A gas chromatographic—mass spectrometric method for the determination of cobalt in biological materials employing stable enriched 62Ni as an internal standard and using lithium bis(trifluoroethyl)dithiocarbamate as a chelating agent is described. The method involves the addition of a known amount (1 μg) of 62Ni to the sample, the formation of the chelate and the determination by selected-ion monitoring of the m/z ratio, which corresponds to . No appreciable memory effect was observed, and an acceptable dynamic range of 100 was found. There was good agreement between the cobalt concentration values determined by gas chromatography—mass spectrometry and electrothermal atomic absorption spectrometry. The present method has high sensitivity and can be used for the quantitation of cobalt at concentrations as low as 1 μg/l. The use of enriched 62Ni circumvents the problem caused by endogenous nickel and simultaneously provides data on the nickel concentration in the biological sample without any additional experimental effort.  相似文献

Cobalt determinations in biological fluids are of great interest in biological or toxicological research programs. Cobalturia is often chosen as an indicator for a biological monitoring program in occupational exposure to cobalt dusts. The method described here derives from the IUPAC reference method for nickel determination. It enables cobaltemia and cobalturia to be measured in small samples (1 mL). The mean usual values for cobalt in biological fluids are very low (2.7 nmol L−1 for serum and 6.7 nmol L−1 for urine), and therefore, thus require an analytical procedure with preconcentration and extraction. The sample is mineralized by wet acid digestion. After digestion, inorganic cobalt is extracted in form of ammonium pyrrolidine dithiocarbamate complex into isobutyl methyl ketone and measured in the organic layer by electrothermal atomic absorption spectrometry. The analytical parameters are described in detail. The extraction output is about 99%. The detection limits are 1.93 and 1.89 nmol L−1 for serum and urine, respectively. Sensitivity (expressed as the concentration that gives a 0.044 absorbance) is 3.4 nmol L−1 for serum and 3.3 nmol L−1 for urine. Within-run precision ranged between 3.9 and 2.5% (coefficients of variation) for serum and 4.2 and 1.1% for urine, at 87 and 136 nmol L−1 levels, respectively. Between-run precision ranged between 4.3 and 3.3% (coefficients of variation) for serum and 4.2 and 2.3% for urine, at 87 and 136 nmol L−1 levels, respectively. At very low concentration, 5.7 nmol L−1 for serum and 2.5 nmol L−1 for urine, the between-run precision is, respectively, 19.5 and 28%. Linearity is effective between 0 and 272 nmol L−1. Interferences and matrix effects are negligible for urine, serum, or plasma samples without hemoglobin. The method is easily applicable for routine determinations.  相似文献

A sediment sample with high organic matter and trace metal content was suspended in synthetic river water for four weeks under an inert gas atmosphere. Subsequently, the anaerobic suspension was reoxidized by bubbling air through it. The concentrations of dissolved oxygen, sulfide, ferrous iron, manganese, cadmium, cobalt and the pH-value were measured at close time intervals during the anaerobic incubation. The anaerobic suspension was a post-oxic or sub-oxic environment with oxygen and total sulfide concentrations less than 1 µmole 1–1. Concentrations of dissolved ferrous iron and manganese were 50–150 µmole 1–1 and 5–30 µmole 1–1, respectively. The total sulfide concentration was measured using a sensitive voltammetric technique, with a detection limit of 1 nmole 1–1. A sequential extraction procedure was applied to two sediment samples taken at the end of the anaerobic incubation and after one week of reoxidation. The extractions indicated that cadmium was bound in sulfide minerals under post-oxic conditions. Thermodynamic equilibrium calculations revealed that the concentrations of dissolved cobalt in the post-oxic suspension were limited by the precipitation of cobalt sulfide minerals.  相似文献

Cryptococcus adeliensis was initially described as a psycrophilic species containing a single strain CBS 8351T isolated from decayed algae in Terre Adelie (Antartida). Later, a second strain of this species was isolated from an immunosuppressed patient affected by leukaemia in Germany and recently several strains from this species have been found in human patients and pigeon droppings of the same country. In this study, we isolated from sheep droppings in Spain a xylanolytic strain named LEVX01 that was phenotypically related to the strain CBS 8351T and showed a 100% similarity in the D1/D2 domain and 5.8S-ITS region sequences with respect to the remaining described strains of C. adeliensis. These findings suggest that this species has a wide geographical distribution and that the animal faeces are a common habitat for C. adeliensis. The chemotaxonomic analyses showed the absence of detectable amounts of xylose in the cell walls of the strains LEVX01 and CBS8351T in contrast to other Cryptococcus species. Interestingly, the ultrastructural study showed the presence of fimbriae in these two strains that could be involved in the attachment to the host cells and, as occurs in Candida albicans, they could also be a pathogenicity factor for the man.  相似文献

