Outer membrane proteins of Brucella have been classified as group 1 (94 or 88 kDa), group 2 (36–38 kDa), and group 3 (31–34 and 25–27 kDa). Two proteins of 25 and 31 kDa with only 34% of identity are included in group 3 and they are coded for by the omp25 and omp31 genes. Proposed study planned to detect antibodies to Brucella melitensis Omp31 in farm goats having history of B. melitensis induced abortions, in B. melitensis-infected goats and sheep. By enzyme-linked immunosorbent assay (ELISA), using recombinant Omp31 as antigen, of 872 farm goats antibodies to Omp31 were detected in 112 (12.8%) cases. Out of 14 naturally infected goats infected with B. melitensis 12 (85.7%) showed anti Omp31 antibodies. Out of 10 naturally infected sheep with Brucella ovis, antibodies to Omp31 were detected only in 6 (60%) cases and in 18 (81.8%) out of 22 cases infected with B. melitensis. Obtained results were also compared with the rose Bengal plate test (RBPT). In controlled experiments, sensitivity and specificity of recombinant Omp31 (rOmp31) ELISA and RBPT were also evaluated and it was found that former test is 100% specific though RBPT has slightly higher sensitivity. In this study, we found a significant difference between the two groups (B. melitensis and B. ovis infected) in terms of the percentage of positive reactions or signal level by an ELISA. The reactivity of the positive sera against the purified rOmp31 was also tested by Western blotting. Sera from B. melitensis-infected animals showed a strong reactivity in comparison to sera from B. ovis-infected animals. The potential diagnostic usefulness of this antigen in combination with other recombinant proteins from B. melitensis would be of great importance in future in eradication of brucellosis.  相似文献

为了阐明木兰围场天然次生林优势种群的结构和动态特征,对群落乔木层组成以及优势种群的径级结构、静态生命表、存活曲线、死亡曲线、生存函数和时间序列预测等进行研究.结果表明: 群落乔木层有9个种群,其中,五角枫、蒙古栎、糠椴和白桦种群为群落的优势种群;五角枫和糠椴种群存活曲线接近于Deevey-Ⅱ型,蒙古栎种群存活曲线趋于Deevey-Ⅲ型,白桦种群存活曲线为Deevey-Ⅱ型;随着径级的增加,各优势种群的死亡率和消失率曲线呈现相似的变化趋势,五角枫、蒙古栎、糠椴种群的消失率和死亡率分别在Ⅳ、Ⅰ和Ⅰ径级下出现最大值,而白桦种群在各径级呈现稳定的变化趋势;4种生存函数曲线分析表明,五角枫、蒙古栎和糠椴种群具有前期锐减、中后期相对稳定的特点;白桦种群数量波动不明显,具有较稳定的特点.种群数量的时间序列预测表明,白桦种群小径级(Ⅰ~Ⅱ级)个体数量逐渐减少,种群未来有趋于衰退的趋势,五角枫、蒙古栎和糠椴种群趋于相对稳定.推测该次生林将逐步演替为以五角枫、蒙古栎和糠椴为主要树种的天然次生林.  相似文献

Forty isolates of entomopathogenic fungi from six genera were assessed against the two-spotted spider mite, Tetranychus urticae, in a single dose, direct application laboratory bioassay on tomato leaflets. Only three isolates caused greater mortality than the control: these were Metarhizium anisopliae 442.99, Hirsutella spp. 457.99, and Verticillium lecanii 450.99. These isolates were assessed in a multiple dose bioassay, together with three isolates cultured from commercial biopesticides as follows: Beauveria bassiana 432.99 (cultured from 'Naturalis-L', Troy Biosciences, Phoenix, TX, USA); Hirsutella thompsonii 463.99 (cultured from 'Mycar', Abbott Laboratories USA); and V. lecanii 19.79 (used in 'Mycotal' Koppert BV, The Netherlands). Beauveria bassiana 432.99, H. thompsonii 463.99, M. anisopliae 442.99, and V. lecanii 450.99 were all pathogenic to T. urticae in this bioassay. In addition, it was found that the mortality caused by B. bassiana 432.99 and Naturalis-L was increased when the mites were exposed to tomato leaflets sprayed previously with conidia suspensions, compared to spraying the mites directly. In a glasshouse experiment, sprays of B. bassiana 432.99, H. thompsonii 463.99, M. anisopliae 442.99, V. lecanii 450.99 and Naturalis-L reduced T. urticae populations in a tomato crop grown according to commercial practice. Naturalis-L reduced T. urticae numbers by up to 97%. In a second glasshouse experiment, single sprays of Naturalis-L and the acaricide fenbutatin oxide (Torq) were compared as supplementary treatments to release of the predatory mite, Phytoseiulus persimilis. Supplementary sprays of fenbutatin oxide reduced the numbers of T. urticae nymphs (80% reduction), while Naturalis-L reduced numbers of T. urticae adults, nymphs and eggs (98% reduction in all three cases). It is concluded that Naturalis-L has the potential to be used against T. urticae on glasshouse tomato crops.  相似文献

