons at the time of abscission. UV irradiation resulted in 14-fold increase in LFP concentration at the end. Ethylene increased 8-fold 2 h after irradiation. Individual cytokinins increased after UV irradiation to a different extent and time-course, nevertheless cotyledon life span was shortened. Decapitation induced LFP decrease. On day 13, LFP abruptly increased and than decreased and stayed lowered until abscission. Ethylene was maximum on day 24, at the time of abscission, it was above 200 % of control. Decapitation produced transient decrease in some cytokinins namely zeatin and isopentenyl adenine riboside.  相似文献

We employed continuous irradiation (CL) for induction of premature senescence caused by enhanced production of reactive oxygen species. As a model plant we used bean (Phaseolus vulgaris L. cv. Jantar) cotyledons because they have well defined and a quite short life span. Senescence of bean cotyledons induced by CL progressed more rapidly than natural senescence: the life span of CL cotyledons was 13 d compared to 16 d in controls (C). Chl content was significantly lower in 10- and 13-d-old CL plants than in C plants and the change with age was not statistically significant. Activities of all antioxidative enzymes declined either with senescence onset or during whole life span. Activity of antioxidative enzymes, except ascorbate peroxidase, was lower in CL plants compared to C plants. On the contrary, contents of non-enzymatic antioxidants -carotene and ascorbate were higher in CL plants than in C plants. No significant difference, except in the youngest cotyledons, was observed in glutathione content.  相似文献

Deducing from the fall of chlorophyll (Chl) concentration expressed per unit protein, senescence started after the 10th day in non-decapitated plants. Decapitation postponed the onset of senescence until the 21st day. Expression of Chl amount per unit dry mass did not detect any changes in LI plants, hence this parameter can not be used for the assessment of senescence. The measurements of Chl a and b concentrations indicated that the light-harvesting complexes (LHCs) proliferated during ageing and were rapidly destroyed at the onset of senescence. Changes of the concentrations of carotenoids supported the hypothesis of free radicals involvement in senescence. The bean cotyledons responded to free radical production induced under higher irradiance by increased β-carotene synthesis. Oxidative damage to galactolipids during senescence was documented by fluorescence measurements. The changes in cotyledon composition were correlated to morphologic changes observed by electron microscopy.  相似文献

Stems of chrysanthemum (Chrysanthemum morifolium Ramat.) cv. Maghi were harvested when half of the buds showed colour and were put in distilled water at 21°C. Flowers showed visible senescence symptoms after 12–15 d. Reactive oxygen species (ROS) concentration and lipid peroxidation increased from young floret stage to the senescent stage. Activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), peroxidase (POD) and catalase (CAT) showed uniform increases from young floret through to the mature stage and thereafter, declined. Among the SOD isoforms, Fe-SOD and Cu/Zn-SOD were induced during the onset of senescence. Similarly different isoforms of APX and glutathione reductase (GR) also appeared during the senescence process. The capacity of the antioxidative defence system increased during the onset of senescence but the imbalance between ROS production and antioxidant defences ultimately led to oxidative damage. It is proposed that a decrease in the activity of a number of antioxidant enzymes that normally prevent the build up of free radicals can at least partially account for the observed senescence of chrysanthemum florets.  相似文献

French bean (Phaseolus vulgaris L.) cotyledons lost most of their reserve substances during several early days of germination and turned green. In cotyledon mesophyll cells of one-week-old seedlings, plastids were represented predominantly by amyloplasts (starch grains) and chloroamyloplasts, and the cells appeared to be metabolically highly active. Cell heterogeneity associated with distance of the cells from cotyledon vascular bundles was evident. Only mesophyll cells near to the bundles were rich in plastids. In two-weeks-old intact bean plants, the cotyledons were yellow and shrunken, and their cells were nearly "empty". The plastids in them were represented by senescent plastids (gerontoplasts) only. In the gerontoplasts as well as freely in cytosol, fluorescent lipoid inclusions were accumulated. This cotyledon development was more or less independent of irradiance. In "decapitated" bean plants, senescence of mesophyll cells and plastids was slowed down considerably, and the life span of the cotyledons was prolonged.  相似文献

