缺氧条件下冻伤对大鼠微循环液血灌流量的影响   总被引:2,自引:0,他引:2  
本文采用体重200±20g健康雄性Wistar大鼠,随机分为平原冻伤(FN)组,急性缺氧冻伤(FAH)组和缺氧习服缺氧冻伤(FHAC)组,实验观察了大鼠右后肢重度冻伤前后各组大鼠双后肢皮肤微循环灌流量的改变,结果表明,平原冻伤使大鼠以后肢微循环灌流量明显减少,提示局部重度冻伤对微循环的影响不只局限于冻区也涉及到对侧肢体,冻冻前FAH组大鼠微循环灌流量已明显低于FN组,表明生缺氧时血容量进行代偿性的  相似文献

目的:探讨急性低氧对血管内皮细胞(VEC)和循环血液中内皮细胞(CEC)的影响.方法:用光镜观察急性低氧情况下血中CEC的形态并记数,用组织化学染色和显微分光光度计对血中年轻型CEC及肺小动脉VEC的乳酸脱氢酶(LDH)活性进行定量分析.结果:与低氧前相比,低氧后各组VEC的LDH活性均下降,且随低氧时间延长,LDH活性下降越明显.而低氧前后各组年轻型CEC的LDH活性是一致的,且明显低于各组VEC的LDH活性.低氧前CEC多为衰老型,低氧后CEC数量增多且多为年轻型,随低氧时间延长年轻型CEC增多越明显.结论:CEC的形态和数量的变化可以反映血管的损伤情况;VEC的LDH活性可以反映VEC向CEC转化的进程;年轻型CEC的LDH活性可以反映VEC脱落时的损伤程度.  相似文献

The lipid composition of the lung plasmatic membrane in rats which have been under the acute emotional pain stress action is studied. These results are compared with the control group of animals. It is shown that at acute stress the changes of lipid composition of the lung plasmatic membranes are manifested in decrease the phospholipids and increase of cholesterol levels. The correlation of phospholipids/cholesterol in plasmic membranes in the lungs decreases at stress. At the same time the decrease of triglyceroles and diglyceroles contents is observed as well as the increase of fat acids' number. The changes that take place in the lipid contents of the lung plasmatic membranes at acute stress can play an essential role in the mechanism of cell damage development.  相似文献

Rats were exposed to a simulated altitude of 25,000 ft for 4 h in a decompression chamber, and the activity of some tissue enzymes estimated. Succinate dehydrogenase activity was significantly decreased and cholinesterase activity significantly elevated in the brain homogenates of the hypoxic rats, succinic dehydrogenase activity was significantly increased. There was no change in the activity of Mg+2-ATPase and Na+-K+-ATPase in the microsomal fractions of liver or brain homogenates of the hypoxic animals.  相似文献

The membrane activity of Na+, K(+)-ATPase, Mg2+, Ca(2+)-ATPase, mitochondrial NAD-isocitrate dehydrogenase, mitochondrial and cytosolic L-glycerol-3-phosphate dehydrogenase was determined in the liver and brain of Wistar rats under acute hypoxic hypoxia against the background of preventive taurine administration. It was shown that preliminary taurine treatment prevented a decrease of hypoxia in activity of Na+. K(+)-ATPase and mitochondrial calcium-dependent enzymes, mostly in the liver. Changes in the intracellular calcium content and biomembrane structure have been discussed as the mechanisms of the taurine effect on the enzymes' activity.  相似文献

目的:观察急性间歇性低氧刺激后大鼠颈动脉体对低氧的敏感性以及多巴胺对颈动脉体低氧敏感性的影响。方法:将分离SD大鼠的颈动脉体-窦神经移入到孵育槽,然后把分离的窦神经吸入到记录的玻璃电极中行电信号记录。记录基线部分缓冲液充入气体为95%O2+5%CO2混合气,低氧应激给予5%O2+5%CO2+90%N2混合气,低氧刺激给予30 s,95%O2+5%CO2给予90 s,共10个循环,每组实验大鼠数量n大于等于5。结果:大鼠离体的颈动脉体,给予急性间歇性低氧应激,再给予低氧刺激,窦神经较之前低氧刺激放电活动增强。但加入多巴胺后,可以抑制窦神经对低氧的反应,急性间歇性低氧后,多巴胺对窦神经的低氧放电活动抑制作用加强。结论:大鼠颈动脉体给予急性间歇性低氧可增强窦神经对低氧的反应,多巴胺可抑制急性低氧诱导的颈动脉体对低氧敏感性的增强。  相似文献

Enzymatic activities were determined in the prosencephalon of rats which had been exposed repeatedly for 8 hrs, either from the age of 1 to 17 days or in adulthood, to an altitude of 7,000 m in a barometric chamber (up to a total of 104 hrs). The activity of enzymes was assayed 20 hrs after the last exposure. The results were compared with values obtained 20 hrs after a single 8-hour exposure to a stimulated altitude of 7,000 m in 17-day-old and adult rats. In young rats a single exposure to hypoxia, the most elevated was the activity of LDH and isoCDH, while GPT and CPK were decreased. After repeated hypoxia, the most significant increasw was noted in the activity of PK and again a decrease in GPT and CPK. In adulthood, a single exposure to hypoxia causes the greatest increase in CPK, while LDH and GOT are reduced. Following repeated hypoxia, none of the enzyme activities were increased by more than 20%, while LDH, GOT, GIDH and CPK were again lower. It is concluded from these results that a single exposure to hypoxia increases anaerobic glycolysis in the immature nervous tissue and improves its oxygen utilization. The relationship between glycid and amino acid metabolism are not appreciably altered. On the other hand, a general reduction of bioenergetics and biosynthesis occurs in adulthood. Repeated hypoxia, on the contrary, has similar after-effects in both age groups, namely inhibition of aerobic metabolism and of the relations between glycid and amino acid metabolism.  相似文献

