Two commercialized essential oils and their constituent compounds were investigated for fumigant and contact activities against two grain storage insects, adults of the maize weevil (Sitophilus zeamais) and the red flour beetle (Tribolium castaneum). The two commercialized basil and orange oils showed strong fumigant and contact activities against S. zeamais and T. castaneum. The constituents of the basil oil were linalool (21.83%), estragole (74.29%), and α-humulene (2.17%), and those of the orange oil were α-pinene (0.54%), sabinene (0.38%), β-myrcene (1.98%), limonene (96.5%), and linalool (0.6%). As a toxic fumigant, the basil oil was more effective (24-h LC50 = 0.014 and 0.020 mg cm 3) than the orange oil (24-h LC50 = 0.106 and 0.130 mg cm 3) against S. zeamais and T. castaneum adults, respectively. Among the constituents of the two essential oils, the toxicity of estragole was the highest (0.004 and 0.013), followed by linalool (0.016 and 0.023), limonene (0.122 and 0.171), α-pinene (0.264 and 0.273), and β-myrcene (0.274 and 0.275) based on 24-h LC50 values (mg cm 3). Similar results were obtained in a contact toxicity test. The contact activity of basil oil was more toxic than orange oil, and estragole and linalool showed pronounced contact toxicity against S. zeamais and T. castaneum adults. Alpha-humulene had no activity as a fumigant at the tested doses, but it did have an effect as a contact poison, having 24-h LD50 values of 0.040 and 0.045 mg adult 1 to S. zeamais and T. castaneum, respectively. Although basil oil, orange oil, and their components displayed both contact and fumigant toxicities, their effects were mainly exerted by fumigant action via the vapor phase. Thus, basil oil, orange oil, and their components could be potential candidates as new fumigants for the control of S. zeamais and T. castaneum adults.  相似文献

During our screening program for agrochemicals from Chinese medicinal herbs and wild plants, the essential oil of Litsea cubeba fruits was found to possess strong contact toxicity against the cigarette beetle Lasioderma serricorne adults and the booklouse Liposcelis bostrychophila, with LD50 values of 27.33 μg/adult and 71.56 μg/cm2, respectively, and also showed strong fumigant toxicity against the two stored product insects with LC50 values of 22.97 and 0.73 mg/L, respectively. The essential oil obtained by hydrodistillation was investigated by GC MS. The main components of the essential oil were identified to be E-citral (geranial) (27.49%), Z-citral (neral) (23.57%) and d-limonene (18.82%) followed by β-thujene (3.34%), β-pinene (2.85%), α-pinene (2.57%), 6-methyl-5-hepten-2-one (2.40%) and linalool (2.36%). Citral (Z/E-citral), d-limonene, β-pinene, α-pinene and linalool were separated and purified by silica gel column chromatography and preparative thin layer chromatography, and further identified by means of physicochemical and spectrometric analysis. Citral and linalool showed strong contact toxicity against L. serricorne and L. bostrychophila (LD50 = 11.76, 12.74 μg/adult and 20.15, 99.97 μg/cm2, respectively) and fumigant toxicity against L. serricorne and L. bostrychophila (16.54, 18.04 mg/L air and 0.14, 0.71 mg/L air, respectively). Otherwise, citral, d-limonene and linalool were strongly repellent against the cigarette beetle L. serricorne as the essential oil whereas β-pinene and α-pinene exhibited weaker repellency against the cigarette beetle compared with the positive control, DEET. Moreover, except α-pinene and linalool, the other three compounds as well as the essential oil exhibited comparable repellency against the booklouse relative to DEET.  相似文献

