Background:

Holothuroids (sea cucumbers) are members of the phylum echinodermata, which produce saponins. Saponins exhibit a wide spectrum of pharmacological and biological activities. In this study, we isolated the crude saponins from the body wall of the dominant Iranian species of sea cucumber, Holothuria leucospilota (H. leucospilota). The purpose of this study was to confirm the presence of saponins in the Persian Gulf H. leucospilota and study the hemolytic and cytotoxic activities of these compounds.

Methods:

The body wall of sea cucumber was dried and powdered and the crude saponins were isolated using various solvents. The crude saponins were further purified by column chromatography using HP-20 resin. The foam test, Thin Layer Chromatography (TLC), hemolytic assay, and Fourier Transform Infrared Spectroscopy (FTIR) confirmed the presence of saponins. Cytotoxicity was analyzed using a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay on A549 cells, a human lung cancer cell line.

Results:

The foam test, hemolytic assay, and TLC supported the presence of saponin compounds in the 80% ethanol fraction of H. leucospilota. The infrared (IR) spectrum of the extract showed hydroxyl (-OH), alkyl (C-H), ether (C-O) and ester (–C=O) absorption characteristic of teriterpenoid saponins. The C-O-C absorption indicated glycoside linkages to the sapogenins. The crude saponin extracted from sea cucumber was cytotoxic to A549 cells.

Conclusion:

The 80% ethanol fraction of saponin isolated from H. leucospilota exhibited hemolytic activity and offers promise as an anti-cancer candidate.Key Words: Sea cucumber, Holothuria leucospilota, Saponin, Hemolytic assay, Cytotoxicity assay  相似文献

The suggestion that saponins in eggs and larvae of Acanthaster planci (L.) serve as chemical defences was tested by feeding groups of planktivorous pomacentrid fish with random series of gelatin food particles, some with and some without crude saponin extract from A. planci. The four fish species discriminated at statistically significant levels against food particles with crude saponin extract at 1× 10?3 and 1× 10?5 parts of wet weight. Three species also discriminated at significant levels against particles with 1× 10?7 parts crude saponin extract per wet weight. Degree of discrimination was strongly influenced by the state of hunger of the fish. Tastiness of particles containing saponins also influenced acceptability. The lower two concentrations of saponins used in these feeding trials were respectively two and four magnitudes less than in A. planci eggs and larvae. Thus, the saponins in eggs and larvae of A. planci are at levels detectable by, and unpalatable to, planktivorous fish and they account, at least in part, for the observed rejection of these early developmental stages by planktivorous fish.  相似文献

Lantana seeds contain 70.6 g crude protein, 12.8 g ether extract, 103.1 g crude fibre, 761.0 g nitrogen-free extract, 52.5 g total ash, 8.4 g calcium and 2.5 g phosphorus/kg dry matter. The replacement, by lantana seeds, of 200 g maize/kg of concentrate mixture for lambs was studied in two experiments lasting 3–4 months. The crude protein concentration in the lantana ration was lower than in the maize, and significantly (P<0.01) so in the second experiment. In the first experiment, the digestibility and intakes of dietary crude protein (DCP) and total dietary nitrogen (TDN) of the lantana group were not significantly lower. In the second experiment, dry matter and crude protein digestibility and intakes of DCP and TDN of the lantana group were significantly (P<0.01) lower than the maize group. Even after adjustment for mean protein concentration in the diets, the intakes of DCP (P<0.01) and TDN (P<0.05) in the lantana group were significantly lower. In a repeat metabolic trial also, the adjusted intakes of both DCP and TDN (P<0.01) of the lantana group were significantly lower. In spite of the depression in feed utilization, the biochemical and haematological parameters of the blood of animals in the lantana group were within the normal range. The apparent non-toxicity of lantana seeds to lambs over long-term feeding trials calls for further work on their possible utilization as a feed ingredient.  相似文献

