We studied the temporal sequence of changes in the photosynthetic CO2/H2O gas exchange intensity, as well as leaf water status, contents of soluble carbohydrates, starch, proline, pigments, and MDA, in maize seedlings (Zea mays L., cv. Luchistaya) under adaptation to increasing water deficit. The duration of drought was 2, 3, 5, and 6 days. Withholding water from maize plants caused gradual increase in the intensity of water deficit: from mild (2 or 3 days) to moderate (5 days) and nearly severe (6 days) water stress. After 6 days, relative leaf water content decreased by 19.8% as compared to the control. On the second day after the onset of drought, slight reduction in the photosynthetic CO2/H2O gas exchange intensity of the treated plants was observed. After 6 days, photosynthesis and transpiration of leaves synchronously reduced almost threefold due to stomatal closure. The progressive soil drought had substantial impact on the carbohydrate metabolism. After 2 days of water deficit, the content of reducing sugars and sucrose increased slightly, whereas after 6 days, it increased ten and four times, respectively. After 2, 3, and 5 days of drought, the starch content declined slightly; however, under severe drought (6 days), it increased by 30% as compared to the control. Simultaneously with the increase in the content of soluble sugars, proline content increased significantly and it was the highest on the sixth day of drought. At all stages of water deficit, the proline content increased more significantly than the content of reducing carbohydrates and sucrose. Under increasing water deficit (5 and 6 days), the content of MDA was found to rise. At the initial drought stage (2 or 3 days) and under severe water deficit (6 days), no significant changes in the pigment content were observed. Thus, at the initial stages of progressive drought, in the leaves of this maize cultivar, a decline in photosynthetic activity proceeded simultaneously with accumulation of reducing sugars, sucrose, and proline. The results obtained showed that, at the first stages of adaptation of maize seedlings to drought, the changes in carbohydrate and proline metabolism have been observed, which have increased upon further plant dehydration.

The net CO2 assimilation by leaves of maize (Zea mays L. cv. Adonis) plants subjected to slow or rapid dehydration decreased without changes in the total extractable activities of phosphoenolpyruvate carboxylase (PEPC), malate dehydrogenase (MDH) and malic enzyme (ME). The phosphorylation state of PEPC extracted from leaves after 2–3 h of exposure to light was not affected by water deficit, either. Moreover, when plants which had been slowly dehydrated to a leaf relative water content of about 60% were rehydrated, the net CO2 assimilation by leaves increased very rapidly without any changes in the activities of MDH, ME and PEPC or phosphorylation state of PEPC. The net CO2-dependent O2 evolution of a non-wilted leaf measured with an oxygen electrode decreased as CO2 concentration increased and was totally inhibited when the CO2 concentration was about 10%. Nevertheless, high CO2 concentrations (5–10%) counteracted most of the inhibitory effect of water deficit that developed during a slow dehydration but only counteracted a little of the inhibitory effect that developed during a rapid dehydration. In contrast to what could be observed during a rapidly developing water deficit, inhibition of leaf photosynthesis by cis-abscisic acid could be alleviated by high CO2 concentrations. These results indicate that the inhibition of leaf net CO2 uptake brought about by water deficit is mainly due to stomatal closure when a maize plant is dehydrated slowly while it is mainly due to inhibition of non-stomatal processes when a plant is rapidly dehydrated. The photosynthetic apparatus of maize leaves appears to be as resistant to drought as that of C3 plants. The non-stomatal inhibition observed in rapidly dehydrated leaves might be the result of either a down-regulation of the photosynthetic enzymes by changes in metabolite pool sizes or restricted plasmodesmatal transport between mesophyll and bundle-sheath cells.

