The influence of short day and low temperature on cold acclimation of A. crispa (Ait.) Pursh, A. glutinosa (L.) Gaertn. and A. rubra Bong, was investigated. Two clones of each species originating from in vitro propagation were exposed to three daylength/temperature treatments. Periodically plantlets were exposed to controlled freezing temperature in order to evaluate their level of frost hardiness.
Short day (SD) and cold temperature (CT) and long day (LD) and cold temperature (CT) were the most effective treatments for the development of frost hardiness in shoots and roots of the three species tested. Short day (SD) and warm temperature (WT) induced a significant increase in hardiness in shoots of all three species. However, this treatment did not trigger root hardening. A. crispa was found to be the hardiest species followed by A. glutinosa and A. rubra . Intraspecific variation was observed between the two A. glutinosa clones. A glutinosa clone AG8, a Russian provenance, showed a greater freezing resistance than A. glutinosa clone AG2, a German provenance.

Root growth, development and frost resistance were examined in winter rye ( Secale cereale L. cv. Puma) plants grown under 6 combinations of temperature and photoperiod (20/16°C or 5/3°C, day/night; 8, 16- or 24-h days). Overall root system growth is influenced by the interaction of temperature and photoperiod. Maximum shoot growth occurs at a 24-h photoperiod in 20°C plants and at a 16-h photoperiod in 5°C plants, and is correlated in both treatments with a high root:shoot ratio. Frost resistance of rye roots is affected by short photoperiods in 2 ways. First, short photoperiod and low temperature delay production of new adventitious roots so that newly developing roots are not exposed to freezing temperatures. Second, short photoperiod alone can induce several degrees of frost tolerance in existing roots during the lag phase of growth. Low temperature alone does not decrease the rate of dry weight accumulation in rye root systems, but cold temperature does retard developmental processes within the roots. Rye roots grown at 5°C develop first order lateral roots, differentiate metaxylem vessels and suberize endodermal cell walls more slowly than roots grown at 20°C.

Sugar-beet plants were subjected to all combinations of two day temperatures (12-5 and 18-5 oC), two night temperatures (8-o and 14-0 oC) and two light intensities (275 and 550 J cm-2 of visible radiation in a 12 h day) during the last month of their growth. Cold day or night temperatures resulted in the plants having slightly smaller leaf areas, final dry weights and amounts of sugar in their roots than plants grown in warm temperatures. Plants grown in the cold also contained less water, particularly those given cold nights, so that they had smaller fresh weights and a greater concentration of sugar in the fresh, but not in the dry roots, than plants grown in warm conditions. Halving the light intensity had little effect on leaf area but decreased the net assimilation rate. The final dry weight of the shoot was not affected by changing the light intensity, but the dry weights of the roots of plants grown in dim light were 20 % smaller than in plants grown in bright light and they contained correspondingly less sugar. There was no effect of varying the light intensity on the concentrations of sugar in the fresh or dry roots. There was no evidence that cold night temperatures, either alone or in conjunction with bright conditions during the day, induced the storage root to accumulate sugar faster relative to non-sugar dry matter, i.e. to 'ripen'.