Herbal medicinal products have proven to be safe, economical and effective alternatives to synthetic chemical pharmaceuticals. The green tea plant (Camellia sinensis) is of profound medicinal value due to the presence of potent bioactive constituents. The purpose of the present work is to investigate saponins from green tea seeds for potential use as anti-angiogenic, antimicrobial, and hemolytic agents. Green tea seed saponins were separated into six fractions by reverse phase HPLC. The presence of three aglycone chains in the saponins of each fraction was confirmed by acid hydrolysis. Anti-angiogenic activity was evaluated using saponin fractions at concentrations in the range of 2.5 ~ 25 μg/mL. Antimicrobial activity was evaluated by thin-layer chromatography (TLC) using E. coli; S. mutans, a zoonotic Salmonella species and the fungal strain, A. niger. Saponin fractions were more potent against E. coli (gram negative bacteria) than against S. mutans (gram positive bacteria) and strongly inhibited six strains of zoonotic Salmonella. Green tea saponins also showed potent anti-angiogenic effects. All saponin fractions exhibited hemolytic activity. Our results confirm that green tea saponins have antimicrobial, anti-angiogenic, and hemolytic activities; indicating their potential as natural pharmaceutical products.  相似文献

Progenin III, one of the most active spirostanol saponins, is a potential candidate for anti-cancer therapy due to its strong antitumor activity and low hemolytic activity. However, the concentration of progenin III is extremely low in natural Dioscorea plants. In this paper, the progenin III production from total steroidal saponins of Dioscorea nipponica Makino was studied using the crude enzyme from Aspergillus oryzae DLFCC-38. The crude enzyme converting total steroidal saponins into progenin III was obtained from the A. oryzae DLFCC-38 culture. For enzyme production, the strain was cultured for 72 h at 30 °C with shaking at 150 rpm in 5 % (w/v) malt extract medium containing 2 % (v/v) extract of D. nipponica as the enzyme inducer. The crude enzyme converted total steroidal saponins into major progenin III with a high yield when the reaction was carried out for 9 h at 50 °C and pH 5.0 with the 20 mg/ml of substrate. In the preparation of progenin III, 117 g of crude progenin III was obtained from 160 g of substrate, and the crude product was purified with silica gel column to obtain 60.3 g progenin III of 93.4 % purity.  相似文献

The effect of the crude extract of Artocarpus lakoocha (70% composition is 2,4,3′,5′- tetrahydroxystilbene -THS) on adult Fasciola gigantica was evaluated after incubating the parasites in M-199 medium containing 250, 500, 750 and 1000 μg/ml of the crude extract, or triclabendazole (TCZ) at the concentrations of 80 and 175 μg/ml as the positive control, for 3, 6, 12 and 24 h, using relative motility (RM) assay and observation by scanning electron microscope (SEM). Decreased contraction and motility were first observed after 3 h incubation with TCZ at the concentration 80 and 175 μg/ml. TCZ markedly reduced the parasite’s motility at the concentration of 175 μg/ml at 6 h, and killed the worms after 12 h exposure. The crude extract of A. lakoocha at all concentrations reduced the parasite’s motility similar to TCZ at 3 h incubation. In 250 and 500 μg/ml of the crude extract, the values were decreased from 3 to 12 h, then they were stable between 12 and 24 h and reduced to the level approximately 30-40% of the control. At 750 and 1000 μg/ml concentrations the crude extract rapidly reduced the RM values from the start to 12 h and killed the parasites between 12 and 24 h incubation. The crude extract also inhibited the larval migration by 75% and 100% at the concentrations of 250-500 and 750-1000 μg/ml, respectively. TCZ and the crude extract caused sequentially changes in the tegument including swelling, followed by blebbings that later ruptured, leading to the erosion and desquamation of the tegument syncytium. As the result, lesion was formed which exposed the basal lamina. The damage appeared more severe on the dorsal than the ventral surface, and earlier on the anterior part and lateral margins when compared to the posterior part. The severity and rapidity of the damages were enhanced with increasing concentration of the crude extract. Hence, the crude extract of A. lakoocha, may exert its fasciolicidal effect against adult F. gigantica by initially causing the tegumental damage.  相似文献

