Charophyte oosporangia and water samples from a highly calcareous lake were measured for stable carbon and oxygen isotopic composition. The time period over which the oosporangia calcify is short, thus any biochemical relationship between the water and oosporangia"s calcite represents only one time window (late Summer in Malham Tam). This important temporal restraint must also apply to interpretations of all fossil material measured. The 18Oc of the charophyte oosporangia is deduced to be in equilibrium with the 18O of the water for a given temperature. The 13 Cc of the charophyte oosporangia was approximately 2.5 per mil lower than the 13CDIC in the water we measured. With the release Of CO2 with phosphoric acid from the charophyte oosporangia, there was no significant difference in the 18Oc values obtained, regardless of whether or not the carbonate was separated from the organic center, however 13Cc values were marginally lower for carbonate plus organic center measurements. Our results indicate that fossil charophyte gyrogonites can be used to elucidate the geochemistry of the ancient water body in which they lived.  相似文献

Despite theories of large-scale movement and assimilation of carbon in estuaries, recent evidence suggests that in some estuaries much more limited exchange occurs. We measured the fine-scale movement and assimilation of carbon by resident macroinvertebrates between adjacent saltmarsh and mangrove habitats in an Australian estuary using 13C analysis of animals at different distances into adjacent patches of habitat. 13C values of crabs (Parasesarma erythrodactyla –15.7 ± 0.1, Australoplax tridentata –14.7 ± 0.1) and slugs (Onchidina australis –16.2 ± 0.3) in saltmarsh closely matched that of the salt couch grass Sporobolus virginicus (–15.5 ± 0.1). In mangroves, 13C values of crabs (P. erythrodactyla –22.0 ± 0.2, A. tridentata –19.2 ± 0.3) and slugs (–19.7 ± 0.3) were enriched relative to those of mangroves (–27.9 ± 0.2) but were more similar to those of microphytobenthos (–23.7 ± 0.3). The 13C values of animals across the saltmarsh-mangrove interface fitted a sigmoidal curve, with a transition zone of rapidly changing values at the saltmarsh-mangrove boundary. The width of this transition indicated that the movement and assimilation of carbon is limited to between 5 and 7 m. The 13C values of crabs and slugs, especially those in saltmarsh habitat, clearly indicate that the movement and assimilation of carbon between adjacent saltmarsh and mangrove habitat is restricted to just a few metres, although some contribution from unmeasured sources elsewhere in the estuary is possible. Such evidence demonstrating the extent of carbon movement and assimilation by animals in estuarine habitats is useful in determining the spatial arrangement of habitats needed in marine protected areas to capture food web processes.  相似文献

We present the results of a 5-year examination of variation in the stable carbon isotope composition () of three C3 graminoid species from a Sandhills prairie: Agropyron smithii, Carex heliophila and Stipa comata. Although consistent species-specific patterns for mean were seen, there was also significant and substantial among-year and within-season variation in . A smaller contribution to variation in came from topographic variation among sampling sites. Effects of species, year, season and topography contribute to variation in in an additive manner. An association between climate and exists that is consistent with previous work suggesting that reflects the interplay between photosynthetic gas exchange and plant water relations. Within the growing season, the time over which integrates plant response to the environment ranges from days to months.  相似文献

Stable carbon (13C) and nitrogen (15N) isotopes were used to elucidate primary food sources and trophic relationships of organisms in Khung Krabaen Bay and adjacent offshore waters. The three separate sampling sites were mangroves, inner bay and offshore. The 13C values of mangrove leaves were –28.2 to –29.4, seagrass –10.5, macroalgae –14.9 to –18.2, plankton –20.0 to –21.8, benthic detritus –15.1 to –26.3, invertebrates –16.5 to –26.0, and fishes –13.4 to –26.3. The 15N values of mangrove leaves were 4.3 to 5.7, seagrass 4.3, macroalgae 2.2 to 4.4, plankton 5.7 to 6.4 , benthic detritus 5.1 to 5.3, invertebrates 7.2 to 12.2 , and fishes 6.3 to 15.9. The primary producers had distinct 13C values. The 13C values of animals collected from mangroves were more negative than those of animals collected far from shore. The primary carbon sources that support food webs clearly depended on location. The contribution of mangroves to food webs was confined only to mangroves, but a mixture of macroalgae and plankton was a major carbon source for organisms in the inner bay area. Offshore organisms clearly derived their carbon through the planktonic food web. The 15N values of consumers were enriched by 3–4 relative to their diets. The 15N data suggests that some of aquatic animals had capacity to change their feeding habits according to places and availability of foods and as a result, individuals of the same species could be assigned to different trophic levels at different places.  相似文献