Water-distilled essential oil from the dried bulbs of Allium chinense (Liliaceae) was analyzed by gas chromatography–mass spectrometry (GC–MS). Eighteen compounds, accounting for 98.4% of the total oil, were identified and the main components of the essential oil of A. chinense were methyl allyl trisulfide (30.7%), dimethyl trisulfide (24.1%), methyl propyl disulfide (12.8%) and dimethyl disulfide (9.6%) followed by methyl allyl disulfide (3.4%) and methyl propyl trisulfide (3.6%). The essential oil exhibited contact toxicity against the booklice (Liposcelis bostrychophila) with an LC50 value of 441.8 μg/cm2 while the two major constituents, dimethyl trisulfide and methyl propyl disulfide had LC50 values of 153.0 μg/cm2 and 738.0 μg/cm2 against the booklice, respectively. The essential oil of A. chinense possessed strong fumigant toxicity against the booklice with an LC50 value of 186.5 μg/l while methyl allyl trisulfide (LC50 = 90.4 μg/l) and dimethyl trisulfide (LC50 = 114.2 μg/l) exhibited stronger fumigant toxicity than methyl propyl disulfide (LC50 = 243.4 μg/l) and dimethyl disulfide (LC50 = 340.8 μg/l) against the booklice. The results indicated that the essential oil and its major constituents have potential for development into natural insecticides or fumigants for control of insects in stored grains.  相似文献

This study was performed to investigate the repellent effect of 5 μl doses of ten essential oils (bergamot, chamomile, clary sage, fennel, lavender, lemongrass, majoram, peanut, pennyroyal, and peppermint) against Lycorma delicatula 4th nymphs using an olfactometer. Only lavender oil exhibited significant repellency. We then tested 10, 5, 2.5, and 1 μl doses of lavender oil against the nymphs and females of L. delicatula. The oil showed significant repellency at 10 and 5 μl, although the latter is less potent to 1st instar nymphs. At the lavender oil dose of 2.5 μl, only 3rd and 4th instar nymphs and females were significantly affected. None of the stages tested were affected by 1 μl. Chromatographic and mass spectrometric analyses of lavender oil detected linalool (42.2%), linalyl acetate (49.4%), terpinen-4-ol (5.0%), and caryophyllene oxide (3.4%). Among the four main components, only linalool showed repellency to all instar nymphs and females. No synergism was detected. Antennae of all instar nymphs and females showed electrophysiological responses only to linalool. In field studies using linalool, 4th nymphs and adults were highly repelled at a dose of 30 μl of lavender oil. The effect differed according to test plot and treatment dose.  相似文献

In this work, essential oils extracted from roots and aerial parts of Inula graveolens by hydrodistillation and their fractions obtained by chromatographic simplification were first investigated for their chemical composition by GC/MS and then evaluated for the first time for their repellency and contact toxicity properties against Tribolium castaneumadults. Twenty-eight compounds were identified in roots essential oil (REO), which accounted for 97.9 % of the total oil composition, with modhephen-8-β-ol (24.7 %), cis-arteannuic alcohol (14.8 %), neryl isovalerate (10.6 %) and thymol isobutyrate (8.5 %) as major constituents. Twenty-two compounds were found in the essential oil from aerial parts (APEO), which accounted for 93.9 % of the total oil, with borneol (28.8 %), caryophylla-4(14),8(15)-dien-6-ol (11.5 %), caryophyllene oxide (10.9 %), τ-cadinol (10.5 %) and bornyl acetate (9.4 %) as main compounds.REO and APEO displayed stronger repellency after 2 h of exposure (80.0 and 90.0 %, respectively) against T. castaneum at the concentration of 0.12 μL/cm2. After fractionation, fractions R4 and R5 exhibited greater effects (83.3 % and 93.3 %, respectively) than the roots essential oil. Furthermore, the fractions AP2 and AP3 showed higher repellency (93.3 and 96.6 %, respectively) than the aerial parts oil. The LD50 values of oils from roots and aerial parts topically applied were 7.44 % and 4.88 %, respectively. Results from contact toxicity assay showed that fraction R4 was more effective than the roots oil with LD50 value of 6.65 %. These results suggests that essential oils of roots and aerial parts from I. graveolens may be explored as potential natural repellent and contact insecticides against T. castaneum in stored products.  相似文献