Plants have evolved on the earth, indicating the morphology, growth and development, and life cycle of plants are highly influenced by gravity as well as other environmental stimuli. Indeed, simulated microgravity on a clinostat or hypergravity on a centrifuge has recently been reported to change the growth and development of plants (Hoson et al. 1992, 1993, 1995, Rasmussen et al. 1994, Kasahara et al. 1995). Senescence is a final drastic phenomenon in life cycle of plants, which is characterized by the loss of total chlorophyll and protein, and/or the formation of the abscission (Osborne 1973, Thimann 1977, Addicott 1982). Many environmental stimuli as well as the qualitative and quantitative changes of plant hormones have been reported to affect plant senescence. Among those stimuli, light is the most important factor to regulate plant senescence (Leopold 1964). Dark condition promotes leaf senescence due to the decrease in endogenous level of cytokinin and/or the increase in that of abscisic acid or ethylene (Tetley and Thimann 1974, Gepstein and Thimann 1980). However, there are few reports concerning the effect of gravity on leaf senescence. Strenuous effort to learn leaf senescence under microgravity condition has been done using a three-dimensional (3-D) clinostat. In this paper, we report that simulated microgravity condition on a 3-D clinostat promoted the senescence of oat leaf segments in the dark. A possible mechanism of microgravity condition on promoting the senescence is also discussed.  相似文献

Several characteristics of mitochondrial respiration and energy status have been studied during growth and senescence of mung bean ( Phaseolus radiatus L.) cotyledons. The results showed that mitochondrial oxygen consumption, respiratory control, ADP:O ratios, and energy charge changed in the cotyledons during germination and growth of the seedlings. The respiration rate of intact cotyledons approximately reflected the trend of the oxidative activities of the isolated mitochondria. An increase was observed in both whole cotyledon respiration and mitochondrial oxygen uptake at the onset of senescence of mung bean cotyledons (day 3 after germination), which thereafter declined gradually. The capacity and activity of the alternative pathway increased markedly in mitochondria isolated from senescent cotyledons. After the onset of senescence, the mung bean cotyledon mitochondria exhibited a decrease both in the respiratory control ratios and ADP:O ratios, and the cotyledons exhibited a gradual decline in energy charge. All these results showed an irreversible deterioration of energy conservation in mung bean cotyledons. The role(s) of the alternative pathway in senescent mung bean cotyledons is discussed.  相似文献

Senescence is a highly orchestrated developmental stage in the life cycle of plants. The onset of senescence is tightly controlled by signaling cascades that initiate changes in gene expression and the synthesis of new proteins. This complement of new proteins includes hydrolytic enzymes capable of executing catabolism of macromolecules, which in turn sets in motion disassembly of membrane molecular matrices, leading to loss of cell function and, ultimately, complete breakdown of cellular ultrastructure. A distinguishing feature of senescence that sets it apart from other types of programmed cell death is the recovery of carbon and nitrogen from the dying tissue and their translocation to growing parts of the plant such as developing seeds. For this to be accomplished, the initiation of senescence and its execution have to be meticulously regulated. For example, the initiation of membrane disassembly has to be intricately linked with the recruitment of nutrients because their ensuing translocation out of the senescing tissue requires functional membranes. Molecular mechanisms underlying this linkage and its integration with the catabolism of macromolecules in senescing tissues are addressed.  相似文献

The objective of the present work was to study biochemical alterations in lime plants infected by the Candidatus Phytoplasma aurantifoliae. Changes in antioxidant activities, content of chlorophylls (Chl), carotenoids (Car), soluble proteins, sugars and auxin (IAA) in infected plant were investigated. The activities of polyphenol oxidase (PPO), peroxidase (POX) and superoxide dismutase (SOD) were observed to be greater in infected leaves than the healthy control. Also according to non-denaturing PAGE, in infected leaves all the antioxidative enzymes isoforms were stronger than that of the healthy control. These results suggest that antioxidant enzymes can be activated in response to infection by phytoplasma. The decrease in content of proteins, total soluble and reducing sugars in infected plants point out changes in host metabolism due to the phytoplasma infection. The reduction in chlorophylls and auxin content shows that the phytoplasma can interfere in photosynthesis and induces senescence in the leaf. In conclusion, this study provides new insights into the lime response to phytoplasma infection.  相似文献

