The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. The responses of net CO2 assimilation to sudden changes in irradiancewere studied in Phaseolus vulgaris L. in the laboratory andthe field. For irradiance changes between 50 µmol m–2s–1 to 350 µmol m–2 s–1 in the laboratory,assimilation rate increased with half-times of 2.7 and 4.1 minin well-watered and water-stressed plants, respectively. Ina field experiment with a change in irradiance from 400 to 1200µmol m–2 s–1 the response was faster (half-time=c.1.2 min). In all cases when irradiance was returned to a lowvalue, assimilation declined rapidly with a half-time of approximately1 min, which approached the time resolution of the gas-exchangesystem. The corresponding changes in stomatal conductance in responseto both increasing and decreasing irradiance were much slowerthan the assimilation responses, indicating that biochemicalprocesses, rather than CO2 supply, primarily determined theactual rate of assimilation in these experiments. The conceptof stomatal limitation to photosynthesis is discussed in relationto these results. A simple model for assimilation in a fluctuating light environmentis proposed that depends on a steadystate light response curve,an ‘induction lag’ on increasing irradiance, andan induction-state memory. The likely importance of taking accountof such induction lags in natural canopy microclimates is considered. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

A comparative study was made of the inhibition of ribulose-1,5-bisphosphatecarboxylase-oxygenase (Rubisco) amongst six cultivars of Glycinemax L. Merr., associated with synthesis of 2-carboxyarabinitol1-phosphate (CA1P) during darkness. Significantly lower meanvalues of dark inhibition of Rubisco were observed in soybeancv. Davis than in cvs Bragg, Cobb, Hardee, Gordon, and Kirby.The CA1P synthesis/degradation cycle during dark/light transitionsremained operational in cv. Bragg plants grown at low irradiance(40 µmol photons m–2 s–1). However, CA1P synthesisand degradation rates were slower in the dark (t0.5 = 240 versus25 min), and light (t0.5 = 20 versus 3.8 min) respectively,as compared to plants grown at higher irradiance (550 µmolphotons m–2 s–1). In addition, the activation stateof Rubisco in low-light-grown plants showed only a small declineafter a transition to darkness. We conclude that (a) cultivar-dependentvariation occurs amongst soybeans with respect to CAlP regulationof Rubisco, and (b) soybeans acclimated to low irradiance maydepend more on CA1P synthesis/degradation to regulate Rubisco,and less on changes in the enzyme activation state. Stability of thylakoid components under supra-high irradiancewas studied with the cyanophyte Synechocystis PCC 6714. Theactivity of overall photosynthesis was quickly inactivated (T1/2=20min) under supra-high irradiance (300 W m–2, white light).In parallel with the inactivation of photosynthesis, QA in PSII was also inactivated. Both inactivations were acceleratedby chloramphenicol (CAP) addition. The reactivation of PS IIrequired weak irradiation and was suppressed by CAP. However,PS I measured as P700 was very stable. The level of PS I measuredas P700 was not significantly reduced by the irradiation for12 h even in the presence of CAP while the level of Cyt b559,component of PS II, was decreased markedly. The function ofPS I before and after supra-high irradiation with CAP was examinedby comparing sizes of P700 oxidation induced by a short flash,by a continuous light, and by determination of O2-and ferredoxin-reduction.No difference was observed in PS I actions before and afterthe irradiation treatment. These results indicate that the PSI complex is very tolerant of supra-high irradiation. However,the cells grown under supra-high irradiance contained much fewerPS I and PS II complexes than Cyt b6–f complexes. Theformer levels were reduced to a half to one fourth of thosebefore growth while the level of Cyt b6–f complex wasnot reduced so much. The Km(CO2) ancl Vmax of ribulose 1,5-bisphosphate (RuBP) carboxylaseand its protein ratio to total soluble protein from Oryza speciesincluding cultivars (25 varieties) and wild types (11 species,21 strains) were surveyed. Their variabilities among cultivarsof O. sativa were very small. The averages of the Km(CO2) andVmax values and the ratio of carboxylase to soluble protein,and their standard errors were 10.2?1.0µM, 1.72?0.13units.mg–1(pH 8.0 and 25?C) and 52?2%, respectively. However, some differencesseemed to exist based on genome constitution in the Oryza genus.RuBP carboxylases from the species with the AgAg genome, O.graberrima and O. breviligulate, exhibited low Km(CO2) values(8.0?0.8 µM). High Vmax was associated with the CC genome,O. eichingeri and O. officinalis (2.08?0.15 units.mg–1).A higher ratio of RuBP