A flow injection chemiluminescent method is described for the determination of choline. The method is based on the production of hydrogen peroxide from choline using on-line covalently bound immobilized choline oxidase column. The product is mixed downstream and detected via the cobalt catalyzed chemiluminescent oxidation of luminol. The detection limit is 1×10−7 mol/L, with rsd 1.8 to 2.8% in the range 2–10×10−5 mol/L. The sample throughput is 30 per hour. The method was applied to the determination of choline produced off-line from phosphatidylcholine using phospholipase-D isolated from cabbage. © 1997 John Wiley & Sons, Ltd.  相似文献

A ?ow injection method is reported for the determination of sulphite‐based on chemiluminescent detection. Hydro‐gen peroxide is produced from sulphite using on‐line covalently bound immobilized sulphite oxidase packed in a mini‐column, which was mixed downstream and detected via cobalt(II)‐catalysed chemiluminescent oxidation of luminol. The limit of detection (2 × standard deviation of the blank) was 1 × 10?3 mmol/L with sample throughput 60 h?1. The kinetics of formation of the complex ion, μ-carbonato-di-μ-hydroxo-bis((1,5-diamino-3-aza-pentane) cobalt(III), from the tri-μ-hydroxo-bis((1,5-diamino-3-aza-pentane(III)cobalt(III)) ion in aqueous buffered carbonate solution have been studied spectrophotometrically at 295 nm over the ranges 20.0θ°C34.8, 8.03pH9.44, 5 mM [CO32−35 mM and at an ionic strength of 0.1 M (LiClO4). On the basis of the kinetic results a mechanism, involving rapid cleavage of an hydroxo bridge followed by carbon dioxide uptake with subsequent bridge formation, has been proposed. At 25 °C, the rate of the carbon dioxide uptake is 0.58 M−1 s−1 with ΔH≠ = (13.2±0.7) kcal mol−1 and ΔS≠ = (−15.1 ± 0.7) cal deg−1 mol−1. The results are composed with those obtained for several mononuclear cobalt(III) and one dinuclear cobalt(III) complexes.  相似文献

The reversed-phase HPLC separation of fluorescent o-phthalaldehyde (OPA) derivatives has been applied to the assay of hepatic γ-glutamylcysteine and glutathione (GSH) levels and the enzymes producing these peptides. The method has been compared to the assay using monobromobimane (MB) as the derivatizing agent. The OPA method has the advantage of faster derivitization, the lack of need to adjust the pH, isocratic separation and selectivity for GSH and γ-glutamylcysteine. The MB method requires pH adjustment following derivatization and gradient elution chromatography. MB is also non-selective, yielding fluorescent derivatives of all biological thiols and more interfering peaks on the chromatogram. MB-based analyses are also approximately sixty times more expensive per sample. MB yields fluorescent degradation products on exposure to light. OPA adducts are stable for up to ten days when stored at −20°C. OPA detection is sensitive to 12.5 pmol in the sample, at a signal-to-noise ratio of 2.5. The two methods correlate well. Hepatic γ-glutamylcysteine synthetase in the same liver preparation was found to be 4.85±0.47 nmol min−1 mg−1 protein by the OPA method and 4.42±0.52 nmol min−1 mg−1 protein by the MB method. GSH concentrations were found to be 90.4±6.5 nmol/mg protein for the OPA method and 92.5±3.4 for the MB method.  相似文献

We tested the impacts of most common sample preservation methods used for aquatic sample materials on the stable isotope ratios of carbon and nitrogen in clams, a typical baseline indicator organism for many aquatic food web studies utilising stable isotope analysis (SIA). In addition to common chemical preservatives ethanol and formalin, we also assessed the potential impacts of freezing on δ13C and δ15N values and compared the preserved samples against freshly dried and analysed samples. All preservation methods, including freezing, had significant impacts on δ13C and δ15N values and the effects in general were greater on the carbon isotope values (1.3–2.2‰ difference) than on the nitrogen isotope values (0.9–1.0‰ difference). However, the impacts produced by the preservation were rather consistent within each method during the whole 1 year experiment allowing these to be accounted for, if clams are intended for use in retrospective stable isotope studies.  相似文献

