To quantify the transmission of tomato spotted wilt virus (TSWV) by Frankliniella occidentalis, the median acquisition access period (AAP50) and median inoculation access period (IAP50) were determined. These parameters were established using transmission rates obtained after AAPs and in IAPs which both ranged from 5 to 2560 min. An AAP50 of 106 min was found when larvae acquired virus from TSWV-infected Impatiens plants. IAP50s of 58 or 137 min, respectively, were calculated when petunia or Datura stramonium leaf disks were used to test the inoculation efficiency of viruliferous thrips. The virus could successfully be acquired or inoculated in periods of 5 min. Transmission reached an optimum after an AAP of 21.3 h (AAPopt) and in an IAP of 42.7 h (IAPopt). These results show that TSWV can be acquired and transmitted efficiently by F. occidentalis in short feeding periods.  相似文献

Phytoviruses including tospoviruses are known to affect the behavior and fitness of their vectors both positively and negatively. In this study, we investigated the effects of Tomato spotted wilt virus (TSWV) (family Bunyaviridae, genus Tospovirus) infection on the fitness and feeding ability of tobacco thrips, Frankliniella fusca (Hinds) (Thysanoptera: Thripidae) using peanut, Arachis hypogaea L. (Fabaceae), as a host. Potentially viruliferous F. fusca laid more eggs than non‐viruliferous F. fusca. In contrast, fewer potentially viruliferous F. fusca developed into adults and required a longer developmental time than non‐viruliferous F. fusca, indicating a direct negative effect of the virus on thrips fitness. In addition, no‐choice feeding tests indicated that non‐viruliferous F. fusca fed more rapidly than potentially viruliferous F. fusca. Typically, phytovirus infections are known to enhance the availability of vital nutrients such as free amino acids in infected host plants and to affect other important physiological processes negatively. Free amino acids are known to play a vital role in egg production and development. Further investigations in this study revealed that leaflets of infected plants had ca. 15 times more free amino acids than non‐infected leaflets. TSWV‐infected leaflets were used to rear potentially viruliferous thrips. Higher amino acid levels in TSWV‐infected leaflets than in non‐infected leaflets could have contributed to increased oviposition by potentially viruliferous F. fusca compared to non‐viruliferous F. fusca. Taken together, these results suggest that increased concentrations of free amino acids in TSWV‐infected plants might serve as an incentive for thrips feeding on otherwise unsuitable hosts, thereby facilitating TSWV acquisition and transmission.  相似文献

Tomato spotted wilt orthotospovirus (TSWV) causes substantial economic losses to vegetables and other crops. TSWV is mainly transmitted by thrips in a persistent and proliferative manner, and its most efficient vector is the western flower thrips, Frankliniella occidentalis (Pergande). In moving from the thrips midgut to the salivary glands in preparation for transmission, the virions must overcome multiple barriers. Although several proteins that interact with TSWV in thrips have been characterized, we hypothesized that additional thrips proteins interact with TSWV and facilitate its transmission. In the current study, 67 F. occidentalis proteins that interact with GN (a structural glycoprotein) were identified using a split-ubiquitin membrane-based yeast 2-hybrid (MbY2H) system. Three proteins, apolipoprotein-D (ApoD), orai-2-like (Orai), and obstructor-E-like isoform X2 (Obst), were selected for further study based on their high abundance and interaction strength; their interactions with GN were confirmed by MbY2H, yeast β-galactosidase and luciferase complementation assays. The relative expressions of ApoD and Orai were significantly down-regulated but that of Obst was significantly up-regulated in viruliferous thrips. When interfering with Obst in larval stage, the TSWV acquisition rate in 3 independent experiments was significantly decreased by 26%, 40%, and 35%, respectively. In addition, when Obst was silenced in adults, the virus titer was significantly decreased, and the TSWV transmission rate decreased from 66.7% to 31.9% using the leaf disk method and from 86.67% to 43.33% using the living plant method. However, the TSWV acquisition and transmission rates were not affected by interference with the ApoD or Orai gene. The results indicate that Obst may play an important role in TSWV acquisition and transmission in Frankliniella occidentalis.  相似文献

