脱落酸作为一种抑制生长的植物激素,是平衡植物内源激素和调节生长代谢的关键因子。脱落酸具有提高作物抗旱耐盐、减少果实褐变的作用,同时可降低疟疾发病率、刺激胰岛素分泌,因此在农业和医药领域有着广阔的应用前景。相较于传统的植物提取法和化学合成法,利用微生物合成脱落酸是一种经济、可持续的来源方式。目前利用天然微生物如灰葡萄孢霉菌、蔷薇色尾孢菌等合成脱落酸的研究已经取得了诸多进展,而脱落酸的异源微生物合成研究相对较少。酿酒酵母、解脂耶氏酵母、大肠杆菌等工程菌株作为天然产物异源合成的常用宿主,具有遗传背景清晰、易于操作、便于工业化生产等优势,因此利用微生物异源合成脱落酸是一种更具潜力的生产方式。本文着重从底盘细胞的选择、关键酶的筛选与表达强化、辅因子的调节、增强前体供应及促进脱落酸外排5个方面对微生物异源合成脱落酸的研究进行综述。最后,对该领域的未来发展方向进行了展望。  相似文献

植物激素包括脱落酸、乙烯和多胺等可调节水稻幼苗的耐冷性.表明在5℃下,稻苗叶片和木质部液体中的脱落酸含量迅速增加;耐冷水稻品种的内源脱落酸含量显著高于感冷品种.用脱落酸预处理感冷水稻,48小时可增强其耐冷性.脱落酸的耐冷机理是可使稻株避免冷害引起的水胁迫;此外,脱落酸还可增加稻株中脯氨酸等溶质的积累,以防止冷害.脱落酸反应基因可在某些植物中表达,且可能与低温的习性有关.  相似文献

Phloem sap collected from Yucca and coconut inflorescence stalks was shown to contain abscisic acid (ABA) and trace amounts of 2-trans ABA. Replacing the 4'-carbonyl group of abscisic acid with a methoxy group does not affect the abscisic acid (ABA)-like activities of the product in barley aleurone protoplasts, although the reduction of ABA to 4'-hydroxyl derivatives significantly reduces the ABA-like activity of the products. We have developed a radioimmunoassay (RIA) for abscisic acid (ABA) in the 0.1 ng to 2.5 ng range. Antibodies were obtained from rabbits immunized with ABA bound via its carboxyl group to bovine serum albumin. Cross-reactivity studies indicate that ABA esters are completely cross-reactive with ABA, while trans, trans abscisic acid (t-ABA) phaseic acid (PA) and dihydrophaseic acid (DPA) have much lower but significant cross-reactivities. Abscisic acid (ABA), conjugated abscisic acid, phaseic acid (PA), and conjugated phaseic acid were determined by enzyme-linked immunosorbent assay (ELISA) and gas chromatography (GC) in xylem sap of well-watered and drought-stressed sunflower plants. Conjugated ABA and conjugated PA were determined indirectly after chemical or enzymatic hydrolysis. Conjugated ABA was found to be the predominant ABA metabolite in xylem sap. In xylem sap from well-watered plants at least five, and in sap from drought-stressed plants at least six alkaline hydrolysable ABA conjugates were found. One of them corresponds chromatographically (HPLC) with abscisic acid glucose ester (ABAGE). Under drought conditions the concentrations of ABA, alkaline hydrolysable ABA conjugates, -glucosidase hydrolysable ABA conjugates, PA, and conjugated PA increased. After rewatering the drought-stressed plants, the ABA and the conjugated ABA content decreased. The possible function of the ABA conjugates in the xylem sap as a source of free ABA is discussed.  相似文献

Aseptically cultured lateral buds of Salix viminalis L. collected from field-grown trees exhibited a clear periodicity in their ability to respond to exogenous abscisic acid (ABA). Buds were kept unopened by ABA only when the plants were dormant or entering dormancy. Short days alone did not induce bud dormancy in potted plants but ABA treatment following exposure to an 8-h photoperiod prevented bud opening although ABA treatment of buds from long-day plants did not. Naturally dormant buds taken from shoots of field-grown trees and cultured in the presence of ABA opened following a chilling treatment. A monoclonal antibody was used to localize abscisic acid in Lavandula stoechas L. plants treated with 1 M abscisic acid. ABA localization was performed using EDC as the only fixative, which preserves antigenicity and improves the specificity of monoclonal antibodies. A relationship was observed between the effect of abscisic acid on chloroplast ultrastructure and ABA accumulation. Gold particles accumulated on swollen chloroplasts. The effects of methyl jasmonate (MJ) and abscisic acid (ABA) on some physiological processes of rice were compared. MJ exhibited ABA-like effects by promoting senescence of detached leaves, by inducing acid phosphatase activity of detached leaves, by inhibiting ethylene production and shoot growth of seedlings, as well as inhibiting callus formation from anthers. However, MJ and ABA had opposite effects on 1-aminocyclopropane-1-carboxylic acid-dependent ethylene production in detached leaves. The regeneration ability of anther-derived callus was inhibited by MJ but not by ABA. MJ but not ABA markedly induced peroxidase activity in senescing detached leaves. A simple and rapid technique was developed to synthesize abscisic acid glucose ester. The free acid of abscisic acid (ABA) was combined with CsHCO3 to form the Cs salt of ABA. The Cs salt of ABA was then combined with acetobromo-α-d-glucose tetraacetate, and the tetraacetate derivative of abscisic acid glucose ester was formed. Acetate groups were selectively removed from the glucose moiety with a crude enzyme preparation derived from Helianthus annuus seeds. Abscisic acid glucose ester was purified via silica gel column chromatography and identified by micro NMR.  相似文献
