Seahorses, sea dragons and pipefishes of the teleost family Syngnathidae are unique in that embryos develop within specialized brooding structures of the male. We enriched brooding Syngnathus fuscus and Syngnathus floridae males with injections of L-lysine-[15N2] and 16:0-palmitic acid 1-[13C] to demonstrate embryonic uptake of paternally-derived nutrients. While all embryos demonstrated amino acid enrichment, late stages showed significantly higher [15N], indicating greater utilization of paternal resources as yolk reserves diminished and embryonic energy demands increased. Limited embryonic [13C] uptake, defined as less than 10% of adult enrichment, in 75 and 81% of S. floridae and S. fuscus respectively signified rapid lipid metabolism and thus the need for greater enrichment. Interspecific differences in embryonic uptake of paternally-derived nutrients were not demonstrated. However, interspecific differences in egg nutrient reserves and fry size but comparable fry nutrient levels along with data from a published paternal exposure study indicate paternal transfer in S. fuscus most likely compensates for the comparative egg nutrient deficiency. This study is the first to our knowledge to provide direct evidence for the functional significance of the brood pouch in nutrient provisioning. These results add comparative information on the diversity of Syngnathid paternal care and further our understanding of paternal influence on development.  相似文献

Analytical results for Aroclor may contain a great deal of uncertainty if the relative ratios of certain PCB congeners are substantially altered following differential environmental degradation. Selective degradation of PCB congeners has been shown to occur in biological tissues, and recent studies have shown that Aroclor analyses may significantly underestimate PCB concentrations in fish. The purpose of this study is to assess whether a similar phenomenon can occur with PCB-impacted sediments. Five hundred and thirty-seven sediment samples from the Passaic River were analyzed for Aroclors and selected non-ortho (PCB #77, 126, and 169) and mono-ortho (PCB #105, 114, 118, 123, 156, 157, 167, and 189) coplanar PCB congeners. Aroclors 1248 and 1254 were detected in 67 and 53% of the samples, respectively; 54 samples (approximately 10%) did not contain detectable levels of any Aroclors. In these 54 samples, the sum of the detected PCB congener concentrations was significantly greater, on average, than the Aroclor limit of detection by approximately threefold. In individual samples the sum of the PCB congeners exceeded the Aroclor limit of detection by up to 36-fold. In those samples in which Aroclors were detected, the summed PCB congener concentrations exceeded the Aroclor 1248 and 1254 concentrations, on average, by 41 and 33%, respectively. Given the fact that only a fraction of the 209 PCB congeners were quan-titated, these findings indicate that Arocolor data may significantly underestimate total PCB concentrations in Passaic River sediments. Total PCB data obtained from total homologue analysis indicated that Aroclor results underestimated total PCB mass by up to 43-fold. These findings suggest that caution is required when comparing Aroclor sediment data from the Passaic River to total PCB sediment criteria.  相似文献

Polychlorinated biphenyls (PCBs) are a group of persistent organochlorine compounds that have the potential to disrupt the homeostasis of thyroid hormones (THs) in fish, particularly juveniles. In this study, thyroid histology, plasma TH levels, and iodothyronine deiodinase (IDs, including ID1, ID2, and ID3) gene expression patterns were examined in juvenile Japanese flounder (Paralichthys olivaceus) following 25- and 50- day waterborne exposure to environmentally relevant concentrations of a commercial PCB mixture, Aroclor 1254 (10, 100, and 1000 ng/L) with two-thirds of the test solutions renewed daily. The results showed that exposure to Aroclor 1254 for 50 d increased follicular cell height, colloid depletion, and hyperplasia. In particular, hypothyroidism, which was induced by the administration of 1000 ng/L Aroclor 1254, significantly decreased plasma TT4, TT3, and FT3 levels. Profiles of the changes in mRNA expression levels of IDs were observed in the liver and kidney after 25 and 50 d PCB exposure, which might be associated with a reduction in plasma THs levels. The expression level of ID2 mRNA in the liver exhibited a dose-dependent increase, indicating that this ID isotype might serve as sensitive and stable indicator for thyroid-disrupting chemical (TDC) exposure. Overall, our study confirmed that environmentally relevant concentrations of Aroclor 1254 cause significant thyroid disruption, with juvenile Japanese flounder being suitable candidates for use in TDC studies.  相似文献

