Alterations of mitochondrial-encoded subunits of the FoF1-ATPsynthase are frequently associated with cytoplasmic male sterility(CMS) in plants; however, little is known about the relationshipof the nuclear encoded subunits of this enzyme with CMS. Inthe present study, the full cDNA of the gene TaFAd that encodesthe putative FAd subunit of the FoF1-ATP synthase was isolatedfrom the wheat (Triticum aestivum) fertility restorer ‘2114’for timopheevii cytoplasm-based CMS. The deduced 238 amino acidpolypeptide is highly similar to its counterparts in dicotsand other monocots but has low homology to its mammalian equivalents.TaFAd is a single copy gene in wheat and maps to the short armof the group 6 chromosomes. Transient expression of the TaFAd–GFPfusion in onion epidermal cells demonstrated TaFAd's mitochondriallocation. TaFAd was expressed abundantly in stem, leaf, anther,and ovary tissues of 2114. Nevertheless, its expression wasrepressed in anthers of CMS plants with timopheevii cytoplasm.Genic male sterility did not affect its expression in anthers.The expression of the nuclear gene encoding the 20 kDa subunitof Fo was down-regulated in a manner similar to TaFAd in theT-CMS anthers while that of genes encoding the 6 kDa subunitof Fo and the subunit of F1 was unaffected. These observationsimplied that TaFAd is under mitochondrial retrograde regulationin the anthers of CMS plants with timopheevii cytoplasm. 以光核桃(Prunus mira)幼苗为材料, 通过控制根系温度研究了根系温度变化与叶片脱落酸(ABA)的关系及其对光合机构热稳定性的影响。结果表明: 1)环境高温(37和40 ℃)胁迫下保持根系温度适宜((25±2) ℃)时, 幼苗叶片相对含水量(Relative water content, RWC)下降较少, 但叶片ABA含量低, 超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)、过氧化物酶(POD)和过氧化氢酶(CAT)活性低, 过氧化氢(H2O2)含量和膜质过氧化水平(丙二醛(Malondialdehyde, MDA)浓度)提高, 最大光化学效率(Fv/Fm)下降程度较大; 2)而同等环境高温(37和40 ℃)条件下根系温度逐步升高时, 幼苗叶片RWC降低, 叶片ABA含量增加, SOD、APX、POD、CAT活性高, H2O2含量高, MDA生成量低, Fv/Fm降低程度较小。与37 ℃相比, 40 ℃处理条件下各生理指标变化趋势相似, 但差异加大。因此认为: 高温胁迫条件下, 根系温度适宜时RWC高, 但导致光合机构伤害较重; 根系感受高温胁迫能够增加叶片ABA含量, 有助于保护光合机构、提高光合机构的抗热性。  相似文献

To evaluate the variations of CO2 and CH4 emissions from FACE (free-air CO2 enrichment, F) soils three years after rice-wheat rotation FACE treatment, incubation experiments in the laboratory with laboratory and elevated CO2 concentration (1000 μl L?1) were carried out under flooded conditions at 25°C. Results show that soil organic carbon is increased by 11% after exposure to FACE treatment for three years. The results indicate that at laboratory and elevated CO2, the cumulative CO2 emissions from FACE soils are 35% and 22% higher than those from the ambient soils, whereas the cumulative CH4 emissions from FACE soils are 2.6 and 2.3 times that of ambient soils. Thus, there is a larger ratio of cumulative emissions of CH4 to CO2 in the soil F. The elevated CO2 concentration during the incubation stimulates the cumulative CO2 emission significantly, but its stimulation on CH4 emission is not statistically significant. The results indicate that the elevated atmospheric CO2 concentration stimulates the turnover rates of soil organic matter, with a net increase in soil organic matter content, and alters the CH4/CO2 ratio.  相似文献

三江平原沼泽湿地受到大气沉降、地表径流、农业排水等外源氮素的输入,对湿地生态系统CH4和N2O通量有重要影响。采用野外原位施肥试验模拟外源氮输入,设0,60,120,240kgN•hm-2 4种试验处理,探讨外源氮对沼泽湿地CH4和N2O通量的影响。结果表明,外源氮促进了CH4和N2O排放。与对照处理比较,各施氮水平CH4平均排放通量分别增加了181%,254%和155%,N2O排放通量分别增加了21%,100%和533%。外源氮输入对CH4排放的季节变化形式影响不大,而N2O的季节变化形式随着氮输入表现出波动变化的趋势。不同施氮水平对CH4排放的促进作用与植物生长阶段和产CH4的微生物过程密切相关,N2O排放通量随氮输入量呈指数增加(R2=0.97,p<0.01)。外源氮通过影响湿地微生物过程来进一步影响CH4和N2O的排放。  相似文献