Weekly estimates of dry matter in rooted dwarf-bean leaves (Phaseolusvulgaris) and rooted runner-bean leaves (Phaseolus multiflorus)were made with roots held at 13°, 17°, 21°, or 25°C. Most dry matter accumulated in the lamina of both speciesat the coldest root temperature. Roots of runner bean grew morethan dwarf bean at all temperatures compared, especially at13° and 17° C and the greater net assimilation rate,i.e. dry matter increase per dm1 per week, of runner-bean leavesat cold temperatures, is attributed to faster root growth. Shading lessened accumulation in the lamina, especially at coldroot temperatures, because a greater proportion of assimilatewas transferred to roots. Rates of root growth are mainly influenced by temperature, andno effect could be detected of carbohydrate accumulation inthe lamina at cold temperatures on root growth after the rootswere kept warmer. Water content of the lamina per unit areaincreased as dry matter increased. For any particular dry-mattercontent, the water content was greater with roots at 20°than at 13°, i.e. the leaves were more succulent with warmerroots. When roots first formed on the petioles the lamina lost sugarand subsequently regained it, faster when roots were cold. Whentransferred from warm to cold root temperature, sugars accumulatedin the lamina. Starch changed more than sugars in response tochanging root temperatures. Starch in the lamina progressivelydeclined when roots were warm, but increased when they werecold. The starch content of the lamina was rapidly influencedby root temperature. Sugars and starch together account forless than half of the dry-matter increase in the lamina. Theunidentified fraction is little influenced by changes of roottemperature; it may be largely structural because the palisadecells continued to grow after the leaves were excised, althoughthe lamina was fully expanded before the leaves were rooted.Palisade cells were larger with roots at 20° than at 13°C.

Two bell pepper (Capsicum annuum) cultivars, differing in their response to chilling, were exposed to three levels of root‐zone temperatures. Gas exchange, shoot and root phenology, and the pattern of change of the central metabolites and secondary metabolites caffeate and benzoate in the leaves and roots were profiled. Low root‐zone temperature significantly inhibited gaseous exchange, with a greater effect on the sensitive commercial pepper hybrid (Canon) than on the new hybrid bred to enhance abiotic stress tolerance (S103). The latter was less affected by the treatment with respect to plant height, shoot dry mass, root maximum length, root projected area, number of root tips and root dry mass. More carbon was allocated to the leaves of S103 than nitrogen at 17°C, while in the roots at 17°C, more nitrogen was allocated and the ratio between C/N decreased. Metabolite profiling showed greater increase in the root than in the leaves. Leaf response between the two cultivars differed significantly. The roots accumulated stress‐related metabolites including γ‐aminobutyric acid (GABA), proline, galactinol and raffinose and at chilling (7°C) resulted in an increase of sugars in both cultivars. Our results suggest that the enhanced tolerance of S103 to root cold stress, reflected in the relative maintenance of shoot and root growth, is likely linked to a more effective regulation of photosynthesis facilitated by the induction of stress‐related metabolism.

Studying Arabidopsis thaliana (L.) Heynh. root development in situ at the whole plant level without affecting shoot development has always been a challenge. Such studies are usually carried out on individual plants, neglecting competition of a plant population, using hydroponic systems or Agar-filled Petri dishes. Those both systems, however, present some limitations, such as difficulty to study precisely root morphogenesis or time-limited culture period, respectively. In this paper, we present a method of Arabidopsis thaliana (L.) Heynh. cultivation in soil medium, named “Ara-rhizotron”. It allows the non-destructive study of shoot and root development simultaneously during the entire period of vegetative growth. In this system, roots are grown in 2D conditions, comparable to other soil cultures. Moreover, grouping several Ara-rhizotrons in a box enables the establishment of 3D shoot competition as for plants grown in a population. In comparison to a control culture grown in pots in the same environmental conditions, the Ara-rhizotron resulted in comparable shoot development in terms of dry mass, leaf area, number of leaves and nitrogen content. We used this new culture system to study the effect of irrigation modalities on plant development. We found that irrigation frequency only affected root partitioning in the soil and shoot nitrogen content, but not shoot or root growth. These effects appeared at the end of the vegetative growth period. This experiment highlights the opportunity offered by the Ara-rhizotron to point out tardy effects, affecting simultaneously shoot development and root architecture of plants grown in a population. We discuss its advantages in relation to root development and physiology, as well as its possible applications.

In experiments with rooted cuttings of aspen (Populus tremula L). with a small leaf area, it was found that the roots grew well as long as there was no shoot growth. The onset of shoot growth was followed by a period of decreased root growth. When the leaf area had increased sufficiently, root growth recovered. Decreasing the shoot growth by removal of growth points in the shoot or by short day treatment increased the fraction of photosynthesis products used for root growth, leading to increased root/shoot ratios. Competition between growing shoots and roots for carbohydrates formed in photosynthesis is considered to cause the effects noted and to be of importance for maintaining the balance between the root and shoot systems.

