Summary Enhanced cellular cAMP levels have been shown to increase apical membrane Cl and HCO 3 conductances in epithelia. We found that the phosphodiesterase inhibitor 3-isobutyl-1-methyl-xanthine (IBMX) increases cAMP levels inNecturus gallbladder. We used conventional open-tip and double-barreled Cl-selective microelectrodes to study the effects of IBMX on membrane conductances and intracellular Cl activities in gallbladders mounted in a divided chamber and bathed with Ringer's solutions at 23°C and pH 7.4. In HCO 3 -free media, 0.1mM IBMX added to the mucosal medium depolarized the apical membrane potentialV a , decreased the fractional resistanceF R , and significantly reduced intracellular Cl activity (a Cl i ). Under control conditions,a Cl i was above the value corresponding to passive distribution across the apical cell membrane. In media containing 25mM HCO 3 , IBMX caused a small transient hyperpolarization ofV a followed by a depolarization not significantly different from that observed in HCO 3 -free Ringer's. Removal of mucosal Cl, Na+ or Ca2+ did not affect the IBMX-induced depolarization inV a . The basolateral membrane ofNecturus gallbladder is highly K+ permeable. Increasing serosal K+ from 2.5 to 80mM, depolarizedV a . Mucosal IBMX significantly reduced this depolarization. Addition of 10mM Ba2+, a K+ channel blocker, to the serosal medium depolarizedV a and, essentially, blocked the depolarization induced by IBMX. These results indicate that mucosal IBMX increases apical HCO 3 conductance and decreases basolateral K+ conductance in gallbladder epithelial cells via a cAMP-dependent mechanism. The latter effect, not previously reported in epithelial tissues, appears to be the major determinant of the IBMX-induced depolarization ofV a .  相似文献

Summary In a previous study we presented evidence that chloride transport across the basolateral membrane inNecturus proximal tubule cells occurs predominantly via exchange for both Na+ and HCO 3 . In this study the regulation of intracellular chloride was further examined in the doubly-perfused kidney preparation using conventional and chloride-sensitive microelectrodes. Application of hypertonic basolateral solutions containing 80mm raffinose stimulated an efflux of chloride such that chloride activity remained unchanged at control levels. Membrane potential did not change in these experiments. Inhibition of Cl exit across the basolateral cell membrane by removal of either HCO 3 or Na+ from the perfusion solution resulted in a significant increase in intracellular chloride activity,a Cl i , when basolateral osmolarity was raised. Hypertonic basolateral solutions also produced a significant rise ina Cl i in the presence of SITS.This study provides further evidence that chloride is transported across the basolateral cell membrane in exchange for both Na+ and HCO 3 . Since this exchange mechanism is activated in response to hypertonic solutions, these studies suggest a functional role for this exchanger in the regulation ofa Cl i in theNecturus proximal tubule cell during volume changes.  相似文献

Summary Two methods, the measurement of the response of the basolateral membrane potential (V bl) of proximal tubule cells ofNecturus to step changes in basolateral K+ concentration, and cellular cable analysis, were used to assess the changes in basolateral potassium conductance (G K) caused by a variety of maneuvers. The effects of some of these maneuvers on intracellular K+ activity (a K i ) were also evaluated using double-barreled ion-selective electrodes. Perfusion with 0mm K+ basolateral solution for 15 min followed by 45 min of 1mm K+ solution resulted in a fall in basolateral potassium (apparent) transference number (t K),V bl anda K i . Results of cable analysis showed that total basolateral resistance,R b , rose. The electrophysiological effects of additional manipulations, known to inhibit net sodium reabsorption across the proximal tubular epithelium ofNecturus, were also investigated. Ouabain caused a fall int K accompanied by large decreases ina K i andV bl. Lowering luminal sodium caused a fall int K and a small reduction inV bl. Selective reduction of peritubular sodium, a maneuver that has been shown to block sodium transport from lumen to peritubular fluid, also resulted in a significant decrease int K. These results suggest thatG K varies directly with rate of transport of the sodium pump, irrespective of the mechanism of change in pump turnover.Part of this material has been presented at the 10th International Conference on Biological Membranes (Cohen & Giebisch, 1984).  相似文献