Twelve monoterpenes were evaluated for larvicidal and adulticidal activities towards Culex pipiens. Geraniol and cuminaldehyde were the most toxic monoterpenes to larvae, with LC50 values of 38.6 and 38.9 mg/l after 24 h of treatment, respectively, whereas cuminaldehyde was the most potent compound after 48 h of treatment, followed by geraniol and thymol. In fumigant toxicity experiments, (R)-carvone and geraniol were the most toxic monoterpenes against the adults at all three tested concentrations and after both 24 and 48 h. When tested at sublethal concentrations (0.5 LC50), (R)-carvone, (S)-limonene and cuminaldehyde decreased hatchability, pupation and adult emergence and induced high larval mortality. Our results suggest that geraniol, cuminaldehyde and (R)-carvone are promising toxicants against Culex pipiens and could be useful in the search for new natural insecticides.  相似文献

Mosquitoes are major arthropod vectors responsible for several pathogenic diseases. In recent years, repellents of botanical origin, particularly essential oils, have been used against mosquitoes and have been found effective and safe. In this study, five different repellent cream formulations (CF1–5) were prepared using combinations of essential oils, including camphor, cinnamon, citronella, lemongrass, lime, orange, neem, basil, Vitex, Lantana, eucalyptus, and clove, and their repellency was tested using Culex quinquefasciatus Say and Aedes aegypti L. under laboratory conditions and compared to the standard synthetic repellent N,N-diethyl-meta-toluamide (DEET-12%, w/w). Among the five cream formulations, CF2 at a dose of 5 mg/cm2 showed the longest protection time of 4.18 h and 3.31 h against C. quinquefasciatus and A. aegypti, respectively, under laboratory conditions. CF3 at a dose of 5 mg/cm2 was moderately effective, with protection times of 3.42 h and 2.58 h against C. quinquefasciatus and A. aegypti, respectively, under laboratory conditions. CF2 at a dose of 5 mg/cm2 was also tested in the field against wild mosquitoes for 3 h, and 100% protection was observed for the entire study period. Thus, CF2 could be used in developing an effective natural repellent as an alternative to the existing synthetic repellents to C. quinquefasciatus and A. aegypti.  相似文献

The natural compounds carvacrol and thymol completely prevented seizures in the 6 Hz, 32 mA partial seizure model. Carvacrol and thymol, both exhibited an ED50 = 35.8 mg/kg, ip and yielded protective indices of 5.3 and 3.4, respectively. At 44 mA current intensity, carvacrol and thymol exhibited ED50s of 88.82 mg/kg (PI = 2.15) and 73.0 mg/kg (PI = 1.65), respectively. Thymol, but not carvacrol showed partial inhibitory activity in the maximal electroshock (MES), sc Metrazol (scMET) and Corneal-kindled models. These results suggest that carvacrol and thymol are more efficacious anticonvulsants than suggested by their lower efficacies in the conventional MES and scMET tests.  相似文献

The efficacy of an essential oil obtained from Pimpinella anisum fruits and its major compound, trans-Anethole, was tested on the eggs, larvae and adults of Culex quinquefasciatus. While causing no significant mortality on eggs, other tested stages were very sensitive to the essential oil and trans-Anethole. LC50 for the 2nd to 4th instar larvae was estimated as 26–27 μL·L 1 and 15–19 μL·L 1 for the essential oil and trans-Anethole, respectively. As for the essential oil applied on adults, LC(LD)50 was estimated as 9.3 μL mL 1 (spray test), 1.9 μL L 1 (fumigation test) and 0.6 μg cm 2 (tarsal test), and for trans-Anethole as 8.1 μL mL 1 (spray test), 2.1 μL L 1 (fumigation test) and 0.4 μg cm 2 (tarsal test). The time needed to achieve 50% mortality after application of LC(LD)99 of the essential oil was significantly different; for example, in larvicidal assays it ranged from 15 to 235 min depending on the larval instar, and from 9 to 180 min when applied to adults, depending on the mode of application. It was also found that temperature had an important effect on the larvicidal efficacy of the essential oil, and oviposition deterrent activity was studied.The essential oil and trans-Anethole were toxic for Daphnia magna (62–92% mortality) and significantly reduced its fertility at high concentrations (35–50 μL mL 1) and long exposure (48 h). However, no negative effect on Daphnia mortality or fertility was found at shorter exposure times (6 h) and/or lower concentrations (20 μL mL 1).Based on the results of this study, we can recommend the essential oil from P. anisum as a suitable active substance for potential botanical insecticides.  相似文献