对低浓度Na2CO3胁迫下星星草幼苗相对电导率、O-2产生速率、H2O2含量以及保护酶CAT、SOD和POD活性的研究结果表明,低盐胁迫1 d后,星星草幼苗细胞膜的通透性、O-2产生速率、H2O2含量及保护酶活性都随着盐胁迫的加剧而升高,其具体的变化规律与盐胁迫强度和幼苗细胞膜的受损伤程度密切相关,但相关关系的性质上具有差异。  相似文献

在150 mmol·L-1 NaHCO3胁迫下,研究了不同浓度硝酸镧对黑麦草幼苗生长的影响及其对叶片光合生理响应的调节作用。结果表明,叶面喷施低浓度硝酸镧(0.05 mmol·L-1)预处理显著减小了碱胁迫下黑麦草幼苗生物量、叶片叶绿素和类胡萝卜素含量、叶绿体希尔反应、光合电子传递、光合磷酸化、Mg2+ ATP酶、RuBP羧化酶活性和光合效率的下降幅度,却对光呼吸调节酶乙醇酸氧化酶活性无明显影响。而高浓度硝酸镧(0.5 mmol·L-1)预处理不仅对碱胁迫没有缓解作用,反而加重了碱胁迫伤害。因此,适宜浓度硝酸镧可以通过改善光合功能从而缓解碱胁迫对黑麦草幼苗生长的抑制作用。  相似文献

The distribution of selection coefficients of new mutations is of key interest in population genetics. In this paper we explore how codon-based likelihood models can be used to estimate the distribution of selection coefficients of new amino acid replacement mutations from phylogenetic data. To obtain such estimates we assume that all mutations at the same site have the same selection coefficient. We first estimate the distribution of selection coefficients from two large viral data sets under the assumption that the viral population size is the same along all lineages of the phylogeny and that the selection coefficients vary among sites. We then implement several new models in which the lineages of the phylogeny may have different population sizes. We apply the new models to a data set consisting of the coding regions from eight primate mitochondrial genomes. The results suggest that there might be little power to determine the exact shape of the distribution of selection coefficient but that the normal and gamma distributions fit the data significantly better than the exponential distribution.  相似文献

人细胞质硫氧还蛋白(hTrx1)在抗氧化和氧化还原调控中起重要作用.如果静脉注射重组hTrx1,动物抗氧化能力将增高.近年来,随着人们对氧化还原调控的关注,hTrx1需求不断增加.为了快速获得高纯度重组hTrx1,N末端亲和标签,如组氨酸标签(His-tag)和谷胱甘肽S-转移酶标签(GST-tag),被用于hTrx1亲和纯化.带N末端标签的hTrx1融合蛋白在实验中用的越来越多.但N末端延长是否会影响hTrx1特性尚不清楚.我们构建与优化了hTrx1原核表达质粒,在大肠杆菌中高效表达了含天然N末端、带His-tag或带GST-tag的3种重组hTrx1.纯化蛋白在SDS-PAGE上呈现1条带,对应的分子量分别为12kD、17kD及38kD.在无氧化剂存在时,它们催化胰岛素还原的能力不分仲伯.当有H2O2存在时,天然N末端hTrx1通过形成可逆二聚体,对H2O2表现出较强的耐受性;而N末端亲和标签有干扰二聚体形成,使hTrx1对H2O2耐受性降低的作用,其中GST-tag干扰作用明显大于His-tag.此外,体内重要的氧化还原对GSH/GSSG,有增进hTrx1及其还原酶催化NADPH氧化的作用,N末端亲和标签可明显扩大GSH/GSSG的这种作用.我们分析了N末端亲和标签对hTrx1活性影响的可能机理.  相似文献