Summary Conventional and Cl-selective liquid ion-exchanger intracellular microelectrodes were employed to study the effects of extracellular ionic substitutions on intracellular Cl activity (aCl i ) inNecturus gallbladder epithelium. As shown previously (Reuss, L., Weinman, S.A., 1979;J. Membrane Biol. 49:345), when the tissue was exposed to NaCl-Ringer on both sidesaCl i was about 30mm, i.e., much higher than the activity predicted from equilibrium distribution (aCleq) across either membrane (5–9mm). Removal of Cl from the apical side caused a reversible decrease ofaCl i towards the equilibrium value across the basolateral membrane. A new steady-stateaCl i was reached in about 10 min. Removal of Na from the mucosal medium or from both media also caused reversible decreases ofaCl i when Li, choline, tetramethylammonium or N-methyl-d-glucamine (NMDG) were employed to replace Na. During bilateral Na substitutions with choline the cells depolarized significantly. However, no change of cell potential was observed when NMDG was employed as Na substitute. Na replacements with choline or NMDG on the serosal side only did not changeaCl i . When K substituted for mucosal Na, the cells depolarized andaCl i rose significantly. Combinations of K for Na and Cl for SO4 substitutions showed that net Cl entry during cell depolarization can take place across either membrane. The increase ofaCl i in depolarized cells exposed to K2SO4-Ringer on the mucosal side indicates that the basolateral membrane Cl permeability, (P Cl) increased. These results support the hypothesis that NaCl entry at the apical membrane occurs by an electroneutral mechanism, driven by the Na electrochemical gradient. In addition, we suggest that Cl entry during cell depolarization is downhill and involves an increase of basolateral membraneP Cl.  相似文献

Summary The characteristics of Cl movement across luminal and basolateral membranes ofAmphiuma intestinal absorptive cells were studied using Cl-sensitive microelectrodes and tracer36Cl techniques. Intracellular Cl activity (a Cl i ) was unchanged when serosal Cl was replaced; when luminal Cl was replaced cell Cl was rapidly lost. Accordingly, the steady statea Cl i could be varied by changing the luminal [Cl]. As luminal [Cl] was raised from 1 to 86mM,a Cl i rose in a linear manner, the mucosal membrane hyperpolarized, and the transepithelial voltage became serosa negative. In contrast, the rate of Cl transport from the cell into the serosal medium, measured as the SITS-inhibitable portion of the Cl absorptive flux, attained a maximum whena Cl i reached an apparent value of 17mm, indicating the presence of a saturable, serosal transport step. The stilbeneinsensitive absorptive flux was linear with luminal [Cl], suggestive of a paracellular route of movement. Intracellulara Cl was near electrochemical equilibrium at all but the lowest values of luminal [Cl] after interference produced by other anions was taken into account.a Cl i was unaffected by Na replacement, removal of medium K, or elevation of medium HCO 3 . Mucosae labeled with36Cl lost isotope into both luminal and serosal media at the same rate and from compartments of equal capacity. Lowering luminal [Cl] or addition of theophylline enhanced luminal Cl efflux. It is concluded that a conductive Cl leak pathway is present in the luminal membrane. Serosal transfer is by a saturable, stilbene-inhibitable pathway. Luminal Cl entry appears to be passive, but an electrogenic uptake cannot be discounted.  相似文献