The predation potential of Haplothrips brevitubus (Karny) for thrips was evaluated in the laboratory. When second stage larvae of Pseudodendrothrips mori (Niwa) were presented to an adult H. brevitubus at densities of 10, 20, 30, and 40 larvae per cage at 25 °C over 24 h, the number of larvae consumed per day increased with an increasing density up to 30. Predation of H. brevitubus exhibited the type II functional response. The mean development time of the egg, larva, and pupa of H. brevitubus were 4.5, 9.6, and 4.8 days, respectively, at 25 °C. The survival rate from egg to adult emergence was 94.7%. One H. brevitubus larva consumed 41.6 P. mori larvae on average during the total larval period. Adult longevity was 35.2 days in females and 34.6 days in males. The pre-oviposition period was 2.7 days and the oviposition period was 31.5 days. The lifetime fecundity was 120.1 eggs and the mean daily oviposition rate was 3.6 eggs. Calculated mean generation time (T) was 29.5 days, intrinsic rate of natural increase (rm) was 0.162, and net reproductive rate (R0) was 56.5. The rm value of H. brevitubus was higher than that of Thrips palmi Karny and almost equal to that of Frankliniella occidentalis (Pergande). These results indicate that H. brevitubus has good potential as a predator of P. mori and is likely to be useful for controlling thrips.  相似文献

Thermal requirements for development and life table statistics ofAphis gossypii Glover (Homoptera, Aphididae) were determined over a range of constant temperatures from 10 to 30°C. The lower development threshold and the sum of effective temperatures were 6.9°C and 90.1°C, respectively, for preimaginal development, and 5.8°C and 113.6°C from birth to the onset of reproduction. Mean total fecundity ranged from 36 larvae per female at 10°C to 76 larvae at 30°C. On a time scale of days, net reproductive rate (R o ) increased with increasing temperature while generation time (T) decreased causing the intrinsic rate of increase (r m ) to increase linearly from 0.115 to 0.465. On a day-degree scaler m only varied from 0.019 to 0.028 because the growth ofR o was compensated by an increase inT with increase in temperature. The nearly constantr m in terms of day-degrees, over a wide range of temperatures, greatly simplifies the prediction of future population numbers ofA. gossypii.  相似文献

Herbivores can profit from vectoring plant pathogens because the induced defence of plants against pathogens sometimes interferes with the induced defence of plants against herbivores. Plants can also defend themselves indirectly by the action of the natural enemies of the herbivores. It is unknown whether the defence against pathogens induced in the plant also interferes with the indirect defence against herbivores mediated via the third trophic level. We previously showed that infection of plants with Tomato spotted wilt virus (TSWV) increased the developmental rate of and juvenile survival of its vector, the thrips Frankliniella occidentalis. Here, we present the results of a study on the effects of TSWV infections of plants on the effectiveness of three species of natural enemies of F. occidentalis: the predatory mites Neoseiulus cucumeris and Iphiseius degenerans, and the predatory bug Orius laevigatus. The growth rate of thrips larvae was positively affected by the presence of virus in the host plant. Because large larvae are invulnerable to predation by the two species of predatory mites, this resulted in a shorter period of vulnerability to predation for thrips that developed on plants with virus than thrips developing on uninfected plants (4.4 vs. 7.9 days, respectively). Because large thrips larvae are not invulnerable to predation by the predatory bug Orius laevigatus, infection of the plant did not affect the predation risk of thrips larvae from this predator. This is the first demonstration of a negative effect of a plant pathogen on the predation risk of its vector.  相似文献