The isolation and culturing of toxic Gambierdiscus species is desirable for obtaining reliable sources of toxins responsible for Ciguatera Fish Poisoning (CFP) in order to provide reference materials and purified standards for chemical analyses and bioassays. Gambierdiscus polynesiensis CAWD212, G. pacificus CAWD213 and G. australes CAWD149 and 216 have been isolated from samples of sea water from the Cook Islands. G. polynesiensis produced ciguatoxins (CTX; 18.2 pg per cell) and G. australes CAWD149 produced maitotoxin (MTX; 8.3 pg per cell) and in both extracts putative MTX-3 was detected. G. pacificus also produced putative MTX-3. The toxicities of extracts of G. polynesiensis and G. pacificus were similar by i.p. injection (LD50s of 1.0 mg/kg and 0.8 mg/kg respectively), but when administered by gavage, the MTX-3 analogue producing G. pacificus (LD50 251 mg/kg) was 30-times less toxic than the CTX-producing G. polynesiensis (LD50 7.9 mg/kg). Consumption of fish contaminated by CFP has caused illness and even death in the Cook Islands and throughout the Pacific region and is now considered a globally neglected disease. Large-scale cultures of these organisms are now available, and isolation of the toxins they produce is in progress.  相似文献

Powders and essential oils were obtained from Achillea biebersteinii, A. fragrantissima and Ageratum conyzoides and tested for their insecticidal activity against Sitophilus oryzae, Rhyzopertha dominica and Tribolium castaneum. Composition of the oils was identified by gas chromatography (GC) and GC/mass spectrometry (MS). The tested plants showed adulticidal activity, where toxicity varied with dosage of the plant product, the insect tested and the exposure period. When mixed with grains as ground powders, a dosage of 20 g/kg grains killed 88.2, 73.6 and 64.4% of S. oryzae after 12 days of exposure with 79.6, 63.3 and 48% reduction in progeny for powders of A. biebersteinii, A. conyzoides and A. fragrantissima, respectively. Under the same assay conditions, A. biebersteinii and A. conyzoides powders killed 100% of R. dominica. T. castaneum was less susceptible to the plant powders. The LC50 values for powders were 22.8, 31.6 and 39.7 mg/g grains for A. biebersteinii, 27.1, 35.4 and 47.8 mg/g grains for A. conyzoides and 36.9, 48.3 and 78.6 mg/g grains for A. fragrantissima against R. dominica, S. oryzae and T. castaneum, respectively. Upon fumigation, a dose of 60 μl/L air of A. biebersteinii and A. conyzoides oils was sufficient to kill 100% of R. dominica after 12 days of adult exposure, while 91.3% adult mortality was recorded in the case of A. fragrantissima oil. S. oryzae and T. castaneum showed a great susceptibility to the plant oils. Powders exhibited a moderate to strong residual activity where A. conyzoides showed the greatest grain protecting activity.  相似文献