A comparative study of renin and erythropoietin content in the blood serum of rats with "endocrine" kidneys and changes in their activity following the action of specific erythropoietic stimulus (4-hour hypoxia) has been conducted. The presence of "endocrine" kidneys increased renin and erythropoietin activity in such animals. Acute hypoxia produced further increase in erythropoietin titre in the blood serum, with renin remaining at the same level. Possible differences in the mechanisms of renin and erythropoietin biogenesis in the kidneys are considered.  相似文献

The aim of the present work was to investigate the mechanisms of oxidative damage of the liver mitochondria under diabetes and intoxication in rats as well as to evaluate the possibility of corrections of mitochondrial disorders by pharmacological doses of melatonin. The experimental (30 days) streptozotocin‐induced diabetes mellitus caused a significant damage of the respiratory activity in rat liver mitochondria. In the case of succinate as a respiratory substrate, the ADP‐stimulated respiration rate V3 considerably decreased (by 25%, p < 0·05) as well as the acceptor control ratio (ACR) V3/V2 markedly diminished (by 25%, p < 0·01). We observed a decrease of the ADP‐stimulated respiration rate V3 by 35% (p < 0·05), with glutamate as substrate. In this case, ACR also decreased (by 20%, p < 0·05). Surprisingly, the phosphorylation coefficient ADP/O did not change under diabetic liver damage. Acute rat carbon tetrachloride‐induced intoxication resulted in considerable decrease of the phosphorylation coefficient because of uncoupling of the oxidation and phosphorylation processes in the liver mitochondria. The melatonin administration during diabetes (10 mg·kg‐1 body weight, 30 days, daily) showed a considerable protective effect on the liver mitochondrial function, reversing the decreased respiration rate V3 and the diminished ACR to the control values both for succinate‐dependent respiration and for glutamate‐dependent respiration. The melatonin administration to intoxicated animals (10 mg·kg−1 body weight, three times) partially increased the rate of succinate‐dependent respiration coupled with phosphorylation. The impairment of mitochondrial respiratory plays a key role in the development of liver injury under diabetes and intoxication. The resistance of male Wistar rats to acute hypoxia was estimated from the lifetime at an "altitude" of 11.5 km above sea level from 13 to 21 p.m. and in different seasons of a year. Identical types of rhythms (circaseptan, circasemiseptan, infradian, circadian, and ultradian) of geomagnetic activity and lifetime were revealed. It was found that the periods of basal rhythms either coincide or are multiple. It was shown that the rhythms of geomagnetic activity (including nonbasal rhythms) affect lifetime rhythms (especially ultradian rhythms). As the periods of the rhythms decrease, the number of rhythms for both parameters increase (ultradian rhythms are most numerous), and relative differences in the values of periods (they are minimal for ultradian rhythms) and the amplitudes of rhythms decrease.  相似文献

The erythrocytes represent an important source of antioxidant capacity of the blood. Catalase (EC 1.11.1.6.) is one of the enzymatic components of their antioxidant defense system. The objective of this study was to follow erythrocyte catalase (CAT) in 7-, 15-, 21-, 35-, 60- and 90-day-old Wistar rats of both sexes in normoxia and after exposure to intensive acute hypobaric hypoxia. During the development CAT activity increases in both sexes, but the rise was usually higher in females. Hypobaric hypoxia increased CAT activity in all studied age groups of both sexes. However, higher CAT activity in females was less affected by hypoxia than the lower activity in males. This was true for nearly all age groups studied. It can be concluded that both ontogenetic aspects and sex differences play a major role in establishing the activity of CAT, which is an important part of the antioxidant defense of the organism.  相似文献

Hypoxia in the neonate is known to alter the activity of hepatic and pancreatic enzymes involved in lipid and carbohydrate metabolism. The purpose of this study was to evaluate the effect of neonatal hypoxia on the activity of intestinal enzymes, and to determine whether the administration of glucocorticoids to neonates can mimic the effects of hypoxia. Hypoxia in neonatal rats (0-7 days) increased protein content, and lactase and maltase activity in the duodenal and the jejunal segments of the small intestine compared with normoxic controls. Hypoxia in juvenile rats (28-35 days) did not change these enzymes. Two weeks after returning hypoxic (0-7 days) pups to normoxia, their body weight remained lower than the age-matched controls. In the group recovering from hypoxia, sucrase, maltase, and leucine aminopeptidase activities were lower in the duodenal and the jejunal segment. Compared with controls, LDH activity was lower only in the jejunal intestine in the group recovering from hypoxia. All enzyme activities returned to control levels 3 weeks after recovery. Neonatal rats treated with dexamethasone had a decrease in body weight, but increases in sucrase and maltase activity in both the duodenal and the jejunal segment. Hypoxia in newborn rats caused a delayed maturation of small intestinal enzymes. Increases in serum glucocorticoids after hypoxic exposure probably do not play a major role in the delayed maturation of the disaccharidase activity in the small intestine.  相似文献