The levels of indol-3yl-acetic acid and gibberellins were determined in shoots and storage roots of radish (Raphanus sativus L.) at various times during the vegetative growth cycle of control plants and plants in which the root to shoot ratio was modified by daminozide treatment. In control plants the onset of storage organ growth was preceded by a change in the hormone root to shoot ratio to favour the root. There was a general reduction in hormone levels in daminozide-treated plants but the pattern of their distribution in roots and shoots was very similar to that in control plants. Thus the effects of daminozide on increased storage root growth cannot be explained in terms of altered root to shoot hormone ratios.

Spatial distributions of root systems of Larix gmelinii (Rupr.) Rupr. trees were examined in two stands in central Siberia: an even-aged stand (ca. 100 yrs-old) and a mature, uneven-aged (240–280 yrs-old) stand. Five larch trees of different sizes were sampled by excavating coarse roots (diameter > 5 mm) in each stand. Dimensions and ages of all first-order lateral roots were measured. Micro-scale conditions of soil temperature and soil water suction (each 10 cm deep) were also examined in relation to earth hummock topography (mound vs. trough) and/or ground floor vegetation types (moss vs. lichens). All larch trees developed superficial root systems, consisting of the aborted short tap root (10–40 cm in soil depth) and some well-spread lateral roots (n= 4-13). The root network of each tree was asymmetric, and its rooting area reached about four times the crown projection area. Lateral roots generally expanded into the upper soil layers of the mounds where summer soil temperature was 1–6°C higher than inside nearby troughs. Chronological analysis indicated that lateral root expansion started successively from lower to upper parts of each aborted tap root, and some lateral roots occurred simultaneously at several decades after tree establishment. The process of root system development was likely to be primarily linked with post-fire dynamics of rhizosphere environment of the permafrost soils.

The long and short term metabolic effects of a shift in root temperature was investigated in Plantago lanceolata L. with special reference to the role of the cyanide resistant alternative pathway in root respiration. After a 10-day period of growth at a 13°C root temperature, a decrease in root as well as shoot growth was observed, compared to control plants grown continuously at 21°C. Apart from an increase in shoot soluble and insoluble sugar level, no changes in metabolism were found, neither in root respiration, shoot photosynthesis, nor in root sugar and plant protein level.
Decreasing the root temperature from 21 to 13°C gave several clear short term changes in metabolism. Within one hour a decrease in cytochrome chain activity of the roots was found together with an increase in activity of the alternative chain. After 24 h a recovery to the initial level of both chains was observed. An increase in root temperature from 13 to 21°C gave an immediate increase in activity of both respiratory chains that was still present 24 h after the switch.
It is concluded that the activity of the alternative respiratory pathway in the root is strongly affected by a sudden temperature change in the root environment. This pathway acts in a way which is described by 'the energy overflow model'. The presence of the alternative electron transport pathway should be taken into account in determinations of the respiratory Q10. Moreover, the length of time between the temperature change and respiration measurements is an important factor.

Brassica rapa plants were exposed for a 52 h period (as pretreatment) to a differential temperature (DT) between roots (5°C) and shoots (20°C), while control plants were maintained with both shoot and roots at 20°C (warm grown = WG). Measured at 20°C, volume flow of xylem exudate from roots of DT plants was enhanced compared with that from WG plants, while transpiration flows were similar in pretreated and control plants. Both transpiration and exudation flows were dependent upon shoot/root ratio. Differences in the volume flow of exudate were principally related to increases in root hydraulic conductance. Anion fluxes (notably nitrate) into xylem exudate of DT plants were significantly greater than those into exudate of WG plants. This enhancement of nitrate flow from the pretreated roots was associated with a two-fold increase in nitrate uptake rate. The relationship of the cold-induced change in nitrate uptake capacity with shoot/root ratio is discussed in terms of control of nitrate absorption by shoot sink strength.