Magnesium (Mg)-deficiency affects productivity and quality in agriculture, yet at a physiological level it is not well understood. Citrus grandis and Citrus sinensis seedlings were irrigated for 12?weeks with 0, 50, 500 or 2,000?μM MgSO4. Thereafter, Mg-deficiency-induced changes in photosynthesis, antioxidant system and carbohydrates were investigated. Mg-deficiency affected CO2 assimilation more in C. grandis leaves than in C. sinensis ones, but Mg-deficiency-induced accumulation of sugars was not higher in the former except for sucrose. Mg-deficiency-induced photoinhibitory impairment occurring on the whole photosynthetic electron transport chain was more severe in C. grandis leaves than in C. sinensis ones. Mg-deficient leaves had higher or similar activities of antioxidant enzymes and contents of antioxidant metabolites except for catalase (CAT) activity and reduced glutathione (GSH) content. However, Mg-deficiency increased leaf malondialdehyde (MDA) content. In conclusion, the greater decrease in CO2 assimilation in Mg-deficient C. grandis leaves may be caused by the greater decrease in the photosynthetic electron transport capacity. Mg-deficiency-induced up-regulation in leaf antioxidant system does not provide enough protection to Mg-deficient leaves against the oxidative damage.

To understand how light quality influences plant photosynthesis, we investigated chloroplastic ultrastructure, chlorophyll fluorescence and photosynthetic parameters, Rubisco and chlorophyll content and photosynthesis-related genes expression in cucumber seedlings exposed to different light qualities: white, red, blue, yellow and green lights with the same photosynthetic photon flux density of 100 μmol m?2 s?1. The results revealed that plant growth, CO2 assimilation rate and chlorophyll content were significantly reduced in the seedlings grown under red, blue, yellow and green lights as compared with those grown under white light, but each monochromatic light played its special role in regulating plant morphogenesis and photosynthesis. Seedling leaves were thickened and slightly curled; Rubisco biosynthesis, expression of the rca, rbcS and rbcL, the maximal photochemical efficiency of PSII (Fv/Fm) and quantum yield of PSII electron transport (ФPSII) were all increased in seedlings grown under blue light as compared with those grown under white light. Furthermore, the photosynthetic rate of seedlings grown under blue light was significantly increased, and leaf number and chlorophyll content of seedlings grown under red light were increased as compared with those exposed to other monochromatic lights. On the contrary, the seedlings grown under yellow and green lights were dwarf with the new leaves etiolated. Moreover, photosynthesis, Rubisco biosynthesis and relative gene expression were greatly decreased in seedlings grown under yellow and green light, but chloroplast structural features were less influenced. Interestingly, the Fv/Fm, ФPSII value and chlorophyll content of the seedlings grown under green light were much higher than those grown under yellow light.

The influence of arbuscular mycorrhizal fungi (AMF), Funneliformis mosseae and Rhizophagus intraradices, on plant growth, leaf water status, chlorophyll concentration, photosynthesis, nutrient concentration, and fractal dimension (FD) characteristics of black locust (Robinia pseudoacacia L.) seedlings was studied in pot culture under well-watered, moderate drought stress, and severe drought stress treatments. Mycorrhizal seedlings had higher dry biomass, leaf relative water content (RWC), and water use efficiency (WUE) compared with non-mycorrhizal seedlings. Under all treatments, AMF colonization notably enhanced net photosynthetic rate, stomatal conductance, and transpiration rate, but decreased intercellular CO2 concentration. Leaf chlorophyll a and total chlorophyll concentrations were higher in AM seedlings than those in non-AM seedlings although there was no significant difference between AMF species. AMF colonization improved leaf C, N, and P concentrations, but decreased C:N, C:P, and N:P ratios. Mycorrhizal seedlings had a larger FD value than non-mycorrhizal seedlings. The FD value was positively and significantly correlated to the plant growth parameters, photosynthesis, RWC, WUE, and nutrient concentration but negatively correlated to leaf/stem ratio, C:N and C:P ratios, and intercellular CO2 concentration. We conclude that AMF lead to an improvement of growth performance of black locust seedlings under all growth conditions, including drought stress via improving leaf water status, chlorophyll concentration, photosynthesis, and nutrient uptake. Moreover, FD technology proved to be a powerful non-destructive method to characterize the effect of AMF on the physiology of host plants during drought stress.