Populations of onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), were shown to differ significantly in their ability to transmit an isolate of tomato spotted wilt virus (Tospovirus: Bunyaviridae) (TSWV) collected from potato [Solanum tuberosum L. (Solanaceae)]. To gain an understanding of the basis for this variation, we generated reciprocal crosses between an efficient and an inefficient transmitting population. The resulting F1 progeny and progeny from the parental populations were tested for their ability to transmit TSWV. Our results indicate that the ability to transmit TSWV efficiently by T. tabaci is inherited as a recessive trait.  相似文献

Tomato spotted wilt virus (TSWV) is an important plant virus that infects a wide range of hosts including weeds making its management difficult. A survey was undertaken to establish the occurrence of weed species in tomato production systems in Kenya and their role as hosts of TSWV and its vectors. Selected weed species were further evaluated for their reaction to TSWV, transmission efficiency by Frankliniella occidentalis and ability to support thrips reproduction. Of the 43 weed species identified in the field, 29 species had been reported as hosts of TSWV, two were non‐hosts and 11 had no record of their status. Among the more common species, Amaranthus hybridus, Solanum nigrum, Tagetes minuta and Datura stramonium were susceptible to the virus and supported high levels of thrips reproduction. The TSWV could not be transmitted to Galinsoga parviflora and Sonchus oleraceus by F. occidentalis despite them being highly susceptible in mechanical transmission tests. There was a significant correlation between feeding damage and number of larvae of F. occidentalis on different weeds. Occurrence of weeds that support thrips reproduction and are good hosts of TSWV is a clear indicator of their role in epidemiology and the importance of their management for disease control.  相似文献

Stratiolaelaps ( = Hypoaspis) miles (Berlese) (Acari: Mesostigmata: Laelapidae) is a polyphagous soil-dwelling predatory mite that is widely marketed for use in greenhouse production systems to manage populations of dark-winged fungus gnats, Bradysia spp. (Diptera: Sciaridae) and for supplemental control of thrips. The suggestion by Walter and Campbell (2003, Biol. Control 26: 253–269) that North American commercial cultures of S. miles may actually be S. scimitus was confirmed. The development and reproduction at 21–23 °C of S. scimitus provided ad libidum with one of three different prey – the fungus gnat Bradysia aff. coprophila (Lintner), potworms (Enchytraeidae), or Sancassania aff. sphaerogaster (Zachvatkin, 1937) (Acari: Astigmata: Acaridae) – were compared. Developmental duration of the egg and non-feeding larval stages were 2.47 and 1.11 days, respectively; mortalities were 8.3 and 5.5%. Stratiolaelaps scimitus failed to develop beyond the protonymphal stage when provided with S. aff. sphaerogaster alone, although some feeding was observed. Development and reproduction of S. scimitus was successful on both fungus gnat larvae and enchytraeids, with no influence of prey on protonymphal duration (4.70 days) and mortality (8.3%), or on deutonymphal duration (4.61 days) and mortality (6.1%). Adult female S. scimitus feeding on potworms, compared to those feeding on fungus gnat larvae, had a significantly shorter pre-oviposition period (2.69 vs. 4.59 days). However, diet did not influence other adult female developmental or reproductive characteristics (oviposition period, 18.6 days; post-oviposition period, 6.2 days; total adult longevity, 27.3 days; total number of eggs, 26.5). S. scimitus reared on potworms tended (p = 0.06) to have a higher intrinsic rate of increase, a higher finite rate of increase and a shorter doubling time (r m = 0.142 day−1, λ = 1.153, Dt = 4.85 days) than those reared on fungus gnat larvae (r m = 0.105 day−1, λ = 1.110, Dt = 6.58 days), but differences in net reproductive rate (R 0) and generation time (G) were not significant.  相似文献