Summary In goldfish intestine chloride was substituted by large inorganic anions (gluconate or glucuronate) either mucosally, serosally or bilaterally. Changes in intracellular activities of chloride (a i Cl), sodium (a i Na+) and potassium (a i K+), pHi, relative volume, membrane and transepithelial potentials, transepithelial resistance and voltage divider ratio were measured. Control values were:a i Cl=35 meq/liter, a i Na+=11 meq/liter and a i K+=95 meq/liter. During bilateral substitution the latter two did not change while a i Cl dropped to virtually zero.Mucosal membrane potentials (ms) were: control,-53 mV; serosal substitution,-51 mV; bilateral substitution,-66 mV; while during mucosal substitution a transient depolarization occurred and the final steady state ms was-66 mV.During control and bilateral substitution the transepithelial potentials (ms) did not differ from zero. During unilateral substitutions ms was small, in the order of magnitude of the errors in the liquid junction potentials near the measuring salt bridges.During bilateral substitution pH i increased 0.4 pH units. Cellular volume decreased during mucosal substitution to 88% in 40 min; after serosal substitution it transiently increased, but the new steady-state value was not significantly above its control value.Three minutes after mucosal substitution ana i Cl of approx. 10 meq/liter was measured.Chemical concentrations of Na, K and Cl were determined under control conditions and bilateral substitution. Cl concentrations were also measured as a function of time after unilateral substitutions.The data indicate an electrically silent chloride influx mechanism in the brush border membrane and an electrodiffusional chloride efflux in the basolateral membrane. A substantial bicarbonate permeability is present in the basolateral membrane. The results are in agreement with the observed changes in membrane resistances, volume changes and pH changes.  相似文献

Summary The Na+ requirement for active, electrogenic Cl absorption byAmphiuma small intestine was studied by tracer techniques and double-barreled Cl-sensitive microelectrodes. Addition of Cl to a Cl-free medium bathingin vitro intestinal segments produced a saturable (K m =5.4mm) increase in shortcircuit current (I sc) which was inhibitable by 1mm SITS. The selectivity sequence for the anion-evoked current was Cl=Br>SCN>NO 3 >F=I. Current evoked by Cl reached a maximum with increasing medium Na concentration (K m =12.4mm). Addition of Na+, as Na gluconate (10mm), to mucosal and serosal Na+-free media stimulated the Cl current and simultaneously increased the absorptive Cl flux (J ms Cl ) and net flux (J net Cl ) without changing the secretory Cl flux (J sm Cl ). Addition of Na+ only to the serosal fluid stimulatedJ ms Cl much more than Na+ addition only to the mucosal fluid in paired tissues. Serosal DIDS (1mm) blocked the stimulation. Serosal 10mm Tris gluconate or choline gluconate failed to stimulateJ ms Cl . Intracellular Cl activity (a Cl i ) in villus epithelial cells was above electrochemical equilibrium indicating active Cl uptake. Ouabain (1mm) eliminated Cl accumulation and reduced the mucosal membrane potential m over 2 to 3 hr. In contrast, SITS had no effect on Cl accumulation and hyperpolarized the mucosal membrane. Replacement of serosal Na+ with choline eliminated Cl accumulation while replacement of mucosal Na+ had no effect. In conclusion by two independent methods active electrogenic Cl absorption depends on serosal rather than mucosal Na+. It is concluded that Cl enters the cell via a primary (rheogenic) transport mechanism. At the serosal membrane the Na+ gradient most likely energizes H+ export and regulates mucosal Cl accumulation perhaps by influencing cell pH or HCO 3 concentration.  相似文献

Summary Forskolin (i.e, cAMP)-modulation of ion transport pathways in filter-grown monolayers of the Cl-secreting subclone (19A) of the human colon carcinoma cell line HT29 was studied by combined Ussing chamber and microimpalement experiments.Changes in electrophysiological parameters provoked by serosal addition of 10–5 m forskolin included: (i) a sustained increase in the transepithelial potential difference (3.9±0.4 mV). (ii) a transient decrease in transepithelial resistance with 26±3 · cm2 from a mean value of 138±13 · cm2 before forskolin addition, (iii) a depolarization of the cell membrane potential by 24±1 mV from a resting value of –50±1 mV and (iv) a decrease in the fractional resistance of the apical membrane from 0.80±0.02 to 0.22±0.01. Both, the changes in cell potential and the fractional resistance, persisted for at least 10 min and were dependent on the presence of Cl in the medium. Subsequent addition of bumetanide (10–4 m), an inhibitor of Na/K/2Cl cotransport, reduced the transepithelial potential, induced a repolarization of the cell potential and provoked a small increase of the transepithelial resistance and fractional apical resistance. Serosal Ba2+ (1mm), a known inhibitor of basolateral K+ conductance, strongly reduced the electrical effects of forskolin. No evidence was found for a forskolin (cAMP)-induced modulation of basolateral K+ conductance.The results suggest that forskolin-induced Cl secretion in the HT-29 cl.19A colonic cell line results mainly from a cAMP-provoked increase in the Cl conductance of the apical membrane but does not affect K+ or Cl conductance pathways at the basolateral pole of the cell. The sustained potential changes indicate that the capacity of the basolateral transport mechanism for Cl and the basal Ba2+-sensitive K+ conductance are sufficiently large to maintain the Cl efflux across the apical membrane. Furthermore, evidence is presented for an anomalous inhibitory action of the putative Cl channel blockers NPPB and DPC on basolateral conductance rather than apical Cl conductance.  相似文献