Vascular corrosion casts of Syngnathus floridae and Syngnathus fuscus brood pouches were examined by scanning electron microscopy. Morphological and quantitative data on the vasculature of the paternal brood pouch during each stage of embryonic development were investigated to explore potential changes during brooding, to consider interspecific differences and to provide structural evidence for previously reported functional roles of the brood pouch. The brood pouches of both species are highly vascularized structures with cup‐like arrangements of brood‐pouch vasculature developing around each embryo shortly after fertilization and breaking down before fry release. The density and size of paternally derived blood vessels in contact with the embryos were found to be consistent for S. fuscus once this structure was established early in development. On the contrary, these vasculature measurements varied with early S. floridae brood stages when the embryo still relied heavily on the yolk sac. Diameter measurements of S. fuscus brood‐pouch blood vessels were also comparatively smaller during these early developmental stages, suggesting that the structural stability and opportunity for greater transport via slower blood flow may contribute to greater paternal allocation. This is the first study to document changes in brood‐pouch vasculature during specific stages of embryonic development, to show regression of this vasculature before fry release and to provide morphological data for two syngnathid species for which information on brood‐pouch physiology is available.  相似文献

Atlantic croaker (Micropogonias undulatus) were exposed to the polychlorinated biphenyl (PCB) mixture (Aroclor 1254) or one of three individual congeners (planar PCB 77 or ortho-substituted PCB 47 and PCB 153) in the diet for 30 days to investigate the effects of PCBs on circulating thyroid hormones, thyroxine (T4) and triiodothyronine (T3). Aroclor 1254 (0.2 and 1.0 mg/kg body mass/day) decreased plasma T3 levels consistently, but the effects on T4 levels were inconsistent from year to year. Exposure to PCB 153 (0.1 and 1.0 mg/kg body mass/day) significantly lowered both T4 and T3, while PCB 47 at the same doses had no effect on thyroid hormone levels. The lower doses of PCB 77 (0.004, 0.01 and 0.02 mg/kg body mass/day) had no effect on T4 or T3, whereas the highest dose (0.1 mg/kg body mass/day) increased T4 levels significantly. The results of the present study demonstrate that exposure to PCBs at environmentally realistic concentrations can have profound effects on the thyroid status of Atlantic croaker. The ortho-substituted PCB 153 appears to contribute at least partially to the deleterious effects of Aroclor 1254 on thyroid status, whereas the planar PCB 77 at concentrations present in the mixture is unlikely to alter thyroid hormone levels.  相似文献

Accumulation and effects of polychlorinated biphenyls (PCBs) in avian species were evaluated at a Superfund site located at Crab Orchard National Wildlife Refuge, Illinois, and seven criteria were used to assess whether there was a causal relationship between PCB exposure and observed reproductive effects. European starlings (Sturnus vulgaris) were monitored at nest boxes constructed at each of two exposed and two reference sites. During the breeding season, starling productivity (number of chicks produced per nest) and adult nest attentiveness behavior (provisioning behavior) were monitored. At 15 days post-hatch, chicks were collected for contaminant and biomarker analyses. Chicks were necropsied, ethoxyresorufin-O-deethylase (EROD) activity measured in liver tissue, and PCB (Aroclor 1254) and 34 chlorinated biphenyl (CB) congener concentrations measured in carcasses using gas chromatography. PCB and CB concentrations also were measured in eggs that failed to hatch. Mean Aroclor 1254 and quantified CB concentrations were greater (P<0.001) in eggs that failed to hatch and 15-day-old chicks collected from PCB sites compared to those collected from reference sites. EROD activity was greater (P=0.005) in 15-day-old chicks collected from PCB sites and was correlated with carcass PCB concentrations. Reduced adult nest attentiveness behavior and decreased chick survival were observed at PCB sites. Six of the seven causal criteria evaluated provided evidence that observed reproductive effects resulted from exposure to PCBs. Using this weight-of-evidence approach provided a means for establishing the likely cause of effects and thus provided managers with information needed in decision-making processes.  相似文献