Strawberry (Fragaria × ananassa) plants were grown in field plots at the current ambient [CO2], and at ambient + 300 and ambient + 600 μmol mol−1 [CO2]. Approximately weekly measurements were made of single leaf gas exchange of upper canopy leaves from early spring through fall of two years, in order to determine the temperature dependence of the stimulation of photosynthesis by elevated [CO2], whether growth at elevated [CO2] resulted in acclimation of photosynthesis, and whether any photosynthetic acclimation was reduced when fruiting created additional demand for the products of photosynthesis. Stimulation of photosynthetic CO2 assimilation by short-term increases in [CO2] increased strongly with measurement temperature. The stimulation exceeded that predicted from the kinetic characteristics of ribulose-1,5-bisphosphate carboxylase at all temperatures. Acclimation of photosynthesis to growth at elevated [CO2] was evident from early spring through summer, including the fruiting period in early summer, with lower rates under standard measurement conditions in plants grown at elevated [CO2]. The degree of acclimation increased with growth [CO2]. However, there were no significant differences between [CO2] treatments in total nitrogen per leaf area, and photosynthetic acclimation was reversed one day after switching the [CO2] treatments. Tests showed that acclimation did not result from a limitation of photosynthesis by triose phosphate utilization rate at elevated [CO2]. Photosynthetic acclimation was not evident during dry periods in midsummer, when the elevated [CO2] treatments conserved soil water and photosynthesis declined more at ambient than at elevated [CO2]. Acclimation was also not evident during the fall, when plants were vegetative, despite wet conditions and continued higher leaf starch content at elevated [CO2]. Stomatal conductance responded little to short-term changes in [CO2] except during drought, and changed in parallel with photosynthetic acclimation through the seasons in response to the long-term [CO2] treatments. The data do not support the hypothesis that source-sink balance controls the seasonal occurrence of photosynthetic acclimation to elevated [CO2] in this species. This revised version was published online in June 2006 with corrections to the Cover Date.

SA treatment effectively ameliorated the negative effect of moderate drought stress on T. grandis Seedlings through increasing the water content, Pn, proline content, antioxidant enzymes activity and reducing MDA.

Abstract

Water availability is one of the most critical factors that limits the growth and development of plants. Salicylic acid (SA) is an important signal molecule that modulates plant responses to abiotic stress. To elucidate the regulating mechanism of exogenous SA on Torreya grandis cv. Merrillii under different water stresses, a pot experiment was conducted in a greenhouse. Exposure of T. grandis seedlings to drought conditions resulted in reduced growth rate that was associated with a decline in water content and CO2 assimilation. Foliar application of SA effectively increased the water content, net CO2 assimilation rate, proline content and antioxidant enzymes activity in the plants, which helped T. grandis to acclimate to moderate drought stress and increase the shoot dry matter. However, when the plants were under severe drought stress, the relative water content and CO2 assimilation in the SA-treated plants were significantly lower than those in the control plants. Therefore, our results indicated that SA can effectively ameliorate the negative effect of moderate drought stress on T. grandis seedling growth.

The hypothesis that net photosynthesis is diminished in many plant species because of a high rate of CO2 evolution in the light has been tested further. High rates of CO2 output in CO2-free air in comparison with dark respiration were found in Chlamydomonas reinhardi, wheat leaves, tomato leaves, and to a lesser extent in Chlorella pyrenoidosa by means of the 14C-photorespiration assay. In tobacco leaves high photorespiration was characteristic of a standard variety, Havana Seed, and a possibly still higher rate was found in a yellow heterozygous mutant, JWB Mutant. However, the dark homozygous sibling of the latter, JWB Wild, had a low photorespiration for the tobacco species. The relative rates of photorespiration were in the same sequence when measured by the 14CO2 released in normal air from leaf disks supplied with glycolate-1-14C in the light.

As would be predicted by the hypothesis, the maximal net rate of photosynthesis at 300 ppm of CO2 in the air in JWB Wild leaves was greater (24%) than in Havana Seed, while JWB Mutant had less CO2 uptake than the standard variety (21%). At 550 ppm of CO2 the differences in net photosynthesis were not as great between the 2 siblings as at 200 ppm. The relative leaf expansion rates of seedlings of the 3 tobacco varieties in a greenhouse had the same relationship as their rates of CO2 assimilation.

Thus within the tobacco species, as in a comparison between tobacco and maize, low photorespiratory CO2 evolution was correlated with higher photosynthetic efficiency. Therefore it seems that increased CO2 uptake should be achieved by genetic interference with the process of photorespiration.