 高等植物大多为C3植物, C4植物和景天酸代谢(Crassulacean acid metabolism, CAM)植物是由C3植物进化而来的。C4途径的多源进化表明, 光合途径由C3途径向C4途径的转变相对简单。该文分析研究了植物光合途径的进化前景, 指出C4植物是从C3植物进化而来的高光效种类, 且地质时期以来降低的大气CO2浓度和升高的大气温度以及干旱和盐渍化是C4途径进化的外部动力。C3植物的C4途径的发现说明植物的光合途径并非是一成不变的, C3和C4植物的光合特征具有极大的可塑性, 某些环境的变化会引起植物光合途径在C3和C4途径之间转变。C3植物具有的C4途径是环境调控的产物, 是对逆境的适应性进化结果, 因而光合途径的转变也适用于干旱地区植被的适应性生存机理研究。该文还利用国外最新的C4光合进化模型介绍了植物在进化C4途径中所经历的7个重要时期(从分子基础到形态基础、结构基础, 再到物质代谢水平、光合酶活水平, 直到C3和C4途径协调运转时期, 最后达到形态与功能最优化阶段), 并结合全球气候变化的特点对国内外相关领域的研究进行了分析, 总结了植物光合途径的适应性转变和进化的研究成果, 为今后的相关工作提出建议。  相似文献

在全球动态植被模型的发展中, 受限于人力、物力和财力使得在物种水平上的研究变得既不可能也无必要。 植物功能群的划分是从生态学的, 而不是系统发育的角度来相互比较地对待不同地区的植物, 从而削减了植被变化研究中植物分类群的数量, 已成为研究植被变化及生物多样性对生态系统功能作用的重要单位。 植物的不同光合途径(C3、C4和CAM)从叶片组织结构到生理功能, 从生态适应到地理分布均表现出对不同水、热、光环境的响应, 是理想的植物功能群分类。 为此,分析了中国东北样带以羊草(Leymus chinensis)为建群种或共建种的草原群落植物光合类型功能群生物量及其与群落初级生产力和环境变化的关系。 结果表明:  (1)C4植物生物量具有明显的变化规律, 且对环境变化的响应显著, 其变异性较高, 更能反映样地间环境变化的差异; (2)C4与C3植物变化具有明显的互补性, 并且多数C4植物常在逆境中起到更大作用, 如干旱化、盐碱化和放牧干扰; (3)C4植物种类少,在所有调查样方中仅出现7种,占总出现种类的9.72%。这些特点说明C4植物可以考虑作为评估和预测我国温带草原植被及其生态系统变化的重要植物功能群。  相似文献

BACKGROUND AND AIMS: Stem and branch respiration, important components of total forest ecosystem respiration, were measured on Norway spruce (Picea abies) trees from May to October in four consecutive years in order (1) to evaluate the influence of temperature on woody tissue CO2 efflux with special focus on variation in Q10 (change in respiration rate resulting from a 10 degrees C increase in temperature) within and between seasons, and (2) to quantify the contribution of above-ground woody tissue (stem and branch) respiration to the carbon balance of the forest ecosystem. METHODS: Stem and branch CO2 efflux were measured, using an IRGA and a closed gas exchange system, 3-4 times per month on 22-year-old trees under natural conditions. Measurements of ecosystem CO2 fluxes were also determined during the whole experiment by using the eddy covariance system. Stem and branch temperatures were monitored at 10-min intervals during the whole experiment. KEY RESULTS: The temperature of the woody tissue of stems and branches explained up to 68% of their CO2 efflux. The mean annual Q10 values ranged from 2.20 to 2.32 for stems and from 2.03 to 2.25 for branches. The mean annual normalized respiration rate, R10, for stems and branches ranged from 1.71 to 2.12 micromol CO2 m(-2)s (-1) and from 0.24 to 0.31 micromol CO2 m(-2) s(-1), respectively. The annual contribution of stem and branch CO2 efflux to total ecosystem respiration were, respectively, 8.9 and 8.1% in 1999, 9.2 and 9.2% in 2000, 7.6 and 8.6% in 2001, and 8.6 and 7.9% in 2002. Standard deviation for both components ranged from 3 to 8% of the mean. CONCLUSIONS: Stem and branch CO2 efflux varied diurnally and seasonally, and were related to the temperature of the woody tissue and to growth. The proportion of CO2 efflux from stems and branches is a significant component of the total forest ecosystem respiration, approx. 8% over the 4 years, and predictive models must take their contribution into account.  相似文献