Summary The ability of early proximal tubule cells of theNecturus kidney to regulate volume was evaluated using light microscopy, video analysis and conventional microelectrodes.Necturus proximal tubule cells regulate volume in both hyperand hyposmotic solutions. Volume regulation in hyperosmotic fluids is HCO 3 dependent and is associated with a decrease in the relative K+ conductance of the basolateral cell membrane and a decrease in the resistance ratio,R a /R bl . Volume regulation in hyposmotic solutions is also dependent upon the presence of HCO 3 but is also inhibited by 2mm Ba2+ in the basolateral solution. Hyposmotic regulation is accompanied by an increase in the relative K+ conductance of the basolateral cell membrane and an increase inR a /R bl . Neither hypo- nor hyposmotic regulation have any affect on the depolarization of the basolateral cell membrane potential induced by HCO 3 removal. We conclude that volume regulation in the early proximal tubule of the kidney involves both HCO 3 -dependent transport systems and the basolateral K+ conductance.  相似文献

Summary A simple numerical model for theNecturus gallbladder epithelium is presented. K+, Na+ and Cl cross the mucosal and serosal membranes as well as the junctions by means of electrodiffusion; furthermore the mucosal membrane contains a neutral entry mechanism for NaCl and the serosal membrane contains an active pump for K+ and Na+. The values which have been used for the model are taken from the literature. The model can only attain steady states if the resistance of the serosal membrane is lower than 1000 cm2. Values reported in the literature for the resistance of this membrane vary from about 3000 to about 100 cm2. We shall argue, however, that the higher estimates are in error because they are derived from a model of the tissue in which each membrane and the junction are modeled by a resistor; this procedure is invalid because the resistance of the lateral intercellular space relative to the resistance of the tight junctions is neglected and consequently the resistance of the serosal membrane is overestimated by a factor of about four. Apart from predicting a realistic steady state at normal external concentrations the model can predict quantitatively several experimental results obtained from the living epithelium. We have focused on the experiments which test the permeabilities of the serosal membrane and the properties of the pump:i) Replacement of serosal Cl by an impermeant ion.ii) Replacement of serosal K+ by Na+.iii) Inhibiting the (Na+, K+)-pump. The best correspondence between model and experiments is obtained when the pump is assumed to be electrogenic (or rheogenic) with a ratio of coupling between Na+ and K+ of 32. In this case both model and direct experiments (also presented in this paper) show an initial abrupt depolarization of 6 to 7 mV. The model also shows that it cannot be concluded fromi andii that the Cl permeability of the serosal membrane is low. The model explains, even with high passive Cl permeabilities, why the intracellular Cl concentration is relatively unaffected by paracellular currents, a fact which in other epithelia has been taken as an implication of a low Cl permeability of the serosal membranes.  相似文献

Summary Intracellular C1, K and Na activities (a Cl i ,a k i anda Na i ) and transmucosal membrane potential (E m) in epithelial cells ofNecturus gallbladder were measured at different external Na concentrations ([Na]o), with liquid ion-exchanger and conventional microelectrodes. Bladders were mounted in a divided chamber at 23°C between identical HCO3-free Ringer solutions containing 5mm K. The pH was 7.2. Tris was substituted for Na. Measurements were made under steady-state conditions as determined by the constancy of the transepithelial potential difference. Both,a Cl i anda Na i increased in a saturable fashion with [Na]o.E m did not change significantly. Average values (±sem) under normal conditions ([Na]o=100mm) fora Cl i ,a Na i andE m were 16.8±0.8mm (n=9), 9.7±0.6mm (n=10) and –52.6±0.6 mV (n=26), respectively. In Na-free mediaa Cl i declined to its equilibrium value.a K i (96±2mm;n=7) did not change when [Na]o was varied between 100 and 10mm but decreased to 80±3mm (n=4) in Na-free media.Transmembrane electrochemical potential differences, , for Cl and Na were calculated at four different [Na]o levels. A highly significant linear relation between and was found, indicating that Cl and Na transport are energetically linked. The results support the view that the energy necessary for intracellular Cl accumulation is derived from the simultaneous dissipation of the chemical potential gradient of Na across the apical membrane and that the coupled entry mechanism is electroneutral.  相似文献