carboxylase protein to soluble proteinwas found for the AA genome, O. sativa and O. perennis. The causes of interspecific differences in the µ-l relationshipare examined in the context of a mechanistic model which relatesµ to irradiance in terms of six factors:, kc photosyntheticquotient (PQ), Chl a:C, respiration and excretion. The effectof cell size on the light saturated growth rate is also considered.It is shown that photosynthetic efficiency and PQ exhibit remarkablylittle interspecific variability, and average 0.024 ±0.005 µg C(µg Chl a)–1 h–1 (µEm–2 s–1)–1 and 1.5 ± 0.2 mol 02 molC–1 (when NO3 is the nitrogen source) respectively.Two useful relationships were derived: (i) between growth efficiency,g and Chl a:C at µ. = 0; (ii) between the compensationintensity, Ic and the Chl a-specific maintenance respirationrate. Both relationships were independent of temperature anddaylength. Species best adapted to growth at low light werefound to exhibit high Chl a:C ratios and low maintenance respirationrates. As a group, diatoms were consistently the best adaptedfor growth at low irradiance. Chiorophytes, haptophytes, chrysophytesand cryptophytes were intermediate in their performance at lowirradiance. Dinoflagellates exhibited extreme diversity, withspecies spanning the spectrum from very good performance atlow irradiance to very poor. A new µmax-cell carbon relationshipis given based on growth rates normalized to 15°C. Evidenceis presented to show that noise in this relationship can besignificantly reduced by using only carbon-specific growth ratesand using only data for species grown at the same daylength.  相似文献

Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. Ribulose bisphosphate carboxylase (EC 4.1.1.39 [EC] ) activity wasvery low in tomato leaf extracts unless prepared in the presenceof Mg2+, and polyclar AT. With young leaves, but not with fully-expanded leaves, the RuBP carboxylase activityextracted was increased by prolonged illumination of the leaves(2 h). The main effect of the light treatment was to increasethe specific activity of the enzyme but there was also a smallincrease in RuBP carboxylase protein. Tomato leaf RuBP carboxylasein extracts had specific activities in the range 0.2–0–6µmol CO2 min–1 mg–-1 total protein extracted,or 0.5–1.2 µmol CO2 min–1 mg–1 RuBPcarboxylase, and an apparent Km (CO2) at 20 ?C of 9.3 ? 1.2µM (using a of 6.407). Intact chloroplasts were isolated from mesophyll and bundlesheath protoplasts of a C4 plant, Panicum miliaceum L., to measurethe uptake of [1-14C]pyruvate into their sorbitol-impermeablespaces at 4?C by the silicone oil filtering centrifugation method.When incubated in the dark, both chloroplasts showed similarslow kinetics of pyruvate uptake, and the equilibrium internalconcentrations were almost equal to the external levels. Whenincubated in the light, only mesophyll chloroplasts showed remarkableenhancement of the uptake, the internal concentration reaching10–30 times of the external level after 5 min incubation.The initial uptake rate of the mesophyll chloroplasts was enhancedabout ten fold by light and was saturated with increasing pyruvateconcentration; Km and Vmax were 0.2–0.4 mM and 20–40µmol(mg Chl)–1 h–1, respectively. The lightenhancement was abolished by DCMU and uncoupling reagents suchas carbonylcyanide-m-chlorophenylhydrazone and nigericin. Theseresults indicate the existence of a light-dependent pyruvatetransport system in the envelope of mesophyll chloroplasts ofP. miliaceum. The uptake activity of mesophyll chloroplastsboth in the light and the dark was inhibited by sulfhydryl reagentssuch as mersalyl and p-chloromercuriphenylsulfonate, but thebundle sheath activity was insensitive to the reagents. Thesefindings are further evidence for the differentiation of mesophylland bundle sheath chloroplasts of a C4 plant with respect tometabolite transport. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) Aschers, Brassica chinensis L., Brassica juncea (L.) Czern. & Coss., Brassica oleracea L. var. gongylodes L., Camelina saliva (L.) Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. Photosynthetic rates of outdoor-grown soybean (Glycine max L.Merr. cv. Bragg) canopies increased with increasing CO2 concentrationduring growth, before and after canopy closure (complete lightinterception), when measured over a wide range of solar irradiancevalues. Total canopy leaf area was greater as the CO2 concentrationduring growth was increased from 160 to 990 mm3 dm–3.Photosynthetic rates of canopies grown at 330 and 660 mm3 CO2dm–3 were similar when measured at the same CO2 concentrationsand high irradiance. There was no difference in ribulose bisphosphatecarboxylase/oxygenase (rubisco) activity or ribulose 1,5-bisphosphate(RuBP) concentration between plants grown at the two CO2 