Polychlorinated biphenyls (PCBs) are stable, lipophilic compounds that accumulate in the environment and in the food chain. Though some studies provided evidence that PCBs had adverse effects on reproductive function, most of these results were from in vitro models. Therefore we investigated the effect of Aroclor 1254 (a commercial PCBs mixture) treatments on in vivo maturation and developmental potential of mouse oocytes. In the present study, female ICR mice were treated with different doses (12.5, 25 and 50 mg/kg) of Aroclor 1254 (a commercial PCB mixture) once every 72 hours by intraperitoneal injection for 9 days. After three treatments of Aroclor 1254, the mice were superovulated to collect oocytes one day after the last exposure. The effects of Aroclor 1254 on oocyte maturation, fertilization, and preimplantation embryonic development were investigated. Immunofluorescence-stained oocytes were observed under a confocal microscope to assess the effects of Aroclor 1254 on spindle morphology. Parthenogenic activation and the incidence of cumulus apoptosis in cumulus-oocyte complexes were observed as well. Oocytes exposed to different doses of Aroclor 1254 in vivo were associated with a significant decrease in outgrowth potential, abnormal spindle configurations, and the inhibition of parthenogenetic activation of ovulated oocytes. Furthermore, the incidence of apoptosis in cumulus cells was increased after exposed to Aroclor 1254. These results may provide reference for the treatment of reproductive diseases such as infertility or miscarriage caused by environmental contaminants.  相似文献

We evaluated the effects of two organochlorinated environmental contaminants, Endosulfan and Aroclor 1254 on peripheral thyroid hormone metabolism and thyroid hormone plasma levels in Nile tilapia (Oreochromis niloticus). Tilapia were exposed through diet to 0.1 and 0.5 microg g(-1) of Endosulfan and 0.5 microg g(-1) of Aroclor 1254 for 21 and 35 days. Decreased plasma T4 and rT3 levels were observed in tilapia exposed to the lower dose of Endosulfan, while treatment with a higher dose and Aroclor 1254 produced no changes. Plasma T3 levels were not affected by these compounds. Hepatic type I deiodinase (D1) activity was depressed by a lower dose of Endosulfan and hepatic type III (D3) activity was increased following 35 days of exposure to the lower dose of Endosulfan and following 21 and 35 days of exposure to Aroclor 1254; while type II (D2) remained unchanged in liver as well as in all other organs analysed. Apart from hepatic D3 activity, Endosulfan and Aroclor 1254 also increased D3 activity in gill, but not in other tested organs. It is concluded that dietary exposure of tilapia to Endosulfan or Aroclor 1254 can lead to changes in circulating thyroid hormone levels and/or in peripheral thyroid hormone metabolism. The changes in hormone metabolism differ between tissues, eventually reflecting tissue-specific differences in adaptation.  相似文献

Territorial defence by Brazilian damselfish Stegastes fuscus was studied in north-east Brazil through underwater observations. The fish mostly fed on Gellidium sp. and strongly defended a territory with a mean size of 2 m2 (range 1·27–3·07 m2); its mean ± s.d. feeding rate was 8·73 ± 2·44 bites min−1. Stegastes fuscus responded similarly to all intruding fish species, with aggressive behaviour becoming more pronounced only with an increase in the number of fishes in the sampled area. Analysis of attacks elicited by different fish species placed within S. fuscus ' territory, however, showed that it launched more attacks on conspecifics than on other fishes.  相似文献