Summary The effects of stepwise concentration changes of K+ and HCO 3 in the basolateral solution on the basolateral membrane potential (V bl) of proximal tubule cells of the doubly-perfusedNecturus kidney were examined using conventional microelectrodes. Apparent transference numbers were calculated from changes inV bl after alterations in external K+ concentration from 1.0 to 2.5mm (t K, 1.0–2.5), 2.5 to 10, and in external HCO 3 concentration (at constant pH) from 5 to 10mm (t HCO3, 5–10), 10 to 20, or 10 to 50.t K, 2.5–10 was 0.38±0.02 under control conditions but was sharply reduced to 0.08±0.03 (P>0.001) by 4mm Ba++. This concentration of Ba++ reducedV bl by 9±1 mV (at 2.5 external K+). Perfusion with SITS (5×10–4 m) for 1 hr hyperpolarizedV bl by 10±3 mV and increasedt K, 2.5–10 significantly to 0.52±0.01 (P<0.001). Ba++ application in the presence of SITS depolarizedV bl by 22±3 mV. In control conditionst HCO3, 10–50 was 0.63±0.05 and was increased to 0.89±0.07 (P<0.01) by Ba++ but was decreased to 0.14±0.02 (P<0.001) by SITS. In the absence of apical and basolateral chloride, the response ofV bl to bicarbonate was diminished but still present (t HCO3, 10–20 was 0.35±0.03). Intracellular pH, measured with liquid ion-exchange microelectrodes, increased from 7.42±0.19 to 7.57±0.17 (P<0.02) when basolateral bicarbonate was increased from 10 to 20mm at constant pH. These data show that the effects of bicarbonate onV bl are largely independent of effects on the K+ conductance and that there is a significant current-carrying bicarbonate pathway in the basolateral membrane. Hence, both K+ and HCO 3 gradients are important in the generation ofV bl, and their relative effects vary reciprocally.  相似文献

Summary The effects of complete substitution of gluconate for mucosal and/or serosal medium Cl on transepithelial Na+ transport have been studied using toad urinary bladder. With mucosal gluconate, transepithelial potential difference (V T) decreased rapidly, transepithelial resistance (R T) increased, and calculated short-circuit current (I sc) decreased. CalculatedE Na was unaffected, indicating that the inhibition of Na+ transport was a consequence of a decreased apical membrane Na+ conductance. This conclusion was supported by the finding that a higher amiloride concentration was required to inhibit the residual transport. With serosal gluconateV T decreased,R T increased andI sc fell to a new steady-state value following an initial and variable transient increase in transport. Epithelial cells were shrunken markedly as judged histologically. CalculatedE Na fell substantially (from 130 to 68 mV on average). Ba2+ (3mm) reduced calculatedE Na in Cl Ringer's but not in gluconate Ringer's. With replacement of serosal Cl by acetate, transepithelial transport was stimulated, the decrease in cellular volume was prevented andE Na did not fall. Replacement of serosal isosmotic Cl medium by a hypo-osmotic gluconate medium (one-half normal) also prevented cell shrinkage and did not result in inhibition of Na+ transport. Thus the inhibition of Na+ transport can be correlated with changes in cell volume rather than with the change in Cl per se. Nystatin virtually abolished the resistance of the apical plasma membrane as judged by measurement of tissue capacitance. With K+ gluconate mucosa, Na+ gluconate serosa, calculated basolateral membrane resistance was much greater, estimated basolateral emf was much lower, and the Na+/K+ basolateral permeability ratio was much higher than with acetate media. It is concluded the decrease in cellular volume associated with substitution of serosal gluconate for Cl results in a loss of highly specific Ba2+-sensitive K+ conductance channels from the basolateral plasma membrane. It is possible that the number of Na+ pump sites in this membrane is also decreased.  相似文献