Nostoc sp. BHU001, a planktonic cyanobacterium isolated from an agricultural pond in India, was examined for its toxicity. Mice, administered intraperitoneally with Nostoc sp. BHU001 crude extract (50 mg kg?1 body weight) died at 4.5 h. Examination of liver and spleen showed microcystin (MC)-like symptoms. Serum enzyme aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities increased by 1.6–1.8 and 2.6–3.0-folds, respectively at 50 and 100 mg crude extract kg?1 body weight. Thin layer chromatography of the crude extract produced five bands (N-1 to N-5). UV absorption maxima of band N-4 corresponded to that of standard microcystin-LR. Further analysis of the band N-4 by high-performance liquid chromatography gave a retention time (R t ) of 4.61 min similar to that of standard microcystin–LR (LR stands for lysine and arginine). Total MC content was quantified by enzyme-linked immunosorbent assay, and was 189.9 μg g?1 of crude extract, 9.8 μg l?1 of spent medium and 5.5 μg l?1 of pond water. Exposure of rice (Oryza sativa var. Sonam) seeds to the crude extract did not affect their germination, but inhibited the root and shoot growth of seedlings by 27.3 and 42.89 folds at 3 mg ml?1 crude extract, respectively.  相似文献

A spirostane with an attached trisaccharide, (25R)-5α-spirostane-2α,3β,5α-triol 3-O-(O-α-l-rhamnopyranosyl-(1 → 2)-O-(β-d-galactopyranosyl-(1 → 3))-β-d-glucopyranoside), was isolated and identified from the aerial parts of Agapanthus africanus by activity-guided fractionation. Fungicidal properties of the crude extract, semi-purified fractions as well as the purified active saponin from A. africanus were screened in vitro against Fusarium oxysporum. At a concentration of 1 mg mL?1, the crude extract and semi-purified ethyl acetate and dichloromethane fractions showed significant antifungal activity. The purified saponin inhibited the in vitro mycelial growth of F. oxysporum completely (100 %) at a concentration of 125 µg mL?1. Furthermore, to verify previously observed induced resistance by crude extracts of A. africanus towards leaf rust, intercellular PR-protein activity was determined in wheat seedlings following foliar application of the purified saponin at 100 µg mL?1. In vitro peroxidase enzyme activity increased significantly (60 %) in wheat seedlings 48 h after treatment with the purified saponin, demonstrating its role as an elicitor to activate a defence reaction in wheat.  相似文献

Fenugreek (Trigonella foenum-graecum L. seed) is a food with traditional medicinal use in diabetes. Beneficial effects have been demonstrated in diabetic animals and both insulin-dependent and non-insulin-dependent diabetic subjects. Effects of a lipid extract A, crude ethanolic extract B, further sub-fractions of B (saponin-free C, saponin D and sapogenin E) and a gum fibre fraction F on intestinal sodium-dependent glucose uptake were investigated in vitro using rabbit intestinal brush border membrane vesicles. All fractions except A inhibited glucose-uptake at 0.33 and/or 3.3mg/mL (p < 0.001). Greatest inhibition was observed with fractions D and E. Diosgenin and trigonelline (compounds reported in fenugreek)also inhibited glucose-uptake (50C values approximately 3mg/ ml, equivalent to 8mM and 19mM respectively) but did not account for the activity of the crude extracts. Fenugreek extracts had no effect on basal levels of glycogen phosphorylase a (HGPa) activity in rat hepatocyte suspensions. However fractions C and E caused a marginal but statistically significant inhibition (18.9 and 15.1% respectively, p < 0.05) of glucagon induction of this enzyme suggesting a glucagon-antagonist effect. Diosgenin (1.65mg/ml; 4mM) inhibited glucagon-induced HGPa activity by 20% (p < 0.05), and was more effective than trigonelline (non significant inhibition of 9.4% at 1.65mg/ml, 10mM).  相似文献