We measured leaf dry matter 18O and 13C in parasitic plants and their hosts growing in southwestern Australia. Parasite/host pairs included two mistletoe species, three species of holoparasites, and five species of root hemiparasites. Among these parasite functional types, significant variation was observed in parasite/host isotopic differences for both 18O (P<0.0001, n=65) and 13C (P<0.0001, n=64). Mistletoes were depleted in both 18O and 13C compared to their hosts; parasite/host differences were –4.0 for 18O (P<0.0001) and –1.9 for 13C (P<0.0001). The lower 18O in mistletoe leaf dry matter compared to their hosts is consistent with the frequently observed high transpiration rates of these parasites. Root hemiparasites were also depleted in 18O and 13C compared to their hosts, but not to the same extent as mistletoes; parasite/host differences were –1.0 for 18O (P=0.04) and –1.2 for 13C (P=0.0006). In contrast to mistletoes and root hemiparasites, holoparasites were enriched in both 18O and 13C compared to their hosts; parasite/host differences were +3.0 for 18O (P<0.0001) and +1.5 for 13C (P=0.02). The enrichment in 18O for holoparasite dry matter did not result from more enriched tissue water; holoparasite tissue water 18O was less than host leaf water 18O by a difference of –3.8 when sampled at midday (P=0.0003). Enrichment of holoparasites in 13C compared to their hosts is consistent with a generally observed pattern of enrichment in heterotrophic plant tissues. Results provide insights into the ecology of parasitic plants in southwestern Australia; additionally, they provide a context for the formulation of specific hypotheses aimed at elucidating mechanisms underlying isotopic variations among plants.  相似文献

Summary Five subunits (-, -, -, - and -subunits) of the six -and -subunits) in the F1 portion (F1ATPase) of sweet potato (Ipomoea batatas) mitochondrial adenosine triphosphatase were isolated by an electrophoretic method. The - and -subunits were not distinguishable immunologically but showed completely different tryptic peptide maps, indicating that they were different molecular species. In vitro protein synthesis with isolated sweet potato root mitochondria produced only the -subunit when analyzed with anti-sweet potato F1ATPase antibody reacting with all the subunits except the -subunit. Sweet potato root poly(A)+RNA directed the synthesis of six polypeptides which were immunoprecipitated by the antibody: two of them immunologically related to the -subunit and the others to the - and -subunits. We conclude that the -subunit of the F1ATPase is synthesized only in the mitochondria and the -, - and -subunits are in the cytoplasm.  相似文献

Arid environments are characterized by spatial and temporal variation in water and nitrogen availability. differences in 15N and D of four co-occurring species reveal contrasting patterns of plant resource acquisition in response to this variation. Mineralization potential and nitrogen concentration of surface soils associated with plant canopies were greater than inter-canopy locations, and values decreased with increasing depth in both locations. Mineralization potential and nitrogen concentration were both negatively correlated with soil 15N. The spatial variation in soil 15N caused corresponding changes in plant 15N such that plant 15N values were negatively correlated with nitrogen concentration of surface soils. Plants occurring on soils with relatively high nitrogen concentrations had lower 15N, and higher leaf nitrogen concentrations, than plants occurring on soils with relatively low nitrogen concentrations. Two general temporal patterns of water and nitrogen use were apparent. Three species (Juniperus, Pinus andArtemisia) relied on the episodic availability of water and nitrogen at the soil surface. 15N values did not vary through the year, while xylem pressure potentials and stem-water D values fluctuated with changes in soil moisture at the soil surface. In contrast,Chrysothamnus switched to a more stable water and nitrogen source during drought. 15N values ofChrysothamnus increased throughout the year, while xylem pressure potentials and stem-water D values remained constant. The contrasting patterns of resource acquisition have important implications for community stability following disturbance. Disturbance can cause a decrease in nitrogen concentration at the soil surface, and so plants that rely on surface water and nitrogen may be more susceptible than those that switch to more stable water and nitrogen sources at depth during drougnt.  相似文献

