The white rot basidiomycete Ganoderma lucidum was evaluated for its capability to tolerate and to degrade the herbicide diuron. Diuron at a subtoxic concentration was added at the start of the cultivation in glucose liquid stationary cultures. Under this condition diuron was a laccase inducer. Almost 50% of the initially present diuron was removed after 15 d of cultivation. Two diuron metabolites were found N′-(3,4-dichlorophenyl)-N-methylurea (DCPMU) and 3,4-dichlorophenylurea (DCPU). The addition of the cytochrome P450 inhibitors 1-aminobenzotriazole and piperonyl butoxide reduced significantly the capability of the fungus in degrading diuron. The activities of superoxide dismutase and catalase were significantly increased in the mycelial extracts by the presence of diuron. On the other hand, diuron did not cause any significant alteration in the levels of reactive oxygen species. Additionally, laccase could also degrade diuron in vitro and this degradation was increased by the addition of synthetic mediators, 3-ethylbenzthiazoline-6-sulphonic acid and acetylacetone. Significant reduction in the toxicity, as evaluated by the Lactuca sativa bioassay, was observed after G. lucidum treatment. In conclusion, G. lucidum is able to metabolize diuron by intra- and extracellular mechanisms, without the accumulation of toxic products.  相似文献

A technique for the propagation of Brussels sprout by means of root cuttings is described. Adventitious shoots arise exogenously on callus tissue which develops around the base of side roots. Cuttings sometimes rot without forming adventitious shoots, and cuttings which remain sound do not all produce shoots. Rotting may largely be prevented by planting cuttings with the proximal end exposed above the surface of the medium, and by allowing the root portions to dry before planting. Surface sterilization with mercuric chloride controls rotting but reduces bud formation. Individual plants differ in their capacity to form buds on root cuttings, and this difference is carried by the clones derived from them. Portions of root form more buds if cut into several pieces than if planted intact.  相似文献

Ascorbate levels and redox states, as well as the activities of the enzymes of ascorbate metabolism, were analyzed in roots of tomato seedlings during the culture on a medium supplemented with auxin and compared to the control cultured on an auxin-free medium. Biochemical parameters were determined separately in the distal part of the root where the inhibitory effect of auxin on root elongation growth is observed and in the proximal half on the organ which reacts to auxin treatment with increased lateral root proliferation. ASC peroxidase activity was found to be stimulated by auxin treatment in the lateral-root forming part of the root. This effect was not observed in the distal part of the organ. On the other hand, ASC oxidase activity was raised by auxin exclusively in the distal part of the root. An inhibitory effect of auxin supplementation to the medium on ASC—reducing enzymes was observed. The dehydroascorbate reductase activity was found to be inhibited by auxin only in the proximal part, while the activity of monodehydroascorbate reductase in both, the proximal and distal parts of the root. Ascorbate content increased in roots during culture irrespective of the presence of auxin. However, auxin treatment resulted in higher DHA levels and more significant participation of DHA in the total ascorbate pool when compared to the control grown on the auxin-free medium. Similar to auxin, adding DHA to the culture medium stimulated lateral root formation and inhibited primary root elongation. In contrast to DHA, ASC treatment affected significantly neither lateral root formation nor primary root growth and partly reversed the stimulatory effect of IAA on root formation and the inhibitory effect on root elongation. These results suggest that auxin induced changes in ascorbate metabolism may be involved in developmental reactions in tomato roots.  相似文献

This study examined the performances of 21 cassava cultivars in two cropping seasons on the field against root rot disease and the yield in cassava-maize intercrop. Data were collected on number of root/plant, weight of root (t/ha) and disease severity (DS) on rotted roots at 12 and 16 months after planting (MAP), respectively. There were significant (P ≤ 0.05) differences for DS at 12 and 16 MAP in both seasons with cultivar TMS 97/JW2 having the least DS score. TMS 97/JW2 was resistant to the root rot pathogen, while eleven other cultivars were moderately resistant to the disease at 16 MAP. There was no consistency in the roots weight for the cultivars over the two cropping seasons but higher roots weight was recorded at 16 MAP than 12 MAP with different cultivars having highest roots weight at these periods. Intercropping maize with cassava does not have any management potential on root rot development.  相似文献

Some 136 isolates of fluorescent pseudomonads were isolated from the rhizosphere of plants growing in 5 different ecosystems. Thirty-four percent of these isolates inhibited the causal agent of cassava stem rot, Erwinia carotovora pv. carotovora, in vitro. One month old plantlets, produced by rooting the shoots of 4 cultivars in distilled water, were inoculated with a suspension (1.1 × 109 cells/ml) of each pseudomonad. Some isolates increased root weight by 95% over uninoculated controls two months after planting when inoculated at planting, and 15, and 30 days afterwards. Inoculated plants were free from symptoms of root pathogens and roots filled earlier than controls. Taxonomic studies showed that these bacterial isolates, were either Pseudomonas putida (90%) or P. fluorescens (10%).  相似文献