Our current understanding of congenital transmission of Toxoplasma gondii from ewe to lamb dictates that infection frequently results in abortion and the death of the developing foetus, that the birth of live infected lambs occurs rarely and that the cat is the predominant source of infection in ewes. Using direct polymerase chain reaction detection of T. gondii, we report high levels of congenital transmission occurring in a commercially managed sheep flock. We sampled foetal-derived placental tissue and tissues from aborted lambs and showed that congenital transmission was detected in these tissues from 61% of all pregnancies. Where pregnancies resulted in the death of one or more lambs, T. gondii was detected in the lamb tissue for all but one of 18 (94%) pregnancies. Of the successful pregnancies resulting in the birth of live lambs we were able to detect T. gondii in foetal-derived placental tissue from 37 of 70 (42%) pregnancies. These results show that congenital transmission is occurring in a high percentage of lambings including normal healthy lambings, at this farm, suggesting that this route of transmission from generation to generation may be much more significant than that reported previously. These results may have implications for sheep husbandry and future epidemiological studies of T. gondii.  相似文献

Adult whiteflies are characterized by the presence of copious amounts of wax particles covering all surfaces of the body except the eyes. The lipid composition was determined for wax particles removed from the surfaces of the sweetpotato whitefly, Bemisia tabaci (Gennadius), and the greenhouse whitefly, Trialeurodes vaporariorum (Westwood). The lipid components in the wax particles of both species were mostly mixtures of long-chain aldehydes and long-chain primary alcohols. The major wax particle components for B. tabaci were C34 aldehyde and C34 alcohol and small amounts of C32 aldhyde and alcohol. For the wax particles from T. vaporariorum, C32 aldehyde and C32 alcohol were the major components with lesser amounts of the C30 components. These findings were compared to the surface lipids of fully-waxed B. tabaci and T. vaporariorum adults that contained, in addition to the major amounts of long-chain aldehydes and alcohols, quantities of long-chain wax esters. Wax esters were not present in lipid extracts from the surface of B. tabaci whiteflies at the time of adult emergence (prior to deposition of wax particles). Thus, the appearance of wax esters on the cuticular surfaces occurred during the period of deposition of wax particles. The quantities of wax esters in the surface lipid extracts of wing tissues separated from the bodies of adult whiteflies indicated that the wing surfaces were a major site of wax ester deposition.  相似文献

Lyme disease is a tick-borne zoonotic infectious disease caused by Borrelia burgdorferi. The present study assessed the infection status of B. burgdorferi among horses reared in Korea using ELISA and PCR. Between 2009 and 2013, blood samples were collected from 727 horses throughout Korea. Data for each animal including age, gender, breed, and region of sample collection were used for epidemiological analysis. Overall, 38 (5.2%; true prevalence: 5.5%) of 727 horses were seropositive by ELISA. There were statistically significant differences according to breed and region (P<0.001) whose differences might be attributed to the ecology of vector ticks and climate conditions. Using 2 nested PCR, none of the samples tested positive for B. burgdorferi. Thus, a positive ELISA result can indicate only that the tested horse was previously exposed to B. burgdorferi, with no certainty over the time of exposure. Since global warming is likely to increase the abundance of ticks in Korea, continuous monitoring of tick-borne diseases in Korean horses is needed.  相似文献