Abstract

A simple, rapid and inexpensive method of the solidified floating organic drop extraction (SFODME) technique coupled with graphite furnace atomic absorption spectrometry (GFAAS) has been developed for the determination of cobalt in water samples. 8-Hydroxyquinoline was used as a complex agent and 1-undecanol was used as the extraction solvent. The factors, including solvent types, solution pH, extractant volume and interfering ions, were investigated and optimised. Under the optimum conditions, the calibration graphs were linear in the range 0.05–10.0 ng mL-1 cobalt, the limit of detection was 0.02 ng mL-1, the limit of quantification was 0.05 ng mL-1 and the relative standard deviation for 10 replicate measurements of 3 ng mL-1 cobalt was 2.8%. The proposed method was successfully applied for the determination of cobalt in different water samples and the results were satisfactory.  相似文献

The marine dinoflagellate Protoceratium reticulatum has been recently identified as a source for the disulfated polyether toxin, yessotoxin (YTX), and may pose a risk to human health, aquaculture development and coastal environments. The requirements of P. reticulatum for selenium, iron and cobalt were assessed in culture. P. reticulatum was grown in nutrient enriched seawater (1/10 GP medium) without selenium or with 0.003 and 0.0003 μM selenium added; without iron or with 0.076 and 0.0076 μM iron added; and without cobalt or with 0.008 μM cobalt added. Test flasks were monitored for growth rate, cell yield and YTX production. P. reticulatum was found to exhibit a strong requirement for both selenium and iron. Growth rate and cell yield in treatments without added selenium were significantly (P<0.05) reduced to 60.2% (μ=0.15 day−1) and 20.2% (4942 cell ml−1), respectively, of those with selenium added (μ=0.23 day−1 and 24, 387 cell ml−1). YTX production was significantly increased by addition of selenium in two of three transfers tested. Cells of P. reticulatum subjected to medium without selenium added showed morphological changes observable at the light microscope level which included enlarged cell size. The diameter of cells in medium without selenium added were significantly (P<0.05) enlarged to 36.7±0.90 μm compared to cells in the medium with selenium added, 27.5±1.25 μm. Growth rate and cell yield in treatments without added iron were also significantly reduced to 70.1% (μ=0.16 day−1) and 34.2% (8003 cells ml−1), respectively, of those with iron added (μ=0.23 day−1 and 23,416 cells ml−1). No significant effect on YTX production was measured. In contrast to selenium and iron, no limitation of growth or cell yield or differences in YTX production were observed for flasks without cobalt as compared to those with cobalt added. The possibility that harmful algal events of P. reticulatum may be influenced by selenium or iron in neritic waters is discussed.  相似文献

Two dinuclear cobalt/copper compounds have been isolated from the reaction between N,N′-ethylenebis(salicylideniminato)cobalt(II), [Co(salen)], and copper(II) chloride in different conditions. The first one is a dinuclear cobalt(III)/copper(II) derivative, [Co(salen)Cl2Cu(EtOH)2Cl], 1, that have the cobalt atom six-coordinated to the four donor atoms of the salen ligand and to two chlorine atoms in a slightly distorted octahedral environment and the copper atom five-coordinated to the two bridging oxygen atoms of the salen ligand, two ethanol molecules and one extra chlorine atom. This compound is the only reported example of a cobalt/copper derivative with the cobalt maintaining the salen coordinative site, since the usual reaction takes place by a transmetalation process. This reaction is observed in the second derivative, [Cu(salen)CoCl2], 2, where the copper atom displaces the cobalt from the salen cavity. The copper atom adopts a square-planar coordinative environment, while the cobalt is tetrahedrically coordinated to the two bridging oxygen and two chlorine atoms. Both compounds present several intermolecular contacts that increase the dimensionality in the crystal and some of which can transmit magnetic interactions. The magnetic properties confirm the structural picture, with isolated copper(II) centres in 1, where the cobalt(III) is in the low spin form, and with antiferromagnetically coupled S = 1/2 and S = 3/2 centres in 2.  相似文献

Summary The localization of carbonic anhydrase in the rat lung has been demonstrated, at light and electron microscopic levels, by the cobalt bicarbonate histochemical method of Hansson. Focal deposits of the cobalt sulfide reaction product were found not only in the capillary endothelium of the alveolar walls, but also in the small and large alveolar cells. The histochemical reaction was abolished by two potent inhibitors, acetazolamide (10–5 to 10–6 M) and KCNO (5×10–3 to 10×10–3 M). Physiological assay with Maren's method indicated that values for carbonic anhydrase activity in rat lung are 4.4±0.8 UA/mg of protein, 25.0±5.5 UA/mg of nitrogen, and 369±86 UA/g of wet weight. In addition, it was calculated that after fixation in glutaraldehyde-formaldehyde-picric acid about 9% activity is retained.  相似文献