Tumour initiating/promoting effect of diuron, a widely used substituted urea herbicide, was studied in rats using liver tumour model. Chronic exposure to diuron at a dose of 250 mg/kg body wt resulted in high mortality and weight loss in treated animals. The animals which received diuron + HCH treatment showed an increase in size and weight of liver as compared to controls. Liver tumours were not observed in any of the treated group whereas some significant histological changes were seen in diuron treated rat liver. Diuron thus has been found to be hepatotoxic albeit neither tumour initiating nor promoting in rat liver tumorigenesis assay system.  相似文献

The incidence in unheated glasshouses of tomato brown root rot (BRR) was recorded on five successive crops starting with the first ever grown on the experimental site. In the protected environment the changes followed a pattern that was defined mathematically, the regression linking angular or logit transforms of per cent BRR, observed at different intervals after planting, accounting for c. 72% of the variation. Rates of build-up, seemingly related to initial amounts of inocula, increased in successive seasons, there being more BRR in the fifth season than in the first. Four per cent BRR recorded 5 weeks after planting increased threefold during the next 3 months to 12 %, but in later years, when amounts of early-season infection were doubled to 8%, the incidence of root rot subsequently increased five times. Tomato roots colonized by Pyrenochaeta lycopersici either brown and rot or become corky. The proportion of end-of-season BRR attributable to corkiness increased from the first to fifth seasons. The monthly increments of fruit yield were inversely proportional to the incidence of BRR.  相似文献

The proportion of sugar-beet roots infected by Helicobasidium purpureum increased most rapidly in September and October. Violet root rot was not controlled by fungicides applied at drilling or in July. Heavily infected roots yielded 31% less sugar than healthy or lightly infected roots. Sugar beet following infected carrots lifted or ploughed in during July had no more violet root rot than when following barley or fallow, but the beet crop became heavily infected when it followed carrots left in the ground until December, whether they were then lifted or ploughed in. Hot‐water drench treatments were evaluated for disinfesting roots of potted anthurium Anthurium andraeanum of the burrowing nematode Radopholus similis. A continuous drench of roots and media in pots with 50°C water for 5 to 20 min eliminated or reduced nematode populations to < 1 g?1 of dry root. A second hot water drench, 2 months after the first drenching, tended to increase the efficacy of the heat treatment. A few survivors persisted in the roots and/or stems of a few plants 2 to 4 months after heat treatment. Non‐treatment of the shoots and possible migration from stem to root tissues are probable causes of nematode survival. Drenching potting media and roots in pot were as effective against R. similis in the roots as hot water dipping bare‐rooted plants. Plant response to hot‐water drenching varied among cultivars, but most exhibited tolerance to the heat treatments. Visual inspection of the plants showed little difference between treated and untreated plants in the heat tolerant cultivars. However, all heat‐treated ‘Marian Seefurth’ plants, especially hot water‐dipped bare‐rooted plants, appeared to suffer some degree of growth reduction as measured by lower root and stem dry weights when compared to untreated plants 3 months after treatment. Conditioning anthurium plants with hot water or hot air prior to hot water drenching did not benefit plant quality when compared to unconditioned, heat‐treated plants.  相似文献

Organ differentiation and growth of tissue cultured bulbscales of Lilium auratum Lindl. was investigated. Benzyladenine stimulated bulb and bulbscale differentiation but inhibited root formation. Addition of activated charcoal to the medium negated the effect of BA on differentiation, while the growth of bulbs was markedly stimulated. Organ formation was also influenced by the physiological age of bulbs. From bulbscales of a 3-week-old bulb, a large number of bulbs and roots was formed but callus was not produced. A high sucrose concentration (90 g/1) reduced the subsequent sprouting after planting in soil. Scale leaf formation was also regulated. Sucrose (30 g/1) stimulated scale leaf formation but 90 g/1 were inhibitory.  相似文献