BACKGROUND AND AIMS: It is well documented that C(4) grasses have a shorter distance between longitudinal veins in the leaves than C(3) grasses. In grass leaves, however, veins with different structures and functions are differentiated: large longitudinal veins, small longitudinal veins and transverse veins. Thus, the densities of the three types of vein in leaves of C(3) and C(4) grasses were investigated from a two-dimensional perspective. METHODS: Vein densities in cleared leaves of 15 C(3) and 26 C(4) grasses representing different taxonomic groups and photosynthetic subtypes were analysed. KEY RESULTS: The C(4) grasses had denser transverse veins and denser small longitudinal veins than the C(3) grasses (1.9 and 2.1 times in interveinal distance), but there was no significant difference in large longitudinal veins. The total length of the three vein types per unit area in the C(4) grasses was 2.1 times that in the C(3) grasses. The ratio of transverse vein length to total vein length was 14.3 % in C(3) grasses and 9.9 % in C(4) grasses. The C(3) grasses generally had greater species variation in the vascular distances than the C(4) grasses. The bambusoid and panicoid C(3) grasses tended to have a denser vascular system than the festucoid C(3) grasses. There were no significant differences in the interveinal distances of the three vein types between C(4) subtypes, although the NADP-malic enzyme grasses tended to have a shorter distance between small longitudinal veins than the NAD-malic enzyme and phosphoenolpyruvate carboxykinase grasses. CONCLUSIONS: It seems that C(4) grasses have structurally a superior photosynthate translocation and water distribution system by developing denser networks of small longitudinal and transverse veins, while keeping a constant density of large longitudinal veins. The bambusoid and panicoid C(3) grasses have a vascular system that is more similar to that in C(4) grasses than to that in the festucoid C(3) grasses.  相似文献

The paper compares randomized rules of the biased-coin type for the sequential allocation of treatments in a clinical trial. An important characteristic is the loss, which measures the increase in the variance of parameter estimates due to the imbalance caused by randomization. Simulations are used to find the small-sample distribution of loss. For some rules a simple chi-squared approximation to the asymptotic distribution holds well down to very small sample sizes.  相似文献

采用单因素试验对甘草地上部分(茎叶)的超临界CO2提取工艺进行了研究。实验考察了压力、萃取时间、温度及CO2流量对甘草地上部分总黄酮提取率的影响,以总黄酮提取率和含量为指标,系统的研究了超临界二氧化碳萃取法提取甘草地上部分总黄酮的提取效果。得出的最佳工艺参数为:采用40~60目原料,80%乙醇为夹带剂,萃取时间:1.5 h;萃取压力:30.0 MPa;萃取温度:50℃;CO2流量:10 kg·h-1;分离压力: 5.8 MPa;分离温度: 40℃。实验结果表明超临界二氧化碳萃取甘草总黄酮的提取率2.09%,含量5.42%,工艺具有提取率高,纯度高的特点,为规模化生产甘草总黄酮的提取提供了研究基础。  相似文献

BACKGROUND AND AIMS: Numerous studies have examined the effects of climatic factors on the distribution of C(3) and C(4) grasses in various regions throughout the world, but the role of seasonal fluctuations in temperature, precipitation and soil N availability in regulating growth and competition of these two functional types is still not well understood. This report is about the effects of seasonality of soil N availability and competition on plant N dynamics and N-use strategies of one C(3) (Leymus chinensis) and one C(4) (Chloris virgata) grass species. METHODS: Leymus chinensis and C. virgata, two grass species native to the temperate steppe in northern China, were planted in a monoculture and a mixture under three different N seasonal availabilities: an average model (AM) with N evenly distributed over the growing season; a one-peak model (OM) with more N in summer than in spring and autumn; and a two-peak model (TM) with more N in spring and autumn than in summer. KEY RESULTS: The results showed that the altered N seasonality changed plant N concentration, with the highest value of L. chinensis under the OM treatment and C. virgata under the TM treatment, respectively. N seasonality also affected plant N content, N productivity and N-resorption efficiency and proficiency in both the C(3) and C(4) species. Interspecific competition influenced N-use and resorption efficiency in both the C(3) and C(4) species, with higher N-use and resorption efficiency in the mixture than in monoculture. The C(4) grass had higher N-use efficiency than the C(3) grass due to its higher N productivity, irrespective of the N treatment or competition. CONCLUSIONS: The observations suggest that N-use strategies in the C(3) and C(4) species used in the study were closely related to seasonal dynamics of N supply and competition. N seasonality might be involved in the growth and temporal niche separation between C(3) and C(4) species observed in the natural ecosystems.  相似文献