Summary Exposing the apical membrane of toad urinary bladder to the ionophore nystatin lowers its resistance to less than 100 cm2. The basolateral membrane can then be studied by means of transepithelial measurements. If the mucosal solution contains more than 5mm Na+, and serosal Na+ is substituted by K+, Cs+, or N-methyl-d-glucamine, the basolateral membrane expresses what appears to be a large Na+ conductance, passing strong currents out of the cell. This pathway is insensitive to ouabain or vanadate and does not require serosal or mucosal Ca2+. In Cl-free SO 4 2– Ringer's solution it is the major conductive pathway in the basolateral membrane even though the serosal side has 60mm K+. This pathway can be blocked by serosal amiloride (K i=13.1 m) or serosal Na+ ions (K i 10 to 20mm). It also conducts Li+ and shows a voltage-dependent relaxation with characteristic rates of 10 to 20 rad sec–1 at 0 mV.  相似文献

Summary The specific activity of the Na+/K+/Cl cotransporter was assayed by measuring the initial rates of furosemide-inhibitable86Rb+ influx and efflux. The presence of all three ions in the external medium was essential for cotransport activity. In cultured smooth muscle cells furosemide and bumetanide inhibited influx by 50% at 5 and 0.2 m, respectively. The dependence of furosemide-inhibitable86Rb+ influx on external Na+ and K+ was hyperbolic with apparentK m values of 46 and 4mm, respectively. The dependence on Cl was sigmoidal. Assuming a stoichiometry of 112 for Na+/K+/Cl, aK m of 78mm was obtained for Cl. In quiescent smooth muscle cells cotransport activity was approximately equal to Na+ pump activity with each pathway accounting for 30% of total86Rb+ influx. Growing muscle cells had approximately 3 times higher cotransport activity than quiescent ones. Na+ pump activity was not significantly different in the gorwing and quiescent cultures. Angiotensin II (ANG) stimulated cotransport activity as did two calcium-transporting ionophores, A23187 and ionomycin. The removal of external Ca2+ prevented A23187, but not ANG, from stimulating the cotransporter. Calmodulin antagonists selectively inhibited86Rb+ influx via the cotransporter. Beta-adrenoreceptor stimulation with isoproterenol, like other treatments which increase cAMP, inhibited cotransport activity. Cultured porcine endothelial cells had 3 times higher cotransport activity than growing muscle cells. Calmodulin antagonists inhibited cotransport activity, but agents which increase cAMP or calcium had no effect on cotransport activity in the endothelial cells.  相似文献

AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). Cl conductance in cultured embryonic chick cardiac myocytes was characterized using whole-cell patch clamp techniques. Following elimination of cation currents in Na+and K+-free internal and external solutions, the basal whole-cell current was predominantly a Cl current. Cl-sensitive current (I Cl) was defined as the difference between the whole-cell currents recorded in normal and low [Cl] o when measured in the same cell. The whole-cell current in the absence or presence of 10 m cAMP was time independent, displayed outward rectification with the pipette [Cl] < 40 mm, and was not saturated with a physiological Cl gradient. The Cl current was also activated by 1 m forskolin and inhibited by 0.3 mm anthracene-9-carboxylic acid (9-AC). Forskolin was less effective than cAMP (internal dialysis) in activating the Cl current. The cAMP- or forskolin-activated and basal Cl current were reasonably fit by the Goldman-Hodgkin-Katz equation. The calculated P Cl in the presence of cAMP was increased by fiveto sixfold over the basal level. In the presence of 5 mm EGTA to decrease free [Ca2+] i , the whole-cell current could not be stimulated by cAMP, forskolin or IBMX (0.1 mm). These data suggest that cultured chick cardiac myocytes have a low basal Cl conductance, which, as in some mammalian cardiac ventricular myocytes, can be activated by cAMP. Internal chloride activity, ai Cl, and membrane potential, Em, were measured simultaneously in 120 R2 giant neurons of Aplysia californica. ai Cl was 37.0 ± 0.8 mM, Em was -49.3 ± 0.4 mv, and E Cl calculated using the Nernst equation was -56.2 ± 0.5 mv. Such values were maintained for as long as 6 hr of continuous recording in untreated neurons. Cooling to 1°–4°C caused ai Cl to increase at such a rate that 30–80 min after cooling began, E Cl equalled Em. The two then remained equal for as long as 6 hr. Rewarming to 20°C caused ai Cl to decline, and E Cl became more negative than Em once again. Exposure to 100 mM K+-artificial seawater caused a rapid increase of ai Cl. Upon return to control seawater, ai Cl declined despite an unfavorable electrochemical gradient and returned to its control values. Therefore, we conclude that chloride is actively transported out of this neuron. The effects of ouabain and 2,4-dinitrophenol were consistent with a partial inhibitory effect. Chloride permeability calculated from net chloride flux using the constant field equation ranged from 4.0 to 36 x 10-8 cm/sec.  相似文献