concentrations.However, photosynthetic rates averaged 87% greater for the canopiesgrown and measured at 660 mm3 CO2 dm–3. A 10°C differencein air temperature during growth resulted in only a 4°Cleaf temperature difference, which was insufficient to changethe photosynthetic rate or rubisco activity in canopies grownand measured at either 330 or 660 mm3 CO2 dm–3. RuBP concentrationsdecreased as air temperature during growth was increased atboth CO2 concentrations. These data indicate that the increasedphotosynthetic rates of soybean canopies at elevated CO2 aredue to several factors, including: more rapid development ofthe leaf area index; a reduction in substrate CO2 limitation;and no downward acclimation in photosynthetic capacity, as occurin some other species. The rate of carbon transport from an old tomato leaf (54 days),grown at 80 W m–2, was measured under light flux densitiesbetween 7 and 90 W m–2. Under low light, the rate of carbontransport over a 6 h period was about 1 mg C dm–2 h–1,well in excess of the concurrent photosynthetic rate. The lossfrom these leaves of 14C-leaf assimilate which was fixed beforethe experimental period amounted to 62 per cent of the totalinitial uptake and was higher than that from leaves with higherconcurrent photosynthetic rates. The higher loss of 14C fromleaves with low photosynthetic rates was due to a greater contributionof 14C from the starch and residue fractions. The rate of transportappeared to be determined by the concentration of the labilesucrose, not the total sucrose concentration. In comparisonwith young fully-expanded tomato leaves (Ho, 1976) the sizeof the labile sucrose pool appeared to decrease with age. Thephotosynthesistranslocation coefficient was low (k1k2=0•21)for an old tomato leaf. Based on these results a scheme of carbonpartitioning in relation to translocation is proposed. Criteriafor assessing the efficiency of translocation in leaves arediscussed.  相似文献

The stomatal response of seedlings grown in 360 or 720 µmolmol–1 to irradiance and leaf-to-air vapour pressure deficit(VPD) at both 360 and 720 µmol mol–1 to CO2 wasmeasured to determine how environmental factors interact withCO2 enrichment to affect stomatal conductance. Seedlings offour species with different conductances and life histories,Cercis canadensis (L.), Quercus rubra (L.), Populus deltoides(Bartr. ex Marsh.) P. nigra (L.), and Pinus taeda (L.), weremeasured in hopes of identifying general responses. Conductanceof seedlings grown at 360 and 720 µmol mol–1 CO2were similar and responded in the same manner to measurementCO2 concentration, irradiance and VPD. Conductance was lowerfor all species when measured at 720 than when measured at 360µmol mol–1 CO2 at both VPDs ({small tilde}1.5 and{small tilde}2.5 kPa) and all measured irradiances greater thanzero (100, 300, 600,>1600 µmol m–2 S–2)The average decrease in conductance due to measurement in elevatedCO2 concentration was 32% for Cercis, 29% for Quercus, 26% forPopulus, and 11% for Pinus. For alt species, the absolute decreasein conductance due to measurement in CO2 enrichment decreasedas irradiance decreased or VPD increased. The proportional decreasedue to measurement in CO2 enrichment decreased in three of eightcases: from 0.46 to 0.10 in Populus and from 0.18 to 0.07 inPinus as irradiance decreased from>1600 to 100 µmolm–2 s–1 and from 0.35 to 0.24 in Cercis as VPD increasedfrom 1.3 to 2.6 kPa. Diurnal series of fluorescence and photosynthesis assays wereconducted in high altitude (3803 m), tropical (16°), LakeTiticaca (Peru/Bolivia). Near-surface diurnal thermoclines formedon typical days of high photon flux density (PFD, {small tilde}2000 µE m–2 s–1). In the depth range of diurnalstratification profiles of in vivo fluorescence, both without(Fa and with (Fb DCMU, exhibited a mean decrease of 64% frommorning to mid-day, but little change (mean increase of 1.5%)through the afternoon. Three times during the day surface, mid-depth(3–5 m) and deep (15–20 m) phytoplankton sampleswere incubated with H14CO3 under short (<2 h) exposuresto a range of in situ PFDs. Comparison of phytoplankton in differentsamples (ANOVA) showed identical photosynthetic response insunrise (isothermal) samples but a significant drop in surfaceand mid-depth photosynthesis at all PFDs during times of diurnalstratification. Similarly, both low-light () and light-saturated(P2 max photosynthetic parameters were lower in mid-day surfacesamples compared to deep samples. In addition, previously photoinhibitedsamples had a higher threshold intensity for photoinhibition,IT. These results, together with diurnal time series of fluorescencefrom in situ incubations, demonstrate that recovery from extendedepisodes of photoinhibition during diurnal stratification isslower than suggested by previous observations in