代谢组学(metabolomics/metabonomics)是系统生物学的重要组成部分之一,主要通过分析生物体受环境刺激、病理生理或基因变异等因素引起的内源性小分子代谢物变化来研究其生理功能与代谢之间的关系,进而揭示代谢物变化背后的代谢调控机制与机理。代谢组学技术具有灵敏度高、选择范围广和分析速度快等特点,逐渐在微藻研究领域发挥出优势。本文介绍了代谢组学的研究流程、分析方法以及代谢组学在微藻领域的应用和发展趋势,并提出了代谢组学在微藻研究过程中面临的问题和挑战,以期为代谢组学技术在微藻生物能源利用、环境保护、生产高价值产物等方面的应用提供新的思路和选择。

Metabolomics: back to basics

Metabolomics has developed into a major tool in functional genomics and plant systems biology. The various methods used for metabolomic analysis will be discussed from the analytical methods back to the preanalytical phase and the biological experiment. Particularly aspects of the preanalytical phase of the analysis is dealt with, including the risks of artefact formation with the various commonly used solvents. Metabolomics is like a snap shot, and conclusions from dynamic systems must be drawn with great care as demonstrated with a biosynthetic study of salicylate in Catharanthus roseus cell cultures.     

Toward the implementation of metabolomic assessments of human health and nutrition

Metabolomics is emerging as an exciting post-genomic science with applications that span the scope of biotechnology and medicine. Although metabolomics is still in its infancy, it has already been used to identify the function of genes, describe the effects of toxicological, pharmaceutical, nutritional and environmental interventions, and to build integrated databases of metabolite concentrations across human and research animal populations. Metabolomics provides nutrition with an invaluable tool for determining the distributions of metabolite concentrations in humans, the relationship of these metabolite concentrations to disease, and the extent to which nutrition can modulate metabolite concentrations.