We determined the 18O and 13C composition of the same fixed growth increment in severalPorites lutea coral skeletons from Phuket, South Thailand. Skeletal growth rate and 18O are inversely related. We explain this in terms of McConnaughey's kinetic isotopic disequilibria model. Annual trends in 18O cannot be solely explained by observed variations in seawater temperature or salinity and may also reflect seasonal variations in calcification rate. Coral tissue chlorophylla content and 13C of the underlying 1 mm of skeleton are positively related, suggesting that algal modification of the dissolved inorganic carbonate pool is the main control on skeletal 13C. However, in corals that bleached during a period of exceptionally high seawater temperatures in the summer of 1991, 13C of the outer 1 mm of skeleton and skeletal growth rate (over 9 months up to and including the bleaching event) are inversely related. Seasonal variations in °13C may reflect variations in calcification rate, zooxanthellae photosynthesis or in seawater 13C composition. Bleached corals had reduced calcification over the 9-month period up to and including the bleaching event and over the event they deposited carbonate enriched in13C and18O compared with unaffected corals. However, calcification during the event was limited and insufficient material was deposited to influence significantly the isotopic signature of the larger seasonal profile samples. In profile, overall decreases in 18O and 13C were observed, supporting evidence that positive temperature anomalies caused the bleaching event and reflecting the loss of zooxanthellae photosynthesis.  相似文献

Winter and spring precipitation that saturates to deep soil layers precedes summer droughts in the Intermountain West. Occasional summer convection storms relieve summer drought, but are infrequent and unreliable from year to year, leading to the hypothesis that dominant tree species might not invest limited carbon reserves to surface roots to take up summer precipitation in these regions. We compared the hydrogen (D) and oxygen (18O) isotope ratios of winter, spring and summer precipitation to that of xylem sap water in Acer grandidentatum and Quercus gambelii, two dominant trees of this region. By this method we could identify water sources utilized throughout the growing season. Xylem D and 18O values changed significantly when each species leafed-out; this change was not associated with changes in either soil or plant water status (as measured by predawn and midday water potentials). This shift is apparently related increased transpirational flux, which may flush out residual stem water from the previous growing season. D values of xylem sap of both species matched winter precipitation input values throughout most of the summer, indicating a reliance on deep-soil moisture sources throughout the growing season. Mature Q. gambelii did not take up summer precipitation, whereas A. grandidentatum responded slightly to the largest summer rain event. Small trees of both species, particularly A. grandidentatum, showed a limited uptake of summer rains.  相似文献

Summary Oxygen isotope ratios were determined in leaf cellulose from two plant species at Barro Colorado (Republic of Panama) in 4 different plots, two of which were undergoing an irrigation treatment during the dry season. There is a gradient in 18O values of leaf cellulose from the understory to canopy leaves, reflecting the differences in relative humidity between these two levels of the forest. This gradient is most pronounced in irrigated plots. For irrigated plots there was a highly significant correlation between 18O and 13C values, which was not observed in control plots. This relationship can be explained by humidity controlling stomatal conductance. Low humidity affects 18O values of leaf water during photosynthesis, which isotopically labels cellulose during its synthesis. Low humidity also decreases stomatal conductance, which affects discrimination against carbon-13 by photosynthetic reactions, thus affecting the 13C values of photosynthates. WUE values calculated by using plant carbon and oxygen isotope ratios were similar to those observed with gas exchange measurements in other tropical and temperate area. Thus the concurrent analysis of carbon and oxygen isotope ratios of leaf material can potentially be useful for long term estimation of assimilation and evapotranspiration regimes of plants.  相似文献