Despite the observed impact of water stress on photosynthesis, some of the most used models of CO2 assimilation in C3 and C4 functional types do not directly account for it. We discuss an extension of these models, which explicitly includes the metabolic and diffusive limitations due to water stress on photosynthesis. Functional relationships describing the photosynthetic processes and CO2 diffusion inside leaves are modified to account for leaf water status on the basis of experimental results available in the literature. Extensive comparison with data shows that the model is suitable to describe the reduction in CO2 assimilation rate with decreasing leaf water potentials in various species. A simultaneous analysis of photosynthesis, transpiration and soil moisture dynamics is then carried out to explore the actual impact of drought on different photosynthesis processes and on the overall plant activity. The model well reproduces measured CO2 assimilation rate as a function of soil moisture and could be useful to formulate hypotheses for detailed experiments as well as to simulate in detail transpiration and photosynthesis dynamics under water stress.

Effects of sulfur on photosynthesis in sugar beets (Beta vulgaris L. cv. F58-554H1) were studied by inducing sulfur deficiency and determining changes in the photosynthesis of whole attached leaves and of isolated chloroplasts. The rates of photosynthetic CO2 uptake by intact leaves, photoreduction of ferricyanide, cyclic and noncyclic photophosphorylation of isolated chloroplasts, and the rate of CO2 assimilation by ribulose diphosphate carboxylase, decreased with decrease in total leaf sulfur from 2500 to about 500 μg g−1 dry weight. Sulfur deficiency reduced photosynthesis through an effect on chlorophyll content, which decreased linearly with leaf sulfur, and by decreasing the rate of photosynthesis per unit chlorophyll. There was only a small effect of sulfur deficiency on stomatal diffusion resistance to CO2 until leaf sulfur decreased below 1000 μg g−1 when stomatal resistance became a more significant proportion of the total diffusion resistance to CO2. Light respiration rates were positively correlated with photosynthesis rates and dark respiration was unchanged as leaf sulfur concentrations declined.

To understand physiological acclimation of psammophyte to repeated soil drought and rewatering, two psammophytes (Setaria viridis and Digitaria ciliaris) were subjected to three cycles of soil drought and rewatering. The response process of leaf relative water content (RWC), membrane permeability, lipid peroxidation, gas exchange characteristics, antioxidant enzymes, soluble protein, and free proline was examined. Leaf RWC, the net photosynthesis rate, stomatal conductance, and water use efficiency decreased, while membrane permeability, lipid peroxidation, intercellular CO2 concentration, soluble protein, and free proline increased during three soil drought periods for both psammophytes. These physiological characteristics were recovered to the control levels following rewatering for 4 days. However, activities of SOD, CAT, and POD were induced continuously under soil drought conditions, and remained higher than those in the control throughout the whole experiment period, which agrees with our hypothesis that drought hardening activates defensive systems of both psammophytes continuously. Decreasing level of leaf RWC and increasing levels of leaf membrane permeability and lipid peroxidation were suppressed with increasing the number of drought cycles, suggesting that drought hardening alleviates damages of both psammophytes and improves their drought tolerance and acclimation to soil drought conditions in the future. Additionally, the photosynthesis decreased more slowly in the subsequent drought cycles than in the first cycle, allowing both psammophytes to maximize assimilation in response to repeated soil drought conditions. Thus, both psammophytes acclimatize themselves to repeated soil drought.

The effects of drought stress and high irradiance and their combination were studied under laboratory conditions using young plants of a very drought-resistant variety, ICMH 451, of pearl millet (Pennisetum glaucum) and three varieties of sorghum (Sorghum bicolor)—one drought-resistant from India, one drought-tolerant from Texas, and one drought-sensitive variety from France. CO2 assimilation rates and photosystem II fluorescence in leaves were analyzed in parallel with photosynthetic electron transport, photosystem II fluorescence, and chlorophyll-protein composition in chloroplasts isolated from these leaves. High irradiance slightly increased CO2 assimilation rates and electron transport activities of irrigated plants but not fluorescence. Drought stress (less than −1 megapascal) decreased CO2 assimilation rates, fluorescence, and electron transport. Under the combined effects of drought stress and high irradiance, CO2 assimilation rates and fluorescence were severely inhibited in leaves, as were the photosynthetic electron transport activities and fluorescence in chloroplasts (but not photosystem I activity). The synergistic or distinctive effect of drought and high irradiance is discussed. The experiments with pearl millet and three varieties of sorghum showed that different responses of plants to drought and light stresses can be monitored by plant physiological and biochemical techniques. Some of these techniques may have a potential for selection of stress-resistant varieties using seedlings.