Phenology is central to understanding vegetation response to climate change, as well as vegetation effects on plant resources, but most temporal production data is based on shoots, especially those of trees. In contrast, most production in temperate and colder regions is belowground, and is frequently dominated by grasses. We report root and shoot phenology in 7‐year old monocultures of 10 dominant species (five woody species, five grasses) in southern Canada. Woody shoot production was greatest about 8 weeks before the peak of root production, whereas grass shoot maxima preceded root maxima by 2–4 weeks. Over the growing season, woody root, and grass root and shoot production increased significantly with soil temperature. In contrast, the timing of woody shoot production was not related to soil temperature (r=0.01). The duration of root production was significantly greater than that of shoot production (grasses: 22%, woody species: 54%). Woody species produced cooler and moister soils than grasses, but growth forms did not affect seasonal patterns of soil conditions. Although woody shoots are the current benchmark for phenology studies, the other three components examined here (woody plant roots, grass shoots and roots) differed greatly in peak production time, as well as production duration. These results highlight that shoot and root phenology is not coincident, and further, that major plant growth forms differ in their timing of above‐ and belowground production. Thus, considering total plant phenology instead of only tree shoot phenology should provide a better understanding of ecosystem response to climate change.

[3H]iso-Pentenyladenine ([3H]iP) was fed for 24 h to the tips of intact and root tip-decapitated Pinus pinea seedlings. Twelve and 24 h after application to the roots of intact plants most of the applied radioactivity (±60%) was transported to the shoot. Root tip removal increased transport of the applied radioactivity to the shoot, but the overall pattern of distribution of radioactivity in the seedling did not change. Large amounts of radioactivity were recovered from the elongation zone of the root. Some radioactivity also accumulated in the older part of the root with well-developed lateral roots. When [3H]iP was applied one day after decapitation, no significant changes in the pattern of radioactivity distribution were found between the intact and decapitated root systems. However, when applied 7 days after decapitation there was a significant increase of radioactivity in the region of the root where lateral roots were emerging. HPLC separation of extracts from the different root sections showed that [3H]iP was extensively metabolized in the root. Six peaks of radioactivity, which co-chromatographed with authentic cytokinin standards, were detected.Abbreviations ABA abscisic acid - ADE adenine - IAA indole-acetic acid - iP iso-pentenyladenine - HPLC high performance liquid chromatography - [OG]DHZ O-glycosyldihydrozeatin - [9R-MP]DHZ ribosyldihydrozeatin monophosphate - [9G]iP iso-pentenyladenine-9-glucoside - [9R]Z ribosylzeatin - [9R]iP iso-pentenyladenosine - TLC thin layer chromatography  相似文献

Rice cultivar evaluation for phosphorus use efficiency
Phosphorus deficiency is one of the most growth-limiting factors in acid soils in various parts of the world. The objective of this study was to screen 25 rice cultivars (Oryza sativa L.) at low, medium, and high levels of soil P. Number of tillers, root length, plant height, root dry weight and shoot dry weight were related to tissue P concentrations, P uptake and P-use efficiency. Shoot weight was found to be the plant parameter most sensitive to P deficiency. Significant cultivar differences in P use efficiency were found. Phosphorus use efficiency was higher in roots than shoots and decreased with increasing levels of soil P. Positive correlations were found among growth parameters such as plant height, tillers, root and shoot weight, and P content of roots and shoots. These results indicate selection of rice cultivars for satisfactory performance under low P availability can be carried out using shoot and root dry weight as criteria.