Plant ageing and senescence are associated with increased levels of reactive oxygen species. Level of cytokinins, the apparent inhibitors of plant senescence, is controlled by their irreversible degradation catalysed by cytokinin oxidase/dehydrogenase (CKX). We investigated the CKX activity, cytokinin concentration, and activities of antioxidative enzymes in tobacco (Nicotiana tabacum L. cv. Samsun NN) overexpressing the Arabidopsis gene for AtCKX2, targeted for extracellular secretion pathway. The control and AtCKX2 plants differed substantially in their phenotypes. When the lowest leaves in controls became yellow all leaves in AtCKX2 tobacco still remained green. Activities of antioxidant enzymes decreased with leaf age in both tobacco plants except for ascorbate peroxidase (APX) in the old leaves and glutathione reductase (GR) in young leaves. Enhancement of GR activity at all leaf stages, an increase of superoxide dismutase and a decline of catalase in young leaves, as well as an increase of APX in the oldest leaves were observed in AtCKX2 plant compared to control. Similar changes were detected after determination of isoenzymes on zymograms. It is evident that AtCKX2 plants had postponed onset of senescence despite the significantly lowered level of cytokinins. Enhanced antioxidant protection, especially in the oldest leaves, could subsidise this phenomenon.  相似文献

Darkness mediates different senescence-related responses depending on the targeting of dark treatment (whole plants or individual leaves) and on the organs that perceive the signal (leaves or cotyledons). As no data are available on the potential role of darkness to promote senescence when applied to individual cotyledons, we have investigated how darkness affects the progression of senescence in either a single or both individually darkened cotyledons of young 10-day-old Cucurbita pepo (zucchini) seedlings. Strong acceleration of senescence was observed when both cotyledons were darkened as judged by the damage in their anatomical structure, deterioration of chloroplast ultrastructure in parallel with decreased photosynthetic rate and photochemical quantum efficiency of PSII. In addition, the endogenous levels of cytokinins (CKs) and IAA were strongly reduced. In a single individually darkened cotyledon, the structure and function of the photosynthetic apparatus as well as the contents of endogenous CKs and IAA were much less affected by darkness, thus suggesting inhibitory effect of the illuminated cotyledon on the senescence of the darkened one. Apparently, the effect of darkness to accelerate/delay senescence in a single darkened cotyledon depends on the light status of the other cotyledon from the pair. The close positive correlation between CK content and the activity of CK oxidase/dehydrogenase (CKX; EC 1.4.3.18/1.5.99.12) suggested that CKX was essentially involved in the mechanisms of downregulation of endogenous CK levels. Our results indicated that CKX-regulated CK signaling could be a possible regulatory mechanism controlling senescence in individually darkened cotyledons.  相似文献

In the present study, we evaluate the protective effect of nitric oxide (NO) against senescence of rice leaves promoted by methyl jasmonate (MJ). Senescence of rice leaves was determined by the decrease of protein content. MJ treatment resulted in (1) induction of leaf senescence, (2) increase in H2O2 and malondialdehyde (MDA) contents, (3) decrease in reduced form glutathione (GSH) and ascorbic acid (AsA) contents, and (4) increase in antioxidative enzyme activities (ascorbate peroxidase, glutathione reductase, peroxidase and catalase). All these MJ effects were reduced by free radical scavengers such as sodium benzoate and GSH. NO donors [N-tert-butyl-α-phenylnitrone (PBN), sodium nitroprusside, 3-morpholinosydonimine, and AsA+NaNO2] were effective in reducing MJ-induced leaf senescence. PBN prevented MJ-induced increase in the contents of H2O2 and MDA, decrease in the contents of GSH and AsA, and increase in the activities of antioxidative enzymes. The protective effect of PBN on MJ-promoted senescence, MJ-increased H2O2 content and lipid peroxidation, MJ-decreased GSH and AsA, and MJ-increased antioxidative enzyme activities was reversed by 2-(4-carboxy-2-phenyl)-4,4,5,5-tetramethyl-imidazoline-1-oxyl-3-oxide, a NO-specific scavenger, suggesting that the protective effect of PBN is attributable to NO released. Reduction of MJ-induced senescence by NO in rice leaves is most likely mediated through its ability to scavenge active oxygen species including H2O2  相似文献