The present work reports on the biological activity of alfalfa (Medicago sativa) saponins on white poplar (Populus alba, cultivar ‘Villafranca’) cell suspension cultures. The extracts from alfalfa roots, aerial parts and seeds were characterized for their saponin content by means of thin layer chromatography (TLC) and electrospray ionisation coupled to mass spectrometry. The quantitative saponin composition from the different plant extracts was determined considering the aglycone moieties and determined by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS) analyses. Only soyasapogenin I was detected in the seed extract while several other saponins were found in the root and leaf extracts. Actively proliferating white poplar cell cultures were challenged with the different saponin extracts. Only alfalfa root saponins, at 50 µg ml?1, induced significant cell death rates (75.00 ± 4.90%). Different cell subpopulations with peculiar cell death morphologies were observed and the programmed cell death (PCD)/necrosis ratio was reduced at increasing saponin concentrations. Enhancement of nitric oxide (NO) production was observed in white poplar cells treated with root saponins (RSs) at 50 µg ml?1 and release of reactive oxygen species (ROS) in the culture medium was also demonstrated. Saponin‐induced NO production was sensitive to sodium azide and NG‐monomethyl‐l ‐arginine, two specific inhibitors of distinct pathways for NO biosynthesis in plant cells.  相似文献

Fifteen alfalfa populations were tested for resistance to the seedling damping-off disease sourced by Rhizoctonia solani, Fusarium solani, and Macrophomina phaseolina. In a laboratory experiment, saponin treatment significantly diminished the mycelial growth of the causal fungi of alfalfa damping-off disease. Roots of the fifteen alfalfa populations varied in saponin and lignin content. Selection for the considerably resistant plants leads to the best growth performance, desirable yield, and high nutritive values such as crude protein (CP), crude fier (CF), nitrogen free extract (NFE), ash, and ether extract (EE) contents. For the PCR reaction, 10 SSR pairs of the JESPR series primers and the cDNA-SCoT technique with seven primers were used. SSR and SCoT revealed some unique markers that could be linked to resistance to damping-off disease in alfalfa that appeared in the considerably resistant alfalfa population (the promised pop.). SSR and SCoT markers can be an excellent molecular method for judging genetic diversity and germplasm classification in tetraploid alfalfa. We recommend breeding for saponin concentration in the alfalfa plant may affect resistance to some diseases like root rot and damping-off because saponin might improve plant growth, yield, and nutritional values.  相似文献

The extracts prepared from various areal parts of the Adenium obesum (Forssk.) Roem. & Schult. (Family: Apocynaceae) including leaves, fruit and seeds ethanolic extracts and seed aqueous extract were evaluated against MCF-7 cells in order to investigate its potential of cytogenotoxicity and induction of apoptosis. The ethanolic seeds extract had comparatively higher cytotoxicity (IC50?~?337?µg/ml). Further, apoptosis and DNA damaging potential of seeds ethanolic extract was analyzed by applying multiple sub-lethal concentrations and durations. Flow cytometry results revealed that maximum percentage of early apoptosis (37%) and late apoptosis (35%) were observed after 12?h exposure in concentrations 200?µg/ml and 300?µg/ml, respectively. Similarly, the higher effect of extract in terms of DNA damage by alkaline comet assay was registered after 12?h treatment at concentrations 200 and 300?µg/mL. The calculated total damage score (TDS) for these concentrations were 614 and 617, respectively. The above findings indicate that A. obesum ethanolic seeds extracts has cytogenotoxic properties that could be further explored for the potential source of chemotherapeutic lead against cancer.  相似文献

Background

The extract from Moringa oleifera seeds is used worldwide, especially in rural areas of developing countries, to treat drinking water. M. oleifera seeds contain the lectins cmol and WSMoL, which are carbohydrate-binding proteins that are able to reduce water turbidity because of their coagulant activity. Studies investigating the ability of natural products to damage normal cells are essential for the safe use of these substances. This study evaluated the cytotoxic and anti-inflammatory properties of the aqueous seed extract, the extract used by population to treat water (named diluted seed extract in this work), and the isolated lectins cmol and WSMoL.