Cobalt is one of the possible contaminants originating from radioactive wastes or from metal mines and refineries. This paper describes sorption of cobalt by the foliose lichen Hypogymnia physodes from CoCl2 solutions spiked with 60Co2+ in laboratory experiments. Maximum uptake was reached within 1 hour; the biosorption after 24 hours is not pH-dependent within the range of pH 4–7, negligible at pH 2 and is not dependent on metabolic activity. The process can be described by the Freundlich adsorption isotherm with ln k = 2.77, 1/n = 0.22 and R 2 = 0.94. Bivalent metal ions showed a concentration-dependent competitive effect on cobalt biosorption, decreasing in the order: Cu > Ni > Ca > Mg. Monovalent ions, such as K+ and Na+, showed only very weak competitive effect. Up to 98% of Co taken up by lichen can be removed by washing with 0.1 M NiCl2 at 20°C. This means that only a small fraction of the cobalt is localized intracellularly. These results can be used for elucidating the behaviour of lichens as bioindicators of cobalt pollution in water systems, including the risk of cobalt leakage from lichen probes under the influence of rain, snow and atmospheric humidity.  相似文献

The physical properties of hydrated multilamellar sample of 1,2-dimyristamido-1,2-deoxyphosphatidylcholine (DDPC) were investigated by means of differential scanning calorimetry (DSC), static X-ray diffraction, and simultaneous DSC and X-ray diffraction. The DDPC is a synthetic sphingomyelin analogue and has two amide bonds in its hydrophobic parts. This paper reports on metastable phase behavior of the hydrated DDPC sample. By cooling from a chain-melted state at the rates of greater than 4 °C min−1, hydrated DDPC bilayers form a metastable gel phase. In the gel phase, the hydrophobic chains are tilted with respect to the bilayer normal, as like the gel phase of glycero-phosphatidylcholines. By heating, the metastable gel phase is transformed in to a stable phase associated with an exothermic heat event at 18.3 °C (ΔH = 14.6 kJ mol−1) and then the stable phase is transformed into a liquid-crystalline phase at 25.6 °C (ΔH = 42 kJ mol−1). The incubation at 17 °C for more than 1 h also induces the formation of the stable phase. In the stable phase, the hydrophobic chains are packed into highly ordered crystal-like structure. However, the X-ray diffraction pattern of the stable phase suggested that the entire DDPC molecules do not form a two-dimensional molecular ordered lattice, differing from normal subgel phase of glycero-phosphatidylcholines. The structure and phase behavior of DDPC revealed by the present study are discussed from the viewpoint of hydrogen bonds.  相似文献

The hyperaccumulator Pycnandra acuminata is a New Caledonian rainforest tree known to have the highest concentration of nickel in any living organism, with 25 wt% nickel in its latex. All trees (with a diameter of >10 cm) and soil profiles in a 0.25-hectare permanent plot were sampled to assess the biogeochemical compartmentalisation of nickel in a dense stand of P. acuminata trees. Nickel stable isotope analysis permitted insights into the cycling of nickel in this ecosystem. The total tree biomass of the plot was calculated to be 281 tonnes ha−1, which contained 0.44 kg of cobalt, 49.1 kg of manganese, 257 kg of nickel and 6.76 kg of zinc. Nickel stable isotope analysis identified the biotic origin of the nickel in the soil upper layers, with P. acuminata shoots enriched in lighter nickel isotopes. The δ60Ni latex signature suggests that long-distance transport, radial xylem and phloem loading are at play in P. acuminata.  相似文献

Cobalt is an important element with magnetic properties used in various industrial applications, but is also needed for biological activity. Very little is known about the cellular response of living systems to cobalt stress. Towards investigating this mechanism, we isolated individual Saccharomyces cerevisiae cells resistant to high cobalt concentrations up to 8 mmol l−1, by employing four different ‘in vivo’ evolutionary engineering strategies: selection under constant or gradually increasing stress levels, and selection under continuous or pulse exposure to cobalt stress. Selection under continuous exposure to gradually increasing cobalt stress levels yielded the most resistant cell population to cobalt. However, the resistance was highly heterogeneous within the mutant populations ranging from 3- to 3700-fold survival rate of isolated individuals to 8 mmol l−1 CoCl2 in the most resistant population. Moreover, cobalt-resistant individual colonies were associated with 2–4-times lower intracellular cobalt contents as compared to wild-type, and with cross-resistance to metals such as nickel, zinc, manganese, but not to copper and chromium ions. Contrary to mutants evolved under continuous exposure to cobalt, those isolated by pulse exposure strategy also exhibited resistance to heat shock and hydrogen peroxide stress. Taken together, this study reinforced the fact that evolutionary engineering is useful in selecting strains with very specific phenotypes, and further illustrated the importance of the strategy chosen to isolate the best evolved strain.  相似文献