Optimization of intravascular shear stress assessment in vivo   总被引:1,自引:0,他引:1  
The advent of microelectromechanical systems (MEMS) sensors has enabled real-time wall shear stress (WSS) measurements with high spatial and temporal resolution in a 3-D bifurcation model. To optimize intravascular shear stress assessment, we evaluated the feasibility of catheter/coaxial wire-based MEMS sensors in the abdominal aorta of the New Zealand white (NZW) rabbits. Theoretical and computational fluid dynamics (CFD) analyses were performed. Fluoroscope and angiogram provided the geometry of aorta, and the Doppler ultrasound system provided the pulsatile flow velocity for the boundary conditions. The physical parameters governing the shear stress assessment in NZW rabbits included (1) the position and distance from which the MEMS sensors were mounted to the terminal end of coaxial wire or the entrance length, (Le), (2) diameter ratios of aorta to the coaxial wire (Daorta /Dcoaxial wire=1.5–9.5), and (3) the range of Reynolds numbers (116–1550). At an aortic diameter of 2.4 mm and a maximum Reynolds number of 212 (a mean Reynolds number of 64.2), the time-averaged shear stress (τave) was computed to be 10.06 dyn cm?2 with a systolic peak at 33.18 dyn cm?2. In the presence of a coaxial wire (Daorta /Dcoaxial wire=6 and Le=1.18 cm), the τave value increased to 15.54 dyn cm?2 with a systolic peak at 51.25 dyn cm?2. Real-time intravascular shear stress assessment by the MEMS sensor revealed an τave value of 11.92 dyn cm?2 with a systolic peak at 47.04 dyn cm?2. The difference between CFD and experimental τave was 18.5%. These findings provided important insights into packaging the MEMS sensors to optimize in vivo shear stress assessment.  相似文献

The purpose of the work was to provide a crystallographic demonstration of the venerable idea that CO photolyzed from ferrous heme-a3 moves to the nearby cuprous ion in the cytochrome c oxidases. Crystal structures of CO-bound cytochrome ba3-oxidase from Thermus thermophilus, determined at ~ 2.8–3.2 Å resolution, reveal a Fe–C distance of ~ 2.0 Å, a Cu–O distance of 2.4 Å and a Fe–C–O angle of ~ 126°. Upon photodissociation at 100 K, X-ray structures indicate loss of Fea3–CO and appearance of CuB–CO having a Cu–C distance of ~ 1.9 Å and an O–Fe distance of ~ 2.3 Å. Absolute FTIR spectra recorded from single crystals of reduced ba3–CO that had not been exposed to X-ray radiation, showed several peaks around 1975 cm? 1; after photolysis at 100 K, the absolute FTIR spectra also showed a significant peak at 2050 cm? 1. Analysis of the ‘light’ minus ‘dark’ difference spectra showed four very sharp CO stretching bands at 1970 cm? 1, 1977 cm? 1, 1981 cm? 1, and 1985 cm? 1, previously assigned to the Fea3–CO complex, and a significantly broader CO stretching band centered at ~ 2050 cm? 1, previously assigned to the CO stretching frequency of CuB bound CO. As expected for light propagating along the tetragonal axis of the P43212 space group, the single crystal spectra exhibit negligible dichroism. Absolute FTIR spectrometry of a CO-laden ba3 crystal, exposed to an amount of X-ray radiation required to obtain structural data sets before FTIR characterization, showed a significant signal due to photogenerated CO2 at 2337 cm? 1 and one from traces of CO at 2133 cm? 1; while bands associated with CO bound to either Fea3 or to CuB in “light” minus “dark” FTIR difference spectra shifted and broadened in response to X-ray exposure. In spite of considerable radiation damage to the crystals, both X-ray analysis at 2.8 and 3.2 Å and FTIR spectra support the long-held position that photolysis of Fea3–CO in cytochrome c oxidases leads to significant trapping of the CO on the CuB atom; Fea3 and CuB ligation, at the resolutions reported here, are otherwise unaltered. Two tetracyanometalate building blocks, [Fe(5,5′-dmbipy)(CN)4]? (2) and [Fe(4,4′-dmbipy)(CN)4]? (3) (5,5′-dmbipy = 5,5′-dimethyl-2,2′-bipyridine; 4,4′-dmbipy = 4,4′-dimethyl-2,2′-bipyridine), and two cyano-bridged heterobimetallic complexes, [Cu2(bpca)2(H2O)2Fe2(5,5′-dmbipy)2(CN)8] · 2[Cu(bpca)Fe(5,5′-dmbipy)(CN)4] · 4H2O (4) and [Cu(bpca)Fe(4,4′-dmbipy)(CN)4]n (5) (bpca = bis(2-pyridylcarbonyl)amidate), have been synthesized and structurally characterized. Complex 4 contains two dinuclear and one tetranuclear heterobimetallic clusters in an asymmetric unit whereas the structure of complex 5 features a one-dimensional heterobimetallic zigzag chain. The Cu(II) ion is penta-coordinated in the form of a distorted square-based pyramid. Magnetic studies show ferromagnetic coupling between Cu(II) and Fe(III) ions with g = 2.28, J1 = 2.64 cm?1, J2 = 5.40 cm?1 and TIP = ?2.36 × 10?3 for complex 4, and g = 2.17, J = 4.82 cm?1 and zJ = 0.029 cm?1 for complex 5.  相似文献