Hammondia hammondi and Toxoplasma gondii are two related coccidian parasites, with cats as definitive hosts and warm-blooded animals as intermediate hosts. It is difficult to differentiate them by morphological and serological parameters. In the present study, primers were designed to specifically amplify the ITS-1 region of H. hammondi to differentiate it from T. gondii. Attempts were made to detect the presence of H. hammondi DNA in the tissues of mice infected with H. hammondi alone, as well as from mixed infections with T. gondii, using the newly designed primers. The de novo primers effectively amplified the H. hammondi-specific target fragment from all samples containing H. hammondi, including those with concomitant T. gondii infection. Further, the primers did not amplify any fragment from the related parasites like T. gondii, Neospora caninum and Hammondia heydorni. The new primers provide simple and efficient means to differentially diagnose H. hammondi from T. gondii even in samples containing both parasites, thus obviating the need for other labourious techniques like mouse bioassay and in vitro cultivation.  相似文献

Lambs were infected with 6000 Trichostrongylus colubriformis L3 per week for 18, 12 or 6 weeks, beginning at ages 14, 20 and 26 weeks, respectively. At the end of the primary infection subgroups had geometric mean adult worm burdens of 5000, 15,000 and 18,000, respectively. In the remaining sheep the worm population was removed with anthelmintic, and sheep had no further larval intake until challenged with T. colubriformis 1,6, 12, 18 or 24 weeks later. In the groups given 18 or 12 weeks primary infection, establishment of challenge doses was low (less than 25% of establishment in helminth naive controls) in most animals at all challenge times. However, for the groups given 6 weeks primary infection, establishment was low only at the first two challenge times. Thereafter it had similar mean to control groups, but much greater variance. Other subgroups were challenged with T. colubriformis, Ostertagia circumcincta and Haemonchus contortus 1 week after worm removal. In these animals T. colubriformis establishment was not different to animals challenged at the same time with T. colubriformis alone, however immunity to T. colubriformis afforded little protection against the other species. The results of this experiment were incorporated into a simulation model of the population dynamics of T. colubriformis.  相似文献

Cattle on two typically managed drylot dairies were serologically monitored from birth through year 1 to year 4 of life to characterise congenital and postnatal Neospora caninum transmission. Of the 456 calves enrolled, 284 were classified as N. caninum negative and 172 were classified as N. caninum positive. Ninety-six percent of congenitally infected calves were seropositive for all samples tested. Seven (4%) of the 172 congenitally infected animals had a period that persisted for 9 to 18 months when they were seronegative; however, all returned to seropositive status by 25 months of age. In N. caninum-negative calves, colostral antibody decayed by 128 days, with an estimated half-life of 19.6 +/- 5.2 days. Of the 284 calves classified as negative, 18% had sporadic, isolated responses to N. caninum, typically between 29 and 35 months of age, without subsequent seroconversion or infection. During the study, 17 animals seroconverted and remained seropositive throughout the follow-up. Thirteen of the seroconversions occurred in the neonatal period; however, in nine of 10 where dam status was available, the dam was N. caninum positive, suggesting late gestation congenital infection rather than postnatal infection. Seroconversion was detected in an additional four animals, between 13 and 22 months of age. The estimate of postnatal infection rate was less than 1% per year despite a high N. caninum seroprevalence in the herds, and the presence of potential definitive and intermediate hosts on the dairy throughout the study. The extremely low rate of postnatal infection, as well as the lifelong persistence of congenital infection, emphasises the importance of congenital transmission in maintaining N. caninum infection in dairies.  相似文献