Respiration and net photosynthetic O2 production by cotton cotyledons were determined from an early age through the senescent stage. Various treatments were applied to cotyledons to assess the importance of current photosynthesis as compared to translocation of reserves to seedling development. Rates of respiration and net photosynthesis per cm2 were high on 1-day-old cotyledons, but the rates decreased sharply with rapid expansion to reach a fairly stable rate. Respiration per cotyledon decreased linearly with age until the onset of senescence, then exhibited a distinct climacteric rise followed by a sharp decrease. Net photosynthesis per cotyledon increased until expansion was completed and then decreased linearly and steeply with age. Excision of cotyledons, inhibition of photosynthesis either chemically or by covering, and removal of the terminal bud indicated that current photosynthesis is a potent force behind early epicotyl growth.  相似文献

Senescence of detached mustard cotyledons was greatest in full light or at low light intensity but was reduced at intermediate light intensities. The location of yellowing in the blade depended on the conditions in which it was maintained after excision. Cotyledons cultured at full light intensity in the absence of exogenous nutrients formed roots at the base and yellowed around the margins. Reduction in light intensity and increasing nutrient concentrations resulted in an increasing proportion of the cotyledons yellowing from the base upwards, and fewer developed roots. At low light intensity with high nutrient levels rooting was suppressed and basal senescence predominated. The location of yellowing in the cotyledon may be associated with mobilisation gradients induced by the differential effects of light and mineral nutrient level on root initiation and blade growth.  相似文献

The flower is the most significant and beautiful part of plants. Flowers are very useful organs in plant developmental phenomenon. During flower bud opening, various events takes place in a well defined sequence, representing all aspects of plant development, such as cell division, cellular differentiation, cell elongation or expansion and a wide spectrum of gene expression. The complexity of flower bud opening illustrates that various biological mechanisms are involved at different stages. Senescence represents the ultimate stage of floral development and results in wilting or abscission of whole flower or flower parts. Senescence is an active process and governed by a well defined cell death program. Once a flower bud opens, the programmed senescence of petal allows the removal of a metabolically active tissue. In leaves, this process can be reversed, but in floral tissue it cannot, indicating that a highly controlled genetic program for cell death is operating. The termination of a flower involves at least two, sometimes overlapping, mechanisms. In one, the perianth abscises before the majority of its cells initiate a cell death program. Abscission may occur before or during the mobilization of food reserves to other parts of the plant. Alternatively, the petals may be more persistent, so that cell deterioration and food remobilization occur while the petals are still part of the flower. The overall pattern of floral opening varies widely between plant genera, therefore, a number of senescence parameters have been used to group plants into somewhat arbitrary categories. Opening and senescence of rose flower is still an unsolved jigsaw in the world of floriculture industry and the mechanism behind the onset of the very early events in the sequence still remains to be elucidated. Hence, for advancing the knowledge on the pertinent aspect of bud opening and senescence the literature has been cited under this review.  相似文献

萝卜离体子叶在光下或暗中衰老及激素调节衰老过程中,作为叶片衰老指标的叶绿素和蛋白质含量的降低,发生在MDA含量增高之前,更早于SOD活性的下降。表明由SOD活性降低所导致的膜脂过氧化的增强,并非衰老的原初反应,而是叶片衰老到一定程度的生理变化。因此,至少在萝卜离体子叶上,不能将其衰老的启动归因于受SOD控制的膜脂过氧化作用导致的膜累积性质变。  相似文献

Selenium (Se) is regarded as an antioxidant in animals and plants, even though considered as non-essential element in plants. To test its ability to counteract senescence related oxidative stress in soybean a pot culture experiment was conducted. The soybean plant was sprayed with sodium selenate (50ppm) at 78days after sowing (DAS). Soybean leaves were harvested at 80 and 90 DAS for analysis of oxidant production and antioxidative enzymes activity. Se positively promoted growth and acted as antioxidant by inhibiting lipid peroxidation and per cent injury of cell membrane. The antioxidative effect was associated with an increase in superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) enzymes activity. Significant increase in antioxidant enzyme activity was positively related to Se content. The decrease in antioxidative enzymes at 90 DAS was much faster in control plants than Se-sprayed plants. The reduction in SOD and GSH-Px may be associated with senescence-induced oxidative burst.  相似文献