The effect of oxygen and storage conditions on the metabolic activities, measured by volatile fatty acid (VFA) degradation and methane production, and by the dechlorinating activity of methanogenic polychlorinated biphenyl (PCB) granules, were studied. Incubation of the granules in air for different periods did not result in significant inhibition in volatile fatty acid degradation and methane production activities. The inhibitory effect of oxygen increased with increased length of exposure. The overall methanogenic activities, however, recovered after a 10-day incubation period in the absence of oxygen. Oxygen exposure did not cause any significant effect on the dechlorinating activity of the granules tested with a PCB mixture, Aroclor 1254. In 6 months, approximately 80% [based on the concentration (M) of chlorine removed] of the Aroclor 1254 was dechlorinated even by granules exposed to oxygen for 168 h. Granules stored at room temperature (20° C) appeared to be more active compared to the granules stored at 4° C C or –20° C. Similarly, granules stored with a nutrient mixture, containing methanol, glucose and yeast extract showed higher metabolic activities. Our results demonstrate that the effect of oxygen exposure was not significant and was reversible. PCB granules could be stored at room temperature with an auxiliary carbon source in the presence of PCB without significant loss of activity. These properties make methanogenic PCB granules suitable candidates for practical use in PCB dechlorination and biodetoxification.  相似文献

Polychlorinated biphenyls (PCBs) are one of the most persistent and widespread groups of endocrine disrupting compounds in the ecosystem. These substances are present in sewage sludge that is spread in increasing amounts on arable land and pasture as fertilizer, and are ingested by farm animals with food and drinking water. This study investigated the effect of different PCB concentrations on pig oocyte in vitro maturation and developmental competence as well as examined the possible mechanisms involved. A concentration ranging from 0 to 1 mg/mL of Aroclor 1254 (A1254), a pool of more than 60 PCB congeners, was added to the maturation medium, as its composition is considered environmentally relevant. A1254 had no effect on maturation of pig oocytes and on the number of oocytes that cleaved following parthenogenetic activation at any of the doses tested. By contrast, a significant decrease in the number of zygotes that developed to blastocyst stage became evident at a concentration of 10 ng/mL. The number of blastocysts obtained decreased significantly, and in a dose response manner with higher concentrations. Exposure to PCBs altered mitochondria relocation during maturation and this was associated with the lack of a cytoplasmic microtubule network. No effect on mitochondria activity was observed. A1254 exposure also perturbed gap-junction mediated communications between oocytes and cumulus cells. In conclusion, PCB exposure of pig oocytes during in vitro maturation significantly decreased oocyte developmental competence, altered both their cytoplasmic remodelling and the communication with the somatic compartment. These data indicated that accumulation of PCBs in the pig organism may have a detrimental effect on the reproductive efficiency in this species.  相似文献

The effect of egg vitamin A (VA) status and egg incubation temperature on the development of spinal disorders was investigated in Atlantic salmon Salmo salar fry. Atlantic salmon eggs were sorted into two groups with high VA (3·3 ± 0·1 μg retinol g−1 dry mass) and low VA (2·2 ± 0·3 μg retinol g−1 dry mass) status before fertilization and incubated at high (14° C) or low (8° C) temperature from 133 day degrees until the onset of feeding. High egg incubation temperatures increased the concentration of retinol in the eggs: the high VA and high temperature group displayed a significantly higher retinol concentration than the high VA and low temperature group ( P  = 0·001). After hatching, all experimental groups increased their retinol concentration. The source of the increased retinol levels was probably retinal, although astaxanthin may also be a VA precursor after hatching. Atlantic salmon fry incubated at high temperatures had increased amounts of notochord tissue. When measuring morphogenic activity in the notochord using the expression of sonic hedgehog ( shh , mRNA), however, no significant difference was found between the experimental groups. No clear effect of VA status or incubation temperature could be found on the formation of the early vertebral column although Atlantic salmon fry incubated at low temperatures had less regular constrictions of the prospective vertebral column than fry incubated at high temperatures.  相似文献

A preliminary laboratory investigation was conducted to understand the relative contributions of major dredge resuspension and residual processes on the releases of polychlorinated biphenyl (PCB) contaminants from sediments to water column. Sediments from New Bedford Harbor were used as test samples. Six sets of experiments were run for simulated resuspension and residual scenarios. During the experiments, water above the sediments was recirculated by peristaltic pumping or orbital shaking and the levels of two PCBs, Aroclor 1248 (PCB-1248) and Aroclor 1254 (PCB-1254), were monitored for 15 days. Analysis of the model predicted data indicated that resulting water column PCB concentrations differed with sediment surface, residual, and resuspension type. Highest PCB water column concentrations were observed for a condition which used a settled fluff from thin sediment slurry as a residual source and the column water was recirculated by orbital shaking. Lowest water column PCB levels were observed for a thick sediment deposit placed over clean sand. The PCB levels in the water column for all six simulated conditions were several orders higher than the USEPA ambient water quality criteria concentrations for aquatic environment and human consumption.  相似文献