Exposures to particulate matter with a diameter of 2.5 μm or less (PM2.5) may influence the risk of birth defects and make you allergic, which causes serious harm to human health. Bamboo charcoal can adsorb harmful substances,that was of benefitto people’s health. In order to figure out the optimal adsorbtion condition and the intrinsic change of bamboo charcoal, five chemicals were adsorbed by bamboo charcoal and were analyzed by FT-IR. The optimal blast time was 80 min of Na2SO3, 100 min of Na2S2O8, 20 min of Na2SO4, 120 min of Fe2(SO4)3 and 60 min or 100 min of S. FT-IR spectra showed that bamboo charcoal had five characteristic peaks of SS stretch, H2O stretch, OH stretch, CO stretch or CC stretch, and NO2 stretch at 3850 cm−1, 3740 cm−1, 3430 cm−1, 1630 cm−1 and 1530 cm−1, respectively. For Na2SO3, the peaks at 3850 cm−1, 3740 cm−1, 3430 cm−1, 1630 cm−1 and 1530 cm−1 achieved the maximum at 20 min. For Na2S2O8, the peaks at 3850 cm−1, 3740 cm−1, 3430 cm−1 and 1530 cm−1 achieved the maximum at 40 min. For Na2SO4, the peaks at 3850 cm−1, 3740 cm−1 and 1530 cm−1 achieved the maximum at 40 min. For Fe2(SO4)3, the peaks at 3850 cm−1, 3740 cm−1, 1630 cm−1 and 1530 cm−1 achieved the maximum at 120 min. For S, the peaks at 3850 cm−1 and 3740 cm−1 achieved the maximum at 40 min, the peaks at 1630 cm−1 and 1530 cm−1 achieved the maximum at 40 min. It proved that bamboo charcoal could remove sulfur powder from air to restrain sulfur allergies.  相似文献

The current study has concentrated on assessment of the radioprotective potential of REC-2001, a semi-purified fraction of rhizomes of Podophyllum hexandrum, in Swiss albino Strain ‘A’ mice exposed to 10 Gy whole-body gamma radiation. Animals were treated with 10 and 15 mg/kg b wt (i.p.) of REC-2001 1 h prior to exposure to a lethal dose of γ-radiation (10 Gy) and observed upto 30 days. For analysis of maximum tolerable dose (MTD), LD50 and acute toxic dose, different concentrations of the extract were administered to animals and their mortality and morbidity status was observed upto 72 h and one week, respectively. Dose reduction factor (DRF) was determined by exposing REC-2001 pre-treated mice to supra-lethal doses of γ-radiation. Endogenous spleen colony forming units (CFU), DNA strand breaks in thymocytes (alkaline halo assay) and lipid degradation was studied to understand the mechanism of radioprotection . A single dose of REC-2001 (10 and 15 mg/kg b wt i.p.) exhibited >90% survival in the pre-treated irradiated group versus no survival in radiation control group. Single doses of upto 75 mg/kg b wt (i.p.) did not cause any mortality (MTD) in mice. REC-2001, a dose of 90 mg/kg b wt, resulted in 50% mortality (LD50), while the LD100 was 115 mg/kg b wt REC-2001 exhibited a DRF of 1.62. CFU counts in the REC-2001 treated group were found significantly high (5.33/spleen) as compared to controls. Exposure of thymocytes to 10 Gy radiation resulted in increased halo diameter (45±3 μm) in comparison to untreated controls (8±1 μm). REC-2001 administration (500 μg/ml) decreased the halo diameter to 15±2 μm. Radiation-induced lipid degradation was also inhibited by REC-2001. The present study has revealed that REC-2001 is a promising radioprotective fraction that can be effectively used against lethal doses of γ-radiation after further investigations in higher animal models.  相似文献