Methodology/Principal Findings

The data showed that the aqueous seed extract and cmol were potentially cytotoxic to human peripheral blood mononuclear cells, while WSMoL and diluted seed extract were not cytotoxic. The M. oleifera aqueous seed extract and the lectins cmol and WSMoL were weakly/moderately cytotoxic to the NCI-H292, HT-29 and HEp-2 cancer cell lines and were not hemolytic to murine erythrocytes. Evaluation of acute toxicity in mice revealed that the aqueous seed extract (2.000 mg/kg) did not cause systemic toxicity. The aqueous seed extract, cmol and WSMoL (6.25 µg/mL) and diluted seed extract at 50 µg/mL exhibited anti-inflammatory activity on lipopolyssaccharide-stimulated murine macrophages by regulating the production of nitric oxide, TNF-α and IL-1β. The aqueous seed extract reduced leukocyte migration in a mouse model of carrageenan-induced pleurisy; the myeloperoxidase activity and nitric oxide, TNF-α and IL-1β levels were similarly reduced. Histological analysis of the lungs showed that the extract reduced the number of leukocytes.

Conclusion/Significance

This study shows that the extract prepared according to folk use and WSMoL may be non-toxic to mammalian cells; however, the aqueous seed extract and cmol may be cytotoxic to immune cells which may explain the immunosuppressive potential of the extract.  相似文献

BackgroundThe antioxidant potential and antiproliferative activity of the extracts of Nigella sativa seeds (Black Cummins) and honey formulations are to be explored.MethodThe gas chromatography-mass spectrum (GC–MS) and Thin Layer Chromatography (TLC) fingerprint of Black Cummins and Honey formulation revealed alkaloid, saponin, volatile oil, flavonoid, glycosides, sugar, and phenolic compound in the extract. GC–MS profiling of the cold extract of Nigella sativa seeds and honey formulation shows peaks for eleven fractions of compounds. Using TLC, the phenolic compounds of Nigella sativa seeds and honey formulations were separated.ResultsThe current study discovers the cytotoxic effect of black Cummins seeds and honey formulation on human ovarian cancer (PA-1) cell line as assessed by MTT assay. PA-1 cells were inhibited with the increasing concentration of Nigella sativa seeds extract and honey formulation.ConclusionThe study validates the importance of the tested extracts in the treatment of cancer.  相似文献

Studies on the seeds of Annona squamosa yielded a novel lipoxygenase inhibitor fatty acid ester, (+) - annonlipoxy (1). Compound 1 was screened for its enzyme inhibitory activity against lipoxygenase (E.C.1.14.18.1), exhibiting activity with IC50 69.05 ± 5.06 μm. Baicalein (IC50 22.6 ± 0.5 μm) was used as a positive control. Crude extracts of Annona squamosa fruit pulp and seeds were screened for its enzyme inhibitory activity against lipoxygenase and acetylcholinesterase. The crude ethanolic extract of fruit pulp and seeds of Annona squamosa also exhibited lipoxygenase activity with 22.2 and 26.7% inhibition, while the pet.ether extract of seeds of A. squamosa exhibited 52.7% inhibition at a concentration of 40 μg/200 ml. The crude ethanolic extract of seeds of Annona squamosa was also bioassayed for acetylcholinesterase inhibition and it was found inactive.  相似文献

Megagametogenesis, the development of a megaspore into an embryo sac, has been identified in the seagrass Halophila johnsonii, a threatened species with no known sexual reproduction or seeds. Megagametogenesis in H. johnsonii was compared with megagametophyte development in Halophila decipiens, a related species known to readily produce viable seeds. In both species, ovules were structurally similar, megaspore mother cells were seen in premeiotic ovules, and linear tetrads and megagametophytes with two to eight nuclei were present in postmeiotic ovules. However, H. decipiens postmeiotic ovules had a chalazal pouch that was absent in the postmeiotic ovules of H. johnsonii. Late-stage H. decipiens ovules also contained embryos, indicating that they had been fertilized, whereas all late-stage H. johnsonii ovules were degrading and showed no signs of fertilization. These observations suggest that meiosis does occur in H. johnsonii megasporocytes, leading to the formation of viable megagametophytes and egg cells that could be fertilized if pollination occurred. Thus, the lack of seed set is due to a lack of pollination rather than any loss of capacity to produce seeds in this species.  相似文献