Following exposure to a predator stimulus (a brown trout Salmo trutta ), the opercular rate of Atlantic salmon Salmo salar fry increased by 35·3 ± 11·0%(mean ± 95% CI). The time taken for opercular rate to decline to baseline levels depended upon the occurrence of any associated locomotory activity. Opercular rates of fish that dashed when exposed remained elevated for 38·2 ± 20·6 min, whereas those of individuals that did not move ('freezers') recovered within 7·2 ± 2·9 min. The duration that opercular rate remained elevated was positively correlated with the magnitude of the elevation, which was higher in 'dashers' than freezers. The maximum opercular rate in 'freezers' was similar between wild fry and hatchery‐reared fry (from wild parents). There was a significant delay, however, in hatchery compared with wild fry in the time until peak ventilatory response and onset in the decline phase. This difference in opercular response suggests that hatchery fish were slower to realize fully the potential danger from the predator. Any delay in response could be directly attributed to the effect of hatchery‐rearing environment, rather than domestication or hatchery selection effects.  相似文献

Exposure of mice for 8 weeks to drinking water containing diethylnitrosamine (DEN) was accompanied by alterations in hepatocyte structure and varying degrees of liver nonparenchymal cell (NPC) proliferation. Eighteen and a half weeks after cessation of DEN exposure, there was a 47% incidence of hepatocellular nodules. Centrilobular hepatocyte hypertrophy occurred consistently in mice given intraperitoneal injections of the polychlorinated biphenyl (PCB) mixture Aroclor 1254. PCB administration to mice previously treated with DEN was not accompanied by increases in gross liver nodule incidence above that induced by DEN, but many more developing microscopic nodules within the liver were observed in DEN-treated mice given Aroclor 1254 than in mice treated only with DEN. Aroclor 1254 administration over a 16-week period to mice previously treated with DEN was accompanied by an 83% incidence of severe distortion of liver structure resulting from nodule formation, uneven patterns of hepatocyte growth, and extensive deposition of scar tissue containing proliferating bile ducts. Morphological evidence of intestinal metaplasia was observed in proliferating bile duct-like structures during an early stage of liver adenofibrosis.  相似文献

Study of the dilate sea-water tolerance of the pipefish Syngnathus acusimilis indicated that it successfully acclimatized to an abrupt change in water salinity from 5 to 32‰. Larvae in the brood pouch also successfully acclimatized to salinity change, and, after several days, juveniles left the brood pouch and continued to live. In freshwater, adult and fry perished during several minutes. In the gill epithelium of S. acusimilis, chloride cells of only a marine type were found. A comparison of the ultrastructure of chloride cells of gill epithelium and mitochondrion-rich cells of the brood pouch of S. acusimilis demonstrated their high similarity. The ultrastructure of cells of the brood pouch indisputably attests to their involvement in maintaining an osmotic and ionic homeostasis in the cavity of the brood pouch.  相似文献