The α-amylase of Streptomyces sp. IMD 2679 was subject to catabolite repression. Four different growth rates were achieved when the organism was grown at 40 °C and 55 °C in the presence and absence of cobalt, with an inverse relationship between α-amylase production and growth rate. Highest α-amylase yields (520 units/ml) were obtained at the lowest growth rate (0.062 h−1), at 40 °C in the absence of cobalt, while at the highest growth rate (0.35 h−1), at 55 °C in the presence of cobalt, α-amylase production was decreased to 150 units/ml. As growth rate increased, the rate of specific utilisation of the carbon source maltose also increased, from 46 to 123 μg maltose (mg biomass)−1 h−1. The pattern and levels of α-glucosidase (the enzyme degrading maltose) detected intracellularly in each case, indicate that growth rate effectively controls the rate of feeding of glucose to the cell, and thus catabolite repression. The triribbed-functionalized cobalt(II,III) and ruthenium(II) clathrochelate derivatives of the vic-dioximes with two nitrogen or sulfur atoms in α-positions to π-conjugated diazomethine chelate fragments of a macrobicyclic framework were obtained in moderate yields under mild and high dilution conditions by nucleophilic substitution of six reactive chlorine atoms of the boron-capped macrobicyclic cobalt and ruthenium(II) precursors with N2- and S2-dinucleophiles (ethylenediamine and the corresponding α-dithiols in the presence of triethylamine, respectively). The complexes obtained were characterized using elemental analysis, MALDI-TOF mass spectrometry, IR, UV-Vis, 1H and 13C{1H} NMR and EPR spectroscopies, magnetochemistry and X-ray crystallography. The MN6-coordination polyhedra of all the X-ray studied clathrochelates possess a slightly distorted trigonal prismatic geometry. The encapsulated cobalt(II) ions are shifted from the centers of the cavities formed by the macrobicyclic ligand due to the Jahn-Teller distortion, while the ruthenium and iron(II) ions in their clathrochelate analogs do occupy these centers. The main geometrical parameters of the macrobicyclic frameworks vary with Shannon radius of an encapsulated metal ion. In the case of the tris-ethylenediamine cobalt(III) clathrochelate, the field strength of the macrobicyclic amine ligand is essentially lower than those for their aromatic and aliphatic analogs because of the negative σpara-effect of the ribbed alkylamine substituents. The magnetometry and EPR data confirmed the low-spin character of the cobalt(II) complexes synthesized. The electrochemically generated oxidized cobalt clathrochelates are stable in the CVA time scale, whereas their ruthenium- and iron-containing analogs as well as the reduced forms of all the cobalt, ruthenium and iron complexes obtained are unstable.  相似文献

The mononuclear cationic cobalt(III) complex 1 [Co(L)2]+ [where HL is H2N(CH2)2NC(Ph)C6H2(Me)(OH)COPh] has been obtained by the condensation of 4-methyl-2,6-dibenzoylphenol and ethane-1,2-diamine in the presence of CoCl2 · 6H2O and isolated as the chloride solvate {1 · Cl · 2H2O · EtOH}. The water and ethanol do not form part of the cobalt(III) pseudo-octahedral [Co(L)2]+ coordination sphere, which is cis-CoN4O2 with the NNO-ligands mer. Compound 1 has been characterised by mass spectrometry, IR, electronic, 1H and 13C NMR spectroscopy, conductivity measurements, elemental analysis, TGA, cyclic voltammetry and an X-ray structural determination. The average Co-N and Co-O distances are, respectively, 1.929(4) and 1.882(3) Å. The N?N intraligand bite distance is 2.73(4) Å and the N-Co-N and O-Co-O angles are, respectively, 86.2(15)° and 93.3(13)°. Cyclic voltammetric studies of complex 1 indicate an irreversible cathodic peak (Epc, ca. −0.61 V) corresponding to the reduction of cobalt(III) to cobalt(II). Absorption titration experiments gave a binding constant for DNA interaction of 1.4 × 104 M−1 and a binding site size 0.16 base pairs.  相似文献