Pike-perch Sander lucioperca is currently considered as one of the most promising candidates for production in freshwater recirculation aquaculture systems (RAS). Here, due to the lack of studies on nitrite (NO2?) toxicity in pike-perch, a flow-through exposure at 0, 0.44, 0.88, 1.75, 3.5, 7, 14 and 28 mg/L NO2?–N was carried out to determine the acute and chronic toxicity over a period of 32 days. In juvenile pike-perch, 120 h LC50 was 6.1 mg/L NO2?–N and at ≥ 14 mg/L NO2?–N all fish had died within 24 h. Chronic exposure revealed a significant build up of NO2? in the plasma as well as in the muscles at ≥ 0.44 mg/L NO2?–N peaking in fish exposed to the highest concentration of 3.5 mg/L NO2?–N after 32 days. Still, due to high individual variation methemoglobin (MetHb) was only significantly increased (p < 0.01) at 3.5 mg/L NO2?–N. No adverse effects on red blood cells (RBC) and hematocrit were observed in any of the treatments. In a second experiment, compensation of NO2? toxicity at increasing chloride concentrations (40 (freshwater), 65, 90, 140, 240, 440 mg/L Cl?) was observed at a constant exposure of 10 mg/L NO2?–N for 42 days. At ≥ 240 mg/L Cl?, NO2? build-up in blood plasma and muscle was completely inhibited. At lower Cl? concentrations (≤ 140 mg/L), NO2? was significantly increased in plasma, but only insignificantly elevated in muscle due to high individual variation. MetHb was increased significantly difference only at 40 mg/L Cl? (freshwater control) compared to the control. Again, high individual variations were observed. As a conclusion, S. lucioperca is moderately sensitive towards NO2? and acceptable levels in RAS should hence not exceed 1.75 mg/L NO2?–N to avoid MetHb formation. However, based on the 120 h LC50 and a factor of 0.01 according to Sprague (1971), a NO2? concentration of ≤ 0.061 mg/L NO2?–N is considered as “safe.” Thereby, no NO2? should accumulate in the plasma or muscle tissue during chronic exposure. For 10 mg/L NO2?–N, ≥ 240 mg/L chloride compensates for NO2? uptake in plasma and muscle.  相似文献

This study was to determine the lethal dose 50 (LD50) of CdCl2 in adult Rhinella arenarum and analyzed the effect of two sublethal doses (0.5 and 5 mg/kg) of the xenobiotic in gonads. The 48 h LD50 were 50.0 and 49.8 mg/kg for males and females respectively. Alterations in the ovary were evidenced by nuclear pleomorphism and cytoplasmic vacuolization of the oocytes at the early stages of development with the highest dose and an increase in the population of atretic oocytes. In the interstitial tissue we noticed congestion, edema and fibroblast proliferation. The nuclear maturation of the oocytes was affected by the xenobiotic in a dose- and time-dependent manner. In males, treatment with 5 mg/kg of cadmium (Cd) caused a decrease in the concentration, viability and straight progressive motility of sperm while there was an increase in immotile sperm. Testis histopathology revealed dilated seminiferous tubules, disappearance of cysts, tissue disorganization and leukocyte infiltration. Numerous germ cells showed hydropic tumefaction or signs of focal necrosis. The Cd content in animals intoxicated gonads with the highest sublethal dose was significantly higher than in the control. Results indicate that R. arenarum gonads are target for the xenobiotic, compromising the formation of gametes competent for fertilization, the effective CdCl2 dose being 5 mg/kg.  相似文献