Tomato spotted wilt virus (TSWV) replicates in both its plant hosts and its thrips vectors. Replication of TSWV within thrips suggests the potential for pathological effects that could affect the fitness of its vectors directly, whereas infection of the plant may alter its suitability as a host for thrips development. This study was undertaken to examine the influence of TSWV isolate, host plant, and temperature on potential direct and host-mediated effects of virus infection of the thrips and the plant on Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), an important vector of TSWV. Neonate F. occidentalis were reared to adult eclosion on excised foliage of Datura stramonium (L.) (Solanaceae) or Emilia sonchifolia (L.) (Compositae) infected with either the CFL or RG2 isolate of TSWV, or not infected. Effects of the TSWV isolates and host plants on thrips were measured at 18.3, 23.9, and 29.4 °C. Results demonstrate significantly improved survival and a small but significant decrease in development time of F. occidentalis on TSWV-infected plants. These effects resulted from the combined influence of the direct effects of the virus on infected thrips and plant-mediated effects resulting from virus infection of the thrips’ host plant. Our results extend previous findings and help to explain inconsistencies among previously published reports by demonstrating that the manifestation and magnitude of effects of TSWV on F. occidentalis are dependent on host plant, virus isolate, and temperature.  相似文献

Tomato spotted wilt virus (TSWV) causes serious diseases of many economically important crops. Disease control has been achieved by breeding tomato and pepper cultivars with the resistance genes Sw‐5 and Tsw, respectively. However, TSWV isolates overcoming these genetic resistances have appeared in several countries. To evaluate the risk of spread of these resistance‐breaking isolates, we tested their ability of transmission by the main vector of TSWV, the thrips Frankliniella occidentalis. We compared the transmission rate by thrips of six TSWV isolates of different biotype (able or unable to overcome this resistance in pepper and tomato), and with divergent genotype (A and B). Our results indicate that the transmission rate was related to the amount of virus accumulated in thrips but not to virus accumulation in the source plants on which thrips acquired the virus. No correlation was found between transmission efficiency by thrips and the genotype or between transmission efficiency and the ability of overcoming both resistances. This result suggests that resistance‐breaking isolates have the same potential to be transmitted as the isolates unable to infect resistant tomato and pepper cultivars.  相似文献

One approach that can be employed in integrated pest management is the use of proteins with antinutritional effects on insect metabolism and development. The antimetabolic properties of soybean proteinase inhibitor (SPI) on growth of neonate larvae of the sugarcane borer, Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) have been evaluated. When incorporated into an artificial diet at 0.5% (w/w), SPI retarded growth rate and development of larvae when compared with larvae fed on artificial diet alone. However, larval survival was not significantly affected. The purpose of our research was to calculate demographic statistics for the sugarcane borer reared on diet either with or without semi-purified extract of SPI. Net reproductive rate (R 0), instantaneous rate of increase (r m), combined age-specific survivorship (l x) and age specific fecundity (m x) provide information about population growth potential. These parameters were measured in order to determine the effects of the proteinase inhibitor on the insect's population dynamics. The observed differences would potentially translate into large reductions in population growth, indicating a potential value of using SPI for protecting sugarcane plants against damage by the sugarcane borer.  相似文献

为探讨醉马草水浸液对豌豆蚜触杀活性及种群增长的影响,采用带虫浸叶法比较不同生育期醉马草带菌(E+)和不带菌(E-)水浸液对豌豆蚜触杀活性及种群生命表,测定了豌豆蚜的死亡率及触杀后对其生殖期,平均繁殖力,繁殖率及生命表参数的影响。结果表明,不同生育期醉马草带菌(E+)水浸液触杀豌豆蚜后对其各项指标均有显著影响。在苗期时,E+水浸液触杀豌豆蚜后校正死亡率最高,繁殖力最低,内禀增长率(rm=0.145 d-1)和净生殖率(R0=4.802头)均为最小值。在成熟期时,E+水浸液触杀豌豆蚜24 h、48 h和72 h后校正死亡率分别为26.15%,19.01%,9.07%;繁殖期(3.87 d),平均繁殖力(8.80头),繁殖率(1.40%),内禀增长率(rm=0.208 d-1)和净生殖率(R0=8.820头)。在枯黄期时,E+水浸液触杀豌豆蚜后校正死亡率最低,繁殖力最强,内禀增长率(rm=0.247 d-1)和净生殖率(R0=13.647头)均为最大值。不同生育期醉马草E-水浸液触杀豌豆蚜后对其种群繁殖无显著影响,与对照差异不显著(P>0.05)。综上,苗期醉马草E+水浸液对豌豆蚜有较好的触杀效果,校正死亡率高,且触杀后当代繁殖力减弱,种群扩建时间延长,不利于其种群繁殖和增长;故苗期醉马草E+水浸液具有很好的杀虫潜力,所采用水浸液方法制备简单,成本低,可为新型植物源农药研发提供重要理论依据。  相似文献