Maize (Zea mays L.) is a chilling (below 15 °C) sensitive plant that shows little capacity to acclimate to low growth temperatures. Maize leaves are extremely sensitive to chilling injury, which usually results in premature leaf senescence. Leaves exposed to temperatures below 10 °C in the light show substantial inhibition of CO2 assimilation and down-regulation of photosynthetic electron transport. However, the intrinsic relationships between the quantum efficiencies of photosystems I and II are not modified by chilling. Moreover, the integral relationships between non-cyclic electron transport and CO2 fixation are similar in chilled and unchilled leaves. In this review we examine the roles and importance of photosynthetic regulation, carbon metabolism and antioxidant metabolism in determining the sensitivity of maize leaf photosynthesis to chilling. The distinct cellular localisation patterns of antioxidant enzymes such as glutathione reductase (EC 1.6.4.2) and dehydroascorbate reductase (EC 1.8.5.1) can restrict the recycling of antioxidants associated with photosynthesis during chilling. Disruption of circadian regulation of metabolism and insufficient antioxidant defence are postulated to cause chilling sensitivity.

Leaf rolling observed in some crops such as maize, rice, wheat and sorghum is an indicator of decreased water status. Moderate leaf rolling not tightly or early increases the photosynthesis and grain yield of crop cultivars under environmental stresses. Moreover, the effects of exogenous abscisic acid (ABA) on stomatal conductance, water status and synthesis of osmotic compounds are a well-known issue in plants subjected to water deficit. However, it is not clear how the cross-talk of ABA with H2O2 and osmolyte compounds affects the leaf rolling mechanism. Regulation mechanism of leaf rolling by ABA has been first studied in maize seedlings under drought stress induced by polyethylene glycol 6000 (PEG 6000) in this study. ABA treatment under drought stress reduced hydrogen peroxide (H2O2) content and the degree of leaf rolling (%) while the treatment-induced ABA synthesis, osmolyte levels (proline, polyamine and total soluble sugars) and some antioxidant enzyme activities in comparison to the plants that were not treated with ABA. Furthermore, exogenous ABA up-regulated the expression levels of arginine decarboxylase (ADC) and pyrroline-5-carboxylate synthase (P5CS) genes and down-regulated polyamine oxidase (PAO), diamine oxidase (DAO) and proline dehydrogenase (ProDH) gene expressions. When endogenous ABA content was decreased by the treatment of fluoridone (FLU) that is an ABA inhibitor, leaf rolling degree (%), H2O2 content and antioxidant enzyme activities increased, but osmolyte levels, ADC and P5CS gene expressions decreased. Finally, the treatment of ABA to maize seedlings exposed to drought stress resulted in the stimulation of the antioxidant system, osmotic adjustment and reduction of leaf rolling. We concluded that ABA can be a signal compound cross-talking H2O2, proline and polyamines and thus involved in the leaf rolling mechanism by providing osmotic adjustment. The results of this study can be used to provide data for the molecular breeding of maize hybrids with high grain yield by means of moderately rolled leaves.

Photosynthetic responses of intact leaves of the desert shrub Encelia farinosa were measured during a long term drought cycle in order to understand the responses of stomatal and nonstomatal components to water stress. Photosynthetic rate at high irradiance and leaf conductance to water vapor both decreased linearly with declining leaf water potential. The intercellular CO2 concentration (ci) remained fairly constant as a function of leaf water potential in plants subjected to a slow drought cycle of 25 days, but decreased in plants exposed to a 12-day drought cycle. With increasing water stress, the slope of the dependence of photosynthesis on ci (carboxylation efficiency) decreased, the maximum photosynthetic rates at high ci became saturated at lower values, and water use efficiency increased. Both the carboxylation efficiency and photosynthetic rates were positively correlated with leaf nitrogen content. Associated with lower leaf conductances, the calculated stomatal limitation to photosynthesis increased with water stress. However, because of simultaneous changes in the dependence of photosynthesis on ci with water stress, increased leaf conductance alone in water-stressed leaves would not result in an increase in photosynthetic rates to prestressed levels. Both active osmotic adjustment and changes in specific leaf mass occurred during the drought cycle. In response to increased water stress, leaf specific mass increased. However, the increases in specific leaf mass were associated with the production of a reflective pubescence and there were no changes in specific mass of the photosynthetic tissues. The significance of these responses for carbon gain and water loss under arid conditions are discussed.