Biodegradation of long chain n-alkanes and crude oil with fast rate and high concentration are desirable for bioremediation, especially in heavily oil-polluted areas, and enhanced oil recovery. We discovered Rhodococcus sp. Moj-3449 with such unique abilities by screening microorganisms for the growth on n-hexadecane at 30 mg/mL. The new strain grew very fast on 120 mg/mL of n-hexadecane giving a cell density of 14.7 g cdw/L after only 2 days’ incubation. During the growth with this strain, the oil–water phases were rapidly emulsified, giving rise to tolerance to high alkane concentration (250 mg/mL) and fast growth rate of 0.10–0.20 h?1 for alkane concentration of 1–180 mg/mL. The degraded concentration of n-hexadecane increased linearly with the initial alkane concentration (1–250 mg/mL). Incubation on n-hexadecane at 250 mg/mL for 7 days gave a cell density of 13.5 g cdw/L and degraded 124 mg/mL of n-hexadecane. The strain grew also fast on n-dodecane (C12), n-tetradecane (C14), and n-octadecane (C18), with degradation preference of C14 (=C16) > C12 > C18. Different from many alkane-degrading strains, Rhodococcus sp. Moj-3449 was found to have subterminal oxidation pathway. Rhodococcus sp. Moj-3449 degraded also crude oil fast at 60–250 mg/mL, with a wide range of n-alkanes (C10–C35) as substrates in which C14–C19 are preferred. The degradation ability increased with initial oil concentration from 60 to 150 mg/mL and slightly decreased afterwards. Incubation on 150 mg/mL of crude oil for 7 days degraded 37% of n-alkanes. The outstanding ability of rapidly degrading long chain n-alkanes and crude oil at high concentration makes Rhodococcus sp. Moj-3449 potentially useful for bioremediation and microbial enhanced oil recovery.  相似文献

Karenia brevisulcata (Chang), a new toxic dinoflagellate of the genus Karenia was isolated from a harmful algal bloom that occurred in Wellington Harbour, New Zealand in 1998. The bloom severely affected most marine biota resulting in long-term ecological damage and causing respiratory distress in harbour bystanders. Cultures of K. brevisulcata produced a range of novel toxins including ten lipid-soluble K. brevisulcata toxins (KBTs) and six water-soluble brevisulcatic acids (BSXs). Brevetoxins were not detected. KBT-F, KBT-G, BSX-1 and BSX-2 were isolated from 1450 L of bulk cultures and purified in mg quantities. Preliminary chemical and toxicological investigations show that KBT-F (M 2054 C107H160O38) and KBT-G (M 2084 C108H162O39) are complex polycyclic ethers with UVmax at 227 nm. NMR data gave characteristics of ladder frame polyether structures and a 2-methylbut-2-enal side chain, similar to gymnocins. The mouse i.p. LD50s for KBT-F and -G were 0.032 and 0.040 mg kg−1, respectively. These KBTs were also highly cytotoxic and haemolytic. BSX-1 (M 916 C49H72O16) and BSX-2 (M 872 C47H68O15) are polycyclic ether dicarboxylates with UVmax 196 nm. BSX-4 and BSX-5, the lactone ring-closed analogues and the presumed primary toxins in the algal cells, were isolated in smaller quantities. Preliminary structural information from NMR and MS showed a carboxylated side chain and some similarities to brevetoxin-A. However, the structures have not yet been fully elucidated due to conformers confounding the NMR. The mouse i.p. LD50 for BSX-1 was 3.9 mg kg−1 while no deaths were seen in mice injected with BSX-2 at 6.6 mg kg−1. The LD50s for the lactones BSX-4 and -5 were 1.4 and 1.6 mg kg−1 respectively. BSX-4 and -5 were agonists of voltage-gated sodium channels but only weakly haemolytic. Activities in the Neuro-2a cytotoxicity assay were ca 10% of dihydrobrevetoxin-2 and were fully antagonised by saxitoxin.  相似文献