The nature of spatial and temporal dynamics of Tomato spotted wilt virus (TSWV) and its vector in a potato crop cv. Innovator without insecticide application is analysed. Seed tuber was analysed for the presence of TSWV as a source of initial inoculum. The presence of plants with symptoms of TSWV was evaluated by visual observation and DAS‐ELISA analysis to confirm the virus infection. Thrips species were collected from leaves and inflorescences and identified under stereomicroscope. The distribution of symptomatic plants and thrips species was recorded five times at 14 days intervals. The initial seed tuber infection was of 1.1%. Disease incidence was 0% at 29 days after planting (DAP), 0.2% at 43 DAP, 2.2% at 56 DAP, 11.6% at 70 DAP and 14.6% at 84 DAP. The progress of the disease was adequately described by a Logistic model [y = 0.15/(1 + 1205372.93 × exp (?0.22 × DAP))]. Thrips vector species identified as resident in the crop during the whole cycle were Thrips tabaci (n = 423), Frankliniella occidentalis (n = 141) and as occasional species, F. schultzei (n = 34) and F. gemina (n = 5) were found. At 43 and 56 DAP a random distribution pattern was observed and the thrips species found were T. tabaci (n = 188) and F. occidentalis (n = 105). An aggregated pattern was determined at 70 and 84 DAP. Spatial patterns of the disease spread suggest a polycyclic epidemic with TSWV secondary spread in the potato crop. Multiple control measures were deduced from these epidemiological results like virus testing in tubers, removal of external virus infection sources and thrips control.  相似文献

In the present studies, life table data and intrinsic rate of natural increase (rm) of the eulophid pupal parasitoid,P. furvus (Gahan) were obtained fromChilo partellus (Swinhoe) at a constant temperature of 26.7±0.6°C, 52.3±2.7% RH in the laboratory. Development of immature stages took 19.1±0.3 days; adult females lived for 7.9±3.3 (range, 2–13) days and produced a mean of 91.9±22.4 progeny per female, with a sex ratio of 1:2.9 (♂:♀). The intrinsic rate of natural increase (rm) was 0.2558/female/day; the net reproductive rate (R0), 237.25; the capacity for increase (rc) of 0.252; and the finite rate of increase (λ) of 1.29/female/day; thus, each female contributed 231.42 individuals to the population in a mean generation time of 21.38 days.   相似文献

Sublethal effects of the growth inhibitor, clofentezine, on life-table parameters of Tetranychus urticae Koch females treated at different developmental stages with a concentration causing ≥90% mortality were investigated. Females which survived treatment as ‘early’ (0–24 h old) eggs produced 12% more offspring than the untreated females during the first five days of oviposition. This resulted in a significant rise in the intrinsic rate of increase (r j ): 0.324, compared to 0.299 in the untreated females. This effect may be interpreted as hormoligosis. Clofentezine treatment at any other developmental stage of T. urticae significantly decreased both longevity and fertility of female survivors. Females which survived treatment either as ‘late’ (72–96 h old) eggs or larvae had 2.6 times lower net reproductive rate (R 0) than the untreated females, and the r j values were significantly lower: 0.242 and 0.215, respectively (0.285 in the untreated females). Females which survived treatment either as protonymphs or deutonymphs had 3.9 times and 6 times lower R 0, respectively. Corresponding r j values were 0.178 and 0.146, respectively (0.247 in the untreated females). The clofentezine treatment at all stages influenced the age distribution of survivors. The sublethal effects of clofentezine and their impact on T. urticae management are discussed. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The thrips Ceratothripoides claratris is an efficient vector of the Capsicum chlorosis virus (CaCV). Transmission studies with a natural population of C. claratris found in a greenhouse ‘GH’ and a ‘colony’ derived from this ‘GH’ population by selection and inbreeding resulted in lowering the percentage of viruliferous individuals within the ‘colony’. After passing through approximately 20 generations, the ‘colony’ lost the ability to transmit the CaCV. When either viruliferous or non‐viruliferous virgin females reproduced parthenogenetically, 81% of F1 arrhenotokous males inherited their viruliferous status from their mothers, whilst, no viruliferous offspring arose from non‐viruliferous virgin mothers. Crosses between viruliferous and non‐viruliferous individuals suggest that the competence of the thrips C. claratris as a vector for this virus is probably a heritable trait controlled by a recessive allele.  相似文献