Partitioning of water resources among plants of a lowland tropical forest   总被引:3,自引:0,他引:3  
Source water used by plants of several species in a semi-evergreen lowland tropical forest on Barro Colorado Island, Panama, was assessed by comparing the relative abundance of deuterium, D, versus hydrogen, H (stable hydrogen isotope composition, D) in xylem sap and in soil water at different depths, during the dry season of 1992. Ecological correlates of source water were examined by comparing xylem water D values with leaf phenology, leaf water status determined with a pressure chamber, and rates of water use determined as mass flow of sap using the stem heat balance method. Soil water D values decreased sharply to 30 cm, then remained relatively constant with increasing depth. Average D values were-13, for 0–30 cm depth and-36.7 for 30–100 cm depth. Soil water D values were negatively associated with soil water content and soil water potential. Concurrent analyses of xylem water revealed a high degree of partitioning of water resources among species of this tropical forest. Xylem water D of deciduous trees (average=-25.3±1.4) was higher than that of evergreen trees (average=-36.3±3.5), indicating that evergreen species had access to the more abundant soil water at greater depth than deciduous species. In evergreen shade-tolerant and high-light requiring shrubs and small trees, D of xylem water was negatively correlated with transpiration rate and leaf water potential indicating that species using deeper, more abundant water resources had both higher rates of water use and more favorable leaf water status.  相似文献

The T-cell receptor (TCR) and loci are particularly interesting because of their unique genomic structure, in that the gene segments for each locus are interspersed. The origin of this remarkable gene segment arrangement is obscure. In this report, we investigated the evolution of the TCR and variable loci and their respective recombination signal sequences (RSSs). Our phylogenetic analyses divided the and variable gene segments into two major groups each with distinguishing motifs in both the framework and complementarity determining regions (CDRs). Sequence analyses revealed that TCR variable segments share similar CDR2 sequences with immunoglobulin light chain variable segments, possibly revealing similar evolutionary histories. Maximum likelihood analysis of the region on Chromosome 14q11.2 containing the loci revealed two possible ancestral TCR / variable segments, TRDV2 and TRAV1-1/1-2, respectively. Maximum parsimony revealed different evolutionary patterns between the variable segment and RSS of the same variable gene arguing for dissimilar evolutionary origins. Two models could account for this difference: a V(D)J recombination activity involving embedded heptamer-like motifs in the germline genome, or, more plausibly, an unequal sister chromatid crossing-over. Either mechanism would have resulted in increased diversity for the adaptive immune system.  相似文献

The diets and trophic interactions among Weddell, crabeater, Ross, and leopard seals in the eastern Ross Sea, Antarctica, were investigated by the use of stable isotope techniques during the 1999–2000 summer seasons. The 13C and 15N values in seal serum clearly distinguished the three Antarctic pack-ice seal species at different trophic positions (Weddell>Ross>crabeater). These patterns appeared to reflect a close linkage to their known foraging ecology and diving behaviors, and agreed well with their presumed dietary diversity. The more enriched 13C and 15N values in male Weddell seals than those in females suggested differences in foraging preferences between them. Significant differences in 15N were also found among different age groups of Weddell seals. A strong correlation between the C:N ratios and serum cholesterol was probably due to extremely high cholesterol levels in phocids. Comparisons of isotope data with harbor seals revealed distinct differences between Antarctic phocids and the northern seal species.  相似文献

Summary The ratio of deuterium to hydrogen (expressed as D) in hydrogen released as water during the combustion of dried plant material was examined. The D value (metabolic hydrogen) determined on plant materials grown under controlled conditions is correlated with pathways of photosynthetic carbon metabolism. C3 plants show mean D values of-132 for shoots and -117 for roots; C4 plants show mean D values of -91 for shoots and-77 for roots and CAM plants a D value of-75 for roots and shoots. The difference between the D value of shoot material from C3 and C4 plants was confirmed in species growing under a range of glasshouse conditions. This difference in D value between C3 and C4 species does not appear to be due to differences in the D value (tissue water) in the plants as a result of physical fractionation of hydrogen isotopes during transpiration. In C3 and C4 plants the hydrogen isotope discrimination is in the same direction as the carbon isotope discrimination and factors contributing to the difference in D values are discussed. In CAM plants grown in the laboratory or collected from the field D values range from-75 to +50 and are correlated with 13C values. When deprived of water, the D value (metabolic hydrogen) in both soluble and insoluble material in leaves of Kalanchoe daigremontiana Hamet et Perr., becomes less negative. These changes may reflect the deuterium enrichment of tissue water during transpiration, or in field conditions, may reflect the different D value of available water in areas of increasing aridity. Whatever the origin of the variable D value in CAM plants, this parameter may be a useful index of the water relations of these plants under natural conditions.  相似文献