Interactions between Fusarium solani and Phytophthora parasitica or F. solani and P. citrophthora influenced the development of root rot of citrus but depended on the temporal order of inoculation with F. solani or the two Phytophthora spp. Inoculation of citrus with either Fusarium solani and Phytophthora parasitica or Phytophthora citrophthora increased root rot compared to inoculation with P. parasitica or P. citrophthora alone when plants were inoculated with Phytophthora by dipping their roots in zoospore suspensions and subsequently transplanted into soil infested with F. solani. However, root rot was not increased by simultaneous co-inoculation of P. parasitica and F. solani or when plants were inoculated with F. solani first. Root rot was not increased when heat-stressed or non-stressed plants were inoculated with P. parasitica 30 days after transplanting into soil infested with F. solani. In most but not all experiments, F. solani alone reduced growth of tops or roots a small but significant amount.Co-inoculation of citrus by root-dipping into zoospore suspensions of P. parasitica and transplanting into soil infested with F. solani reduced feeder root length by 62% and root weight by 61% but did not significantly reduce the percentage of living roots when compared to inoculation with P. parasitica alone. When citrus roots were immersed in zoospore suspensions of P. citrophthora and transplanted into soil infested with F. solani, feeder root length was reduced by 68%, but feeder root weight and the percentage of living roots were not significantly reduced when compared to plants inoculated with P. citrophthora alone.Propagule densities of both P. parasitica and P. citrophthora in the rhizosphere of plants inoculated by root-immersion and then transplanting into soil infested with F. solani were not significantly different than propagule densities from plants transplanted into non-infested soil. Propagule densities of P. parasitica were suppressed an average of 41% when citrus was inoculated with P. parasitica 30 days after transplanting into soil infested with F. solani and by 41% when citrus was co-inoculated by transplanting into soil infested with both F. solani and P. parasitica.  相似文献

Pythium irregulare andPythium coloratum were isolated consistently from roots of onion plants exhibiting root rot and associated foliar symptoms in fields located in Organe County, NY.P. coloratum predominated following extremely wet weather in 1984 and 1986, whileP. irregulare was prevalent following moderately wet weather in 1985. Both species produced root rot symptoms similar to field symptoms when 12 week old onion plants (cv Downing Yellow Globe) were inoculated and incubated in a growth chamber at 14°C for 10 days. This is the first report ofP. irregulare andP. coloratum as causes of root rot of onion.  相似文献

 The effect of Tiron (disodium 1,2-dihydroxybenzene-3,5-disulfonate) on the growth, morphology and alkaloid content of adventitious roots in Atropa belladonna was investigated. High concentrations of Tiron had an inhibitory effect on growth of the root. The appearance of cultured roots was significantly changed from rough roots accompanied with callus-like tissue in control cultures to fine roots without callus formation. Alkaloid content was drastically increased by the addition of 1 mM Tiron to the medium. The influence of NAA, which has an inhibitory effect on alkaloid production, was partially restored by Tiron treatment, indicating that this radical scavenger may affect the production of alkaloids through modulation of the mode of action of auxin. The interaction of lesion nematodes, black root rot disease caused by Rhizoctonia fragariae, and root damage caused by feeding of the scarab larva, Maladera castanea, was determined in greenhouse studies. Averaged over all experiments after 12 weeks, root weight was reduced 13% by R. fragariae and 20% by M. castanea. The percentage of the root system affected by root rot was increased by inoculation with either R. fragariae (35% more disease) or P. penetrans (50% more disease) but was unaffected by M. castanea. Rhizoctonia fragariae was isolated from 9.2% of the root segments from plants not inoculated with R. fragariae. The percentage of R. fragariae-infected root segments was increased 3.6-fold by inoculation with R. fragariae on rye seeds. The presence of P. penetrans also increased R. fragariae root infection. The type of injury to root systems was important in determining whether roots were invaded by R. fragariae and increased the severity of black root rot. Pratylenchus penetrans increased R. fragariae infection and the severity of black root rot. Traumatic cutting action by Asiatic garden beetle did not increase root infection or root disease by R. fragariae. Both insects and diseases need to be managed to extend the productive life of perennial strawberry plantings.  相似文献

The influences of Gaeumannomyces graminis var. tritici (which causes take-all of wheat), Rhizoctonia solani AG-8 (which causes rhizoctonia root rot of wheat), Pythium irregulare, P. aristosporum, and P. ultimum var. sporangiiferum (which cause pythium root rot of wheat) on the population dynamics of Pseudomonas fluorescens 2-79 and Q72a-80 (bicontrol strains active against take-all and pythium root rot of wheat, respectively) in the wheat rhizosphere were examined. Root infection by either G. graminis var. tritici or R. solani resulted in populations of both bacterial strains that were equal to or significantly larger than their respective populations maintained on roots in the absence of these pathogens. In contrast, the population of strain 2-79 was significantly smaller on roots in the presence of any of the three Pythium species than on noninfected roots and was often below the limits of detection (50 CFU/cm of root) on Pythium-infected roots after 40 days of plant growth. In the presence of either P. aristosporum or P. ultimum var. sporangiiferum, the decline in the population of Q72a-80 was similar to that observed on noninfected roots; however, the population of this strain declined more rapidly on roots infected by P. irregulare than on noninfected roots. Application of metalaxyl (which is selectively inhibitory to Pythium spp.) to soil naturally infestated with Pythium spp. resulted in significantly larger rhizosphere populations of the introduced bacteria over time than on plants grown in the same soil without metalaxyl. It is apparent that root infections by fungal pathogens may either enhance or depress the population of fluorescent pseudomonads introduced for their control, with different strains of pseudomonads reacting differentially to different genera and species of the root pathogens.  相似文献