Adult, laboratory-bred squirrel monkeys (Saimiri sciureus) previously infected with either Leishmania braziliensis braziliensis or L. b. panamensis were challenge infected with blood-form trypomastigotes of Trypanosoma cruzi (Brazil strain). Monkeys previously infected with T. cruzi were challenged with stationary phase promastigote forms of L. b. braziliensis. Monkeys were examined during the course of challenge for evidence of infection, electrocardiographic alterations and parasite-specific antibody responses. T. cruzi epimastigotes were cultured from the blood of monkeys up to 3 months after challenge with this parasite. Unulcerated cutaneous lesions appeared and persisted in monkeys challenged with L. b. braziliensis. The formation of satellite lesions was observed in one monkey. Increased QRS intervals were not observed in T. cruzi challenged monkeys without prior cardiac irregularities and QRS left axis shifts were observed in only two of these monkeys. Elevated titers of parasite binding IgM and IgG specific for both T. cruzi and L. braziliensis were observed in all monkeys following challenge. These results indicate that prior infection with T. cruzi or L. braziliensis does not protect against heterologous challenge infection with these organisms. However, prior infection with Leishmania parasites may provide some protection against chagasic cardiopathies.  相似文献

Conidial suspensions and cell-free filtrates of Trichoderma harzianum isolates were evaluated for their effectiveness in reducing perithecial and ascospore production of Gibberella zeae on wheat straw. Isolate T-22, which is registered in the US as a biological control agent (Plant Shield™), was included in the study as a positive control. When co-inoculated with G. zeae all 11 isolates of T. harzianum significantly reduced perithecial numbers on wheat straw. Five T. harzianum isolates, including T-22, reduced perithecial formation by 70% or greater. All isolates of G. zeae, varied in their ability to produce perithecia. Isolate 192132 produced the greatest number of perithecia and was used to further evaluate the effect of application time of the T. harzianum isolates. Perithecial reduction was highest (96-99%) when T. harzianum conidial suspension or cell-free filtrate was applied to straw 24 h prior to inoculation with G. zeae. Control was less effective when T. harzianum was applied at the same time (co-inoculated) or 24 h after G. zeae. Treatments which reduced perithecial numbers also reduced ascospore numbers; however, the average numbers of ascospores per perithecia were not significantly lowered. Field trials showed significant reduction of perithecia on residues treated with T. harzianum prior to placement on the soil surface. Both T. harzianum and G. zeae were re-isolated from residues sampled in July and August after 30 and 60 days of exposure to the environment.  相似文献

生物农药是当前棉花黄萎病绿色防治的重要途径.本研究设置田间小区试验,研究了4种生物药剂不同滴施用量(枯草芽孢杆菌粉剂15、30、45 kg·hm-2,“施倍健”哈茨木霉菌剂15、18、24 kg·hm-2,渝峰“99植保”15、22.5、30 kg·hm-2,中农绿康30、45 和60 kg·hm-2)对棉花黄萎病的防治效果及土壤微生物群落组成和多样性的影响.结果表明: 各施药处理均不同程度降低了棉花黄萎病发病率和发病指数,全生育期平均防病效果达到50.0%~77.4%;木霉菌剂施药量18 kg·hm-2的防效显著高于15 和24 kg·hm-2,其他3种药剂防病效果均与施药量呈正相关.施药后土壤中病原菌大丽轮枝菌的相对丰度显著降低,且与防病效果呈极显著负相关.土壤细菌的数量与物种丰度随着施药量的增加而显著增加.施药后土壤放线菌的数量与物种丰度也显著增加,但放线菌数量在不同施药量间相差较大.土壤真菌的数量与物种丰度除木霉菌剂随施药量增加而增加外,其他3种药剂随着施药量的增加而减少.Pearson相关性分析显示,施药组的防病效果与细菌和放线菌数量呈正相关关系,而与真菌数量呈显著负相关;与放线菌、硝化螺旋菌门细菌、子囊菌门真菌、壶菌门真菌丰度呈现显著正相关,而与半知菌门真菌丰度呈显著负相关.建议在滴施用量上渝峰“99植保”30 kg·hm-2、中农绿康60 kg·hm-2、枯草芽孢杆菌可湿粉剂45 kg·hm-2、“施倍健”哈茨木霉菌剂18 kg·hm-2.  相似文献