Fusarium moniliforme was grown on a carob aqueous extract in a chemostat for fungal protein production. The substrate was adjusted to provide 0.5% carob sugars supplemented with inorganic salts. The dilution rate varied from 0.086 to 0.227 hr?1 under constant conditions of temperature (30°C), pH (4.5), and oxygen saturation (60–80%). A yield of 0.709 g dry mycelium/g consumed carob sugar and a productivity value of 0.687 g dry mycelium/liter hr?1 were obtained at μ = 0.205 hr?1. The maintenance coefficient was 0.077 g carob sugar/g dry mycelium hr?1. While the carbohydrate and purine content of dry mycelium increased at μ values from 0.114 to 0.205 hr?1 both true (Lowry) and crude (N × 6.25) protein contents decreased at the same μ range. Maximum values of 36.3% true and 47.9% crude protein of dry mycelium were obtained at μ = 0.114 hr?1, whereas a minimum purine content of 99.8 μmol/g corresponding to 6.42% nucleic acids was recorded at μ = 0.086 hr?1. It was concluded that a continuous fermentation of carob aqueous extract using F. moniliforme should be operated at growth rates of approximately 0.205 hr?1 in order to maximize protein production.  相似文献

Abstract

Soyasapogenols, aglycones of soyasaponins, can be produced from crude soybean saponin extract by acid or enzymatic hydrolysis. Soyasapogenol B is known to have hepatoprotective, antimutagenic, antivirus, and anti-inflammatory activities. Hydrolysis of soyabean saponin extract for 72 h with 2 M HCl in methanol gave three soyasapogenols, namely: soyasapogenol D, soyasapogenol B1 and soyasapogenol A. However, the microbial hydrolysis of soybean saponin extract by Aspergillus terreus led to isolation of soyasapogenol B as a major product. A systematic evaluation of the effect of key operational parameters on the microbial transformation was performed. Maximum production of soyasapogenol B (about 152.3 mg/50ml) was observed using 1.5% (w/v) soybean saponin and 1.5% (w/v) glucose, 32°C after 72 h at pH 7 using phosphate buffer. Under these optimal conditions, the cells’ bioconversion efficiency increased from 20.5 to 85.3%. The isolation of soyasapogenols was performed using chromatographic methods and their structures identified on the basis of spectroscopic tools.  相似文献

A new glycosylated triterpene 1 was identified as 3-O-[β-d-xylopyranosyl-(1→3)-α-l-rhamnopyranosyl-(1→4)-β-d-glucopyranosyl]-11-methoxy-16-hydroxy-17-acetoxy hederagenin from an ethanolic extract of seeds of Nigella sativa Linn. Identification of the naturally acetylated saponin was based on chemical and spectroscopic analyses including FABMS, 1H, 13C, and 2D NMR and DEPT. The saponin was a penta hydroxy pentacyclic triterpene, in which one hydroxyl group was acetylated and other one was methylated naturally.  相似文献

Millepora complanata is a normal resident of coral reefs in the Mexican Caribbean. In this study, we describe for the first time the vasoconstrictor, phospholipase A2 (PLA2), and hemolytic activities elicited by a crude extract obtained from M. complanata. This extract caused a concentration-dependent contraction of isolated rat aortic rings (EC50=22.4+/-1.1 microg protein/mL). This effect was endothelium independent and significantly reduced in the absence of extracellular Ca2+ and when the intracellular Ca2+ stores were depleted. In addition, the crude extract obtained from M. complanata showed PLA2 activity (7.231+/-0.092 mmol min(-1) mg(-1)) and hemolysis of rat erythrocytes (HU50=1.64+/-1.04 mug protein/mL). The hemolysis increased in the presence of Ca2+ and decreased in the presence of cholesterol. Furthermore, this hemolysis was significantly reduced after incubation with an inhibitor of PLA2 enzymes. The hemolytic and vasoconstrictor effects were abolished after incubating the extract under denaturing conditions. Reverse phase chromatography of the M. complanata extract afforded 19 fractions (F1 to F19). F4 induced hemolysis and contained mainly a protein of 30 kDa, probably a PLA2 enzyme, while F8 and F11, containing mainly proteins of 15 and 20 kDa respectively, produced vasoconstrictor effects mediated by different mechanisms of action.  相似文献