The reduction in growth of maize (Zea mays L.) seedling primary roots induced by salinization of the nutrient medium with 100 millimolar NaCl was accompanied by reductions in the length of the root tip elongation zone, the length of fully elongated epidermal cells, and the apparent rate of cell production: Each was partially restored when calcium levels in the salinized growth medium were increased from 0.5 to 10.0 millimolar. We investigated the possibility that the inhibition of elongation growth by salinity might be associated with an inhibition of cell wall acidification, such as that which occurs when root growth is inhibited by IAA. A qualitative assay of root surface acidification, using bromocresol purple pH indicator in agar, showed that salinized roots, with and without extra calcium, produced a zone of surface acidification which was similar to that produced by control roots. The zone of acidification began 1 to 2 millimeters behind the tip and coincided with the zone of cell elongation. The remainder of the root alkalinized its surface. Kinetics of surface acidification were assayed quantitatively by placing a flat tipped pH electrode in contact with the elongation zone. The pH at the epidermal surfaces of roots grown either with 100 millimolar NaCl (growth inhibitory), or with 10 millimolar calcium ± NaCl (little growth inhibition), declined from 6.0 to 5.1 over 30 minutes. We conclude that NaCl did not inhibit growth by reducing the capacity of epidermal cells to acidify their walls.  相似文献

Pot experiments were used to investigate the effect of root-feeding larvae of the soldier fly Inopus rubriceps (Macquart) on shoot production from sugarcane planting pieces (setts) and on growth and ratooning of sugarcane plants. Shoot elongation was inhibited while setts were exposed to larvae, and it resumed when larvae were removed. Infested setts produced a greater weight of roots than uninfested setts. Similar symptoms were induced by mechanical root pruning, suggesting that the effect of soldier fly larvae on setts may be a redirection of growth from the shoot to roots due to root damage. Larvae had a greater effect on shoot production at lower temperature, particularly in cultivar 'Q151', which had a higher temperature threshold than 'CP44-101'. Temperature and cultivar may influence the harmful effect of soldier fly larvae on sett germination by changing the differential rates of plant growth and larval feeding. When growing plants were exposed to larvae, the infested plants were slightly smaller at harvest and subsequently produced many fewer ratoon shoots from underground buds than uninfested plants. Shoot elongation from buds was also inhibited in setts cut from the above-ground stalks of infested plants. Analysis of nutrient levels in plants did not indicate the mechanism for ratooning inhibition, because levels of time 10 elements analyzed were at least as high or higher in infested plants. Infestation was associated with an increased level of sucrose and a reduced level of fructose in stalks. The inhibitory effect of larval feeding on ratooning was not reversed when larvae were removed from pots 10 wk before harvest. However, new stubble produced from infested plants then ratooned normally after a second harvest, provided the new roots were not attacked. The symptoms of larval feeding in growing plants are unexplained, but may be caused by the prolonged withdrawal of sap from roots or the injection of some inhibitory substance by larvae.  相似文献

A split-root technique was used to examine the interaction between Pratylenchus penetrans and the cortical root-rotting pathogen Rhizoctonia fragariae in strawberry black root rot. Plants inoculated with both pathogens on the same half of a split-root crown had greater levels of root rot than plants inoculated separately or with either pathogen alone. Isolation of R. fragariae from field-grown roots differed with root type and time of sampling. Fungal infection of structural roots was low until fruiting, whereas perennial root colonization was high. Isolation of R. fragariae from feeder roots was variable, but was greater from feeder roots on perennial than from structural roots. Isolation of the fungus was greater from structural roots with nematode lesions than from non-symptomatic roots. Rhizoctonia fragariae was a common resident on the sloughed cortex of healthy perennial roots. From this source, the fungus may infect additional roots. The direct effects of lesion nematode feeding and movement are cortical cell damage and death. Indirect effects include discoloration of the endodermis and early polyderm formation. Perhaps weakened or dying cells caused directly or indirectly by P. penetrans are more susceptible to R. fragariae, leading to increased disease.  相似文献