The biotransformations of a series of substituted sulfides were carried out with the filamentous fungi Botrytis cinerea, Eutypa lata and Trichoderma viride. Several products underwent microbial oxidation of sulfide to sulfoxide with medium to high enantiomeric purity. With regard to sulfoxide enantioselectivity, the (R)-enantiomer was favoured in biotransformations by T. viride and E. lata while the (S)-enantiomer was favoured in those by B. cinerea. Hybrids between female Trichinella spiralis and male Trichinella britovi were constructed. Then, hybrid genotype was characterized by DNA markers including mitochondrial cytochrome c oxidase subunit I (CO I) gene, the gene encoding the 43-kDa excretory–secretory (ES) protein, and genomic DNA fragments specific for T. spiralis and T. britovi identified from random amplified polymorphism DNA (RAPD). PCR-restriction fragment length polymorphism (PCR-RFLP) analysis of the mitochondrial CO I gene revealed that all hybrids carried a T. spiralis pattern. The same analysis of the gene encoding the 43-kDa ES protein showed that each hybrid carried both T. spiralis and T. britovi gene type simultaneously. In the analysis of genomic DNA using RAPD-derived PCR primers, some hybrids carried T. spiralis and T. britovi-specific RAPD markers, while others carried the RAPD marker of T. spiralis only.  相似文献

The interaction between Trypanosoma congolense and Haemonchus contortus was studied in 5 groups of 8 Djallonké sheep. Two groups received a single infection with either H. contortus or T. congolense, and 2 groups were infected with T. congolense followed by H. contortus (TH) or vice versa (HT). One group was kept as uninfected controls. Mortality due to infection was observed only in the dual infection groups. In the TH group, the effects were more acute whereas in the HT group they were more chronic. No significant differences in weight gain could be demonstrated between infected and control groups. Djallonké sheep are able to withstand a single infection with either T. congolense or H. contortus, which confirms their trypanotolerant nature and provides preliminary indication of resistance against helminth infections. However, when exposed to successive infections with both parasites, some of the animals lose this tolerance.  相似文献

Despite evidence of West Nile virus (WNV) activity in Colombia, Venezuela and Argentina, this virus has not been reported in most South American countries. In February 2009, we commenced an investigation for WNV in mosquitoes, horses and caimans from the Pantanal, Central-West Brazil. The sera of 168 horses and 30 caimans were initially tested using a flaviviruses-specific epitope-blocking enzyme-linked immunosorbent assay (blocking ELISA) for the detection of flavivirus-reactive antibodies. The seropositive samples were further tested using a plaque-reduction neutralisation test (PRNT90) for WNV and its most closely-related flaviviruses that circulate in Brazil to confirm the detection of specific virus-neutralising antibodies. Of the 93 (55.4%) blocking ELISA-seropositive horse serum samples, five (3%) were seropositive for WNV, nine (5.4%) were seropositive for St. Louis encephalitis virus, 18 (10.7%) were seropositive for Ilheus virus, three (1.8%) were seropositive for Cacipacore virus and none were seropositive for Rocio virus using PRNT90, with a criteria of ≥ four-fold antibody titre difference. All caimans were negative for flaviviruses-specific antibodies using the blocking ELISA. No virus genome was detected from caiman blood or mosquito samples. The present study is the first report of confirmed serological evidence of WNV activity in Brazil.  相似文献

白及是地生兰科多年生草本植物,是常见的中草药之一。白及根是白及植株吸收养分和水分的主要场所,开展其根部内生真菌的群落组成及多样性分析,对了解云南原生境白及与根部内生真菌间的共生关系具有重要意义。本研究采用形态学和ITS rDNA分子系统发育分析相结合的方法进行菌株鉴定。结果表明,3个采样点的白及根部分离到可培养内生真菌32株,包括9个目、13个科、16个属。其中木霉属Trichoderma、镰刀菌属Fusarium、拟盘多毛孢属Pestalotiopsis为优势类群,分别占总物种数的21.88%、18.75%、9.38%;中碳垫菌属Nemania、丛赤壳属Nectria、炭角菌属Xylaria、紫霉属Purpureocillium、刺盘孢属Colletotrichum、黑孢子菌属NigrosporaBiscogniauxia、腐质霉属Humicola、脉孢菌属Neurospora、拟茎点霉属Phomopsis、植物腐霉Phytopythium、毛霉属Mucor、伞状霉属Umbelopsis为常见内生真菌类群。α多样性指数分析表明:各采样点的白及根部内生真菌物种多样性最高的是昆明市水源保护区采样点(D=0.799,H’=1.698),保山市龙陵县采样点物种多样性最低(D=0.787,H’=1.580)。β多样性分析采样点之间的Jaccard(0.912-0.993)、Bray-Curtis(0.838-0.986)和UniFrac(加权0.618-0.631;不加权0.770-0.799)距离,结果表明3个采样点之间的内生真菌群落组成存在差异。3个采样点的不同地理环境和土壤类型给白及提供了不同的生境条件,是影响白及根部内生真菌物种丰度和群落组成差异的主要因素之一。  相似文献