Summary Electrophysiologic and tracer experiments have shown that Cl entersNecturus proximal tubule cells from the tubule lumen by a process coupled to the flow of Na+, and that Cl entry is electrically silent. The mechanism of Cl exit from the cell across the basolateral membrane has not been directly studied. To evaluate the importance of the movement of Cl ions across the basolateral membrane, the relative conductance of Cl to K+ was determined by a new method. Single-barrel ion-selective microelectrodes were used to measure intracellular Cl and K+ as a function of basolateral membrane PD as it varied normally from tubule to tubule. Basolateral membrane Cl conductance was about 10% of K+ conductance by this method. A second approach was to voltage clamp the basolateral PD to 20 mV above and below the spontaneous PD, while sensing intracellular Cl activity with the second barrel of a double-barrel microelectrode. An axial wire electrode in the tubule lumen was used to pass current across the tubular wall and thereby vary the basolateral membrane PD. Cell Cl activity was virtually unaffected by the PD changes. We conclude that Cl leavesNecturus proximal tubule cells by a neutral mechanism, possibly coupled to the efflux of Na+ or K+.  相似文献

Summary Anion exchange transport in the mouse lacrimal gland acinar cell membrane was studied by measuring the intracellular H+ (pHi) and Cl (aCli) activities with double-barreled ion-selective microelectrodes. In a HCO 3 -free solution of pH 7.4 (HEPES/Tris buffered), pHi was 7.25 andaCli was 33mm. By an exposure to a HCO 3 (25mm HCO 3 /5% CO2, pH 7.4) solution for 15 min,aCli was decreased to 25mm and pHi was transiently decreased to about 7.05 within 1 min, then slowly relaxed to 7.18 in 15 min. Intracellular HCO 3 concentration [HCO 3 ]i, calculated by the Henderson-Hasselbalch's equation, was 11mm at 1 min after the exposure and then slowly increased to 15mm. Readmission of the HCO 3 -free solution reversed the changes inaCli and pHi. The intracellular buffering power was about 40mm/pH. An addition of DIDS (0.2mm) significantly inhibited the rates of change inaCli, pHi, and [HCO 3 ]i caused by admission/withdrawal of the HCO 3 , solution and decreased the buffer value. Replacement of all Cl with gluconate in the HCO 3 solution increased pHi, and readmission of Cl decreased pHi. The rates of these changes in pHi were reduced by DIDS by 32–45% but not by amiloride (0.3mm). In the HCO 3 solution, a stimulation of intracellular HCO 3 production by exposing the tissue to 25mm NH 4 + increasedaCli significantly. While in the HCO 3 -free solution or in the HCO 3 , solution containing DIDS, exposure to NH 4 + had little effect onaCli. All of these findings were consistent with the presence of a reversible, disulfonic stilbene-sensitive Cl/HCO 3 exchanger in the basolateral membrane of the acinar cells. The possibility of anion antiport different from one-for-one Cl/HCO 3 exchange is discussed.  相似文献