The overlapping fragments of the chromosomal DNA from black widow spider Latrodectus mactans carrying genes for high-molecular-mass protein neurotoxins, - and -latroinsectotoxins (-LIT and -LIT) and -latrotoxin (-LTX), were PCR-amplified and cloned. Restriction analysis of the PCR products showed that the distribution and sizes of the restriction fragments coincided with those deduced from the earlier sequencing of cDNAs of the corresponding genes. It thus followed that the -LIT and -LIT genes are intronless. Along with our data on the structure of the -latrocrustotoxin (-LCT), this implies that the lack of introns is a common feature of the black widow spider genes encoding high molecular mass neurotoxins.  相似文献

During a multipurpose survey we examined electrophoretic mobilities of major (A, i.e., 22) and minor (A2, i.e., 22) adult hemoglobins from populations of nine primate genera representing a total of 440 New World monkeys and apes. Sequences of hemoglobin chains were inferred from differences in amino acid composition between homologous tryptic peptides supplemented by detailed placement of more than 270 residues. Beta sequences were thus analyzed in five genera (Aotus, Ateles, Hylobates, Saimiri, and Saguinus) and sequences in seven (foregoing plus Gorilla and Pan). In most genera, sequences from several individuals, often from several species, were delineated. Fifteen kinds of intraspecies mutants were detected; 10 of these were precisely characterized. Five of the 15 mutants form electrophoretically detected genetic polymorphisms of ; none such occur in . Six electrophoretically detected mutants, four in and two in , are uncommon. One of these represents the complete absence of minor component. Three kinds of variants, two in and one in , are electrophoretically neutral and chance findings during sequence analysis of the equivalent of 38 allele products. Two of the neutral variants are not especially common; one may have polymorphic frequency. Several general conclusions stem from these and supplementary findings. First, comparisons of sequences suggest that and genes in all primates either arose from a single event in a common ancestor or from two approximately coincident events. Either assumption allows reconstruction of a reasonably accurate archetype sequence that is effectively common to all descendants. Second, there is a pancellular quantitative disproportion between major and minor hemoglobins ranging from 16:1 to 220:1 in species studied. Delta is consequently presumed to be functionally and adaptively less vital than . When these premises are adopted, is expected to be relatively invisible to natural selection, and, where darwinism is the principal arbiter of evolution and polymorphism, is expected to show fewer fixed changes and fewer genetic polymorphisms than . The opposite is observed. Delta exhibits as many or more changes from archetype than . This finding and the comparative abundance of polymorphism are attributed to nonadaptive factors which are thus considered the source of much evolutionary change. Third, particular sequence positions in various species are the site of recurrent mutations in both and . One such area is occupied by the majority of genetic polymorphisms found in man and other primates. The overall distribution of mutations arising in evolution is remarkably nonrandom in , , and a pool of both. These results are quite unlike most other observations in higher organisms. The sources of such nonrandomness are either selection and/or differential mutability. We rely on our prior assumption of relative selective invisibility for and, in part, ascribe the nonrandom distribution of changes to microzones of enhanced mutability. Fourth, the six uncommon electrophoretically detected mutants provide an estimate of heterozygosity (1/73) at hemoglobin loci that is tenfold greater than observed in man. Fifth, the unprecedented chance detection of three kinds of electrophoretically neutral intraspecies mutants among the equivalent of 38 characterized allele products suggests that neutral changes are as common as electrostatically active ones and at least tenfold more common than expected in extrapolation from human variant surveys. Sixth, analyses from three kinds of gibbon (Hylobates) hemoglobin suggest that one of these is a potentially unchanged relict of the ancient archetype and, further, indicate a degree of homozygous diversity within a species that nearly equals the difference between gibbon and man.This investigation received support from grants to S.H.B., HD-02508-04 and K3-GM-6308-03, from the National Institutes of Health.  相似文献