vitro. Photosynthesisby near-surface phytoplankton is different in light increasingup to IT than light decreasing from IT. It has been hypothesized that nitrogen-replete diatoms, butnot flagellates, may release NO2, NH4+ or dissolved organicnitrogen (DON) following rapid increases in irradiance (andconsequently an increase in cellular electron energy), as mightbe expected to occur in a vertically well mixed estuarine system.Just as the increase in irradiance leads to an increase in cellularenergy, so too would a decrease in temperature, due to the temperaturedependency of biosynthetic enzymes. This hypothesis was testedby comparing the response of nitrogen-replete diatoms (Skeletomenacostatum, Thalassiosira weissflogii and Chaetoceros sp.) andflagellates (Dunaliella tertiolecta, Pavlova lutheri and Prorocentrumminimum) to rapid increases in irradiance and decreases in temperature.Short-term (<3 h) changes in extracellular NO2 andNH4+ concentrations were measured in cultures following theseexperimental shifts, as well as in cultures retained at thegrowth irradiance. Net rates of NO2 and NH4+ releasewere calculated from the time course of extracellular nitrogenconcentrations. As a fraction of NO3 uptake, NO2release rates under the increased irradiance increased marginallyrelative to NO2 release rates under the growth irradiance.Release rates of NH4+ under the increased irradiance increasednearly fivefold over release rates at the growth irradiance,and accounted for 84% of the NO3 uptake rate. In directcontrast to the diatom species, the flagellate species releasedNO2 under the higher experimental irradiance at ratesone half those of the release rates under the growth irradiance,and continued to take up NH4+ under both irradiance conditions.Within the experimental boundaries, these findings have importantphysiological and ecological implications. The magnitude ofthe observed nitrogen release represents a significant physiologicalsink for electrons and, in fact, calculations suggest that upto 62% of the total electrons harvested could be consumed. Froman ecological perspective, these findings add to the body ofliterature which suggests that a significant fraction of thenitrogen that is taken up is ultimately released in dissolvedform. More importantly, these data suggest that DON is not theonly compound that phytoplankton may release in the aquaticenvironment.  相似文献

The activities of three Calvin cycle enzymes, RuBPc (E.C. 4.1.1.39 [EC] ),3PGA phosphokinase (E.C. 2.7.2.3 [EC] ) and NADP-G3P dehydrogenase(E.C. 1.2.1.13 [EC] ), and the cytoplasmic enzyme PEPc (E.C. 4.1.1.31 [EC] )together with soluble protein and chlorophyll were measuredin extracts from young tomato leaves during acclimation to achange in irradiance. Leaf area and fresh weight were also measuredto show changes due to growth during treatments. Soluble proteinhad doubled on a unit leaf area basis 7 d after transfer from100 µmol quanta m–2s–1 PAR (low light) to400 µmol quanta m–2s–1 PAR (high light). Duringthis period the protein/chlorophyll ratio rose from 4•6to 10, RuBPc activity almost doubled and PEPc almost trebled.Following the reverse transfer from high to low light, solubleprotein decreased by 30% after 7 d and the protein/chlorophyllratio fell from 12 to 5•6. There was no change in RuBPcactivity 3 d after transfer from high to low light while PEPcactivity decreased by over 30%. There was no decrease in theactivity of 3PGA phosphokinase or NADP-G3P dehydrogenase 1 dafter transfer to low light, but decreases were apparent after3 d. The extracted kinase and dehydrogenase when fully activatedwere able to phosphorylate and reduce 3PGA at more than 2•5-foldits calculated rate of synthesis in the leaf. The CO2 compensation point at 25 °C and 250 µEinsteinsm–2 s–1 wasmeasured for 27 bryo-phyte species, andwas found to be in the range of 45–160 µl CO2 I–1air. Under the same conditions Zea mays gave a value of 11 µlI–1 and Horde um vulgare 76 µI–1. The rate of loss of photosyntheticallyfixed 14CO2 in the light and dark in six bryophytes (three mosses,two leafy liverworts, one thalloid liverwort) was determinedin CO2-free air and 100% O2. The rate of 14CO2 evolution inthe light was less than that in the dark in CL2-free air, butin 100% O2 the rate in the light increased, so that in all butthe leafy liverworts it was greater than that in the dark. Raisingthe temperature tended to increase the rate of 14CO2 evolutioninto CO2-free air both in the light and dark, so that the light/dark(L/D) ratio did not greatly vary. The lower rate of loss of14CO2 in the light compared tothe dark could be due to partialinhibition of ‘dark respiration’ reactions in thelight, a low rate of glycolate synthesis and oxidation, or partialreassimilation of the 14CO2 produced, or a combination of someor all of these factors.  相似文献