Anthracnose disease caused by Colletotrichum gloeosporioides in Jingkwang mango grown in Taiwan was significantly reduced by the integration of fruit bagging with either B. subtilis strain LB5 or fungicides. The combined treatments were most effective in reducing early infection during the 2004 season, leading to 56.4 and 58.3% reduction, respectively, while in 2003 reduction accounted for 51 and 52.3%, respectively. Post-harvest application of B. subtilis strain LB5 cell suspensions on fruits already treated by bagging, bagging+LB5 and baggingfungicides in the field reduced anthracnose incidence significantly at all tested concentrations. These results indicate that biocontrol efficacy of B. subtilis LB5 may be due to the prevention of early fruit infection, thereby reducing significantly anthracnose incidence in ripening fruits to much lower levels than those obtained by using a conventional single post-harvest treatment.  相似文献

Distal ileum, caecum and proximal colon of 470 horses were examined for helminths during 1 year at an abattoir in central Sweden. The infection levels of the horse tapeworm Anoplocephala perfoliata, their stage of development, site of attachment and gross pathological lesions caused by the worm were recorded. Faecal samples from 395 of the horses were examined specifically for tapeworm segments and eggs in order to correlate these findings with the numbers in the alimentary canal. In total 65% of the horses were infected with A. perfoliata and the mean intensity of infection was 79 worms per infected horse with a maximum of 912. The level of infection was significantly higher in (1) 3rd and 4th than in 1st and 2nd quarter of the year; (2) older horses than in yearlings; (3) females than in males and geldings; (4) thoroughbreed and cold-blooded horses than in Swedish standardbreeds and ponies. The level of infection was unaffected by the usage of anthelminthics against nematodes. Of the horses examined 51% had 1-100 worms whereas 14% were infected with more than 100 worms. Of the tapeworm positive horses 72% had mixed infections with both adult and juvenile worms, 20% solely juveniles, and 8% solely adults. The severity of intestinal lesions exacerbated by increasing numbers of A. perfoliata. About 11% of the intestines examined had severe lesions, but there was no history of acute abdominal distress in any of the horses included in this study. Although the number of detectable eggs was significantly higher for horses heavily infected with A. perfoliata, the egg recovery among infected horses was only 35%. An additional field survey comprising 218 horses on 88 premises in central and southern parts of Sweden showed that the prevalence of A. perfoliata egg positive horses was the same as found on faecal examination during the abattoir survey.  相似文献

Observations are reported on seasonal changes in the age structure of populations of nematodes of the family Trichonematidae recovered at regular intervals from the lumens of the large intestines of horses slaughtered in S.W. England. The results show that changes in the size of parasitic populations of Trichonema nassatum follow seasonal variations in the rate of infection, more individuals maturing during summer/autumn than during winter/spring because of proportional differences in the numbers of infective larvae ingested from the pasture. In contrast, larvae of T. longibursatum, T. catinatum and T. goldi ingested by grazing horses during summer accumulate in the gut wall rather than promptly returning to the lumen and developing to maturity. Their development is apparently inhibited until the following spring when 4th-stage larvae emerge en masse and quickly reach the adult stage. The relationship between fluctuations in the size of the adult populations of the 4 species studied and the characteristic seasonal variations in horse faecal egg counts is discussed.  相似文献