Two pea (Pisum sativum L.) symbiotic mutants SGEFix(-)-1 (sym40) and SGEFix(-)-2 (sym33) with abnormalities in infection thread development and function in symbiotic root nodules have been characterised in terms of mycorrhizal colonisation of roots, shoot and root biomass accumulation and shoot and root phosphorus (P) content. The mutation in gene sym33 decreased mycorrhizal colonisation of roots (except arbuscule abundance in mycorrhizal root fragments, which increased) but did not change the effectiveness of mycorrhiza function. The mutation in sym40 did not affect either of these processes. Both mutants showed differences in plant development compared with the wild-type line SGE. The mutants had delayed flowering and pod ripening, and shoot/root biomass ratios and P accumulation also differed from those of SGE. These observations suggest that the gene mutations cause systemic changes in plant development.

Bare-root seedlings of pedunculate oak (Quercus robur L.) and northern red oak (Quercus rubra L.) were lifted in January and stored at 1.8°C, at 82% relative humidity, until their fresh weight declined by 33%. Root growth potential (RGP), fine root electrolyte leakage (REL), fine root water content (RWC), shoot tip water content (SWC), starch and metabolic solute contents in root and shoot, were measured just after lifting and after treatment. Survival of treated seedlings was also assessed in a field trial. RWC, SWC, REL, RGP were dramatically affected by desiccation during cold storage. In both species, root soluble carbohydrate level, inositol level and isocitrate level increased, whereas root starch level and shoot soluble carbohydrate level decreased. In northern red oak, treated seedlings had higher root contents of soluble carbohydrates, inositol and proline than in pedunculate oak. Moreover, treatment induced proline accumulation only in northern red oak roots. These differences could explain why field survival of treated seedlings was significantly better in northern red oak than in pedunculate oak.

Soil temperature and flooding effects on two species of citrus
Summary Rough lemon (Citrus jambhiri Lush.) and sour orange (C. aurantium L.) seedlings were grown at constant soil temperatures of 16, 24, and 33 C for 3 months. Shoot and root growth of rough lemon was greatest at 33 C while growth of sour orange was greatest at 24 C. There were no significant effects of soil temperature on shoot: root ratio, leaf water potential or stomatal conductance. The hydraulic conductivity of intact root systems of both species was highest when seedlings were grown at 16 C. Thus, acclimation through greater root conductivity at low soil temperature may have compensated for decreased root growth at 16 C and negated effects of soil temperature on plant water relations. Half the plants growing at each soil temperature were subsequently flooded. Within 1 week, the soil redox potential (Eh) dropped below zero mV, reaching a minimum Eh of –250mV after 3 weeks of flooded conditions. Flooded plants exhibited lower root conductivity, a cessation of shoot growth, lower leaf water potentials, lower stomatal conductances, and visual sloughing of fibrous roots. Decreases in root conductivity in response to flooding were large enough to account for the observed decreases in stomatal conductance.Florida Agricultural Experiment Stations Journal Series No. 4080.

Primary and secondary induction requirements for flowering of Festuca rubra
Root and shoot temperatures were varied independently to determine the importance of root temperature during cold acclimation. Spinach ( Spinacia oleracea L. cvs Harbin and Bloomsdale) plants were subjected to 20/20°C. 20/5°C, 5/20°C, and 5/5°C (shoot/root) temperature treatments. Leaf freezing tolerance, water potential, stomatal resistance, osmotic potential, and water content were measured at 0.25. 1.25, 3.25, and 7.25 days of treatment. There was no change in freezing tolerance or the water relations of the 20/20°C treated plants during the course of the experiment. Freezing tolerance was increased by the 5°C shoot temperature treatments, but was not enhanced by water stress induced by the low root temperature. Leaf water potential and water content decreased and stomatal resistance increased within 6 h in the 20/5°C plants. By day 3, osmotic potential began decreasing in the 20/5°C plants. Leaf water content, osmotic potential, and water potential decreased more gradually in plants grown with 5°C shoot temperature, irrespective of root temperature. Decreased water content and osmotic potential were not correlated with increased freezing tolerance as reported for other herbaceous crop plants.