Here we report on the use of iodination of the membrane-bound nicotinic acetylcholine receptor (nAChR) from Torpedo californica electric tissue in order to define surface-exposed portions of the receptor molecule. Membrane-bound nAChR was 125I-iodinated using the oxidation agent Iodo-Gen. The iodinated subunits were separated by preparative gel electrophoresis, desalted, and cleaved with trypsin. The resulting peptides were separated by reverse-phase HPLC and the radioactive peptides were identified by mass spectrometry and protein sequencing. For the -subunit, we identified five iodinated peptides containing the tyrosine residues Tyr17, Tyr74, Tyr365, Tyr372, and Tyr428. The surface exposition of these amino acids is in agreement with the four-transmembrane-segment model (4TM model) of the nAChR, but the assignment to the intra- or extracellular surface is doubtful. According to this model, the N-terminal portion of the receptor subunits including the iodinated residues Tyr17 and Tyr74 is extracellular and Tyr372 as a site of tyrosine phosphorylation is located on the cytoplasmic side. But since this latter residue is among the first to be iodinated using an immobilized iodination agent, its true position with respect to the membrane bilayer is not clear.  相似文献

The effect of interspecific competition and element additions (N and P) on four grassland species (Poa pratensis, Lolium perenne, Festuca valida, Taraxacum officinale) grown under field conditions was studied. Two grasses (L. perenne, F. valida) grown in monoculture (absence of competition) showed lower carbon isotope discrimination (13C) and enriched 15N values. Nitrogen addition (as urea) had inconsistent effects on species 13C while caused enrichment of 15N of P. pratensis and F. valida but strong depletion of 15N of T. officinale. Phosphorous had no significant effect on 13C but depleted 15N of all species.  相似文献

The 15N composition of the dominant form of dissolved inorganic nitrogen (DIN) was determined in upland groundwater, riparian groundwater, and stream water of the Barro Branco catchment, Amazônas, Brazil. The 15N composition of organic nitrogen in riparian and upland leaf litter was also determined. The data for these waters could be divided into three groups: upland groundwater DIN predominately composed of NO3 with 15N values averaging 6.25 ± 0.9 riparian groundwater DIN primarily composed of NH4 + with 15N values averaging 9.17 ± 1.0 and stream water DIN predominately composed of NO3 with 15N values averaging 4.52 ± 0.8 Nitrate samples taken from the stream source and from the stream adjacent to the groundwater transects showed a downstream increase in 15N from 1.0to 4.5 Leaf litter samples averaged 3.5 ± 1.2The observed patterns in isotopic composition, together with previously observed inorganic nitrogen species and concentration shifts between upland, riparian and stream waters, suggest that groundwater DIN is not the primary source of DIN to the stream. Instead, the isotopic data suggest that remineralization of organic nitrogen within the stream itself may be a major source of stream DIN, and that the majority of DIN entering the stream via groundwater flowpaths is removed at the riparian-stream interface.  相似文献

Water relations were analysed in a semi-deciduous forest grove occurring in the oxisols of the Orinoco savannas. This grove has a shallow unconsolidated ironstone cuirass, which is overlaid by a sandy loam layer (0.0–0.5 m) that contains more than 90% of the grove forest root phytomass. Evapotranspiration and through drainage were calculated by using data from the soil profile as related to physical characteristics of the site root zone, hydraulic conductivity, volumetric water content and potential hydraulic gradient. Mean annual evapotranspiration was 783 mm year–1 and annual through drainage below the root zone was 14% (162 mm year–1) of the gross rainfall. This drainage recharged the 42% of the annual saturation deficit of the water table. Similar mean annual evapotranspiration (770 mm year–1) was also calculated by using the water balance components. The mean daily coupling omega factor () between the grove canopy and the surrounding atmosphere indicated that a high degree of coupling (=0.14±0.16) occurs in the grove and evapotranspiration was mainly controlled by surface conductance. As the dry season proceeded, the soil saturation deficit () increased rapidly resulting in a threshold surface conductance (0.030–0.005 m s–1) for ranging from 0.05 to 0.10. Hypotheses to explain the omnipresence of perennial species in the wide range of physical conditions in neotropical savannas are discussed.  相似文献