The effects of parasitism by the nematode Thripinema nicklewoodi Siddiqi (Tylenchida: Allantonematidae) on tospovirus infection and feeding behavior of Frankliniella occidentalis Pergande (Thysanoptera: Thripidae) were studied in the laboratory. In an initial experiment, nematode parasitism reduced the numbers of adult thrips that were positive to Impatiens necrotic spot virus (INSV) by enzyme-linked immunosorbent assay (ELISA) compared to controls. Three hypotheses on possible mechanisms causing this reduction were tested. H1: nematodes have a tendency to penetrate healthy thrips rather than INSV-infected thrips; H2: parasitized first instar thrips are less able to acquire virus during feeding; or H3: INSV replication is suppressed in parasitized thrips. H1 and H2 were proven false as we found no difference in nematode attack rate between healthy thrips and thrips that have taken up INSV and no difference in virus uptake or feeding activity between parasitized and non-parasitized larval thrips. H3 was not supported by data from our tests (P=0.07) but remains the explanation most worthy of future investigation. Interestingly, INSV transmission was not affected by nematode parasitism even though it reduced feeding activity of adult female thrips by 81% on leaves, 38% on pollen, and 22% on honey. However, despite lowered total feeding, probing by parasitized thrips (in honey) was not reduced, and this may explain why lowered feeding did not result in lowered virus transmission.  相似文献

Spherical viruslike particles (VLP) were found in the tissues of apparently healthy tobacco thrips, Frankliniella fusca. The particles occurred in abundance in thrips from Ontario but were absent in thrips from Oklahoma reared under identical conditions. The VLP were not transmissible to any of the seven plant hosts (in four families) of F. fusca suggesting that they may be an insect virus. Transmission of tomato spotted wilt virus (TSWV) by F. fusca, a known vector, was not affected by the presence or absence of the VLP. No TSWV particles were detected in tissues of F. fusca that transmitted TSWV to test plants. The VLP occurred in several internal organs and hemocoele of the thrips and were isolated in vitro by preparing homogenates of gut tissues. Infection of oocytes and presence of VLP in young nymphs suggested transovarial transmission of the particles. The VLP measured 62 ± 4 nm in diameter and usually occurred in dense viroplasms in the cell cytoplasm. Development of the particles within the viroplasms is discussed.  相似文献

Six species of the genus Panonychus are known from Japan. Life-history parameters of all six species were investigated at 25 °C, and for three species two strains of different geographical origin were included. The intrinsic rate of natural increase (r m ) ranged from 0.172/day for the P. osmanthi albino strain to 0.209/day for P. citri, while the net reproductive rate (R 0) varied from 23.98 in the thelytokous species P. thelytokus to 46.61 in P. citri. Both values were higher in the polyphagous species (P. ulmi, P. mori and P. citri), which are considered crop pests, than those in the oligophagous species (P. thelytokus and P. osmanthi), considered non-pests. The only exception was P. bambusicola, an oligophagous non-pest species with R 0- and r m -values closely resembling those of the three polyphagous species.