多氯联苯是一种典型的持久性有机污染物。研究表明多氯联苯具有毒物兴奋效应, 但其影响植物生长发育的机制尚不清楚。以毛白杨(Populous tomentosa)组培苗为材料, 探讨3 mg·L-1 Aroclor1254对不定根分化、植物激素水平、与生长素相关的P009g125900P006g142600P002g222700基因表达、与细胞分裂素相关的P005g2489P005g2376基因表达的影响。结果显示, Aroclor1254可促进毛白杨组培苗不定根分化, 缩短不定根初根时间与分化率达100%的时间, 提高不定根数目; 在不定根诱导期, 用Aroclor1254单独诱导, IAA/(ZR+dhZR)比值与阳性对照无显著差异, P006g142600P002g222700P009g125900P005g2489P005g2376基因表达变化趋势与IBA单独诱导下各基因表达变化趋势一致。为验证Aroclor1254是否具有生长素效应, 以玉米(Zea mays)和转生长素报告基因DR5::GUS的拟南芥(Arabidopsis thaliana)为材料, 观察Aroclor1254对胚芽鞘生长及DR5::GUS基因表达的影响。结果显示, 一定浓度的Aroclor1254对胚芽鞘的生长无显著影响, 但可诱导生长素报告基因表达。以上结果表明, 多氯联苯类化合物Aroclor1254虽不属于植物生长调节剂, 但具有毒物兴奋效应, 在一定浓度下具有类似生长素的生物学活性。  相似文献

The sub-chronic (28–56 days) effects of exposure to low concentrations of cadmium (Cd; 0·05, 0·25, 0·50 and 2·50 μg l−1) shortly following fertilization on embryos, larvae and juvenile rainbow trout Oncorhynchus mykiss were examined. Premature hatching occurred at lower concentrations (0·05 and 0·25 μg l−1 Cd), however, delayed hatching was seen in the 2·50 μg l−1 Cd group, with >90% of hatching occurring on the last day of the hatching period. Larval growth was negatively affected by Cd exposure in a concentration-dependent manner. Larvae exposed to 2·50 μg l−1 Cd were 13·9 ± 0·8% shorter in total length ( L T) and weighed 22·4 ± 3·5% (mean ± s . e .) less than controls at the end of the exposure period. Plasma sex steroid concentrations (oestradiol in juvenile females and 11-ketotestosterone in juvenile males) were elevated (four- to 10-fold over controls) in exposed fish in both males and females, following 28 days of exposure to 0·05, 0·25 and 0·50 μg l−1 Cd, respectively. These results suggest that environmentally realistic concentrations (in the μg l−1 range) of Cd can affect the development of O. mykiss impacting embryos, larvae and juvenile fish.  相似文献

The mutagenic activity of the pyrolysis products 2-amino-3-methylimidazo[4,5-f]-quinoline and 2-amino-3,4-dimethylimidazo[4,5-f]-quinoline in Salmonella typhimurium TA98 using rat intestinal and renal subcellular fractions as activation systems was approximately 1 and 5 revertants per nmol, respectively. This was 1,000 times less than the activity with a subcellular fraction from rat liver. The mutagenic activity of both compounds was considerably increased using intestinal, renal and hepatic preparations isolated from PCB (Aroclor 1254)-pretreated rats, compared to preparations from control animals. In addition, both compounds displayed a moderate direct-acting mutagenic activity at concentrations above 10-5 M. Isolated cells from small intestine, kidney and liver incubated in nucleopore chambers were able to convert both compounds into products which mutated bacteria outside the chambers. The concentrations of chemicals required to yield responses of a similar magnitude were approximately 3 orders of magnitude higher in the intestinal and renal systems compared to the hepatic system. Effects of 2 toxicants, Mirex and Aroclor 1254, on Tetrahymena pyriformis strain W in axenic cultures were investigated. Mirex is a chlorinated hydrocarbon effective against the fire ant, and Aroclor 1254 is a compound structurally related to DDT and used extensively in various industrial processes. Both toxicants reduced growth rates and population densities of T. pyriformis grown at 26 C generally in proportion to concentrations of the chemicals, their effects becoming statistically significant (P < 0.05) at 0.9 μg/liter for Mirex and 1.0 and 10.0 μg/liter for Aroclor 1254. Ciliates exposed to the toxicants for 7 days concentrated Mirex 193 × and Aroclor 60 × as compared to the initial concentrations of these compounds. It is suggested that the chief effect of the 2 toxicants on populations of T. pyriformis and of similarly responding ciliates in nature would be to reduce the availability of these protozoa as food organisms and nutrient regenerators. The ability of the ciliates to concentrate the tested compounds would permit the toxicants to enter into and to be translocated through aquatic food chains. In this manner the compounds could exert toxic effects at higher trophic levels.  相似文献