The effects of extreme phosphate (Pi) deficiency during growth on the contents of adenylates and pyridine nucleotides and the in vivo photochemical activity of photosystem II (PSII) were determined in leaves of Helianthus annuus and Zea mays grown under controlled environmental conditions. Phosphate deficiency decreased the amounts of ATP and ADP per unit leaf area and the adenylate energy charge of leaves. The amounts of oxidized pyridine nucleotides per unit leaf area decreased with Pi deficiency, but not those of reduced pyridine nucleotides. This resulted in an increase in the ratio of reduced to oxidized pyridine nucleotides in Pi-deficient leaves. Analysis of chlorophyll a fluorescence at room temperature showed that Pi deficiency decreased the efficiency of excitation capture by open PSII reaction centres (φe), the in vivo quantum yield of PSII photochemistry (φPSII) and the photochemical quenching co-efficient (qP), and increased the non-photochemical quenching co-efficient (qN) indicating possible photoinhibitory damage to PSII. Supplying Pi to Pi-deficient sunflower leaves reversed the long-term effects of Pi-deficiency on PSII photochemistry. Feeding Pi-sufficient sunflower leaves with mannose or FCCP rapidly produced effects on chlorophyll a fluorescence similar to long-term Pi-deficiency. Our results suggest a direct role of Pi and photophosphorylation on PSII photochemistry in both long-and short-term responses of photosynthetic machinery to Pi deficiency. The relationship between φPSII and the apparent quantum yield of CO2 assimilation determined at varying light intensity and 21 kPa O2 and 35 Pa CO2 partial pressures in the ambient air was linear in Pi-sufficient and Pi-deficient leaves of sunflower and maize. Calculations show that there was relatively more PSII activity per mole of CO2 assimilated by the Pi-deficient leaves. This indicates that in these leaves a greater proportion of photosynthetic electrons transported across PSII was used for processes other than CO2 reduction. Therefore, we conclude that in vivo photosynthetic electron transport through PSII did not limit photosynthesis in Pi-deficient leaves of sunflower and maize and that the decreased CO2 assimilation was a consequence of a smaller ATP content and lower energy charge which restricted production of ribulose, 1-5, bisphosphate, the acceptor for CO2.

The photosynthetic properties of a yellow lethal mutant, Oy/oy, and two yellow-green mutants of maize which are allelic (a homozygous recessive oy/oy and a heterozygous dominant Oy/+) were examined. Although Oy/oy had little or no chlorophyll or capacity for CO2 fixation compared to normal siblings, it had 28% as much ribulose-1,5-bisphosphate carboxylase oxygenase (Rubisco) activity, and from 40% to near normal activities of C4 cycle enzymes. Both yellow-green mutants had only half as much chlorophyll per leaf area as normal green seedlings in greenhouse-grown plants in winter and spring. However, the absorbance of light by the mutants was relatively high, as their transmittance was only 5 to 8% greater than normal leaves. In winter-grown greenhouse plants, the activities of Rubisco and several C4 cycle enzymes in the mutants were unaffected and similar to those of normal seedlings on a leaf area basis. After allowing for small differences in leaf absorbance, the light response curves for photosynthesis in the mutants were similar on a leaf area basis but much higher on a chlorophyll basis than those of the normal seedlings. In spring-grown greenhouse plants the enzyme activities and photosynthesis rates were about 30% lower per leaf area in the yellow-green mutant leaves compared to the wild type. The maximum carboxylation efficiency (measured under low CO2 and 1000 mol quanta m-2 s-1) in the mutants and normal leaves was similar on a Rubisco protein basis. The results indicate that maize can undergo a 50% reduction in chlorophyll content without a corresponding reduction in enzymes of carbon assimilation, and still maintain a high capacity for photosynthesis.Abbreviations Chl chlorophyll - PEP phosphoenolypruvate - Rubisco ribulose-1,5-bisphosphate carboxylase oxygenase This research was supported by CSIRO and by USDA Competitive Grant 86-CRCR-1-2036.