为探讨不同浓度NaHCO3胁迫下,马蔺叶片的膜透性、可溶性蛋白及膜脂过氧化物含量的变化规律,以1年生马蔺实生苗为研究对象,通过对细胞膜透性、膜质过氧化产物MDA、SOD、POD和可溶性蛋白等指标进行测定,研究在盆栽控盐条件下,马蔺对NaHCO3胁迫的适应与响应。结果表明:随着NaHCO3浓度和胁迫时间的增加,对马蔺叶片可溶性蛋白含量的影响总体是先下降后上升的趋势,在30 d时,0.4%和0.6%盐浓度胁迫下的马蔺的可溶性蛋白含量上升较大,分别比对照高:21.7%,38.6%;在NaHCO3盐胁迫的10~20 d,马蔺叶片的的膜透性略有降低;在20~30 d时,膜透性迅速上升;NaHCO3胁迫20 d时,除对照外其他盐浓度胁迫下的马蔺叶片的POD活性都达到了最大值,分别比对照高95%,103%,496%;相同盐浓度胁迫下,随着时间的延长,SOD活性变化趋势是先下降后上升;MDA作为膜脂过氧化作用的主要产物之一,随着盐浓度的增加和盐胁迫时间的延长,在30d时0.6%浓度的马蔺叶片中MDA含量大量积累。综上所述,马蔺有抗盐(NaHCO3)胁迫的能力,是重要的盐碱地生植物,本研究为马蔺的抗盐碱性研究又提供了一有力证据,同时为今后将马蔺应用于治理盐碱土地增加了理论基础。  相似文献

为探明CaCl2处理对金叶女贞(Ligustrum vicaryi)茎抗寒性和电阻抗图谱参数的影响,找出抗寒性和电阻抗图谱参数的关系,2006年9月15日开始对金叶女贞当年生扦插苗进行CaCl2处理,对照为清水;分别用电阻抗图谱法和电导法估测其抗寒性,并进行比较分析。结果表明:CaCl2处理能在抗寒锻炼初期使金叶女贞茎的抗寒性提高3.5℃,金叶女贞茎的抗寒性与未冷冻处理茎的弛豫时间τ、冷冻处理后茎的胞外电阻率re、含水量之间均存在较高的相关性。  相似文献

Background and Aims: The grass Alloteropsis semialata is the only plant species withboth C3 and C4 subspecies. It therefore offers excellent potentialas a model system for investigating the genetics, physiologyand ecological significance of the C4 photosynthetic pathway.Here, a molecular phylogeny of the genus Alloteropsis is constructedto: (a) confirm the close relationship between the C3 and C4subspecies of A. semialata; and (b) infer evolutionary relationshipsbetween species within the Alloteropsis genus. Methods: The chloroplast gene ndhF was sequenced from 12 individuals,representing both subspecies of A. semialata and all four ofthe other species in the genus. ndhF sequences were added tothose previously sequenced from the Panicoideae, and used toconstruct a phylogenetic tree. Key Results: The phylogeny confirms that the two subspecies of A. semialataare among the most recently diverging lineages of C3 and C4taxa currently recognized within the Panicoideae. Furthermore,the position of the C3 subspecies of A. semialata within theAlloteropsis genus is consistent with the hypothesis that itsphysiology represents a reversion from C4 photosynthesis. Thedata point to a similar evolutionary event in the Panicum stenodesP.caricoidesP. mertensii clade. The Alloteropsis genusis monophyletic and occurs in a clade with remarkable diversityof photosynthetic biochemistry and leaf anatomy. Conclusions: These results confirm the utility of A. semialata as a modelsystem for investigating C3 and C4 physiology, and provide moleculardata that are consistent with reversions from C4 to C3 photosynthesisin two separate clades. It is suggested that further phylogeneticand functional investigations of the Alloteropsis genus andclosely related taxa are likely to shed new light on the mechanismsand intermediate stages underlying photosynthetic pathway evolution.  相似文献
